In the process of software development,the ability to localize faults is crucial for improving the efficiency of *** speaking,detecting and repairing errant behavior at an early stage of the development cycle considerably reduces costs and development *** have tried to utilize various methods to locate the faulty ***,failing test cases usually account for a small portion of the test suite,which inevitably leads to the class-imbalance phenomenon and hampers the effectiveness of fault ***,in this work,we propose a new fault localization approach named *** obtaining dynamic execution through test cases,ContextAug traces these executions to build an information model;subsequently,it constructs a failure context with propagation dependencies to intersect with new model-domain failing test samples synthesized by the minimum variability of the minority feature *** contrast to traditional test generation directly from the input domain,ContextAug seeks a new perspective to synthesize failing test samples from the model domain,which is much easier to augment test *** conducting empirical research on real large-sized programs with 13 state-of-the-art fault localization approaches,ContextAug could significantly improve fault localization effectiveness with up to 54.53%.Thus,ContextAug is verified as able to improve fault localization effectiveness.

Predictive trace analysis （PTA）, a static trace analysis technique for concurrent programs, can offer power- ful capability support for finding concurrency errors unseen in a previous program execution. Existing PTA techniques always face considerable challenges in scaling to large traces which contain numerous critical events. One main reason is that an analyzed trace includes not only redundant memory accessing events and threads that cannot contribute to dis- covering any additional errors different from the found can- didate ones, but also many residual synchronization events which still affect PTA to check whether these candidate ones are feasible or not even after removing the redundant events. Removing them from the trace can significantly improve the scalability of PTA without affecting the quality of the PTA results. In this paper, we propose a biphasic trace filter ap- proach, BIFER in short, to filter these redundant events and residual events for improving the scalability of PTA to expose general concurrency errors. In addition, we design a model which indicates the lock history and the happens-before his- tory of each thread with two kinds of ways to achieve the efficient filtering. We implement a prototypical tool BIFER for Java programs on the basis of a predictive trace analysis framework. Experiments show that BIFER can improve the scalability of PTA during the process of analyzing all of the traces.

1 Introduction For developers,fault localization is the most tedious and time-consuming *** order to reduce the burden of developers,researchers have developed various techniques to help reduce the cost of locating faults based on possibility of containing a fault in program statements[1].Among them,coverage-based fault localization is one of the most studied automated fault localization ***-based fault localization methods utilize a type of preliminary information to localize the faults in programs,which is a matrix of statements’coverage *** value of each element is 1 or 0,in which 1 means a statement is executed and 0 denotes a statement is not executed[1].

The prognosis of brain metastases(BM)is traditionally *** are mainly treated by local radiotherapy,including stereotactic radiosurgery(SRS)or whole brain radiation therapy(WBRT).Recently,immunotherapy(i.e.,immune checkpoint inhibitors,ICI)has demonstrated a survival advantage in multiple malignancies commonly associated with ***,radiotherapy and ICI both treat BM efficiently;hence,their combination seems *** this review,we summarize the existing preclinical and clinical evidence that supports the applicability of radiotherapy as a sensitizer of ICI for ***,we discuss the optimal timing at which radiotherapy and ICI should be administered and review the safety of the combination *** from a few clinical studies suggest that combining SRS or WBRT with ICI simultaneously rather than consecutively potentially enhances brain abscopal-like responses and ***,there is a lack of conclusion about the definition of"simultaneous";the cumulative toxic effect of the combined therapies also requires further ***,ongoing and planned prospective trials are needed to further explore and validate the effect,safety,and optimal timing of the combination of immunotherapy with radiotherapy for patients with BM.

Immunotherapy represented by anti-PD-(L)1 and anti-CTLA-4 inhibitors has revolutionized cancer treatment,but challenges related to resistance and toxicity still *** to the advancement of immuno-oncology,an increasing number of novel immunoregulatory targets and mechanisms are being revealed,with relevant therapies promising to improve clinical immunotherapy in the foreseeable ***,comprehending the larger picture is *** this review,we analyze and summarize the current landscape of preclinical and translational mechanistic research,drug development,and clinical trials that brought about next-generation pharmacological immunoregulatory anti-cancer agents and drug candidates beyond classical immune checkpoint *** with further clarification of cancer immunobiology and advances in antibody engineering,agents targeting additional inhibitory immune checkpoints,including LAG-3,TIM-3,TIGIT,CD47,and B7 family members are becoming an important part of cancer immunotherapy research and discovery,as are structurally and functionally optimized novel anti-PD-(L)1 and anti-CTLA-4 agents and agonists of co-stimulatory molecules of T *** by bispecific T cell engagers,newly emerging bi-specific and multi-specific antibodies targeting immunoregulatory molecules can provide considerable clinical ***-generation agents also include immune epigenetic drugs and cytokine-based *** therapies,cancer vaccines,and oncolytic viruses are not covered in this *** comprehensive review might aid in further development and the fastest possible clinical adoption of effective immuno-oncology modalities for the benefit of patients.

Lately,the success of ICIs has drastically changed the landscape of cancer treatment,and several immune checkpoint inhibitors(ICIs)have been approved by the US Food and Drug Administration(FDA)for advanced non-small cell lung cancer(NSCLC).However,numerous patients are resistant to ICIs and require additional procedures for better efficacy ***,combination therapy is urgently needed to strengthen the anti-tumor immunity.A variety of preclinical and clinical studies combining ICIs with radiotherapy(RT)have demonstrated that the combination could induce synergistic effects,as RT overcomes the resistance to ***,the underlying mechanism of the synergistic effect and the optimal arrangement of the combination therapy are indecisive ***,this review was conducted to provide an update on the current clinical trial results and highlighted the ongoing *** also discussed the optimal parameters in clinical trials,including radiation dose,radiation fractionation,radiation target field,and sequencing of combination *** this review,we found that combination therapy showed stronger anti-tumor immunity with tolerable toxicities in clinical ***,the best combination mode and potential biomarkers for the target patients in combination therapy are still unclear.

CD38 is the main enzyme for nicotinamide adenine dinucleotide(NAD)degradation in mammalian *** NAD levels are closely related to metabolic syndromes and aging-related *** study showed that CD38 deficiency significantly alleviated angiotensin II(Ang ll)-induced vascular remodeling in mice,as shown by decreased blood pressures;reduced vascular media thickness,media-to-lumen ratio,and collagen deposition;and restored elastin ***,our bone marrow transplantation assay showed that CD38 deficiency in lymphocytes led to lack of protection against Ang ll-induced vascular remodeling,suggesting that the effects of CD38 on Ang ll-induced vascular remodeling might rely primarily on vascular smooth muscle cells(VSMCs),not *** addition,we observed that CD38 deficiency or NAD supplementation remarkably mitigated Ang ll-induced vascular senescence by suppressing the biogenesis,secretion,and internalization of senescence-associated small extracellular vesicles(SA-sEVs),which facilitated the senescence of neighboring non-damaged ***,we found that the protective effects of CD38 deficiency on VSMC senescence were related to restoration of lysosome dysfunction,particularly with respect to the maintenance of sirtuin-mediated mitochondrial homeostasis and activation of the mitochondria-lysosomal axis in *** conclusion,our findings demonstrated that CD38 and its associated intracellular NAD decline are critical for Ang ll-induced VSMC senescence and vascular remodeling.
