Analysis of the ratio of mitchondrial DNA with A1555G mutant to wild type in deaf patients of Fujian province in China by a new method and its relationship with the severity of hearing loss

Background It has been suggested that the ratio of mutant and wild type mitochondrial DNA may be related to its clinical phenotype. In this study, we developed a high sensitive real-time amplification refractory mutation system-quantitative polymerase chain reaction （RT-ARMS-qPCR） assay for quantitation of the mitochondrial DNA （mtDNA） with a mutated 1555 site, and explored the relationship between the ratio of mutated mtDNA and the severity of hearing loss of mitochondrial deafness （MD） patients of Fujian province in China. Methods An amplified mtDNA fragment containing the 1555 site was ligated into a vector to construct a plasmid DNA standard. An RT-ARMS-qPCR system was used to measure the mtDNA copy number containing wild-type and mutant sequences in a cohort of 126 MD patients of Fujian province in China. Combined with the clinical data, we explored the relationship between the ratio of mutated mtDNA and the severity of hearing loss of MD. Results The variation coefficient in the cohort was 1.21%, the interassay variation coefficient was 1.78%, and the linear range was 102-10e copies/pl for detecting a recombinant, wild-type plasmid. The primers amplified only the intended sequences. Mutation copy number correlated with the degree of deafness in sporadic cases with heteroplasmic mutations of mtDNA A1555G （R=0.811, P=0.003）, but not in sporadic cases with homoplasmic mutations （R=0.007, P=0.989）. The copy number of homoplasmic or heteroplasmic mutations of mtDNA A1555G in familial cases correlated with degree of deafness （R=0.352, P=0.023 and R=0.90, P=0.012, respectively）. Conclusions The RT-ARMS-qPCR system is suitable for determining the copy number of mtDNA fragments containing the A1555G mutation. The ratio of mutated mtDNA correlates with the severity of hearing loss of MD.

Loss of bifurcation patency after cross-over stenting of ostial lesions in superficial femoral artery： possible causes, prevention and reintervention

Background Crossover stenting across the origin of the profunda femoral artery （PFA） and occasionally into the common femoral artery （CFA） is commonly used after suboptimal balloon angioplasty of ostial occlusive lesions of the superficial femoral artery （SFA） involving the bifurcation. Late stent occlusion at the bifurcation is not rare and results in severe lower extremity ischemia. Therefore, we tried to assess its possible causes, prevention and reintervention. Methods Using a prospectively maintained single-center database, 12-month femoral bifurcation patency was retrospectively compared and lesion and procedural predictors of stent occlusion were determined among 63 patients （64 lesions） who between July 2011 and February 2013 underwent crossover （36 non-jailed and 15 jailed SFA, and 12 distal and 1 complete CFA） stenting of de novo ostial SFA lesions. Results Twelve-month overall patency rate at the femoral bifurcation was 88%, with no significant difference between jailed-ostial SFA （80%） and distal CFA （67%） stenting （P=0.731）, and significant differences between either and non-jailed ostial stenting （100%,P=0.035 and 0.002）. When PFA ostium was jailed by the stent, patients with preexisting CFA or PFA lesions had a 12-month bifurcation patency rate of 20%, significantly lower than those with simple ostial SFA lesions （83%, P=0.015）. Stent induced intimal hyperplasia caused bifurcation occlusion in 6 surgical reintervention cases. Conclusions In crossover stenting of ostial lesions in SFA, bifurcation patency loss was significantly higher in distal CFA and jailed ostial SFA stenting than non-jailed ostial SFA stenting. Preexisting CFA or PFA lesion is a significant risk factor for bifurcation patency loss when PFA ostium is jailed by crossover stenting.

In this paper,we propose an active reconfigurable intelligent surface(RIS)enabled hybrid relaying scheme for a multi-antenna wireless powered communication network(WPCN),where the active RIS is employed to assist both wireless energy transfer(WET)from the power station(PS)to energyconstrained users and wireless information transmission(WIT)from users to the receiving station(RS).For further performance enhancement,we propose to employ both transmit beamforming at the PS and receive beamforming at the *** formulate a sumrate maximization problem by jointly optimizing the RIS phase shifts and amplitude reflection coefficients for both the WET and the WIT,transmit and receive beamforming vectors,and network resource *** solve this non-convex problem,we propose an efficient alternating optimization algorithm with the linear minimum mean squared error criterion,semidefinite relaxation(SDR)and successive convex approximation ***,the tightness of applying the SDR is *** results demonstrate that our proposed scheme with 10 reflecting elements(REs)and 4 antennas can achieve 17.78%and 415.48%performance gains compared to the single-antenna scheme with 10 REs and passive RIS scheme with 100 REs,respectively.

