Metabolic basis of solute carrier transporters in treatment of type 2 diabetes mellitus

Solute carriers(SLCs) constitute the largest superfamily of membrane transporter proteins.These transporters, present in various SLC families, play a vital role in energy metabolism by facilitating the transport of diverse substances, including glucose, fatty acids, amino acids, nucleotides, and ions.They actively participate in the regulation of glucose metabolism at various steps, such as glucose uptake(e.g., SLC2A4/GLUT4), glucose reabsorption(e.g., SLC5A2/SGLT2), thermogenesis(e.g., SLC25A7/UCP-1), and ATP production(e.g., SLC25A4/ANT1 and SLC25A5/ANT2). The activities of these transporters contribute to the pathogenesis of type 2 diabetes mellitus(T2DM). Notably, SLC5A2 has emerged as a valid drug target for T2DM due to its role in renal glucose reabsorption, leading to groundbreaking advancements in diabetes drug discovery. Alongside SLC5A2, multiple families of SLC transporters involved in the regulation of glucose homeostasis hold potential applications for T2DM therapy. SLCs also impact drug metabolism of diabetic medicines through gene polymorphisms, such as rosiglitazone(SLCO1B1/OATP1B1) and metformin(SLC22A1-3/OCT1-3 and SLC47A1, 2/MATE1, 2). By consolidating insights into the biological activities and clinical relevance of SLC transporters in T2DM, this review offers a comprehensive update on their roles in controlling glucose metabolism as potential drug targets.

Enhancement of heavy metals desorption from the soil by eddy deep leaching in hydrocyclone

An eddy deep leaching technology was developed in this paper to address the challenge of treating heavy metal contaminants in industrial mining areas.The desorption effect of As,Cd,Sb and Pb was investigated utilizing chemical leaching and physical eddy techniques.It was found that the heavy metals concentration increased with decreasing particle size.The highest proportion of Cd in the form distribution of soil was in the bound to iron and manganese oxides,while the maximum proportion of As,Sb and Pb were in the residual.The optimal solid-liquid ratio of the hydrocyclone was 1:20,and the corresponding separation efficiency and flow rate were 84.7%and 1.76 m^(3)/hr,respectively.The grade efficiency of soil particle separation increases with particle size and exceeds 99%for particles above 1,000μm.Leaching experiments have revealed that oxalic acid(OA)and a combination of oxalic acid and EDTA(OAPE)were more efficient than citric acid(CA)and a combination of citric acid and EDTA(CAPE)for the desorption of heavy metals,respectively.The comparison of OAPE and eddy leaching found that the latter improved the desorption efficiency by 9.4%,7.5%,7.2%and 7.8%for As,Cd,Sb and Pb compared to the former,respectively.The results demonstrated that the eddy leaching technique could further enhance the desorption efficiency of heavy metals.It is expected to provide technical support for soil remediation with reduced usage of leaching agents.

DNA metabarcoding analysis of fungal community on surface of four root herbs

Objective: Angelicae Sinensis Radix(ASR, Danggui in Chinese), Cistanches Herba(CH, Roucongrong in Chinese), Ginseng Radix et Rhizoma(PG, Renshen in Chinese), and Panacis Quinquefolii Radix(PQ,Xiyangshen in Chinese), widely used as medicine and dietary supplement around the world, are susceptible to fungal and mycotoxin contamination. In this study, we aim to analyze their fungal community by DNA metabarcoding.Methods: A total of 12 root samples were collected from three main production areas in China. The samples were divided into four groups based on herb species, including ASR, CH, PG, and PQ groups. The fungal community on the surface of four root groups was investigated through DNA metabarcoding via targeting the internal transcribed spacer 2 region(ITS2).Results: All the 12 samples were detected with fungal contamination. Rhizopus(13.04%-74.03%),Aspergillus(1.76%-23.92%), and Fusarium(0.26%-15.27%) were the predominant genera. Ten important fungi were identified at the species level, including two potential toxigenic fungi(Penicillium citrinum and P. oxalicum) and eight human pathogenic fungi(Alternaria infectoria, Candida sake, Hyphopichia burtonii, Malassezia globosa, M. restricta, Rhizopus arrhizus, Rhodotorula mucilaginosa, and Ochroconis tshawytschae). Fungal community in ASR and CH groups was significantly different from other groups,while fungal community in PG and PQ groups was relatively similar.Conclusion: DNA metabarcoding revealed the fungal community in four important root herbs. This study provided an important reference for preventing root herbs against fungal and mycotoxin contamination.

