DNA metabarcoding analysis of fungal community on surface of four root herbs

Objective: Angelicae Sinensis Radix(ASR, Danggui in Chinese), Cistanches Herba(CH, Roucongrong in Chinese), Ginseng Radix et Rhizoma(PG, Renshen in Chinese), and Panacis Quinquefolii Radix(PQ,Xiyangshen in Chinese), widely used as medicine and dietary supplement around the world, are susceptible to fungal and mycotoxin contamination. In this study, we aim to analyze their fungal community by DNA metabarcoding.Methods: A total of 12 root samples were collected from three main production areas in China. The samples were divided into four groups based on herb species, including ASR, CH, PG, and PQ groups. The fungal community on the surface of four root groups was investigated through DNA metabarcoding via targeting the internal transcribed spacer 2 region(ITS2).Results: All the 12 samples were detected with fungal contamination. Rhizopus(13.04%-74.03%),Aspergillus(1.76%-23.92%), and Fusarium(0.26%-15.27%) were the predominant genera. Ten important fungi were identified at the species level, including two potential toxigenic fungi(Penicillium citrinum and P. oxalicum) and eight human pathogenic fungi(Alternaria infectoria, Candida sake, Hyphopichia burtonii, Malassezia globosa, M. restricta, Rhizopus arrhizus, Rhodotorula mucilaginosa, and Ochroconis tshawytschae). Fungal community in ASR and CH groups was significantly different from other groups,while fungal community in PG and PQ groups was relatively similar.Conclusion: DNA metabarcoding revealed the fungal community in four important root herbs. This study provided an important reference for preventing root herbs against fungal and mycotoxin contamination.

The accessible seismological dataset of a high-density 2D seismic array along Anninghe fault

The scientific goal of the Anninghe seismic array is to investigate the detailed geometry of the Anninghe fault and the velocity structure of the fault zone.This 2D seismic array is composed of 161 stations forming sub-rectangular geometry along the Anninghe fault,which covers 50 km and 150 km in the fault normal and strike directions,respectively,with~5 km intervals.The data were collected between June 2020 and June 2021,with some level of temporal gaps.Two types of instruments,i.e.QS-05A and SmartSolo,are used in this array.Data quality and examples of seismograms are provided in this paper.After the data protection period ends(expected in June 2024),researchers can request a dataset from the National Earthquake Science Data Center.

Objective:To provide high-quality clinical evidence of the efficacy of Tibetan medicine Honghua Ruyi(HHRY)pills for endometriosis-associated dysmenorrhea.Methods:This study constitutes a multicenter,randomized,double-blind,placebo-controlled trial encompassing a three-menstrual cycle intervention followed by a three-menstrual cycle follow-up period.A total of 164 eligible females with endometriosis-associated dysmenorrhea were randomly divided into HHRY pills and placebo groups in a 1:1 ratio.The primary outcome included dysmenorrhea symptoms assessed using Visual Analog Scale(VAS)scores and quality of life,whereas the secondary outcome measures included the maximum VAS for non-menstrual pelvic pain,duration of pain episodes(in days),frequency and quantity of the consumption of ibuprofen sustained-release capsules(or other non-steroidal anti-inflammatory drugs),and days off work/study for staff/student due to dysmenorrhea,ovarian cyst,and/or pelvic nodule size.The safety was monitored throughout the treatment period.All the analyses were based on the intention-to-treat principle.For continuous outcomes,simple or multiple linear regressions were used to estimate the differences between the HHRY pills and placebo groups,with categorical data expressed as the number and percentage of occurrences.Differences were compared using the chi-square test or Fisher's exact test.The predefined analysis was adjusted for concomitant treatment,a variable considered to be associated with outcomes but unaffected by treatment allocation.Estimates of treatment effects were reported with 95%confidence intervals.Two-tailed P values≤.05 were considered statistically significant.Conclusion:Positive results from this trial,upon completion would provide robust evidence for the efficacy and safety of HHRY pills in treating dysmenorrhea in patients with endometriosis.

＜正＞Aim of the study:Firstly to identify Panax genus by SNP sites,and to set up a special molecular identification method of P.ginseng,P.notoginseng,P.quinquefolius and P.japonicus in Pharmacopoeia of China.Materials and methods:Three regions（ITS2,matK and psbA-trnH）were chosen to investigate intraspecific variation by K2P（Kimura-two-parameter）model in medicinal plants of Panax genus from different populations based on DNA barcoding analysis.Total of 167 ITS2 sequences of the four species were analyzed,including 87 samples of P.ginseng,27 of P.quinquefolius,31 of P.notoginseng and 22 of P.japonicus.SNPs in ITS2 region were detected and the phylogenetic tree using maximum parsimony method based on ITS2 sequence was built to identify Panax species with their adulterants.Patterns of intragenomic and intraspecific variation of ITS2 in the four Panax species were studied by comparing all of the multiple copies in genome by 454 high throughout sequencing with the sequences obtained from large number of samples by the direct PCR product sequencing.Results:Intraspecific genetic distance in P.ginseng,P.quinquefolius and P.notoginseng were pretty low.Zero variation of ITS2 occured in p.ginseng and p.quinquefolius, while the intragenomic of P.ginseng and P.quinquefolius also shared little variation.The dominant variation in P.ginseng,P.quinquefolius possessed about 97.61%and 98.64%respectively of the whole variation in genome,which showed that the ITS2 could be considered as a single locus in genome of P.ginseng and P.quinquefolius.Based on 87 samples of P.ginseng and 27 samples of P. quinquefolius analysis,two SNPs in ITS2 sequences were proved to exist stably between them. What’s more,one of the two SNPs existing in ITS2 of P.notoginseng and one SNP in ITS2 of P.quinquefolius could be used to distinguish them from all of the Panax species respectively.From MP tree of ITS2,the three of the four species aligned in its own clade except P.japonicus,separated with their closely related species and adulterants,indicating that IT

