Mitochondria-targeted nanoparticles overcome chemoresistance via downregulating BACH1/CD47 axis in ovarian carcinoma

The platinum-based chemotherapy is a routine strategy for the treatment of ovarian cancer,while it is prone to chemoresistance in clinical,which hinders the treatment.Therefore,it is urgently needed to elucidate the underlying mechanism of drug resistance and form the appropriate strategy.The sequencing results showed that cisplatin(DDP)resistant ovarian cancer overexpressed BTB and CNC homology 1(BACH1),and up-regulated the“don’t eat me”signal CD47.We identified that hemin,a BACH1 inhibitor,could effectively down-regulate BACH1 and simultaneously inhibit CD47.Moreover,hemin has a synergistic effect with DDP.We designed a pH-responsive nanoparticle(H/D@FA-CaP-NPs)in which folic acid(FA)ensured targeting of ovarian cancer cells,while hemin inhibited BACH1 as well as down-regulated CD47,achieving the promotion of apoptosis of tumor cells and inducing phagocytosis of tumors by macrophages.Moreover,hemin has a synergistic effect with DDP to promote apoptosis of tumor cells.Structurally,hemin and DDP was encapsulated within hydrophobic 1,2-distearoyl-sn-glycero-3-phosphoethanolamine(DSPE)to form a tight core,and hydrophilic polyethylene glycol 2000(PEG2000)and calcium phosphate(CaP)formed the outside shell,and FA was modified on the surface of nanoparticles.In terms of function,(a)FA enhanced the active targeting of nanoparticles to tumors;(b)NPs targeted mitochondria to induce reactive oxygen species(ROS)production;(c)hemin encapsulated in nanoparticles could specifically target BACH1,thereby down regulating CD47;(d)hemin had a synergistic effect with DDP,thus augmenting the chemotherapy.Altogether,mitochondria-targeted nanoparticles H/D@FA-CaP-NPs promoted tumor apoptosis and mobilized phagocytosis to treat tumor,providing a novel scheme for clinical treatment of cisplatin-resistant ovarian carcinoma.

Crustal structure in the Anyuan Coal Mine and its adjacent areas of Jiangxi Province by P-wave receiver functions

We collected high-quality teleseismic events recorded by 12 broadband seismographs deployed in the Anyuan Coal Mine and its adjacent areas in Pingxiang City,Jiangxi Province for nearly two years.The H-κ-c stacking method was employed to obtain the crustal thickness and Poisson's ratio distribution,then the characteristics of crustal structure below the stations were obtained by using the time-domain linear inversion method.The crustal thickness in the Anyuan Coal Mine and its adjacent areas ranges from approximately 32~35 km,with an average thickness of 33 km,which is consistent with the crustal thickness results in South China from previous studies using the receiver function method.The average Poisson's ratio of the crustal bulk composition in the study area varies between 0.22 and 0.25,which is lower than the global value with a 0.27 average,indicating a predominantly intermediate-acidic or felsic crustal composition.There is a weak negative correlation between Poisson's ratio and crustal thickness estimates in the Anyuan Coal Mine and its adjacent areas,suggesting that the absence of mafic-ultramafic materials in the lower crust is associated with the process of crustal delamination.The velocity inversion results indicate that the crustal structure including three velocity discontinuity interfaces,with the first at a depth of approximately 1.5 km,the second at about 10~15 km,and the third being the Moho.The study also indicates that the results obtained by the H-κ-c stacking method are significantly better than those obtained by the H-κmethod,effectively reducing the standard deviation and dispersion of crustal thickness and vP/vSratio.

