Electrocatalytic CO_(2)reduction(ECR)offers an attractive approach to realizing carbon neutrality and producing valuable chemicals and fuels using CO_(2)as the feedstock.However,the lack of cost-effective electrocatalysts with better performances has seriously hindered its application.Herein,a one-step co-electrodeposition method was used to introduce Zn,a metal with weak^(*)CO binding energy,into Cu to form Cu/Zn intermetallic catalysts(Cu/Zn IMCs).It was shown that,using an H-cell,the high Faradaic efficiency of C_(2+)hydrocarbons/alcohols()could be achieved in ECR by adjusting the surface metal components and the applied potential.In suitable conditions,FEC_(2+)and current density could be as high as 75%and 40 mA/cm^(2),respectively.Compared with the Cu catalyst,the Cu/Zn IMCs have a lower interfacial charge transfer resistance and a larger electrochemically active surface area(ECSA),which accelerate the reaction.Moreover,the^(*)CO formed on Zn sites can move to Cu sites due to its weak binding with*CO,and thus enhance the C–C coupling on the Cu surface to form C_(2+)products.

Plastic waste is causing serious environmental problems. Developing efficient, cheap and stable catalytic routes to convert plastic waste into valuable products is of great importance for sustainable development, but remains to be a challenging task. Zeolites are cheap and stable, but they are usually not efficient for plastic conversion at a low temperature. Herein a series of microporous and mesoporous zeolites were used to study the influence of porosity and acidity of zeolite on catalytic activity for plastics conversion. It was observed that H-Beta zeolite was an efficient catalyst for cracking high-density polyethylene to gasoline at 240℃, and the products were almost C_(4)–C_(12) alkanes. The effect of porosity and acidity on catalytic performance of zeolites was evaluated, which clearly visualized the good performance of H-Beta due to high surface area, large channel system, large amount accessible acidic sites. This study provides very useful information for designing zeolites for efficient conversion of plastics.

Background： Porcine reproductive and respiratory syndrome virus （PRRSV）, and particularly its highly pathogenic genotype （HP-PRRSV）, have caused massive economic losses to the global swine industry. Results： To rapidly identify HP-PRRSV, we developed a direct reaL-time reverse transcription polymerase chain reaction method （dRT-PCR） that could detect the virus from serum specimen without the need of RNA purification Our dRT-PCR assay can be completed in 1.5 h from when a sample is received to obtaining a result. Additionally, the sensitivity of dRT-PCR matched that of conventional reverse transcription PCR （cRT-PCR） that used purified RNA The lowest detection limit of HP-PRRSV was 6.3 TCIDs0 using dRT-PCR. We applied dRT-PCR assay to 144 field samples and the results showed strong consistency with those obtained by cRT-PCR. Moreover, the dRT-PCR method was able to tolerate 5-20% （v/v） serum. Conclusions： Our dRT-PCR assay allows for easier, faster, more cost-effective and higher throughput detection of HP-PRRSV compared with cRT-PCR methods. To the best of our knowledge, this is the first report to describe a real-time RT-PCR assay capable of detecting PRRSV in crude serum samples without the requirement for purifying RNA. We believe our approach has a great potential for application to other RNA viruses.

The high intraspecies heterogeneity of Baciillus coagulans leads to significant phenotypic differences among different strains.Thus,6 B.coagulans strains were tested in the present study using an irritable bowel syndrome(IBS)animal model to determine whether the IBS-alleviating effects of B.coagulans strains are strain-specific.The results of this study showed that the ingestion of B.coagulans GBI-30,6086,and B.coagulans CCFM1041 significantly alleviated IBS symptoms in mice.In contrast,other B.coagulans strains showed no or limited alleviating effects on IBS symptoms.According to our experimental results,the two main common features of these strains were as follows:1)The resistance of vegetative cells to bile salts,and 2)ability to synthesize specific lipids and secondary metabolites.Screening strains based on these two indicators may greatly reduce costs and provide a basis for mining new functional B.coagulans strains.Our results also suggest that administration of B.coagulans could significantly regulate microbiota dysbiosis in animal models.Moreover,the close relationships between the gut microbiota,gut microbiota metabolites,and IBS were further confirmed in this study.

Akkermansia muciniphila play an important in ameliorating obesity but is not allowed for direct consumption in most countries.To date,microbiota-directed foods selectively promote the targeted human gut microbes,providing a strategy for A.muciniphila enhancement.Multiple studies have indicated the potential regulation of the polyphenol on A.muciniphila.Therefore,a polyphenol screening based on A.muciniphila upregulation was performed in mice.Chlorogenic acid(CGA)exhibited a greater response to A.muciniphila upregulation.Furthermore,we found that CGA did not directly promote A.muciniphila growth or mucin secretion.Microbiome and metabolomics revealed that the increased abundance of A.muciniphila resulted from the inhibition of CGA on Desulfovibrio and Alistipes and the influence of docosahexaenoic acid,β-hydroxybutyrate,and N-acetyl-lactosamine.Finally,to confirm the regulation of CGA on A.muciniphila under disease conditions,high-fat diet-fed mice were established.The results showed CGA promoted A.muciniphila growth,and we expectedly found that CGA suppressed the augment in body weight of mice,significantly attenuated adipose tissue abnormality,provided liver protection and improved gut barrier integrity.These results suggest that CGA inhibits the development of obesity.Overall,our results indicate that microbiota-directed food is a promising approach for the treatment of obesity.