Fabrication of AZ31/Mg3Y Composites with Excellent Strength and Plasticity via Accumulated Rolling Bonding and Diffusion Annealing

AZ31/Mg3Y composites with a layer thickness of 100-200μm were fabricated by accumulated rolling bonding(ARB),which was followed by diffusion annealing at 300℃ for 0-32 *** interface layer,containing numerous Al-Y precipitates,is formed in the Mg3Y layer that is adjacent to the interface as a result of Al diffusing from the AZ31 layers into the Mg3Y *** thickness of the interface layer gets increased and more precipitates are formed in the interface layer with the extension of the annealing *** microhardness of the AZ31 and Mg3Y layer decreases firstly and then reaches a stable value,while the microhardness of the interface layer increases gradually with the extension of the annealing *** AZ31/Mg3Y composites exhibit equivalent strength but increased ductility after diffusion annealing,in comparison to the as-rolled AZ31/Mg3Y *** addition,the AZ31/Mg3Y composites after annealing always present higher strength and ductility than AZ31/AZ31 composite,which was fabricated by the same process as that for the AZ31/Mg3Y ***-deformation induced strengthening also plays an important role in the excellent strength and ductility of the annealed AZ31/Mg3Y *** study can provide a direction for improving the plasticity and strength of magnesium alloys synergistically.

Objective:To observe the regulatory effects of RhoA/ROCK pathway on the apoptosis of cardiac myocyte induced by anoxia and its ***:The model of cardiac myocyte anoxia was *** beat pulsations and apoptosis rales after 1 h,3 h,6 h,9 h and 12 h of anoxia were recorded and the expressions of RhoA,ROCK1/2,p-PI3 K,p-AKT and caspae-3 were detected,*** apoptosis and the expressions of related proteins were detected after RNAi of RhoA and the inhibition of ROCK by ***:The beat pulsations after 1 h,3 h,6 h.9h and 12 h decreased gradually but the apoptosis rates increased gradually,and the expressions of RhoA,ROCK1/2,p-P13 K,p-AKT and caspase-3 were increasing along with the increasing duration of *** apoptotic rales after 1 h,3 h,6 h.9 h and 12 h of anoxia were(4.36±0.98)%,(8.36±2.12)%,(15.32±3.62)%,(18.68±4.83)%and(24.56±6.22)%.respectively and decreased more significantly than control group in different time points of anoxia(P<0.05).and the expressions of RhoA,ROCK1/2,p-PI3 K,p-AKT and caspase-3 decreased significantly(P<0.05).The apoptosis rate and the expressions of RhoA,ROCK1/2,p-PI3 K,p-AKT and caspase-3 decreased significantly(P<0.05) after the inhibition of ROCK by Y-27632(P<0.05).Conclusions:RhoA/ROCK pathway plays a critical role in the regulation of the apoptosis of cardiac myocyte induced by anoxia,which may be accompanied by regulating the activity of PI3K/AKT/Caspase-3 pathway.

The effects of nano-CaO contents on the microstructure,mechanical properties and corrosion resistance of lean Mg-1Zn alloy were *** results showed that the addition of nano-CaO significantly refined the grain size and improved mechanical properties of the Mg-1Zn *** the same time,CaO reacted with molten Mg in situ to form nano-MgO,whose corrosion product in SBF solution was the same with the degradation product of Mg matrix,resulting in the enhanced compactness of the Mg(OH)_(2) layer and reduced corrosion rate of *** Mg-1Zn alloy had lower corrosion resistance due to excessively large grain size and shedding of corrosion *** composite with 0.5 wt.%CaO had the best corrosion resistance with a weight loss of 9.875 mg·y^(-1)·mm^(-2)due to the small number of Ca_(2)Mg_(6)Zn_(3) phase and suitable grain *** for composites with high content of CaO(0.7 wt.%and 1.0 wt.%),they had lower corrosion resistance due to the coexistence of large number of Ca_(2)Mg_(6)Zn_(3) and Mg_(2)Ca at grain boundaries,especially for 1.0 wt.%CaO composite,resulting from the strong micro-galvanic corrosion.

The introduction of entropy concept into thermoelectric materials recently has attracted broad ***,we showed the phase diagram of the ternary systems of Bi_(2)Se_(3)-Sb_(2)Se_(3)-In_(2)Se_(3).Among the high entropy compounds (HECs),we have identified an intrinsic p-type region near our previously reported Bi_(0.8)Sb_(0.8)In_(0.4)Se_(3) in the phase diagram.