Tandem engineering for CO_(2)electrolysis toward multicarbon products

Electrocatalysis of CO_(2)toward multicarbon(C_(2+))products have multifaceted applications in the energy and chemical industries,offers an attractive route to mitigate carbon emissions and abate the depletion of fossil fuels.However,the productivity of CO_(2)-to-C_(2+)products suffers from a low selectivity and reaction rate owing to the difficulty in C-C coupling and the multiple electronproton transfer steps.Recently,numerous tandem catalysts have been developed to improve the selectivity and formation rate of CO_(2)-to-C_(2+)products via coupled multiple reaction steps,exhibiting high industrial practicability.This review summarized recent progresses in the formation of C_(2+)products from CO_(2)electrolysis on tandem catalysts.In this review,we highlight the cooperative regulation strategy of tandem catalysts formed by introducing different types of new components and reveal the relationships between*CO intermediate mass transport and the selectivity of C_(2+)products.Moreover,theoretical insight into the tandem catalytic mechanisms underlying the enhanced C_(2+)selectivity is also provided.Finally,the remaining challenges and opportunities for the electrocatalytic CO_(2)toward C_(2+)products are discussed.

Gout,a prevalent and painful metabolic disease often associated with obesity and aging,is caused by the deposition of urate crystals in joints,bones,or soft tissues1.Urate is an intermediate metabolite within the purine degradation pathway,predominantly derived from uric acid under physiological pH levels.Hyperuricemia occurs due to excessive uric acid production or insufficient excretion2,which is associated with various chronic diseases,including type 2 diabetes.

High-Volume Mineral Admixtures Cement: The Effects of Particle Size Distribution

The effects of high-volume slag-fly ash cement with different particle sizes on hydration degree,microstructure and mechanical properties were systematically studied,by means of laser particle size(DLS),X-ray diffraction (XRD),comprehensive thermal analysis (TG-DTA),scanning electron microscopy(SEM) and mechanical properties tests.The results show that suitable particle size distribution of cementitious material has significantly promoting effects on hydration reaction rate and mechanical properties.Compared with slag without further grinding,the slag after ball milling for 4 h has an obvious improvement in reactivity,which also provides a faster hydration rate and higher compressive strength for the cementitious material.When the slag milled for 1 and 4 h is mixed at a mass ratio of 2:1 (i e,slag with D_(50) of 7.4μm and average size of 9.9μm,and slag with D_(50) value of 2.6μm and average size of 5.3μm),and a certain amount of fly ash is added in,the most obvious improvement of compressive strength of cement is achieved.

A novel approach to minimizing material loss for computer numerical control flank-regrinding of worn end mills

Flanks of end mills are prone to wear in a long machining process.Regrinding is widely used in workshops to restore the flank to an original-like state.However,the traditional method involves material waste by trial and error and dramatically decreases the potential regrinding.Moreover,over-cut would happen to the flutes of worn cutters in the regrinding processes because of improper wheel path.This study presented a new approach to planning the wheel path for regrinding worn end mills to minimize material loss and recover the over-cut.In planning,a scaling method was developed to determine the maximum size of the new cutter according to the similarity of cutter shapes before and after regrinding.Then,the wheel path is first generated by envelope theory to regrind the worn area with a four-axis computer numerical control grinder according to the new size of cutters.Moreover,a second regrinding strategy is applied to recover the flute shape over-cut in the first grinding.Finally,the proposed method is verified by an experiment.Results showed that the proposed approach could save 25%of cutter material compared with the traditional method and ensure at least three regrinding times.This work effectively provides a general regrinding solution for the worn flank with maximum material-saving and regrinding period.

Hosts evolve defensive strategies to minimize the reproductive costs of brood parasitism,in turn,the hosts'defense promote the optimization of cuckoo parasitism strategies(Soler 2017).Recognizing foreign eggs and selectively removing them from the nest can minimize the fitness costs associated with rearing parasitic fledglings(Soler 2017).Since Rothstein(1971)first experimented with the addition of a model egg to a host nest in the early incubation period to study host egg recognition,the use of model eggs has become one of the most important methods for testing the egg recognition abilities of hosts(Hauber et al.2019).Some studies have looked at various egg colors and their effect on host egg rejection and found differential responses to egg colors(for more details,see Supplementary Materials),highlighting that more work needs to examine the effect of model egg color on host egg rejection behavior.
