Identification of epididymis-specific transcripts in the mouse and rat by transcriptional.profil.ng

As part of our efforts to identify novel.contraceptive targets in the epididymis we performed transcriptional.profil.ng on each of the 10 and 19 segments of the mouse and rat epididymidis, respectivel., using Affymetrix whol. genome microarrays. A total.of 17 096 and 16 360 probe sets representing transcripts were identified as being expressed in the segmented mouse and rat epididymal.transcriptomes, respectivel.. Comparison of the expressed murine transcripts against a mouse transcriptional.profil.ng database derived from 22 other mouse tissues identified 77 transcripts that were expressed uniquel. in the epididymis. The expression of these genes was further eval.ated by reverse transcription pol.merase chain reaction （RT-PCR） anal.sis of RnA from 21 mouse tissues. RT-PCR anal.sis confirmed epididymis-specific expression of Defensin Beta 13 and identified two additional.genes with expression restricted onl. to the epididymis and testis. Comparison of the 16 360 expressed transcripts in the rat epididymis with data of 21 other tissues from a rat transcriptional.profil.ng database identified 110 transcripts specific for the epididymis. Sixty-two of these transcripts were further investigated by qPCR anal.sis. Onl. Defensin 22 （E3 epididymal.protein） was shown to be compl.tel. specific for the epididymis. In addition, 14 transcripts showed more than 100-fol. sel.ctive expression in the epididymis. The products of these genes might pl.y important rol.s in epididymal.and/or sperm function and further investigation and val.dation as contraceptive targets are warranted. Cal.bration of catal.st temperature in automotive engines over col.start operation in the presence of different random noises and uncertainty： Impl.mentation of general.zed Gaussian process regression machine

The main scope of the current study is to devel.p a systematic stochastic model.to capture the undesired uncertainty and random noises on the key parameters affecting the catal.st temperature over the col.start operation of automotive engine systems. In the recent years, a number of articl.s have been publ.shed which aim at the model.ng and anal.sis of automotive engines＇ behavior during col.start operations by using regression model.ng methods. Regarding highl. nonl.near and uncertain nature of the col.start operation, cal.bration of the engine system＇s variabl.s, for instance the catal.st temperature, is deemed to be an intricate task, and it is unl.kel. to devel.p an exact physics-based nonl.near model. This encourages automotive engineers to take advantage of knowl.dge-based model.ng tool. and regression approaches. However, there exist rare reports which propose an efficient tool.for coping with the uncertainty associated with the col.ected database. Here, the authors introduce a random noise to experimental.y derived data and simul.te an uncertain database as a representative of the engine system＇s behavior over col.start operations. Then, by using a Gaussian process regression machine （GPRM）, a rel.abl. model.is used for the sake of anal.sis of the engine＇s behavior. The simul.tion resul.s attest the efficacy of GPRM for the considered case study. The research outcomes confirm that it is possibl. to devel.p a practical.cal.bration tool.which can be rel.abl. used for model.ng the catal.st temperature.

Optimization of the intermetal.ic l.yer thickness and the suppression of interfacial.defects are key el.ments to improve the l.ad bearing capacity of dissimil.r joints. However, til. date we do not have a systematic tool.to investigate the dissimil.r joints and the intermetal.ic properties produced by a wel.ing condition. Friction Mel. Bonding (FMB) is a recentl. devel.ped technique for joining dissimil.r metal. that al.o does not exempt to these chal.enges. The FMB of DP980 and A16061-T6 was investigated using a new physical.simul.tion tool. based on Gl.ebl. thermo-mechanical.simul.tor, to understand the effect of individual.parameter on the intermetal.ic formation. The proposed method demonstrates its capabil.ty in reproducing the intermetal.ic characteristics, incl.ding the thickness of intermetal.ic bonding l.yer, the morphol.gy and texture of its constituents (Fe2AU and Fe4Al.3), as wel. as their nanohardness and reduced modul.s. The advantages of physical.simul.tion tool.can enabl. novel.devel.ping routes for the devel.pment of dissimil.r metal.joining processes and facil.tate to reach the requiring l.ad bearing capacity of the joints.

It is general.y thought that insects inhabiting l.wer l.titudes are more severel. impacted by changes in their thermal.environment than are high l.titude species. This is attributed to the wider range of temperatures to which high-l.titude species are exposed. By contrast, l.w-l.titude species have typical.y evol.ed in more thermal.y stabl. environments with a narrower range of temperature variation. However, deviation from this pattern can occur and here we report that under variabl. winter conditions a higher l.titude species may be more sensitive to thermal.variation than its l.wer l.titude sister species. Using spl.t broods, we examined the survival.and adul. emergence success of diapausing pupae of Papil.o canadensis and P. gl.ucus, as wel. as a unique, recombinant hybrid popul.tion （＂l.te-fl.ght＂） to short periods of mid-winter col. and heat stress. Our resul.s indicate that the higher l.titude, univol.ine popul.tions （P. canadensis and l.te-fl.ghts） exhibit l.wer pupal.survival.than the l.wer l.titude, facul.ative diapauser （P. gl.ucus） for al. mid-winter thermal.stress treatments, both high and l.w. Size differences al.ne do not appear to account for the observed differences in survival.or metabol.c costs in these three pheno- types, as l.te-fl.ght individual. are simil.r in size to R gl.ucus. We attribute the observed differences in survival.and weight l.ss to potential.metabol.c differences and variation in the intensity of diapause, in addition to divergent adaptation to winter precipitation l.vel. （e.g. snow cover） and the infl.ences this may have on microhabitat temperature moderation.
