Early predic.ion of severe ac.te panc.eatitis by urinary trypsinogen ac.ivation peptide

Objec.ive: To investigate the value of urinary trypsi-nogen ac.ivation peptide (TAP) in the early predic.tion of severe ac.te panc.eatitis and to c.mpare itwith ac.te physiology and c.ronic.health evaluationⅡ (APAc.E Ⅱ).Methods: We assessed the predic.ive value of urinaryTAP c.nc.ntrations measured by a c.mpetitive en-zyme-linked immunosorbent assay. Urine sampleswere c.llec.ed for detec.ing TAP c.nc.ntrations atadmission, and 24, 48, and 72 h from 41 patientswith ac.te panc.eatitis (12 with severe disease, 29with mild disease) who presented within 48 h the on-set of symptoms and from 11 c.ntrol patients, whileAPAc.E Ⅱ sc.res were rec.rded at 48 h after ***: The peak median urinary TAP c.nc.ntrationwas seen at admission. The median urinary TAPc.nc.ntration at admission for severe panc.eatitis (95nmol/L) was signific.ntly higher than the medianfor patients with mild panc.eatitis (20 nmol/L, Pc.ntrols (15 nmol/L, Pc.nc.ntrations were signific.ntly higher in patientswith severe ac.te panc.eatitis than the median in pa-tients with mild panc.eatitis (Pc.ntrols(Pc.E Ⅱsc.res of severe patients were signific.ntly differentfrom those of mild patients (10.5 vs 6.0, Pc.fic.ty, positive predic.ive, andnegative predic.ive values of an admission urinaryTAP≥35 nmol/L for severe panc.eatitis were91.7%, 89.7%, 78.6% and 96.3%, whereas 48 hafter admission the values for APAc.E Ⅱ sc.res(≥9) were 75.0%, 72.7%, 52.9% and 87.5%. Inpredic.ion of disease severity, the urine TAP c.nc.n-tration was muc. better than APAc.E Ⅱ at 48 ***: Urinary TAP obtained at the first 48 hof the onset of symptoms c.n predic. severe ac.tepanc.eatitis. SIZ1-mediated SUMOylation of c.SF100 promotes plant thermomorphogenesis by c.ntrolling alternative polyadenylation

Under warm temperatures, plants adjust their morphologies for environmental adaption via prec.se gene expression regulation. However, the func.ion and regulation of alternative polyadenylation (APA), an important fine-tuning of gene expression, remains unknown in plant thermomorphogenesis. In this study, we found that SUMOylation, a c.itic.l post-translational modific.tion, is induc.d by a long-term treatment at warm temperatures via a SUMO ligase SIZ1 in Arabidopsis. Disruption of SIZ1 altered the global usage of polyadenylation signals and affec.ed the APA dynamic.of thermomorphogenesis-related genes. c.SF100, a key subunit of the c.SF c.mplex for polyadenylation regulation, is SUMOylated by SIZ1. Importantly, we demonstrated that SUMOylation is essential for the func.ion of c.SF100 in genome-wide polyadenylation site c.oic. during thermomorphogenesis. Further analyses revealed that the SUMO c.njugation on c.SF100 attenuates its interac.ion with two isoforms of its partner c.SF30, inc.easing the nuc.ear ac.umulation of c.SF100 for polyadenylation regulation. Three novel sesterterpenoids glasesterterpenoids A−c.(1−3), featuring an unprec.dented 7-c.c.ohexyldec.hydronaphthalene c.rbon skeleton, were isolated from the root of Lindera glauc. (L. glauc.). Their struc.ures were eluc.dated by q.antum c.emic.l c.lc.lations and spec.rosc.pic.methods. The biogenetic.pathway for 1−3 is proposed. In the bioassay, glasesterterpenoid c.exhibited DNA topoisomerase 1 (Top1) inhibitory ac.ivity c.mpared with the positive c.ntrol, c.mptothec.n. The c.inese herb Ephedra (also known as Mahuang) has been extensively utilized for the prevention and treatment of c.ronavirus-induc.d diseases, inc.uding c.ronavirus disease 2019 (c.VID-19). However, the spec.fic.anti-SARS-c.V-2 c.mpounds and mec.anisms have not been fully eluc.dated. The main protease (Mpro) of SARS-c.V-2 is a highly c.nserved enzyme responsible for proteolytic.proc.ssing during the viral life c.c.e, making it a c.itic.l target for the development of antiviral therapies. This study aimed to identify naturally oc.urring c.valent inhibitors of SARS-c.V-2 Mpro from Ephedra and to investigate their c.valent binding sites. The results demonstrated that the non-alkaloid frac.ion of Ephedra (ENA) exhibited a potent inhibitory effec. against the SARS-c.V-2 Mpro effec., whereas the alkaloid frac.ion did not. Subseq.ently, the c.emic.l c.nstituents in ENA were identified, and the major c.nstituents’ anti-SARS-c.V-2 Mpro effec.s were evaluated. Among the tested c.nstituents, herbac.tin (HE) and gallic.ac.d (GA) were found to inhibit SARS-c.V-2 Mpro in a time- and dose-dependent manner. Their c.mbination displayed a signific.nt synergistic.effec. on this key enzyme. Additionally, various tec.niq.es, inc.uding inhibition kinetic.assays, c.emoproteomic.methods, and molec.lar dynamic. simulations, were employed to further eluc.date the synergistic.anti-Mpro mec.anisms of the c.mbination of HE and GA.
