The emergence and spread of the mobile colistin-resistance gene,mcr-1,and its variants pose achallenge to the use of colistin,a last-resort antibiotic used to treat severe infections caused by extensively drug-resistant(XDR)Gram-negative pathogens.Antibiotic adjuvants are a promising strategy to enhance the efficacy of colistin against colistin-resistant pathogens;however,few studies have considered the effects of adjuvants on limiting resistance-gene transmission.We found that chelerythrine(4 mg·L^(-1))derived from Macleaya cordata extract,which is used as an animal feed additive,reduced the minimal inhibitory concentration(MIC)of colistin against an mcr-1 positive Escherichia coli(E.coli)strain by 16-fold(from 2.000 to 0.125 mg·L^(-1)).eliminated approximately 10^(4) colony-forming units(CFUs)of an mcr-1-carrying strain in a murine intestinal infection model,and inhibited the conjugation of an mcr-1-bearing plasmid in vitro(by>100-fold)and in a mouse model(by up to 5-fold).A detailed analysis revealed that chelery-thrine binds to phospholipids on bacterial membranes and increases cytoplasmic membrane fluidity,thereby impairing respiration,disrupting proton motive force(PMF),generating reactive oxygen species(ROS),and decreasing intracellular adenosine triphosphate(ATP)levels,which subsequently downregu-lates mcr-1 and conjugation-associated genes.These dual effects of chelerythrine can expand the use of antibiotic adjuvants and may provide a new strategy for circumventing mobile colistin resistance.

Yet More Elementary Proof of Matrix-Tree Theorem for Signed Graphs

A signed graph G˙=(G,σ)is a graph G=(V(G),E(G))with vertex set V(G)and edge set E(G),together with a functionσ:E→{+1,−1}assigning a positive or negative sign to each edge.In this paper,we present a more elementary proof for the matrix-tree theorem of signed graphs,which is based on the relations between the incidence matrices and the Laplcians of signed graphs.As an application,we also obtain the results of Monfared and Mallik about the matrix-tree theorem of graphs for signless Laplacians.

Distribution law and susceptibility of geohazards across a gradient belt of the Western Sichuan Plateau

Across a gradient belt of the Western Sichuan Plateau,geohazards have seriously limited economic and social development.According to incomplete statistics,15,673 geohazards have been recorded in the study area.In order to mitigate the threat of geohazards to human engineering activities in the region,an overall understanding of the distribution pattern of geohazards and susceptibility assessment are necessary.In this paper,a gradient belt of the Western Sichuan Plateau and its zoning criteria were defined.Subsequently,on the basis of relief amplitude,distance to faults,rainfall,and human activities,three indicators of endogenic process were introduced:Bouguer gravity anomaly gradient,vertical deformation gradient,and horizontal deformation gradient.Thereafter,the distribution patterns of geohazards were investigated through mathematical statistics and ArcGIS software.By randomly selecting 10,449 hazards,a geohazard susceptibility map was generated using the Information Value(IV)model.Finally,the IV model was validated against 5224 hazards using the Area Under Curve(AUC)method.The results show that 47.6%of the geohazards were distributed in the zone of steep slope.Geohazards showed strong responses to distance to faults,human activities,and annual rainfall.The distribution of geohazards in the gradient belt of the Western Sichuan Plateau is more sensitive to vertical internal dynamics factors(such as vertical deformation gradient and Bouguer gravity anomaly gradient)without any apparent sensitivity to horizontal internal dynamics factors.The areas of high and very-high risk account for up to 32.22%,mainly distributed in the Longmenshan and Anning River faults.According to the AUC plot,the success rate of the IV model for generating the susceptibility map is 76%.This susceptibility map and geohazard distribution pattern can provide a reference for geological disaster monitoring,preparation of post-disaster emergency measures,and town planning.

