“Zero ischemia”laparoscopic partial nephrectomy by high-power GreenLight laser enucleation for renal carcinoma:A single-center experience

BACKGROUND Laparoscopic partial nephrectomy has been widely used in renal cell carcinoma *** efficacy of GreenLight laser on Laparoscopic partial nephrectomy is still *** To present the first series of laparoscopic partial nephrectomy(LPN)by GreenLight laser enucleation without renal artery *** to the excellent coagulation and hemostatic properties of the laser,laser-assisted LPN(LLPN)makes it possible to perform a“zero ischemia”*** Fifteen patients with T1a exogenous renal tumors who received high-power GreenLight laser non-ischemic LPN in our hospital were retrospectively *** clinical information,surgical and post-operative data,complications,pathological and functional outcomes were *** Surgery was successfully completed in all patients,and no open or radical nephrectomy was *** renal artery was not clamped,leading to no ischemic *** blood transfusions were required,the average hemoglobin level ranged from 96.0 to 132.0 g/L and no postoperative complications *** mean operation time was 104.3±8.2 *** postoperative removal of negative pressure drainage time ranged from 5.0 to 7.0 d,and the mean postoperative hospital stay was 6.5±0.7 *** serious complications *** pathological results showed clear cell carcinoma in 12 patients,papillary renal cell carcinoma in 2 patients,and hamartoma in 1 *** mean creatinine level was 75.0±0.8μmol/L(range 61.0-90.4μmol/L)at 1 mo after surgery,and there were no statistically significant differences compared with pre-operation(P>0.05).The glomerular filtration rate ranged from 45.1 to 60.8 mL/min,with an average of 54.0±5.0 mL/min,and these levels were not significantly different from those before surgery(P>0.05).CONCLUSION GreenLight laser has extraordinary cutting and sealing advantages when used for small renal tumors(exogenous tumors of stage T1a)during ***,use of this technique can lead to the generation

BACKGROUND Complement overactivation is a major driver of lupus nephritis(LN).Impaired interactions of C-reactive protein(CRP)with complement factor H(CFH)have been shown as a pathogenic mechanism that contributes to the overactivation of complement in ***,genetic variations of neither CRP nor CFH show consistent influences on the risk of *** To examine whether genetic variations of CRP and CFH in combination can improve the risk stratification in Chinese *** We genotyped six CRP single nucleotide polymorphisms(SNPs)(rs1205,rs3093062,rs2794521,rs1800947,rs3093077,and rs1130864)and three CFH SNPs(rs482934,rs1061170,and rs1061147)in 270 LN patients and 303 healthy *** No linkage was found among CRP and CFH SNPs,indicating lack of genetic interactions between the two ***,CRP and CFH SNPs,neither individually nor in combination,are associated with the risk or clinical manifestations of *** the unambiguous pathogenic roles of the two *** These findings suggest that the biological effects of most genetic variations of CRP and CFH on their expressions or activities are not sufficient to influence the disease course of LN.

C-reactive protein（CRP） is a prototypical acute phase reactant in humans, whose serum level can increase up to 1000-fold in response to tissue injury or infection. CRP is a secretory protein composed of five identical, non-covalently associated subunits with no glycosylation modification. However, it is unclear how CRP folds and assembles in live cells, leading to its efficient secretion. We show that the cellular folding and assembly of CRP follow a predetermined, stepwise pathway with obligatory intermediates. Conformation folding and disulfide bonding drive distinct folding steps. These results provide new insight into the mechanism of efficient CRP production, and may also help to clarify some controversies in protein folding.

Internal alkynes are widespread skeletons in bioactive molecules and materials which are also used as important building blocks in chemical synthesis. Herein, we report a novel and efficient copper-catalyzed Sonogashira reaction between sulfonium salts and alkynes through an anti-Markovnikov ring-opening pathway promoted by visible light. The coexistence of the nitrogen and phosphorus ligands in the system is the key to the success of this reaction. This radical type ring-opening reaction affords a new strategy for the synthesis of internal alkynes. Furthermore, this study is also an effective complement to the diverse reaction patterns of sulfonium salts.