Heparin， a glycosaminoglycan， is a stable source of carbon that supports the growth of microorganisms in the human intestine. It is also a commonly used anticoagulant drug in clinical practice， with significant therapeutic effects. Low molecular weight heparin （LMWH） is a highly active low molecular weight fragment obtained via enzymatic reaction or the chemical degradation of heparin. LMWH has been applied globally in the prevention and treatment of venous thromboembolism in thrombosis patients. Simultaneously， as a potential prebiotic， because of its low molecular weight， LMWH can be well degraded by the gut microbiota to maintain intestinal balance. Enzymatic heparin degradation has recently emerged as a viable disposal method for LMWH preparation; however， only very few benchmark enzymes have been thoroughly described and subjected to protein engineering to improve their properties over the past few years. The commercialization of enzymes will require the development of robustly engineered enzymes that meet the demands of industrial processes. Herein， we report a rational protein engineering strategy that includes molecular dynamic simulations of flexible amino acid mutations and disulfide bond screening. Several Bt-HepI mutants were obtained and screened for high thermal stability. We obtained the Bt-HepID204C/K208C/H189W/Q198Rvariant， which features a stabilized protein surface structure， with a 1.3-fold increase in kcat/Km， a 2.44-fold increase in thermal stability at 50℃， and a 1.8-fold decrease in the average molecular weight of LMWH produced at 40℃ compared with that seen with Bt-HepIWT. Our study establishes a strategy to engineer thermostable HepI to underpin its industrial applications.

与传统现浇框架结构相比,装配式钢节点混凝土框架结构建造速度更快,但框架钢连接节点处存在应力突变,因此,有必要采用基于数字孪生的手段监测该实际工程结构的施工过程,以获得钢节点对结构性能的影响。基于BIM、有限元和传感器等技术搭建了基于数字孪生的新型装配式钢节点混凝土框架结构的智能建造框架,从物理数据收集、虚拟模型建立、模型信息交互3个方面提出基于数字孪生的建筑结构智能建造实现方法。在新型装配式钢节点混凝土框架实际工程建造阶段,采用传感器技术实现工程中关键点数据的实时监测,并将其与BIM和有限元数据进行比较,进一步调整与修正物理模型中的框架结构受力情况,最终实现装配式钢节点混凝土框架数字孪生模型的建立和应用。研究表明,该数字孪生模型能有效对新型装配式钢节点混凝土框架结构进行实时监测,实现基于传感器网络和时空参数分析的危险点预测,有效减少资源消耗,为该结构的应用提供有效数据信息。

目的评价透明帽辅助无痛胃镜下食管异物取出术的临床疗效。方法抽取登封市人民医院2018年5月至2021年5月接收的80例消化道异物患者,将采用常规无痛胃镜下食管异物取出术治疗的40例患者纳入对照组,采用透明帽辅助无痛胃镜下食管异物取出术治疗的40例患者纳入观察组。比较两组患者术前(T0)、置入胃镜时(T1)、取异物时(T2)的心率与平均动脉压(MAP)变化;比较两组患者内镜下视野清晰度、内镜下操作时间与手术成功情况,比较两组并发症发生率。结果T1、T2时,两组心率与MAP均较T0时升高,且T2时的心率与MAP均较T1时升高,但观察组各时点心率与MAP均低于对照组(P均0.05)。结论透明帽辅助无痛胃镜下食管异物取出术可提高消化道异物患者内镜下视野清晰度与手术成功率,缩短内镜下操作时间,利于稳定患者血流动力学。

建立了一种利用"双探针夹心银染"技术快速检测戊型肝炎病毒(HEV)的基因芯片方法.提取的病毒核酸经过RT-PCR扩增,PCR体系中用掺入生物素修饰的dUTP扩增得到带有生物素的核酸片段,5′末端-NH2修饰的同源寡核苷酸作为捕获探针固定到处理后的玻片上,该探针识别病毒的带有生物素的核酸片段,而链霉亲和素标记的胶体金与生物素紧密结合,通过银沉积在金颗粒表面放大信号可以目视结果.采用该方法对已经确证的86个血清样本进行检测,检出52个阳性和34个阴性样本,结果与荧光定量PCR检出一致.

将锂辉石加入中性硼硅玻璃配方中,用锂辉石中的Li_(2)O等量替代原配方化学组成中的B_(2)O_(3)、Na_(2)O,研究Li_(2)O等量替代B_(2)O_(3)、Na_(2)O后Li_(2)O加入量对中性硼硅玻璃黏度性能的影响。试验结果表明,随着Li_(2)O替代B_(2)O_(3)、Na_(2)O质量分数的增加,中性硼硅玻璃低温黏度点均有不同程度的下降。对于相同含量的Li_(2)O、B_(2)O_(3)、Na_(2)O,Li_(2)O降低中性硼硅玻璃低温黏度点的能力比B_(2)O_(3)、Na_(2)O强。因此,加入Li_(2)O可以降低中性硼硅玻璃低温热处理温度。对于相同含量的Li_(2)O、B_(2)O_(3)、Na_(2)O,Li_(2)O降低中性硼硅玻璃高温黏度点的能力远比B_(2)O_(3)、Na_(2)O强。因此,引进Li_(2)O可以显著降低玻璃加工对应的温度。Li_(2)O可以显著降低中性硼硅玻璃低温、高温黏度点,因此,可以降低低温热处理及高温玻璃加工对应的温度,达到降低生产成本和节能减排的目的。
