3D bioprinting of in vitro porous hepatoma models:establishment,evaluation,and anticancer drug testing

Traditional tumor models do not tend to accurately simulate tumor growth in vitro or enable personalized treatment and are particularly unable to discover more beneficial targeted drugs.To address this,this study describes the use of threedimensional(3D)bioprinting technology to construct a 3D model with human hepatocarcinoma SMMC-7721 cells(3DP-7721)by combining gelatin methacrylate(GelMA)and poly(ethylene oxide)(PEO)as two immiscible aqueous phases to form a bioink and innovatively applying fluorescent carbon quantum dots for long-term tracking of cells.The GelMA(10%,mass fraction)and PEO(1.6%,mass fraction)hydrogel with 3:1 volume ratio offered distinct pore-forming characteristics,satisfactorymechanical properties,and biocompatibility for the creation of the 3DP-7721 model.Immunofluorescence analysis and quantitative real-time fluorescence polymerase chain reaction(PCR)were used to evaluate the biological properties of the model.Compared with the two-dimensional culture cell model(2D-7721)and the 3D mixed culture cell model(3DM-7721),3DP-7721 significantly improved the proliferation of cells and expression of tumor-related proteins and genes.Moreover,we evaluated the differences between the three culture models and the effectiveness of antitumor drugs in the three models and discovered that the efficacy of antitumor drugs varied because of significant differences in resistance proteins and genes between the three models.In addition,the comparison of tumor formation in the three models found that the cells cultured by the 3DP-7721 model had strong tumorigenicity in nude mice.Immunohistochemical evaluation of the levels of biochemical indicators related to the formation of solid tumors showed that the 3DP-7721 model group exhibited pathological characteristics of malignant tumors,the generated solid tumors were similar to actual tumors,and the deterioration was higher.This research therefore acts as a foundation for the application of 3DP-7721 models in drug development research.

The key role of myostatin b in somatic growth in fishes derived from distant hybridization

The basic mechanism of heterosis has not been systematically and completely characterized.In previous studies,we obtained three economically important fishes that exhibit rapid growth,WR(WCC♀×RCC♂),WR-Ⅱ(WR♀×WCC♂),and WR-Ⅲ(WR-Ⅱ♀×4nAU♂),through distant hybridization.However,the mechanism underlying this rapid growth remains unclear.In this study,we found that WR,WR-Ⅱ,and WR-Ⅲshowed muscle hypertrophy and higher muscle protein and fat contents compared with their parent species(RCC and WCC).Candidate genes responsible for this rapid growth were then obtained through an analysis of 12 muscle transcriptomes.Notably,the mRNA level of mstnb(myostatin b),which is a negative regulator of myogenesis,was significantly reduced in WR,WR-Ⅱ,and WR-Ⅲcompared with the parent species.To verify the function of mstnb,a mstnb-deficient mutant RCC line was generated using the CRISPR-Cas9 technique.The average body weight of mstnb-deficient RCC at 12 months of age was significantly increased by 29.57%compared with that in wild-type siblings.Moreover,the area and number of muscle fibers were significantly increased in mstnb-deficient RCC,indicating hypertrophy and hyperplasia.Furthermore,the muscle protein and fat contents were significantly increased in mstnb-deficient RCC.The molecular regulatory mechanism of mstnb was then revealed by transcription profiling,which showed that genes related to myogenesis(myod,myog,and myf5),protein synthesis(PI3K-AKT-mTOR),and lipogenesis(pparγand fabp3)were highly activated in hybrid fishes and mstnb-deficient RCC.This study revealed that low expression or deficiency of mstnb regulates somatic growth by promoting myogenesis,protein synthesis,and lipogenesis in hybrid fishes and mstnb-deficient RCC,which provides evidence for the molecular mechanism of heterosis via distant hybridization.