The development of green and convenient methods for C–S bond formation has received significant attention because C–S bond widely occurs in many important pharmaceutical and biological ***, visible-light photoredox catalysis has been established as an efficient and general tool for the construction of C–C and C-heteroatom bonds. In this review, we have focused on the research on recent advances in C–S bond formation via visible-light photoredox catalysis, and the growing opportunities they present to the construction of complex chemical scaffolds for applications encompassing bioactive molecules synthesis, synthetic methodology development, and sulfur-containing drugs. We hope that this review will provide chemists with a synthetic tool that will open the door to further development of organsulfur chemistry.

Comparative analysis of volatile oils in the stems and roots of Ephedra sinica via GC-MS-based plant metabolomics

With a great difference in therapeutic effects of Mahuang(MH, the stems of Ephedra sinica) and Mahuanggen(MHG, the roots of Ephedra sinica), chemical differences between MH and MHG should be investigated. In the present study, gas chromatography-mass spectrometry(GC-MS)-based plant metabolomics was employed to compare volatile oil profiles of MH and MHG. The antioxidant activities of volatile oils from MH and MHG were also compared. 32 differential chemical markers were identified according to the variable importance in the projection(VIP) value of orthogonal partial least squares discriminant analysis(OPLS-DA) and P value of Mann-Whitney test. Among them, chemical markers of tetramethylpyrazine(TMP) and α-terpineol were quantified. Their contents were much higher in most MH samples compared with MHG. The antioxidant assay demonstrated that MH had significantly higher free radical-scavenging activity than MHG. Although MH and MHG derived from the same medicinal plant, there was much difference in their volatile oil profiles. MH samples had significantly higher content of two reported pharmacologically important chemical markers of TMP and α-terpineol, which may account for their different antioxidant activities.

Prediction of shelled shrimp weight by machine vision

The weight of shelled shrimp is an important parameter for grading *** weight prediction of shelled shrimp by contour area is not accurate enough because of the ignorance of the shrimp *** this paper,a multivariate prediction model containing area,perimeter,length,and width was established.A new calibration algorithm for extracting length of shelled shrimp was proposed,which contains binary image thinning,branch recognition and elimination,and length reconstruction,while its width was calculated during the process of length *** model was further validated with another set of images from 30 shelled *** a comparison purpose,artificial neural network(ANN) was used for the shrimp weight *** ANN model resulted in a better prediction accuracy(with the average relative error at 2.67%),but took a tenfold increase in calculation time compared with the weight-area-perimeter(WAP) model(with the average relative error at 3.02%).We thus conclude that the WAP model is a better method for the prediction of the weight of shelled red shrimp.

N-Methyl-D-aspartate receptors（NMDARs） play a central role in various physiological and pathological processes in the central nervous *** they are commonly composed of four subunits,two GluN1 subunits and two GluN2 or GluN3 *** different subunit compositions make NMDARs a heterogeneous population with distinct electrophysiological and pharmacological properties and thus with different abilities to conduct neuronal *** subunit composition,assembly process,and final structure of assembled NMDARs have been studied for years but no consensus has been *** this study,we investigated the role of the amino terminal domain（ATD） of GluN2A in regulating NMDAR *** ATD of GluN2A was first expressed in heterogeneous cells and the homodimer formation was investigated by fluorescent resonance energy transfer and non-reducing SDS- PAGE *** of the three cysteine residues located in the ATD was mutated into alanine,and the homodimerization of the ATD or GluN2A,as well as the heteromeric assembly of NMDARs was assessed by non-reducing SDS- PAGE electrophoresis,co-immunoprecipitation and *** found that two cysteine residues,C87 and C320,in the ATD of the GluN2A subunit were required for the formation of disulfide bonds and GluN2A ATD ***,the disruption of GluN2A ATD domain dimerization had no influence on the assembly and surface expression of *** results suggest that the two ATD domains of GluN2A are structurally adjacent in fully-assembled ***,unlike GluN1,the homomerization of the ATD domain of GluN2A is not required for the assembly of NMDARs,implying that GluN2A and GluN1 play unequal roles in NMDAR assembly.
