Efficient production of pronuclear embryos in breeding and nonbreeding season for generating transgenic sheep overexpressing TLR4

Background： Brucella is a zoonotic Gram-negative pathogen that causes abortion and infertility in ruminants and humans. TLR4 is the receptor for LPS which can recognize Brucella and initiate antigen-presenting cell activities that affect both innate and adaptive immunity. Consequently, transgenic sheep over-expressing TLR4 are an suitable model to investigate the effects of TLR4 on preventing Brucellosis. In this study, we generated transgenic sheep overexpressing TLR4 and aimed to evaluate the effects of different seasons（breeding and non-breeding season） on superovulation and the imported exogenous gene on growth.Results： In total of 43 donor ewes and 166 recipient ewes in breeding season, 37 donor ewes and 144 recipient ewes in non-breeding season were selected for super-ovulation and injected embryo transfer to generate transgenic sheep.Our results indicated the no. of embryos recovered of donors and the rate of pronuclear embryos did not show any significant difference between breeding and non-breeding seasons（P 〉 0.05）. The positive rate of exogenous TLR4 tested were 21.21 % and 22.58 % in breeding and non-breeding season by Southern blot. The expression level of TLR4 in the transgenic sheep was 1.5 times higher than in the non-transgenic group（P 〈 0.05）. The lambs overexpressing TLR4 had similar growth performance with non-transgenic lambs, and the blood physiological parameters of transgenic and non-transgenic were both in the normal range and did not show any difference.Conclusions： Here we establish an efficient platform for the production of transgenic sheep by the microinjection of pronuclear embryos during the whole year. The over-expression of TLR4 had no adverse effect on the growth of the sheep.

Analysis on Factors Related with Slender Spindle Shaped Apple Trees

[Objectives] This study was to determine the correlations of stem taperingness with small central shaft total roughness,average roughness,total length,and average length by slender spindle pruning mode on different apple cultivars. [Methods]A total of 2 tree structures were selected,namely,the slender spindleshaped Zhongqiuwang from Shen County of Hebei Province and slender spindle-shaped short shoot Fuji from Xingtang County of Hebei Province,to investigate the growth indicators of the small central shafts. SPSS analysis was used to compare the correlations between the taperingness with the growth factors of the small central shafts. [Results]The stem taperingness on slender spindle-shaped Zhongqiuwang was positively correlated with the various growth factors of the small central shaft,and the difference in the positive correlation with average roughness reached the extremely significant level. The stem taperingness of slender spindle-shaped short shoot Fuji was negatively correlated with the total roughness of small central shaft,but was positively correlated with the average roughness,total length and average length. [Conclusions] The results provided scientific and theoretical basis for guiding the dense planting of dwarfing apple tree pruning technology.

[Objectives]The purpose was to standardize the scientific prevention and control of yellow aphids and effectively control their occurrence and spread.[Methods]On the basis of consulting domestic and foreign data and standards extensively,a series of related trials were carried out.[Results]Technical specifications for integrated prevention and control of yellow aphids were formulated.[Conclusions]It is of great significance to the healthy and sustainable development of the fruit trees’industry,the implementation of standardized management,the reduction of pesticide uses,and the reduction of damage caused by yellow aphids to fruit trees,and also provides urgent-needed technical support and guarantee for the prevention and control of yellow aphids.

Analysis of Short-Shoot Fuji Apple Tree Structure and Correlative Factors

In order to clarify the correlation between short-shoot Fuji apple tree structure and different factors under different trimming and pruning modes,we investigate the trunk taperingness of free-spindle short-shoot Fuji and slender-spindle short-shoot Fuji,respectively,as well as the total thickness,average thickness,total length and average length of small main branches in the standard demonstration apple garden in Xingtang County of Hebei Province. By SPSS analysis,we study the correlation between trunk taperingness of trees with different shapes and the growth indices of their small main branches. The results show that the trunk taperingness of free-spindle short-shoot Fuji apple is negatively correlated with the total thickness,average thickness,total length and average length of small main branches,but the correlation is not significant; the trunk taperingness of slender-spindle short-shoot Fuji apple is negatively correlated with the total thickness but positively correlated with other factors,and the correlation with average length reaches a significant level. The results of this study can provide a scientific basis for guiding the high-density dwarf rootstock short-shoot Fuji apple tree trimming technology.

In order to clarify the correlations between the free spindle pruning mode and various factors in Fuji apple,the growth data of the free spindle-shaped short branch Fuji apple Tianhong No. 2 and the free spindle-shaped long branch Fuji apple Changfu No. 2 in the Apple Demonstration Park of Xingtang County,Hebei Province were investigated. Using SPSS statistical software,the correlations between the stem tapering grade and various growth traits were analyzed. The results showed that in Tianhong No. 2,the stem tapering grade was negatively correlated with various growth traits of the central shaft,and the correlation coefficients between the stem tapering grade and the central shaft total thickness,the central shaft average thickness,the central shaft total length and the central shaft average length were-0. 228,-0. 033,-0. 411 and-0. 430,respectively; and in Changfu No. 2,the stem tapering grade was positively correlated with various growth traits of the central shaft,and the correlation coefficients between the stem tapering grade and the central shaft total thickness,the central shaft average thickness,the central shaft total length and the central shaft average length were 0. 475,0. 181,0. 120 and 0. 258,respectively. This paper can provide a scientific basis for guiding the construction of free spindle pruning mode in densely planted dwarf apple trees.

Apple‘Jiping 1’is a new early maturing apple variety bred by Shijiazhuang Pomology Institute,Hebei Academy of Agriculture and Forestry Sciences.To further normalize the standardized production technology of‘Jiping 1’,realize improvement and upgrading of fruit quality,and improve sustainable development of apple industry,according to national and industrial standards,specific requirements on selection of production orchard,planting,soil,fertilizer and water management,shaping and pruning,flower and fruit management,pest control,and fruit harvesting of‘Jiping 1’are put forward,and technical regulation for production of‘Jiping 1’(standard number:DB 13/T 5167-2020)is made.The regulation has important guiding significance for the production of early maturing apple in Hebei Province.