Breast cancer is the most common cancer among women worldwide.Postmastectomy radiotherapy(PMRT)is an essential component of combined therapy for early-stage,high-risk breast cancer.Breast reconstruction(BR)is often considered for patients with breast cancer who have undergone mastectomy.There has been a considerable amount of discussion about the optimal approach to combining PMRT with BR in the treatment of breast cancer.PMRT may increase the risk of complications and prevent good aesthetic results after BR,while BR may increase the complexity of PMRT and the radiation dose to surrounding normal tissues.The purpose of this review is to give a broad overview and summary of the current controversies and trends in PMRT and BR in the context of the most recent literature available.

Genetic Analysis of Structural Styles in the Makran Accretionary Wedge–Insight from Physical Simulations

The Makran accretionary wedge has the smallest subduction angle among any accretionary prism in the world. The factors controlling the spacing and morphological development of its deep thrust faults, as well as the formation mechanism of shallow normal faults, remain unclear. Meanwhile, the factors affecting the continuity of plane faults must be comprehensively discussed. Clarifying the development characteristics and deformation mechanisms of the Makran accretionary wedge is crucial to effectively guide the exploration of gas hydrate deposits in the area. This study aims to interpret seismic data to identify typical structures in the Makran accretionary wedge, including deep imbricate thrust faults, shallow and small normal faults, wedge-shaped piggyback basins, mud diapirs with fuzzy and disorderly characteristics of reflection, décollements with a northward tilt of 1° – 2°, and large seamounts. Physical simulation-based experiments are performed to comprehensively analyze the results of the plane, section, and slices of the wedge. Results reveal that the distances between and shapes of thrust faults in the deep parts of the Makran accretionary wedge are controlled by the bottom décollement. The uplift of the thrust fault-related folds and the upwelling of the mud diapirs primarily contribute to the formation of small normal faults in the shallow part of the area. The mud diapirs originate from plastic material at the bottom, while those that have developed in the area near the trench are larger. Seamounts and mud diapirs break the continuity of fault plane distribution.

Driving safety and accident prevention are attracting increasing global interest.Current safety monitoring systems often face challenges such as limited spatiotemporal coverage and accuracy,leading to delays in alerting drivers about potential hazards.This study explores the use of edge computing for monitoring vehicle motion and issuing accident warnings,such as lane departures and vehicle collisions.Unlike traditional systems that depend on data from single vehicles,the cooperative vehicle-infrastructure system collects data directly from connected and automated vehicles(CAVs)via vehicle-to-everything communication.This approach facilitates a comprehensive assessment of each vehicle’s risk.We propose algorithms and specific data structures for evaluating accident risks associated with different CAVs.Furthermore,we examine the prerequisites for data accuracy and transmission delay to enhance the safety of CAV driving.The efficacy of this framework is validated through both simulated and real-world road tests,proving its utility in diverse driving conditions.

Paeoniflorin alleviates depression by inhibiting the activation of NLRP3 inflammasome via promoting mitochondrial autophagy

Depression ranks among the most common neuropsychiatric disorders globally.Current studies examining the roles of inflammation and mitochondrial autophagy in the antidepressant efficacy of paeoniflorin(PF)are sparse.This study aimed to elucidate PF’s antidepressant mechanism by promoting autophagy and inhibiting NLRP3 inflammasome activation using chronic unpredictable mild stimulation(CUMS)-induced C57BL/6 mouse models in vivo and corticosterone(CORT)-induced HT22 cell models in vitro.Results demonstrated that PF enhanced the viability of HT22 cells following CORT exposure,restored mitochondrial membrane poten-tial(MMP),reduced reactive oxygen species accumulation,increased LC3 fluorescence intensity,and suppressed inflammatory cy-tokine secretion and inflammation activation.Additionally,PF ameliorated depressive behaviors induced by CUMS and improved damage in hippocampal neurons.It also reduced the expression of NLRP3,ASC,Caspase-1,IL-1β,and the assembly of the NLRP3 in-flammasome.Moreover,PF upregulated the expression of autophagy-related proteins in the hippocampus,facilitating the clearance of damaged mitochondria and enhancing autophagy.The role of autophagy in PF’s antidepressant effects was further confirmed through the use of the autophagy inhibitor 3-methyladenine(3-MA),which reduced the efficacy of PF.In conclusion,PF effectively improved depressive behaviors in CUMS-induced mice and reduced NLRP3-mediated inflammation both in vivo and in vitro,likely via the in-duction of autophagy.

