Salvianolic acid B protects hepatocytes from H2O2 injury by stabilizing the lysosomal membrane

AIM To investigate the capability of salvianolic acid B(Sal B) to protect hepatocytes from hydrogen peroxide(H_2O_2)/carbon tetrachloride(CCl_4)-induced lysosomal membrane permeabilization. METHODS Cell Counting Kit-8 assay was used to measure cell viability. Apoptosis and death were assayed through flow cytometry. Brd U incorporation was used to detect cell proliferation. Serum alanine aminotransferase activity and liver malondialdehyde(MDA) content were measured. Liver histopathological changes were evaluated using hematoxylin-eosin staining. Lysosomal membrane permeability was detected with Lyso Tracker Green-labeled probes and acridine orange staining. The levels of protein carbonyl content(PCC), cathepsins(Cat)B/D, and lysosome-associated membrane protein 1(LAMP1) were evaluated through western blotting. Cytosol Cat B activity analysis was performed with chemiluminescence detection. The m RNA level ofLAMP1 was evaluated through quantitative real-time polymerase chain reaction. RESULTS Results indicated that H_2O_2 induced cell injury/death. Sal B attenuated H_2O_2-induced cell apoptosis and death, restored the inhibition of proliferation, decreased the amount of PCC, and stabilized the lysosome membrane by increasing the LAMP1 protein level and antagonizing Cat B/D leakage into the cytosol. CCl_4 also triggered hepatocyte death. Furthermore, Sal B effectively rescued hepatocytes by increasing LAMP1 expression and by reducing lysosomal enzyme translocation to the *** Sal B protected mouse embryonic hepatocytes from H_2O_2/CCl_4-induced injury/death by stabilizing the lysosomal membrane.

Comparative analysis of frictional behavior and mechanism of molybdenum ditelluride with different structures

Two-dimensional(2D)transition metal dichalcogenides(TMDCs)have layered structures with excellent tribological *** the energy difference between hexagonal-molybdenum ditelluride(2H-MoTe_(2))and distorted octahedral-molybdenum ditelluride(1T′-MoTe_(2))is very small among the transition metal dichalcogenides(TMDCs),MoTe_(2) becomes one of the most promising candidates for phase *** our experiment,we found that the friction force and friction coefficient(COF)of 2H-MoTe_(2) were an order of magnitude smaller than those of 1T′-MoTe_(2) by the atomic force microscope(AFM)*** friction difference between 1T′-MoTe_(2) and 2H-MoTe_(2) was further verified in molecular dynamics(MD)*** density functional theory(DFT)calculations suggest that the friction contrast is related to the difference in sliding energy barrier of the potential energy surface(PES)for a tip sliding across the *** PES obtained from the DFT calculation indicates that the maximum energy barrier and the minimum energy path(MEP)energy barrier of 2H-MoTe_(2) are both smaller than those of 1T′-MoTe_(2),which means that less energy needs to be dissipated during the sliding *** difference in energy barrier of the PES could be ascribed to its larger interlayer spacing and weaker Mo–Te interatomic interactions within the layers of 2H-MoTe_(2) than those of 1T′-MoTe_(2).The obvious friction difference between 1T′-MoTe_(2) and 2H-MoTe_(2) not only provides a new non-destructive means to detect the phase transition by the AFM,but also provides a possibility to tune friction by controlling the phase transition,which has the potential to be applied in extreme environments such as space lubrication.

In this paper,we propose a doping approach to lower the error floor of Low-Density Parity-Check(LDPC)*** doping component is a short block code in which the information bits are selected from the coded bits of the dominant trapping sets of the LDPC ***,an algorithm for selecting the information bits of the short code is proposed,and a specific two-stage decoding algorithm is *** results demonstrate that the proposed doped LDPC code achieves up to 2.0 dB gain compared with the original LDPC code at a frame error rate of 10^(-6)Furthermore,the proposed design can lower the error floor of original LDPC Codes.

A Novel Fracturing Fluid with High-Temperature Resistance for Ultra-Deep Reservoirs

Ultra-deep reservoirs play an important role at present in fossil energy *** to the related high temperature,high pressure,and high formation fracture pressure,however,methods for oil well stimulation do not produce satisfactory results when conventional fracturing fluids with a low pumping rate are *** response to the above problem,a fracturing fluid with a density of 1.2~1.4 g/cm^(3)was developed by using Potassium formatted,hydroxypropyl guanidine gum and zirconium crosslinking *** fracturing fluid was tested and its ability to maintain a viscosity of 100 mPa.s over more than 60 min was verified under a shear rate of 1701/s and at a temperature of 175℃.This fluid has good sand-carrying performances,a low viscosity after breaking the rubber,and the residue content is less than 200 mg/*** with ordinary reconstruction fluid,it can increase the density by 30%~40%and reduce the wellhead pressure of 8000 m level reconstruction ***,the new fracturing fluid can significantly mitigate safety risks.

Achieving high maize yields and efficient phosphorus(P)use with limited environmental impacts is one of the greatest challenges in sustainable maize *** plant density is considered an effective approach for achieving high maize ***,the low mobility of P in soils and the scarcity of natural P resources have hindered the development of methods that can simultaneously optimize P use and mitigate the P-related environmental footprint at high plant *** this study,meta-analysis and substance flow analysis were conducted to evaluate the effects of different types of mineral P fertilizer on maize yield at varying plant densities and assess the flow of P from rock phosphate mining to P fertilizer use for maize production in China.A significantly higher yield was obtained at higher plant densities than at lower plant *** application of single superphosphate,triple super-phosphate,and calcium magnesium phosphate at high plant densities resulted in higher yields and a smaller environmental footprint than the application of diammonium phosphate and monoammonium *** scenario analyses suggest that combining the optimal P type and application rate with a high plant density could increase maize yield by 22%.Further,the P resource use efficiency throughout the P supply chain increased by 39%,whereas the P-related environmental footprint decreased by 33%.Thus,simultaneously optimizing the P type and application rate at high plant densities achieved multiple objectives during maize production,indicating that combining P management with cropping techniques is a practical approach to sustainable maize *** findings offer strategic,synergistic options for achieving sustainable agricultural development.

Metronidazole(MNZ)is a type of antibiotic that can help people and animals cure bacterial in fections,however,abuse of MNZ has posed a threat to human ***,portable and vi sual detection of MNZ is meaningful for food safety and rational administration of drugs,bu full of ***,a porous three-dimensional Tb-based metal-organic framework(MOF{(CH_(3))_(2)NH_(2)·[Tb_(5)(TDA)_(8)(H_(2)O)_(2)]·6DMF·2C_(2)H_(5)OH}_n(TDA-Tb)with good solvent and p H stabilities was pre pared,and the framework possesses one-dimensional channels with a diameter of 12 A along the c-axis Experiment results suggest that the synthesized TDA-Tb can selectively and sensitively detect MNZ in water,and the limit of detection(LOD)is as low as 4.1×10^(-7)mol/***,a flexible sensor TDA Tb-M was also constructed by incorporating TDA-Tb into membrane materials for convenient *** the TDA-Tb-M firstly realized portable and visual detection of MNZ through smartphone scanning,which may inspire more probes with wide application ranges.