The current therapeutic drugs for Alzheimer's disease only improve symptoms,they do not delay disease progression.Therefo re,there is an urgent need for new effective drugs.The underlying pathogenic factors of Alzheimer's disease are not clear,but neuroinflammation can link various hypotheses of Alzheimer's disease;hence,targeting neuroinflammation may be a new hope for Alzheimer's disease treatment.Inhibiting inflammation can restore neuronal function,promote neuro regeneration,reduce the pathological burden of Alzheimer's disease,and improve or even reverse symptoms of Alzheimer's disease.This review focuses on the relationship between inflammation and various pathological hypotheses of Alzheimer's disease;reports the mechanisms and characteristics of small-molecule drugs(e.g.,nonsteroidal anti-inflammatory drugs,neurosteroids,and plant extracts);macromolecule drugs(e.g.,peptides,proteins,and gene therapeutics);and nanocarriers(e.g.,lipid-based nanoparticles,polymeric nanoparticles,nanoemulsions,and inorganic nanoparticles)in the treatment of Alzheimer's disease.The review also makes recommendations for the prospective development of anti-inflammatory strategies based on nanocarriers for the treatment of Alzheimer's disease.

Dual-band dynamic reconfigurable intelligent surface for signal enhancement in millimeter wave

Millimeter wave with large bandwidth,high transmission rate,and low delay is considered a reliable alternative to cope with the spectrum shortage.However,the fast attenuation and narrow beam characteristics make it difficult to achieve long-distance or wide-range applications.Here,a 1-bit dual-band reflective reconfigurable intelligent surface(RIS)for signal enhancement in millimeter wave with 16×16 elements is designed,fabricated,and measured.Different from most existent RIS,dynamic programming is realized at two separate frequency bands by integrating the PIN diodes and field-programmable gate array(FPGA).Particularly,the beam deflection,dual-beam,and multi-beam are created based on the coding theory and convolution operation,proving the effectiveness of wavefront manipulation.Moreover,the far-field patterns and signal power with different coding sequences are measured and compared.It is indicated that the received signal power is 6–7 dB stronger than that without coding,which shows good agreement with the desired expectations.The proposed reconfigurable metasurface exhibits great potential in beam forming,making it a promising candidate for progressive wireless communication applications.

A comprehensive and up-to-date web-based interactive 3D emergency response and visualization system using Cesium Digital Earth: taking landslide disaster as an example

As with the fast advances in the technologies of big Earth data and information communication,Web-based 3D GIS system has come a long way from a few years ago.These advances reflect in many aspects of 3D GIS such as higher real-time performance,enhanced interactivity,more realistic 3D visualization effect and improved user interface.This paper aims to present a comprehensive and upto-date 3D Web GIS for Emergency Response using the current vue.js web application framework and the well-known Cesium APl,taking landslide disaster as an example.Building upon recent advances in WebGL technology,we developed a suite of enhanced 3D spatial analysis functions,including interactive route planning,instant text/image/video messaging being incorporated into both 3D WebGL page and mobile GIS applications,and progressive 3D construction and AR visualization using LiDAR and camera over local emergency network or internet.Moreover,professional functions such as landslide susceptibility mapping,landslide monitoring,spatial temporal contingency plan management,landslide information management,personnel and equipment management,and communication are all implemented and integrated in the 3D GIS system.Most of the functions of the system are implemented using open-source projects,which is beneficial to the development of the 3D GIS research community.

A monocular visual SLAM system augmented by lightweight deep local feature extractor using in-house and low-cost LIDAR-camera integrated device

Simultaneous Localization and Mapping(SLAM)has been widely used in emergency response,self-driving and city-scale 3D mapping and navigation.Recent deep-learning based feature point extractors have demonstrated superior performance in dealing with the complex environmental challenges(e.***.extreme lighting)while the traditional extractors are struggling.In this paper,we have successfully improved the robustness and accuracy of a monocular visual SLAM system under various complex scenes by adding a deep learning based visual localization thread as an augmentation to the visual SLAM framework.In this thread,our feature extractor with an efficient lightweight deep neural network is used for absolute pose and scale estimation in real time using the highly accurate georeferenced prior map database at 20cm geometric accuracy created by our in-house and low-cost LiDAR and camera integrated device.The closed-loop error provided by our SLAM system with and without this enhancement is 1.03m and 18.28m respectively.The scale estimation of the monocular visual SLAM is also significantly improved(0.01 versus 0.98).In addition,a novel camera-LiDAR calibration workflow is also provided for large-scale 3D mapping.This paper demonstrates the application and research potential of deep-learning based vision SLAM with image and LiDAR sensors.

