The present study observed hemodynamic changes in 26 patients with cerebral arteriosclerosis using a cerebral circulation dynamics detector and transcranial Doppler. In patients with cerebral arteriosclerosis the blood supply and flow rate in the bilateral carotid arteries and the blood flow rate in the anterior cerebral and middle cerebral arteries were similar to normal controls, but the cerebral vascular resistance, critical pressure and pulsatility index were increased, and cerebral arterial elasticity and cerebral blood flow autoregulation were decreased. Compared with the lesioned hemisphere of patients with cerebral infarction, the total blood supply and blood flow rate of patients with cerebral arteriosclerosis were higher. Compared with normal subjects, patients with cerebral arteriosclerosis exhibited cognitive disturbances, mainly in short-term memory, attention, abstract capability, and spatial and executive dysfunction. Results showed that cerebral arteriosclerosis does not directly affect the blood supply of a cerebral hemisphere, but affects cognitive function. The increased cerebral vascular resistance and reduced autoregulation of cerebral blood vessels may be important hemodynamic mechanisms of arteriosclerosis-induced cerebral infarction.

背景:研究发现,腰椎间盘突出症患者的疼痛机制与炎症相关,自噬与椎间盘疾病及炎症反应密切相关,miR-206表达异常可促进骨骼疾病的发生。目的:探讨miR-206对腰椎间盘突出大鼠髓核炎症、镇痛及自噬相关蛋白的作用机制。方法:SPF级雄性SD大鼠60只按照随机数字方法分为对照组、模型组、miR-206 mimics-NC组、miR-206 mimics组、miR-206 inhibitor-NC组及miR-206 inhibitor组。除对照组外均建立腰椎间盘突出症大鼠模型,建模后第10天miR-206 mimics-NC组、miR-206 mimics组、miR-206 inhibitor-NC组及miR-206 inhibitor组分别在损伤处注射miR-206 mimics-NC、miR-206 mimics(miR-206模拟物)、miR-206 inhibitor-NC及miR-206 inhibitor(miR-206抑制物),均20μmol/L,10μL,1次/d,连续注射4 d;对照组及模型组均注射等剂量生理盐水。Von Frey纤维丝测定大鼠双侧后足机械性缩足反射阈值,热痛测试仪检测大鼠双侧后足热刺激缩足反射潜伏期;苏木精-伊红染色观察背根神经节组织形态;qPCR检测各组大鼠髓核组织炎症因子磷脂酶A2、环氧化酶2、前列腺素E2、肿瘤坏死因子α、白细胞介素1β表达;免疫印迹检测自噬相关蛋白LC3Ⅰ、Beclin-1表达。结果与结论:①造模后3,7及14 d时,与对照组相比,模型组大鼠机械性缩足反射阈值、热刺激缩足反射潜伏期降低,磷脂酶A2、环氧化酶2、前列腺素E2、肿瘤坏死因子α、白细胞介素1β、LC3Ⅰ、Beclin-1表达升高(P0.05);②与miR-206 mimics-NC组相比,miR-206 mimics组大鼠机械性缩足反射阈值、热刺激缩足反射潜伏期降低,磷脂酶A2、环氧化酶2、前列腺素E2、肿瘤坏死因子α、白细胞介素1β、LC3Ⅰ、Beclin-1水平升高(P<0.05);与miR-206 inhibitor-NC组相比,miR-206 inhibitor组大鼠上述指标呈相反变化,差异均有显著性意义(P<0.05);③结果说明,抑制miR-206可以显著改善腰椎间盘突出症大鼠髓核炎症因子水平,提高疼痛阈值,降低细胞自噬,其作用机制与抑制LC3Ⅰ、Beclin-1表达相关。

Most of the important agronomic traits in crop plants, such as yield, quality and stress response, are quantitative and jointly controlled by many genomic loci or major genes. Improving these complex traits depends on the combination of beneficial alleles at the quantitative trait loci （QTLs）. However, the conventional cross breeding method is extremely time-consuming and laborious for pyramiding multiple QTLs. In certain cases, this approach might be technically difficult because of close linkage between genes separately responsible for desirable and undesirable traits.

