Testosterone replacement therapy improves insulin sensitivity and decreases high sensitivity C-reactive protein levels in hypogonadotropic hypogonadal young male patients

Background Many clinical studies suggest the inverse relationship between testosterone levels and insulin sensitivity in men, however the causative relationship of these two events is still not determined. The purpose of this study was to investigate the effects of testosterone replacement therapy （TRT） on insulin sensitivity, body composition, serum lipid profiles and high sensitivity C-reactive protein （hsCRP） in hypogonadotropic hypogonadal （HH） puberty undeveloped male patients. Methods In this prospectively designed study, we compared homeostasis model assessment of insulin resistance （HOMA-IR）, insulin areas under the curves （AUC） of 3-hour oral glucose tolerance test （OGTT） and other metabolic parameters between 26 HH patients and 26 healthy men. The patients＇ HOMA-IR, insulin AUC, body composition, lipid profiles, hsCRP and other parameters were compared before and after nine-month TRT. Results The average levels of total testosterone （TT） in HH and healthy group were （0.9±0.6） nmol/L and （18.8±3.4） nmol/L, respectively. HOMA-IR in HH group was significantly higher than the healthy group （5.14±5.16 vs 2.00±1.38, P 〈0.005）. Insulin AUC in 3-hour OGTT in HH group was significantly higher than the healthy group （698.6±414.7 vs 414.2±267.5, P 〈0.01）. Fasting glucose level in H H group was significantly higher than control group （（5.1±0.6） mmol/L vs （4.7±0.3） mmol/l, P 〈0.005）. Height, weight and grasp strength of the patients were significantly increased after 9-month TRT. Significant reductions in HOMA-IR （from 5.14±5.16 to 2.97±2.16, P 〈0.01）, insulin AUC （from 698.6±414.7 to 511.7±253.9, P 〈0.01） and hsCRP （from （1.49±1.18） mg/L to （0.70±0.56） mg/L, P 〈0.05） were found after TRT. Serum total cholesterol, LDL-C, HDL-C and triglyceride were all decreased, albeit with no significant difference compared to the level prior to TRT. Expression of the Arabidopsis thaliana Histone Gene AtHTA1 Enhances Rice Transformation Efficiency

We expressed the Arabidopsis thaliana histone AtHTA1 in rice under the control of the maize ubiquitin promoter. Transformation efficiencies of rice plants that constitutively expressed AtHTA 1 were 28-44% higher than calli containing an empty vector control. Furthermore, co-infection of rice calli with a vector containing AtHTA 1 and another vector with the target gene increased transformation by 27-50%. Thus, expression of AtHTA 1 either transiently or in stably transformed cells improved rice transformation efficiency.

Oceanic submesoscales can significantly influence phytoplankton production and export owing to their similar timescales of *** on two-year Biogeochemical Argo(BGC-Argo) observations,this study investigated the development of submesoscale instabilities,particularly symmetric and mixed-layer baroclinic instabilities,and their impacts on biological production and export in the oligotrophic South China Sea *** the northern basin,near-surface winter blooms consistently cooccurred with seasonally deepened mixed ***,significantly stronger and weaker winter blooms were observed over two consecutive winters within the BGC-Argo observation *** the first winter,symmetric-instability-induced upward nutrient entrainment played a crucial role in initiating the strong winter bloom in early December,when the mixed layer was approximately 20–30 m shallower than the *** bloom occurred approximately 20–30 days earlier than that anticipated owing to the contact between the seasonally deepened mixed layer and mesoscale-cyclone-induced uplifted *** symmetric instability also facilitated the export of fixed phytoplankton carbon from the surface to deeper ***,during the second winter,remarkably intense mixed-layer baroclinic instability associated with an intense mesoscale anticyclone led to more significant shoaling of the mixed layer compared to the nutricline,thus increasing the vertical distance between the two *** this condition,upward nutrient injection,phytoplankton bloom,and carbon export were *** contrast,the BGC-Argo float in the central basin revealed significantly inhibited seasonality of phytoplankton biomass and submesoscale instabilities compared to those in the northern basin,primarily owing to the significantly shallower winter mixed layer.

The role of vitamin E in polyunsaturated fatty acid synthesis and alleviating endoplasmic reticulum stress in sub-adult grass carp (Ctenopharyngodon idella)

Vitamin E (VE) is an essential lipid-soluble vitamin that improves the fish flesh quality. However, the underlying molecular mechanisms remain unclear. This study aimed to investigate the effects of VE on growth performance and flesh quality in sub-adult grass carp (Ctenopharyngodon idella). A total of 450 fish (713.53 ± 1.50 g) were randomly divided into six treatment groups (three replicates per treatment) and fed for nine weeks with different experimental diets (dietary lipid 47.8 g/kg) that contained different levels of VE (5.44, 52.07, 96.85, 141.71, 185.66, and 230.12 mg/kg diet, supplemented as dl-α-tocopherol acetate). Notably, the treatment groups that were fed with dietary VE ranging from 52.07 to 230.12 mg/kg diet showed improvement in the percent weight gain, special growth rate, and feed efficiency of grass carp. Moreover, the treatment groups supplemented with dietary VE level of 141.71, 185.66, and 230.12 mg/kg diet showed enhancement in crude protein, lipid, and α-tocopherol contents in the muscle, and the dietary levels of VE ranging from 52.07 to 141.71 mg/kg diet improved muscle pH24hand shear force but reduced muscle cooking loss in grass carp. Furthermore, appropriate levels of VE (52.07 to 96.85 mg/kg diet) increased the muscle polyunsaturated fatty acid content in grass carp. Dietary VE also increased the mRNA levels of fatty acid synthesis-related genes, includingfas,scd-1,fad,elovl,srebp1,pparγ, andlxrα, and up-regulated the expression of SREBP-1 protein. However, dietary VE decreased the expression of fatty acid decomposition-related genes, includinghsl,cpt1,acox1, andpparα, and endoplasmic reticulum stress-related genes, includingperk,ire1,atf6,eif2α,atf4,xbp1,chop, andgrp78, and down-regulated the expression of p-PERK, p-IRE1, ATF6, and GRP78 proteins. Dynamic Crosslinked Phosphorescent Poly(vinyl alcohol)-Terpyridine Films with Enhanced Mechanical Properties and Tunable Shape Memory

