Rapid accuracy determining DNA purity and concentration in heavy oils by spectrophotometry methods

DNA analysis is the core of biotechnology applied in petroleum resources and engineering. Traditionally accurate determination of DNA purity and concentration by spectrometer is the first and critical step for downstream molecular biology research. In this study, three different spectrophotometry methods, BPM, NDTT and NPMTTZ were compared for their performance in determining DNA concentration and purity in 32 oil samples, and molecule methods like quantitative real-time PCR (qPCR) and high-throughput sequence were also performed to help assess the accuracy of the three methods in determining DNA concentration and purity. For ordinary heavy oil (OHO), extra heavy oil (EHO) and super heavy oil (SHO), the characteristics of high viscosity (η), density (ρ) and resin plus asphaltene content will affect the DNA extraction and UV determination. The DNA concentration was decreased as density increased: OHO (11.46 ± 18.34 ng/μL), EHO (6.68 ± 9.67 ng/μL) and SHO (6.20 ± 7.83 ng/μL), and the DNA purity was on the reverse: OHO (1.31 ± 0.27), EHO (1.54 ± 0.20), and SHO (1.83 ± 0.32). The results suggest that spectrophotometry such as BPM and NPMTTZ are qualitatively favorite methods as the quick non-consumable methods in determining DNA concentration and purity of medium oil and heavy oil.

目的探讨显微镜辅助经Quadrant通道结合经皮椎弓根钉固定行微创小切口经椎间孔腰椎椎间融合术(MIS-TLIF)治疗单节段腰椎退行性滑脱症的疗效。方法回顾性分析2018年4月至2021年3月行椎体间融合术治疗单节段腰椎退行性滑脱症患者的临床资料,其中采用开放经椎间孔腰椎椎间融合术(TLIF)治疗患者41例(开放组),经Wiltse入路Quadrant通道直视下TLIF治疗患者25例(微创组),显微镜下减压并采用经皮椎弓根钉TLIF治疗患者38例(显微组)。记录3组患者手术时间、术中出血量、X线透视次数、引流量、术后2天和7天切口周围疼痛视觉模拟评分(VAS)、并发症情况,术后CT检查评估椎弓根钉置钉准确率,记录术前及术后6个月、24个月日本骨科协会(JOA)评分和Oswestry功能障碍指数(ODI),末次随访时采用改良MacNab标准评价临床疗效。结果所有手术均顺利完成。显微组术中出血量、引流量、术后2天和7天切口周围疼痛VAS评分均小于微创组和开放组(P0.05)。术后6个月和24个月融合率3组间比较无统计学差异(P>0.05)。按改良MacNab标准,3组患者疗效优良率无统计学差异(P>0.05)。结论显微镜辅助通道下MIS-TLIF治疗单节段腰椎退行性滑脱症,术中出血少、切口疼痛感小、并发症少,采用经皮椎弓根钉置钉准确率高,融合率、疗效优良率与开放手术一致,临床疗效好。

Boron doping-induced interconnected assembly approach for mesoporous silicon oxycarbide architecture

Despite desirable progress in various assembly tactics, the main drawback associated with current assemblies is the weak interparticle connections limited by their assembling protocols. Herein, we report a novel boron doping-induced interconnection-assembly approach for fabricating an unprecedented assembly of mesoporous silicon oxycarbide nanospheres, which are derived from periodic mesoporous organosilicas. The as-prepared architecture is composed of interconnected, strongly coupled nanospheres with coarse surfaces. Significantly, through delicate analysis of the as-formed boron doped species, a novel melt-etching and nucleation-growth mechanism is proposed, which offers a new horizon for the developing interconnected assembling technique. Furthermore, such unique strategy shows precise controllability and versatility, endowing the architecture with tunable interconnection size, surface roughness and switchable primary nanoparticles. Impressively, this interconnected assembly along with tunable surface roughness enables intrinsically dual(both structural and interfacial) stable characteristics, achieving extraordinary long-term cycle life when used as a lithium-ion battery anode.

