Small interfering RNA(siRNA)has a promising future in the treatment of ocular diseases due to its high efficiency,specificity,and low toxicity in inhibiting the expression of target genes and proteins.However,due to the unique anatomical structure of the eye and various barriers,delivering nucleic acids to the retina remains a significant challenge.In this study,we rationally design PACD,an A-B-C type non-viral vector copolymer composed of a hydrophilic PEG block(A),a siRNA binding block(B)and a pH-responsive block(C).PACDs can self-assemble into nanosized polymeric micelles that compact siRNAs into polyplexes through simple mixing.By evaluating its pH-responsive activity,gene silencing efficiency in retinal cells,intraocular distribution,and anti-angiogenesis therapy in a mouse model of hypoxia-induced angiogenesis,we demonstrate the efficiency and safety of PACD in delivering siRNA in the retina.We are surprised to discover that,the PACD/siRNA polyplexes exhibit remarkable intracellular endosomal escape efficiency,excellent gene silencing,and inhibit retinal angiogenesis.Our study provides design guidance for developing efficient nonviral ocular nucleic acid delivery systems.

Dynamic response mechanism and precursor characteristics of gneiss rockburst under different initial burial depths

To investigate the influence mechanism of geostress on rockburst characteristics,three groups of gneiss rockburst experiments were conducted under different initial geostress conditions.A high-speed photography system and acoustic emission(AE)monitoring system were used to monitor the entire rockburst process in real time.The experimental results show that when the initial burial depth increases from 928 m to 1320 m,the proportion of large fracture scale in rockburst increases by 154.54%,and the AE energy increases by 565.63%,reflecting that the degree and severity of rockburst increase with the increase of burial depth.And then,two mechanisms are proposed to explain this effect,including(i)the increase of initial geostress improves the energy storage capacity of gneiss,and then,the excess energy which can be converted into kinetic energy of debris ejection increases,consequently,a more pronounced violent ejection phenomenon is observed at rockburst;(ii)the increase of initial geostress causes more sufficient plate cracks of gneiss after unloading ofσh,which provides a basis for more severe ejection of rockburst.What’s more,a precursor with clear physical meaning for rockburst is proposed under the framework of dynamic response process of crack evolution.Finally,potential value in long term rockburst warning of the precursor obtained in this study is shown via the comparison of conventional precursor.

Rockburst are often encountered in tunnel construction due to the complex geological conditions.To study the influence of unloading rate on rockburst,gneiss rockburst experiments were conducted under three groups of unloading rates.A high-speed photography system and acoustic emission(AE)system were used to monitor the entire process of rockburst process in real-time.The results show that the intensity of gneiss rockburst decreases with decrease of unloading rate,which is manifested as the reduction of AE energy and fragments ejection velocity.The mechanisms are proposed to explain this effect:(i)The reduction of unloading rate changes the crack propagation mechanism in the process of rockburst.This makes the rockbursts change from the tensile failure mechanism at high unloading rate to the tension-shear mixed failure mechanism at low unloading rate,and more energy released in the form of shear crack propagation.Then,less strain energy is converted into kinetic energy of fragments ejection.(ii)Less plate cracking degree of gneiss has taken shape due to decrease of unloading rate,resulting in the destruction of rockburst incubation process.The enlightenments of reducing the unloading rate for the project are also described quantitatively.The rockburst magnitude is reduced from the medium magnitude at the unloading rate of 0.1 MPa/s to the slight magnitude at the unloading rate of 0.025 MPa/s,which was judged by the ejection velocity.

Interdecadal Variations of the March Atmospheric Heat Source over the Southeast Asian Low-Latitude Highlands

Based on the fifth-generation reanalysis dataset from the European Centre for Medium-Range Weather Forecasts for 1979–2019,we investigated the effects of the circumglobal teleconnection(CGT)on the interdecadal variation of the March atmospheric heat source(AHS)over the Southeast Asian low-latitude highlands(SEALLH).The dominant mode of the March AHS over the SEALLH features a monopole structure with an 8–11-year period.Decadal variations in the AHS make an important contribution to the 11-year low-pass filtered component of the AHS index,whichexplains 54.3%of the total variance.The CGT shows a clear interdecadal variation,which explains 59.3%of the total variance.The March AHS over the SEALLH is significantly related to the CGT on interdecadal timescales.When the CGT is optimally excited by a significant cyclonic vorticity source near northern Africa(i.e.,in its positive phase),the SEALLH is dominated by anomalous southerly winds and ascending motions on the east of the anomalous cyclone.The enhanced advection and upward transfer result in a high-enthalpy air mass that converges into and condenses over the SEALLH,leading to a largerthan-average March AHS over this region.The key physical processes revealed by this diagnostic analysis are supported by numerical experiments.

Solid-state flexible supercapacitors(SCs)have many advantages of high specific capacitance,excellent flexibility,fast charging and discharging,high power density,environmental friendliness,high safety,light weight,ductility,and long cycle stability.They are the ideal choice for the development of flexible energy storage technology in the future,and provide a good prospect for energy storage applications.At present,solid-state flexible SCs are widely used for portable electronic equipment and wearable energy storage equipment,the research of them has become the focus of a growing number of researchers.Electrode material is the key part of SCs and always determines the electrochemical performance of SCs.It has been a hotspot and focus of research.Vanadium-based compounds are considered to be a promising electrode material for SCs because of variable valence,open structure,high theoretical capacity,and low price.Therefore,this study first gives an overview of solid-state flexible SCs,then reviews the current research status of vanadium-based electrode materials in solid-state flexible SCs,and proposes some strategies to solve some problems of vanadium-based electrode materials.

