The profiles of chiral pesticides in peri-urban areas near Yangtze River: Enantioselective distribution characteristics and correlations with surface sediments

Chiral pesticides account for 30% of pesticides. Pesticides are inevitably leached into the groundwater by runoff. At the watershed level, the distribution characteristics of enantiomers in sediments collected from the river network of an agricultural area near the middle and lower reaches of the Yangtze River were tested, and their potential correlations with the physicochemical properties and microbial communities of the sediments were analyzed. The sediment pollution was serious at sites 8 and 9, with their pollution source possibly being agricultural or industrial sewage. Moreover, there were higher cumulative contents of pesticide residues at sites 4, 8, and 9. Specifically, Cycloxaprid was the most detected chiral pesticide in the study area, followed by Dinotefuran and Diclofop-methyl. Additionally,Ethiprole and Difenoconazole had strong enantioselectivity in the study area. Interestingly,the enantiomers of some chiral pesticides, such as Tebuconazole, had completely different distributions at different sites. Pearson correlation analysis showed that sediment catalase and microbial biomass carbon were important factors for enantioselectivity of chiral pesticides. The effect of sediment physicochemical properties on enantioselective distribution was achieved by influencing the microorganisms in the sediment. Furthermore, the enantioselective distribution of Tebuconazole was closely related to the genus Arenimonas.Overall, the enantioselective distribution of most of the chiral pesticides was positively correlated with the prokaryotic microbial community. This study provides empirical support for agricultural non-point source pollution caused by chiral pesticides, and also lays a research foundation for exploring the factors that affect the fate of chiral pesticides in the environment.

Gastric cancer is highly prevalent among digestive tract tumors.Due to the intri-cate nature of the gastric cancer immune microenvironment,there is currently no effective treatment available for advanced gastric cancer.However,there is promising potential for immunotherapy targeting the prostaglandin E2 receptor subtype 4(EP4)in gastric cancer.In our previous study,we identified a novel small molecule EP4 receptor antagonist called YY001.Treatment with YY001 alone demonstrated a significant reduction in gastric cancer growth and inhibited tumor metastasis to the lungs in a mouse model.Furthermore,adminis-tration of YYo01 stimulated a robust immune response within the tumor microenvironment,characterized by increased infiltration of antigen-presenting cells,T cells,and M1 macro-phages.Additionally,our research revealed that YYo01 exhibited remarkable synergistic ef-fects when combined with the PD-1 antibody and the clinically targeted drug apatinib,rather than fluorouracil.These findings suggest that YYo01 holds great promise as a potential therapeutic strategy for gastric cancer,whether used as a standalone treatment or in combination with other drugs.

In the present study,the chronic heat stress （CHS） broiler model was developed to investigate the potential protection mechanism of organic selenium （selenomethionine,SeMet） on CHS-induced skeletal muscle growth retardation and poor meat quality.Four hundred Arbor Acres male broilers （680±70 g,21 d old） were grouped into 5 treatments with 8 replicates of 10 broilers per replicate.Broilers in the control group were raised in a thermoneutral environment （22±2°C） and fed with a basal diet.The other four treatments were exposed to hyperthermic conditions （33±2°C,24 h in each day） and fed on the basal diet supplied with SeMet at 0.0,0.2,0.4,and 0.6 mg Se/kg,respectively,for 21 d.Results showed that CHS reduced （P<0.05） the growth performance,decreased （P<0.05） the breast muscle weight and impaired the meat quality of breast muscle in broilers.CHS induced protein metabolic disorder in breast muscle,which increased （P<0.05） the expression of caspase 3,caspase 8,caspase 9 and ubiquitin proteasome system related genes,while decreased the protein expression of P-4EBP1.CHS also decreased the antioxidant capacity and induced mitochondrial stress and endoplasmic reticulum （ER） stress in breast muscle,which increased （P<0.05） the ROS levels,decreased the concentration of ATP,increased the protein expression of HSP60 and CLPX,and increased （P<0.05） the expression of ER stress biomarkers.Dietary SeMet supplementation linearly increased （P<0.05） breast muscle Se concentration and exhibited protective effects via up-regulating the expression of the selenotranscriptome and several key selenoproteins,which increased （P<0.05） body weight,improved meat quality,enhanced antioxidant capacity and mitigated mitochondrial stress and ER stress.What's more,SeMet suppressed protein degradation and improved protein biosynthesis though inhibiting the caspase and ubiquitin proteasome system and promoting the mTOR-4EBP1 pathway.In conclusion,dietary SeMet supplementation increases the expression of several key selenoproteins,alleviates mitochondr

Divergent response of vegetation phenology to extreme temperatures and precipitation of different intensities on the Tibetan Plateau