Recently,graphene oxide(GO),MXene,carbon nanotubes(CNTs) have been used for compounding with other materials as anodes and cathodes to achieve excellent electrochemical properties for metal-ion ***,few researches have focused on the differences between the three ***,silicon,as a typical anode,is selected to integrate with MXene,GO and CNTs in carbon nanofibers(CNFs) and form Si/MXene@CNFs,Si/GO@CNFs and Si/CNTs@CNFs,*** with the results,it can be realized that these CNFs with a significant improved performance compared with pure Si@CNFs show superiority in different aspects of electrochemical ***,the reasons for the superiority are also discussed in this *** addition of MXene can improve the cycle stability of the electrodes,thereby obtaining a high capacity retention rate,CNTs are favorable for the enhancement of rate performance,and the electrodes reversible capacity can be increased due to the addition of ***,the studies on three additives may contribute to the rational design of silicon-based and other anode materials.

上海市中小学教室采光照明现况及监管效果分析

目的了解上海市中小学教室采光照明现况及监管效果,为改善学校教室视觉环境提供依据。方法于2021年4月至2023年12月,对上海市正常开设中的全部普通中小学校1 735所进行教室采光照明监测,并计算各指标合格率。采用χ^(2)检验或Fisher确切概率法比较不同学段、区域、办学性质及采取监管措施(包括约谈、宣教、责令改正、发监督意见书、承诺整改期限、依法处罚)前后合格率的差异。结果中小学教室采光照明合格率为30.1%,其中采光合格率85.6%,照明合格率32.9%;小学和中学教室采光照明合格率(32.4%,28.1%)差异无统计学意义(χ^(2)=3.76,P>0.05),城市与乡镇(32.6%,26.7%)、公办与民办学校(31.4%,20.6%)教室采光照明合格率差异均有统计学意义(χ^(2)值分别为6.99,9.92,P值均<0.05)。采取监管措施整改后,教室采光照明合格率由30.1%提升至83.2%,其中采光合格率由85.6%提升至91.1%,照明合格率由32.9%提升至90.5%;整改前与整改后比较,教室采光照明合格率、采光合格率和照明合格率差异均有统计学意义(χ^(2)值分别为995.29,25.34,1 219.87,P值均<0.01)。结论有效的监管措施可促进教室采光照明的改善。应重视改善学校教室照明,为学生提供良好的视觉环境。

儿童胫骨短缩延长矿化期钢板辅助内固定的临床效果

目的探讨儿童胫骨短缩畸形延长矿化期更换钢板辅助内固定的临床疗效。方法回顾性分析2019年11月至2022年10月天津市天津医院小儿骨科利用环形外固定架延长后更换钢板辅助内固定治疗的胫骨短缩10例患儿资料(钢板辅助组),并以同期单纯采用环形外固定架延长的患儿16例作为对照(外固定架组)。26例中男15例、女11例,左侧10例、右侧16例,手术时年龄为(8.8±4.7)岁(范围5.1~13.8岁)。对比两组患儿的性别、侧别、手术时年龄,记录两组患儿延长长度、外固定架指数、愈合指数、Kolcaba舒适状况量表评分、膝和踝关节活动度、并发症及平均住院总费用。结果钢板辅助组与外固定架组性别、侧别、手术时年龄的差异无统计学意义,延长长度分别为(5.44±1.25)、(5.78±1.11)cm,差异无统计学意义(t=0.096,P=0.096)。钢板辅助组外固定架指数及愈合指数为(17.86±2.94)、(40.94±6.63)d/cm,均短于外固定架组的(50.97±7.03)、(45.24±5.98)d/cm,差异有统计学意义(t=13.299,P<0.001;t=1.289,P=0.033)。矿化期更换钢板辅助内固定后患儿Kolcaba舒适状况量表评分(103.35±4.60)分明显高于外固定架组的(70.65±15.06)分,差异有统计学意义(t=6.821,P=0.001)。术前、延长结束及末次随访时两组膝和踝关节活动度比较的差异均无统计学意义。延长端愈合时钢板辅助组膝关节活动度(137.89°±4.40°)及踝关节活动度(64.35°±5.50°)均大于外固定架组(114.09°±13.60°和56.65°±8.86°),差异均有统计学意义(t=17.235,P<0.001;t=7.821,P=0.002)。钢板辅助组4例出现针道感染,无再骨折发生;外固定架组13例出现针道感染,2例拆除外固定架后发生再骨折。钢板辅助组平均住院总费用相对较高,差异有统计学意义(t=3.745,P=0.036)。结论儿童胫骨短缩延长矿化期更换钢板辅助内固定临床效果可靠,可显著缩短外固定架佩戴时间,利于截骨延长端愈合及膝、踝关节功能恢复,患儿舒适度高,并发症少,适用于大距离延长、多次延长、关节功能受限、外固定架耐受性差及治疗依从性差的患儿。