Environmental standards and beneficial uses of waste-to-energy(WTE)residues in civil engineering applications

The waste-to-energy(WTE)technologies are now recovering energy and materials from over 300 million tonnes of municipal solid wastes worldwide.Extensive studies have investigated substituting natural construction materials with WTE residues to relieve the environmental cost of natural resource depletion.This study examined the beneficial uses of WTE residues in civil engineering applications and the corresponding environmental standards in Europe,the U.S.,and China.This review presents the opportunities and challenges for current technical approaches and the environmental standards to be met to stabilize WTE residues.The principal characteristics of WTE residues(bottom ash and fly ash)and the possible solutions for their beneficial use in developed and developing countries are summarized.The leaching procedures and environmental standards for pH,heavy metals,and polychlorinated dibenzo-p-dioxins/furans(PCDD/Fs)are compared.The current practice and engineering properties of materials using WTE residues,including mixtures with stone aggregate or sand,cement-based or hot-mix asphalt concrete(pavement),fill material in the embankments,substitute of Portland cement or clinker production,and ceramic-based materials(bricks and lightweight aggregate)are comprehensively reviewed.

Self-Exfoliation of Flake Graphite for Bioinspired Compositing with Aramid Nanofiber toward Integration of Mechanical and Thermoconductive Properties

Flexible yet highly thermoconductive materials are essential for the development of next-generation flexible electronic devices.Herein,we report a bioinspired nanostructured film with the integration of large ductility and high thermal conductivity based on self-exfoliated pristine graphene and three-dimensional aramid nanofiber network.A self-grinding strategy to directly exfoliate flake graphite into few-layer and few-defect pristine graphene is successfully developed through mutual shear friction between graphite particles,generating largely enhanced yield and productivity in comparison to normal liquid-based exfoliation strategies,such as ultrasonication,high-shear mixing and ball milling.Inspired by nacre,a new bioinspired layered structural design model containing three-dimensional nanofiber network is proposed and implemented with an interconnected aramid nanofiber network and high-loading graphene nanosheets by a developed continuous assembly strategy of sol-gel-film transformation.It is revealed that the bioinspired film not only exhibits nacre-like ductile deformation behavior by releasing the hidden length of curved aramid nanofibers,but also possesses good thermal transport ability by directionally conducting heat along pristine graphene nanosheets.

Capability of discrete element method to investigate the macro-micro mechanical behaviours of granular soils considering different stress conditions and morphological gene mutation

Discrete element method(DEM)has been widely utilised to model the mechanical behaviours of granular materials.However,with simplified particle morphology or rheology-based rolling resistance models,DEM failed to describe some responses,such as the particle kinematics at the grain-scale and the principal stress ratio against axial strain at the macro-scale.This paper adopts a computed tomography(CT)-based DEM technique,including particle morphology data acquisition from micro-CT(mCT),spherical harmonic-based principal component analysis(SH-PCA)-based particle morphology reconstruction and DEM simulations,to investigate the capability of DEM with realistic particle morphology for modelling granular soils’micro-macro mechanical responses with a consideration of the initial packing state,the morphological gene mutation degree,and the confining stress condition.It is found that DEM with realistic particle morphology can reasonably reproduce granular materials’micro-macro mechanical behaviours,including the deviatoric stressevolumetric straineaxial strain response,critical state behaviour,particle kinematics,and shear band evolution.Meanwhile,the role of multiscale particle morphology in granular soils depends on the initial packing state and the confining stress condition.For the same granular soils,rougher particle surfaces with a denser initial packing state and a higher confining stress condition result in a higher degree of shear strain localisation.

Constitutive modelling of idealised granular materials using machine learning method

Predicting the constitutive response of granular soils is a fundamental goal in geomechanics.This paper presents a machine learning(ML)framework for the prediction of the stress-strain behaviour and shearinduced contact fabric evolution of an idealised granular material subject to triaxial shearing.The MLbased framework is comprised of a set of mini-triaxial tests which provide a benchmark for the setup and validation of the discrete element method(DEM)model of the granular materials,a parametric DEM simulation programme of virtual triaxial tests which provides datasets of micro-and macro-mechanical information,as well as a multi-layer perceptron(MLP)neural network which is trained and tested using the DEM-based datasets.The ML model only requires the initial void ratio of the granular sample as the input for predicting its constitutive response.The excellent agreement between the ML model prediction and experimental test and DEM simulation results indicates that the MLebased modelling approach is capable of capturing accurately the effects of initial void ratio on the constitutive behaviour of idealised granular materials,bypassing the need to incorporate the complex micromechanics underlying the macroscopic mechanical behaviour of granular materials.Lastly,a detailed comparison between the used MLP model and long short-term memory(LSTM)model was made from the perspective of technical algorithm,prediction accuracy,and computational efficiency.