Quantifying how climate factors affect vegetation phenology is crucial for understanding climate-vegetation interactions and carbon and water cycles under a changing climate.However,the effects of different intensities of extreme climatic events on vegetation phenology remain poorly understood.Using a long-term solar-induced chlorophyll fluorescence dataset,we investigated the response of vegetation phenology to extreme temperatures and precipitation events of different intensities across the Tibetan Plateau(TP)from 2000 to 2018.We found that the effect of maximum temperature exposure days(TxED)and minimum temperature exposure days(TnED)on the start of the growing season(SOS)was initially delayed and shifted to advance along the increasing temperature gradients.However,the response of the end of the growing season(EOS)to TxED and TnED shifted from an advance to a delay with increasing temperature gradients until the temperature thresholds were reached,above which thresholds produced an unfavorable response to vegetation growth and brought the EOS to an early end.The corresponding maximum and minimum temperature thresholds were 10.12 and 2.54℃,respectively.In contrast,cumulative precipitation(CP)was more likely to advance SOS and delay EOS as the precipitation gradient increased,but the advance of SOS is gradually weakening.Four vegetation types(i.e.,forest,shrubland,meadow,and steppe)showed similar trends in response to different climates,but the optimal climatic conditions varied between the vegetation types.Generally,meadow and steppe had lower optimal temperatures and precipitation than forest and shrubland.These findings revealed the divergent responses of vegetation phenology to extreme climate events of different intensities,implying that the SOS will continue to advance with warming,whereas the EOS may undergo a partial transformation from delayed areas to advanced areas with continued warming.

With the rapid development of 5G information technology,thermal conductivity/dissipation problems of highly integrated electronic devices and electrical equipment are becoming prominent.In this work,“high-temperature solid-phase&diazonium salt decomposition”method is carried out to prepare benzidine-functionalized boron nitride(m-BN).Subsequently,m-BN/poly(pphenylene benzobisoxazole)nanofiber(PNF)nanocomposite paper with nacremimetic layered structures is prepared via sol–gel film transformation approach.The obtained m-BN/PNF nanocomposite paper with 50 wt%m-BN presents excellent thermal conductivity,incredible electrical insulation,outstanding mechanical properties and thermal stability,due to the construction of extensive hydrogen bonds andπ–πinteractions between m-BN and PNF,and stable nacre-mimetic layered structures.Itsλ∥andλ_(⊥)are 9.68 and 0.84 W m^(-1)K^(-1),and the volume resistivity and breakdown strength are as high as 2.3×10^(15)Ωcm and 324.2 kV mm^(-1),respectively.Besides,it also presents extremely high tensile strength of 193.6 MPa and thermal decomposition temperature of 640°C,showing a broad application prospect in high-end thermal management fields such as electronic devices and electrical equipment.

High-throughput assays play an important role in the fields of drug discovery,genetic analysis,and clinical diagnostics.Although super-capacity coding strategies may facilitate labeling and detecting large numbers of targets in a single assay,practically,the constructed large-capacity codes have to be decoded with complicated procedures or are lack of survivability under the required reaction conditions.This challenge results in either inaccurate or insufficient decoding outputs.Here,we identified chemical-resistant Raman compounds to build a combinatorial coding system for the high-throughput screening of cell-targeting ligands from a focused 8-mer cyclic peptide library.The accurate in situ decoding results proved the signal,synthetic,and functional orthogonality for this Raman coding strategy.The orthogonal Raman codes allowed for a rapid identification of 63 positive hits at one time,evidencing a high-throughput-out capability in the screening process.We anticipate this orthogonal Raman coding strategy being generalized to enable efficient high-throughput-out screening of more useful ligands for cell targeting and drug discovery.

Germinal matrix hemorrhage is one of the leading causes of morbidity,mortality,and acquired infantile hydrocephalus in preterm infants in the United States,with little progress made in its clinical management.Blood clots have been shown to elicit secondary brain injury after germinal matrix hemorrhage,by disrupting normal cerebrospinal fluid circulation and absorption after germinal matrix hemorrhage causing post-hemorrhagic hydrocephalus development.Current evidence suggests that rapid hematoma resolution is necessary to improve neurological outcomes after hemorrhagic stroke.Various articles have demonstrated the beneficial effects of stimulating the polarization of microglia cells into the M2 phenotype,as it has been suggested that they play an essential role in the rapid phagocytosis of the blood clot after hemorrhagic models of stroke.N-formyl peptide receptor 2(FPR2),a G-protein-coupled receptor,has been shown to be neuroprotective after stroke.FPR2 activation has been associated with the upregulation of phagocytic macrophage clearance,yet its mechanism has not been fully explored.Recent literature suggests that FPR2 may play a role in the stimulation of scavenger receptor CD36.Scavenger receptor CD36 plays a vital role in microglia phagocytic blood clot clearance after germinal matrix hemorrhage.FPR2 has been shown to phosphorylate extracellular-signal-regulated kinase 1/2(ERK1/2),which then promotes the transcription of the dual-specificity protein phosphatase 1(DUSP1)gene.In this review,we present an intrinsic outline of the main components involved in FPR2 stimulation and hematoma resolution after germinal matrix hemorrhage.

Juvenile idiopathic arthritis (JIA) is one of the most common chronic inflammatory rheumatic diseases in children,with onset before age 16 and lasting for more than 6 weeks.JIA is a highly heterogeneous condition with various consequences for health and quality of life.For some JIA patients,early detection and intervention remain challenging.As a result,further investigation of the complex and unknown mechanisms underlying JIA is required.Advances in technology now allow us to describe the biological heterogeneity and function of individual cell populations in JIA.Through this review,we hope to provide novel ideas and potential targets for the diagnosis and treatment of JIA by summarizing the current findings of single-cell RNA sequencing studies and understanding how the major cell subsets drive JIA pathogenesis.