Vulnerability and adaptation of an oasis social–ecological system affected by glacier change in an arid region of northwestern China

The Hexi Inland River Basin in an arid region of northwestern China was chosen as the study area for this research. The authors define the vulnerability of an oasis social-ecological system to glacier change; select 16 indicators from natural and socioeconomic systems according to exposure, sensitivity, and adaptive capacity; and construct a vulnerability-assessment indicator system aimed at an inland river basin in the arid region of Northwestern China. Vulnerability of the oasis socialecological system affected by glacier change in the study area is evaluated by Spatial Principal Component Analysis(SPCA) under the circumstance of glacier change. The key factors affecting the vulnerability are analyzed. The vulnerability of the oasis social-ecological system in the Hexi Inland River Basin affected by glacier change is of more than medium grade, accounting for about 48.0% of the total number of counties in the study area. In terms of the spatial pattern of the vulnerability, the oasis economic belt is the most vulnerable. With the rapid development of the area's society and economy, the exposure of the system to glacial changes is significantly increased; and an increase in glacial meltwater is not enough to overcome the impact of increased exposure, which is the main reason for the high vulnerability. Based on the result of the vulnerability analysis and combined with the present industrial structure in the Hexi Inland River Basin, near-,medium-, and long-term adaptation initiatives are put forward in the article.

Adaptation Management of Mountain Tourism Service: The Case of the Source Regions of the yangtze and Yellow River

Mountain areas are often rich in ecological diversity and recreational opportunities. Mountain tourism is thought to be an effective and important means for maintaining and expanding rural economies and, thus, improving the living conditions of rural societies. As mountain tourism service research is a professional field with several disciplines involved, a multi-disciplinary management pIatform is needed and it facilitates participation in sustainable mountain development by diverse stakeholders. With the source regions of the yangtze and the Yellow River as a case study, this paper presents a conceptual framework for an adaptation management of mountain tourism services according to technical, policy, social and economic dimensions. The framework is based on a vulnerability assessment of mountain ecosystems, and can serve as a reference for the development of tourism service in other mountain areas.

Electrochemical reduction of CO_(2)into high-value hydrocarbons and alcohols by using Cu-based catalysts is a promising and attractive technology for CO_(2)capture and utilization,resulting from their high catalytic activity and *** mobility and accessibility of active sites in Cubased catalysts significantly hinder the development of efficient Cu-based catalysts for CO_(2)electrochemical reduction reaction(CO_(2)RR).Herein,a facile and effective strategy is developed to engineer accessible and structural stable Cu sites by incorporating single atomic Cu into the nitrogen cavities of the host graphitic carbon nitride(g-C_(3)N_(4))as the active sites for CO_(2)-to-CH_(4)conversion in CO_(2)*** regulating the coordination and density of Cu sites in g-C_(3)N_(4),an optimal catalyst corresponding to a one Cu atom in one nitrogen cavity reaches the highest CH_(4)Faraday efficiency of 49.04%and produces the products with a high CH_(4)/C_(2)H_(4)ratio over *** work provides the first experimental study on g-C_(3)N_(4)-supported single Cu atom catalyst for efficient CH_(4)production from CO_(2)RR and suggests a principle in designing highly stable and selective high-efficiency Cu-based catalysts for CO_(2)RR by engineering Cu active sites in 2D materials with porous crystal structures.