为了解决传统摩擦摆隔震支座(friction pendulum bearing, FPB)刚度和阻尼特性固定不能适用于更为广泛的地震和结构条件等问题,研发了变频(曲率)变摩擦的摩擦摆隔震支座(friction pendulum bearing with variable frequency and friction, FPB-VFF),其刚度和阻尼特性随支座位移变化。分析了该支座的力学性能和工作原理,推导其理论本构模型和滞回特性参数表达式;建立FPB-VFF精细化有限元模型,得到其等效刚度、等效阻尼比和耗能等,研究滞回特性随支座参数的变化规律;进行了FPB-VFF力学性能试验,验证理论分析和有限元模拟的正确性。研究表明:FPB-VFF的等效刚度和耗能随滑移区2摩擦系数线性增加,等效阻尼比随滑移区2的摩擦系数呈反比例函数关系变化;相比FPB,FPB-VFF的等效刚度、耗能和等效阻尼比增大且最大增幅分别为146%、230%和30%。

Corona virus disease 2019(COVID-19)has exerted a profound adverse impact on human health.Studies have demonstrated that aerosol transmission is one of the major transmission routes of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).Pathogenic microorganisms such as SARS-CoV-2 can survive in the air and cause widespread infection among people.Early monitoring of pathogenic microorganism transmission in the atmosphere and accurate epidemic prediction are the frontier guarantee for preventing large-scale epidemic outbreaks.Monitoring of pathogenic microorganisms in the air,especially in densely populated areas,may raise the possibility to detect viruses before people are widely infected and contain the epidemic at an earlier stage.The multi-scale coupled accurate epidemic prediction system can provide support for governments to analyze the epidemic situation,allocate health resources,and formulate epidemic response policies.This review first elaborates on the effects of the atmospheric environment on pathogenic microorganism transmission,which lays a theoretical foundation for the monitoring and prediction of epidemic development.Secondly,the monitoring technique development and the necessity of monitoring pathogenic microorganisms in the atmosphere are summarized and emphasized.Subsequently,this review introduces the major epidemic prediction methods and highlights the significance to realize a multi-scale coupled epidemic prediction system by strengthening the multidisciplinary cooperation of epidemiology,atmospheric sciences,environmental sciences,sociology,demography,etc.By summarizing the achievements and challenges in monitoring and prediction of pathogenic microorganism transmission in the atmosphere,this review proposes suggestions for epidemic response,namely,the establishment of an integrated monitoring and prediction platform for pathogenic microorganism transmission in the atmosphere.

OBJECTIVE: To summarize the evidence from Traditional Chinese Medicine(TCM) practice in the treatment of acute primary headache and provide clinical practice guidance. METHODS: The guidelines were developed in accordance with the World Health Organization guideline development manual. After the establishment of steering committee, panel and the registration and protocol formulation, the evidence on TCM for acute primary headache from published guidelines, clinical evidence, and expert experience and consensus were collected. The grading of recommendations assessment, development and evaluation method was used to grade the evidence and make the recommendations. RESULTS: Based on the available evidence, the guidelines recommended three TCM herbal decoctions, six Chinese patent medicines, and two kinds of external application of Chinese herbal medicines. Diagnostic recommendations based on the expert experience and consensus were also included in the guidelines. CONCLUSION: TCM diagnosis and treatment of decoction, Chinese patent medicine and external application for treating acute primary headache were recommended. We hope these guidelines will be helpful in standardize the TCM acute treatment of primary headache.

生态脆弱性评价是生态治理和修复的重要前提。柴达木盆地是对全球气候变化和人类活动干扰最为敏感的区域之一。通过SRP模型构建柴达木盆地生态脆弱性评价指标体系,基于地理信息技术和遥感技术,采用层次分析法,系统、定量地对柴达木盆地生态脆弱性进行评价。结果表明:1)柴达木盆地微度,轻度类型脆弱区面积为79326 km^(2),占区域总面积的29.28%;中度类型脆弱区面积为67044 km^(2),占区域总面积的24.74%;重度,极度类型脆弱区面积为124613 km^(2),占区域总面积的45.98%。2)柴达木盆地东中西部生态脆弱性表现为“低-中-高”的分布特征。3)柴达木盆地生态脆弱性呈现出强烈的空间正相关特征。

安大简《仲尼曰》的部分章节与今本《论语》存在同源关系,更多章节则或与传世文献相异,或不见于传世文献,《孟子》所引孔子言论亦是如此。可见,由于孔子弟子各有所记,在战国时期形成了多种“孔子语录”汇编本,其中多数在流传中亡佚,部分散落在《礼记》等儒家文献中。《论语》的早期传本源于“孔子语录”汇编本,并在汉代武、昭、宣时期逐渐经典化,形成多种抄本。汉代后期,张禹所传“张侯《论》”逐渐淘汰其他抄本,成为今本《论语》的主要源头。