铅基固废是含铅的固体废物,主要来源于锌、铅、铜冶炼过程和废旧铅酸蓄电池回收,含有大量有价金属,同时对环境有一定的污染,因此摸清铅基固废的来源结构、产量及区域分布显得尤为重要。本文在实地企业调研的基础上分析了锌、铅、铜主要冶炼技术及其产生的铅基固废,根据单吨精炼金属的固废产出比及精炼金属产量测算了铅基固废产量,基于锌、铅、铜产量分布描绘铅基固废的区域分布,依照铅基固废污染元素含量和铅基固废产量测算有价元素的产量,以期为铅基固废处置技术开发研究及产业布局提供基础支撑。

Increasing fidelity and efficiency by modifying cytidine base-editing systems in rice

The efficiency of plant cytidine base-editing systems is limited, and unwanted mutations frequently occur in transgenic plants. We increased the cytidine editing frequency and fidelity of the plant base editor 3(BE3) and targeted activation-induced cytidine deaminase(CDA)(target-AID) systems by coexpressing three copies of free uracil–DNA glycosylase(UDG) inhibitor(UGI). The editing efficiency of the improved BE3 and CDA systems reached as high as 88.9% and 85.7%, respectively, in regenerated rice plants, with a very low frequency of unwanted mutations. The low editing frequency of the BE3 system in the GC context could be overcome by the modified CDA system. These results provide a highfidelity and high-efficiency solution for rice genomic base editing.

针对部分线性变系数模型的参数估计问题,采用B样条方法逼近非参数部分的未知光滑函数,进而利用非对称拉普拉斯分布实现贝叶斯分位数回归,并基于Gibbs抽样推导出所有未知参数的后验分布,通过数值模拟比较分析了贝叶斯分位数回归与分位数回归参数估计的优劣,结果表明,在均方误差准则下,贝叶斯分位数回归的估计效果更优。最后,通过实例分析说明了所提方法的有效性。

This study reports an investigation of the chemical constituents of Balanophora polyandra Griff.Fifteen compounds were isolated by column chromatography on silica gel,Toyo-pearl HW-40C,Sephadex LH-20 and by HPLC.Their structures were elucidated as 1,4-di-O galloy-2-O-[(E)-p-coumaroyl]-β-D-glucopyranose(1),1-O-galloyl-β-D-pyranglucose(2),1-p-coumaryl-β-D-pyranglu-cose(3),1-O-(E)-caffeoyl-β-D-pyranglucose(4),1,3-di-O-galloyl-β-D-pyranglucose(5),1,6-di-O-galloyl-β-D-pyranglucose(6),1-O-(E)-caffeoyl-4-O-galloyl-β-D-pyranglucose(7),1-O-(E)-caffeoy1-6-O galloyl-β-D-pyranglucose(8),1-0-(E)-caffeoy1-4,6-di-O-galloyl-β-D-pyranglucose(9),1-O-(E)-caffeoyl-4,6-(S)-HHDP-B-D-pyranglucose(10),1,23,6-tetra-O-galloyl-β-D-pyranglucose(11),4,6-(S)-hexahydroxydi-phenoyl-(α/β)-D-glucose(12),1-O-(E)-caffeoyl-4,6-[1′,1″-3′,3″,4′,4″-tetrahydroxydibenzofurandicar-boxyl)]-β-D glucopyranose(13),flavogallonic acid(14),and phloretin-4′-O-β-D-glucoside(15)on the basis of spectral analysis.Compound 1 was a new hydrolyzable tannin,9 was obtained from this genus for the first time,and compounds 5,6 and 11-14 were isolated from this plant for the first time.

Three novel polycyclic polyprenyled acylphloroglucinols,Hyperscabins A-C,were obtained from the aerial parts of Hypericum scabrum.They featured an unprecedented 5,5-spiro ketal subunit with the loss of C-2’carbonyl in the phloroglucinol ring.Their structures were characterized by extensive spectroscopic analyses,NMR calculations with DP4+analysis,calculated electronic circular dichroism(ECD)spectra and the application of modified Mosher’s methods.In the assay of[^(3)H]-5-HT and[^(3)H]-NE reuptake inhibition,compounds 1 and 2 showed good inhibito ry activity(81.8%and 83.2%)in 10μmol/L.In addition,compound 1 significantly increased cell viability in the experiment of oxygen and glucose deprivation/deoxygenation.