Achieving versatile room temperature phosphorescence （RTP） materials, especially with tunable mechanical properties and shape memory is attractive and essential but rarely reported. Here, a strategy was reported to realize multi-functional RTP films with multicolor fluorescence, ultralong afterglow, adjustable mechanical properties, and shape memory through the synergistic dynamic interaction of lanthanide（LnⅢ）-terpyridine coordination, borate ester bonds, and hydrogen bondings in a poly（vinyl alcohol）(PVA) matrix. By varying the amount of borax,the mechanical properties of the films could be finely controlled due to the change of crosslinking degree of dynamic borate ester bonds in *** assembly and disassembly of borate ester bonds upon the trigger of borax and acid were applied as reversible linkage to achieve programmable shape memory behavior. In addition, the films displayed both fascinating multicolor fluorescence and ultralong afterglow characteristics due to the presence of Ln III doping and confinement of terpyridine in PVA. This study provides a new avenue to impart modulable mechanical strength and shape memory to RTP materials.

CMIP 6 models simulation of the connection between North/South Pacific Meridional Mode and ENSO

The subtropical North and South Pacific Meridional Modes(NPMM and SPMM)are well known precursors of El Niño-Southern Oscillation(ENSO).However,relationship between them is not *** the early 1980,the relationship experienced an interdecadal *** in this connection can be attributed mainly to the phase change of the Pacific decadal oscillation(PDO).During the positive phase of PDO,a shallower thermocline in the central Pacific is responsible for the stronger trade wind charging(TWC)mechanism,which leads to a stronger equatorial subsurface temperature *** dynamic process strengthens the connection between NPMM and *** with the negative phase of PDO,a shallower thermocline over southeastern Pacific allows an enhanced wind-evaporation-SST(WES)feedback,strengthening the connection between SPMM and *** 35 Coupled Model Intercomparison Project Phase 6(CMIP6)models,we examined the NPMM/SPMM performance and its connection with ENSO in the historical *** great majority of CMIP6 models can reproduce the pattern of NPMM and SPMM well,but they reveal discrepant ENSO and NPMM/SPMM *** intermodal uncertainty for the connection of NPMM-ENSO is due to different TWC mechanism.A stronger TWC mechanism will enhance NPMM *** SPMM,few models can simulate a good relationship with *** intermodel spread in the relationship of SPMM and ENSO owing to SST bias in the southeastern Pacific,as WES feedback is stronger when the southeastern Pacific is warmer.

TRIM21-dependent ultrasmall chiral gold nanoparticles for preventing microglia senescence against Alzheimer's disease

Tripartite motif 21(TRIM21)is an E3 ubiquitin ligase that shows great promise for protein degradation through ***,ultrasmall chiral gold nanoparticles(D-or L-NPs)modified by D-or L-glutathione ligands were *** conjugated with an NLR family pyrin domain-containing protein 3(NLRP3)antibody(D-NP-a NLRP3 or L-NP-a NLRP3),DNP-a NLRP3 showed effective delivery efficiency of the antibody into microglia and prevented Aβ-mediated microglia senescence,and p16^(ink4a),a marker of senescence,in the microglia was reduced by 90.3%±7.8% while L-NP-a NLRP3 decreased by 48.01%±3.1%.Mechanistic investigations revealed that the D-NP-a NLRP3((1.5±0.3)×10^(7)M^(-1))exhibited sixteen-fold larger binding affinity to transmembrane glycoprotein SLC3A2 than L-type((9.5±2.7)×10^(5)M^(-1)),which led to a high efficiency of antibody delivery and TRIM21-dependent NLRP3 ***,the blood-brain barrier(BBB)-crossing ability of chiral NP-a NLRP3 as well as NLRP3 degradation was demonstrated in *** APP/PS1 Alzheimer's disease(AD)model mice experiments exhibited a reduction of 89.7%±6.8% for the NLRP3 protein and 84.2±7.5% for p16^(ink4a)following the intravenous administration of D-NP-a NLRP3 once a week for 60 ***,the levels of the AD markers Aβ and phosphorylatedTau in the brains were reduced by 86.2%±8.2% and 81.6%±9.1%;these were 2.1-fold and 1.9-fold higher than those treated with L-NP-a NLRP3,*** studies provide a method to rescue AD-like pathologies and prevent senescence.