Deep reinforcement learning and 3D physical environments applied to crowd evacuation in congested scenarios

To avoid crowd evacuation simulations depending on 2D environments and real data,we propose a framework for crowd evacuation modeling and simulation by applying deep reinforcement learning(DRL)and 3D physical environments(3DPEs).In 3DPEs,we construct simulation scenarios from the aspects of geometry,semantics and physics,which include the environment,the agents and their interactions,and provide training samples for DRL.In DRL,we design a double branch feature extraction combined actor and critic network as the DRL policy and value function and use a clipped surrogate objective with polynomial decay to update the policy.With a unified configuration,we conduct evacuation simulations.In scenarios with one exit,we reproduce and verify the bottleneck effect of congested crowds and explore the impact of exit width and agent characteristics(number,mass and height)on evacuation.In scenarios with two exits and a uniform(nonuniform)distribution of agents,we explore the impact of exit characteristics(width and relative position)and agent characteristics(height,initial location and distribution)on agent exit selection and evacuation.Overall,interactive 3DPEs and unified DRL enable agents to adapt to different evacuation scenarios to simulate crowd evacuation and explore the laws of crowd evacuation.

本文依据增强现实数字地图与地理场景在时空、尺度和语义等的上下文关联性,研究并划分了包括存在上下文关联的地理实体空间载体、模型实体空间载体,以及不存在上下文关联的虚位空间载体三类AR地图载体。选取二维纸质地形晕渲图、三维打印地形实体模型、以及桌面虚位空间3种不同类型载体的AR地图为研究对象,以目标搜索和路径规划为任务目标,开展不同载体AR地图空间认知实验研究。结果表明,在2组实验中三维实体模型载体AR地图的认知负荷指数(CLI)分别为2.53和4.84均显著低于二维地图和虚位载体,具有认知负荷低、沉浸在场感强的认知特征;二维地图载体的认知负荷指数略低于虚位空间载体,但两者并无显著差异;同时,两组实验对比分析结果显示,随着任务难度提升,二维地图载体和虚位空间载体AR地图的认知负荷显著增加,但三维实体模型载体AR地图的认知负荷未显著增加,表明实体模型载体在面向复杂空间认知任务时具有较低认知负荷的优势。此外CLI和任务难度的同步变化也表明脑电数据对空间认知实验敏感,能有效反映空间认知实验过程中的认知特征变化。

Graphics processing unit-based dynamic volume rendering for typhoons on a virtual globe

Typhoons are usually accompanied by strong winds,heavy rain,storm surges and other disastrous events,and they are the most severe weather phenomena in the western part of the North Pacific Ocean.Visualisation is a key technology for analysing and presenting climate simulations and observations together with related social and ecological data.Virtual globes are a logical platform for visualising such geospatial data via the Internet.To implement interactive visualisation of typhoon data within virtual globes,this study provides a 3D texture-mapping algorithm to render volume textures on a globe.In addition,a systematic framework is proposed in which a hierarchical octree-based multiresolution data structure is implemented to organise the large volume dataset,and a graphics processing unit-based dynamic interpolation approach is proposed to obtain a smooth effect.Finally,the structure is combined with a level of detail strategy to enable the rendering of dynamic volumetric data at an acceptable interactive frame rate.To demonstrate the capabilities of this framework,data for a simulated typhoon event were rendered in World Wind,an open-source virtual globe.The experimental results demonstrate that the application of the proposed strategies can result in an interactive visualisation performance that renders typhoon data on virtual globes.

传统地理学/GIS对人群时空行为的研究集中在中宏观尺度,对微观时空行为的研究相对不足。针对这一问题,提出了微观人群数字孪生的概念及其基础逻辑框架,从人群行为实验方法、基础行为/场景行为建模、典型应用及案例分析等方面总结了微观人群数字孪生的技术基础及其研究现状,并从地理学的视角,对基于系统论的人群实验思路和可信人群实验方法、以及利己—利他、个体—群体、确定—随机统一的人群行为建模方法、微观行为地理学和微观人地关系理论体系构建等未来研究进行探讨,希望推动地理学、GIS和虚拟地理环境对微观人群时空行为理论、方法和技术的研究。