Objective: To investigate the main components and potential mechanism of Shuxuening Injection(SXNI) in the treatment of myocardial ischemia-reperfusion injury(MIRI) through network pharmacology and in vivo research. Methods: The Traditional Chinese Medicine Systems Pharmacology(TCMSP) and Pharm Mapper databases were used to extract and evaluate the effective components of Ginkgo biloba leaves, the main component of SXNI. The Online Mendelian Inheritance in Man(OMIM) and Gene Cards databases were searched for disease targets and obtain the drug target and disease target intersections. The active ingredient-target network was built using Cytoscape 3.9.1 software. The STRING database, Metascape online platform, and R language were used to obtain the key targets and signaling pathways of the anti-MIRI effects of SXNI. In order to verify the therapeutic effect of different concentrations of SXNI on MIRI in rats, 60 rats were first divided into 5 groups according to random number table method: the sham operation group, the model group, SXNI low-dose(3.68 mg/kg), medium-dose(7.35 mg/kg), and high-dose(14.7 mg/kg) groups, with 12 rats in each group. Then, another 60 rats were randomly divided into 5 groups: the sham operation group, the model group, SXNI group(14.7 mg/kg), SXNI+LY294002 group,and LY294002 group, with 12 rats in each group. The drug was then administered intraperitoneally at body weight for 14 days. The main biological processes were validated using in vivo testing. Evans blue/triphenyltetrazolium chloride(TTC) double staining, hematoxylin-eosin(HE) staining, terminal deoxynucleotidyl transferase d UTP nick end labeling(TUNEL) assay, enzyme-linked immunosorbent assay(ELISA), and Western blot analysis were used to investigate the efficacy and mechanism of SXNI in MIRI rats. Results: Eleven core targets and 30 Kyoto Encyclopedia of Genes and Genomes(KEGG) pathways were selected. Background:Ischemia-reperfusion injury(IRI)poses a significant challenge to liver transplantation(LT).The underlying mechanism primarily involves overactivation of the immune system.Heat shock protein 110(HSP110)functions as a molecular chaperone that helps stabilize protein structures.Methods:An IRI model was established by performing LT on Sprague-Dawley rats,and HSP110 was silenced using siRNA.Hematoxylin-eosin staining,TUNEL,immunohistochemistry,ELISA and liver enzyme analysis were performed to assess IRI following LT.Western blotting and quantitative reverse transcription-polymerase chain reaction were conducted to investigate the pertinent molecular changes.Results:Our findings revealed a significant increase in the expression of HSP110 at both the mRNA and protein levels in the rat liver following LT(P<0.05).However,when rats were injected with siRNAHSP110,IRI subsequent to LT was notably reduced(P<0.05).Additionally,the levels of liver enzymes and inflammatory chemokines in rat serum were significantly reduced(P<0.05).Silencing HSP110 with siRNA resulted in a marked decrease in M1-type polarization of Kupffer cells in the liver and downregulated the NF-κB pathway in the liver(P<0.05).Conclusions:HSP110 in the liver promotes IRI after LT in rats by activating the NF-κB pathway and inducing M1-type polarization of Kupffer cells.Targeting HSP110 to prevent IRI after LT may represent a promising new approach for the treatment of LT-associated IRI.

Fog computing is considered as a solution to accommodate the emergence of booming requirements from a large variety of resource-limited Internet of Things(IoT)devices.To ensure the security of private data,in this paper,we introduce a blockchain-enabled three-layer device-fog-cloud heterogeneous network.A reputation model is proposed to update the credibility of the fog nodes(FN),which is used to select blockchain nodes(BN)from FNs to participate in the consensus process.According to the Rivest-Shamir-Adleman(RSA)encryption algorithm applied to the blockchain system,FNs could verify the identity of the node through its public key to avoid malicious attacks.Additionally,to reduce the computation complexity of the consensus algorithms and the network overhead,we propose a dynamic offloading and resource allocation(DORA)algorithm and a reputation-based democratic byzantine fault tolerant(R-DBFT)algorithm to optimize the offloading decisions and decrease the number of BNs in the consensus algorithm while ensuring the network security.Simulation results demonstrate that the proposed algorithm could efficiently reduce the network overhead,and obtain a considerable performance improvement compared to the related algorithms in the previous literature.

In this study,online water-soluble inorganic ions were detected to deduce the formation mechanism of secondary inorganic aerosol in Xianyang,China during wintertime.The dominant inorganic ions of sulfate(SO_(4)^(2-)),nitrate(NO_(3)^(-)),and ammonium(NH_(4)^(+))(the sum of those is abbreviated as SNA)accounted for 17%,21%,and 12% of PM_(2.5)mass,respectively.While the air quality deteriorated from excellent to poor grades,the precursor gas sulfur dioxide(SO_(2))of SO_(4)^(2-)increased and then decreased with a fluctuation,while nitrogen dioxide(NO_(2))and ammonia(NH_(3)),precursors of NO_(3)^(-)and NH_(4)^(+),and SNA show increasing trends.Meteorological factors including boundary layer height(BLH),temperature,and wind speed also show decline trends,except relative humidity(RH).Meanwhile,the secondary conversion ratio shows a remarkable increasing trend,indicating that there was a strong secondary transformation.From the perspective of chemical mechanisms,RH is positively correlated with sulfur oxidation ratios(SOR),nitrogen oxidation ratios(NOR),and ammonia conversion ratios,representing that the increase of humidity could promote the generation of SNA.Notably,SOR and NOR were also positively related to the ammonia.On the one hand,the low wind speed and BLH led to the accumulation of pollutants.On the other hand,the increases of RH and ammonia promoted more formations of SNA and PM_(2.5).The results advance our identification of the contributors to the haze episodes and assist to establish more efficient emission controls in Xianyang,in addition to other cities with similar emission and geographical characteristics.