Enhancing Fe content tolerance in A356 alloys for achieving low carbon footprint aluminum structure castings

Low carbon footprint aluminum structure castings are the mainstream development direction of alu-minum alloys in the future.Enhancing the Fe content tolerance upper limit in casting aluminum alloys is considered an effective way to promote the application of recycled aluminum production.In this work,it was found that with the Ce and TiCN NPs(nanoparticles)addition simultaneously to A356 alloys,the acicular Fe-rich phases(β-Al 5 FeSi)which damages the properties can be changed into beneficial phases(core-shell heterostructures)for the alloy,and hence the mechanical properties of A356 alloys can be im-proved.The TiCN nanoparticles and rare element Ce play crucial roles in the formation of core-shell het-erostructures.The formation mechanism of core-shell heterostructure was also systematically researched from the perspective of thermodynamics and kinetics.This study provides a simple and feasible strategy to eliminate the harmful effects of Fe impurity and contributes to the industrialization of low carbon footprint aluminum structure castings.

Low infrared emitter from Ti3C2Tx MXene towards highly-efficient electric/solar and passive radiative heating

Realizing all-day and all-weather energy-saving heating is crucial for mitigating the global energy and ecology crisis.Electric/solar heating are two promising heating approaches,yet materials with high elec-trical conductivity,high solar absorptivity,and low infrared emissivity at the same time are rare in na-ture,which are highly anticipated and of great significance for highly efficient electric/solar heating.In this work,we demonstrate that Ti_(3)C_(2)T_(x) MXene with low IR emissivity(14.5%)fills the gap in the absence of the above materials,exhibiting a remarkable electric/solar heating performance.The saturated heating temperature of Ti_(3)C_(2)T_(x) film reaches a record-high value of 201°C at a low driving voltage of 1.5 V,and reaches 84.3°C under practical solar irradiation(750 W/m^(2))with a high solar to the thermal conversion efficiency of 75.3%,which is far superior to other reported materials.Meanwhile,the low IR emissivity endows Ti_(3)C_(2)T_(x) with a remarkable passive radiative heating capability of 7.0°C,ensuring zero-energy heating without electric/solar energy supply.The intrinsic characteristic of high electrical conductivity,high solar absorptivity,and low IR emissivity makes Ti_(3)C_(2)T_(x) unique existence in nature,which is highly promising for all-day and all-weather energy-saving heating.

The Puccinia striiformis effector Hasp98 facilitates pathogenicity by blocking the kinase activity of wheat TaMAPK4

The obligate biotrophic fungus Puccinia striiformis f.***.tritici(Pst)employs virulence effectors to disturb host immunity and causes devastating stripe rust disease.However,our understanding of how Pst effectors regulate host defense responses remains limited.In this study,we determined that the Pst effector Hasp98,which is highly expressed in Pst haustoria,inhibits plant immune responses triggered by flg22or nonpathogenic bacteria.Overexpression of Hasp98 in wheat(Triticum aestivum)suppressed avirulent Pst-triggered immunity,leading to decreased H2O2accumulation and promoting P.striiformis infection,whereas stable silencing of Hasp98 impaired P.striiformis pathogenicity.Hasp98 interacts with the wheat mitogenactivated protein kinase TaMAPK4,a positive regulator of plant resistance to stripe rust.The conserved TEY motif of TaMAPK4 is important for its kinase activity,which is required for the resistance function.We demonstrate that Hasp98inhibits the kinase activity of TaMAPK4 and that the stable silencing of TaMAPK4 compromises wheat resistance against P.striiformis.These results suggest that Hasp98 acts as a virulence effector to interfere with the MAPK signaling pathway in wheat,thereby promoting P.striiformis infection.