Timing and Tectonic Setting of the Gaoaobei TungstenMolybdenum Deposit in Nanling Range, South China

The Gaoaobei tungsten-molybdenum deposit is a newly discovered large-scale quartzvein-type deposit in the Nanling metallogenic belt in South China.The ore bodies are hosted in the Indosinian granites and the Cambrian Xiangnan Group slates and are controlled by NWW-oriented faults,which are obviously different from the“five-story building”model in southern Jiangxi Province.The magmatic rocks in the study area are dominated by medium-to coarse-grained biotite monzogranite,with a few NW-oriented fine-grained granite dykes.The medium-to coarse-grained biotite monzogranite and fine-grained granite dykes have zircon U-Pb ages of 229.4±1.9 Ma(MSWD=1.5)and 164.9±3.3 Ma(MSWD=0.75),respectively,corresponding to the Indosinian and Yanshanian magmatism.The monzogranites have higher contents of FeO,CaO,K2O,P2O5,and TiO2,while the granite dykes have slightly higher contents of SiO2,Al2O3,MnO,and Na2O.Their A/CNK values are 1.11-1.75 and 1.19-2.25,and the contents of CIPW normative corundum are 1.71%-6.66%and 2.41%-9.50%,suggesting both the monzogranites and granite dykes are S-type granite.The total amount of rare earth elements in the monzogranites(from 84.7 ppm to 129 ppm)is slightly lower than that in the granite dykes(from 128 ppm to 133 ppm).The Eu/Eu*values range from 0.12 to 0.30 in monzogranites and from 0.0011 to 0.0013 in granite dykes,indicating the fine-grained granites underwent more intense fractional crystallization.The monzogranite and granite dykes have high 87Sr/86Sri values of 0.7169-0.7193 and 0.72825-0.72880,lowεNd(t)values ranging from-10.2 to-9.6 and from-11.5 to-11.4,and TDM2 ages of 1835-1785 and 1957-1946 Ma,respectively.These isotope data indicate their origin from the remelting of the Paleoproterozoic crustal materials.Combined with regional geology,it is concluded that the medium-to coarse-grained biotite monzogranite was formed in a post-collisional extensional environment.In addition,40Ar-39Ar dating of the greisen type tungsten-molybdenum ore gave consistent plateau age of 164.0±1.2 Ma,isochrona

A sharp interface approach for cavitation modeling using volume-of-fluid and ghost-fluid methods

This paper describes a novel sharp interface approach for modeling the cavitation phenomena in incompressible viscous flows. A one-field formulation is adopted for the vapor-liquid two-phase flow and the interface is tracked using a volume of fluid（VOF） method. Phase change at the interface is modeled using a simplification of the Rayleigh-Plesset equation. Interface jump conditions in velocity and pressure field are treated using a level set based ghost fluid method. The level set function is constructed from the volume fraction function. A marching cubes method is used to compute the interface area at the interface grid cells. A parallel fast marching method is employed to propagate interface information into the field. A description of the equations and numerical methods is presented. Results for a cavitating hydrofoil are compared with experimental data.

家支制度被认为是凉山彝族社会政治制度的重要组成部分,它向内凝聚了血缘共同体的团结,向外抵御了共同体潜在的风险。因此,一般认为家支仅仅是以血缘世系为纽带进行整合的。通过对近代以来云南小凉山的考察发现,在有“等级团结”关系的彝族社会中,家支具有延展性,它既指一个具有血缘世系纽带的群体,也指具有等级涵盖关系的家支联合体。在本土概念中将其归纳为“地界”(mu ddix)。“地界”是指在小凉山彝族社会内部以诺伙家支为名号,诺伙、曲诺、阿加和呷西这三层身份关系为基础,以家支地界利益为最高原则来组成和维系共同体运转的法权机制,以此来讨论小凉山彝族社会中的家支构成关系、运转逻辑与分化原则。只有理解了“地界”中家支分化与整合的关系才能真正厘清小凉山彝族社会运行的基本逻辑。
