On implications of somatostatin in diabetic retinopathy

Somatostatin,a naturally produced neuroprotective peptide,depresses excitatory neurotransmission and exerts anti-proliferative and anti-inflammatory effects on the retina.In this review,we summarize the progress of somatostatin treatment of diabetic retinopathy through analysis of relevant studies published from February 2019 to February 2023 extracted from the PubMed and Google Scholar databases.Insufficient neuroprotection,which occurs as a consequence of declined expression or dysregulation of retinal somatostatin in the very early stages of diabetic retinopathy,triggers retinal neurovascular unit impairment and microvascular damage.Somatostatin replacement is a promising treatment for retinal neurodegeneration in diabetic retinopathy.Numerous pre-clinical and clinical trials of somatostatin analog treatment for early diabetic retinopathy have been initiated.In one such trial(EUROCONDOR),topical administration of somatostatin was found to exert neuroprotective effects in patients with pre-existing retinal neurodysfunction,but had no impact on the onset of diabetic retinopathy.Overall,we concluded that somatostatin restoration may be especially beneficial for the growing population of patients with early-stage retinopathy.In order to achieve early prevention of diabetic retinopathy initiation,and thereby salvage visual function before the appearance of moderate non-proliferative diabetic retinopathy,several issues need to be addressed.These include the needs to:a)update and standardize the retinal screening scheme to incorporate the detection of early neurodegeneration,b)identify patient subgroups who would benefit from somatostatin analog supplementation,c)elucidate the interactions of somatostatin,particularly exogenously-delivered somatostatin analogs,with other retinal peptides in the context of hyperglycemia,and d)design safe,feasible,low cost,and effective administration routes.

Buckling Properties of Water-Drop-Shaped Pressure Hulls with Various Shape Indices Under Hydrostatic External Pressure

The water-drop-shaped pressure hull has a good streamline,which has good application prospect in the underwater observatory.Therefore,this study conducted analytical,experimental and numerical investigation of the buckling properties of water-drop-shaped pressure hulls under hydrostatic pressure.A water-drop experiment was conducted to design water-drop-shaped pressure hulls with various shape indices.The critical loads for the water-drop-shaped pressure hulls were resolved by using Mushtari’s formula.Several numerical simulations including linear buckling analysis and nonlinear buckling analysis including eigenmode imperfections were performed.The results indicated that the critical loads resolved by Mushtari's formula were in good agreement with the linear buckling loads from the numerical simulations.This formula can be extended to estimate the buckling capacity of water-drop-shaped pressure hulls.In addition,three groups of pressure hulls were fabricated by using stereolithography,a rapid prototyping technique.Subsequently,three groups of the pressure hulls were subjected to ultrasonic measurements,optical scanning,hydrostatic testing and numerical analysis.The experimental results were consistent with the numerical results.The results indicate that the sharp end of the water-drop-shaped pressure hulls exhibited instability compared with the blunt end.This paper provides a new solution to the limitations of experimental studies on the water-drop-shaped pressure hulls as well as a new configuration and evaluation method for underwater observatories.

Pd-catalyzed asymmetric carbonyl alkynylation:Synthesis of axial chiral ynones

Ynones are important skeletons in bioactive molecules and valuable building blocks for organic synthesis,thus great efforts have been devoted to their preparation.While,introducing prochiral substrates to construct ynones bearing a chiral framework is unrealized to date.Herein,we reported the first example of Pd/SOP-catalyzed asymmetric carbonylative alkynylation via a non-classical carbonylative Sonogashiratype approach(acyl-Pd(Ⅱ)species generated from nucleophiles).By using cyclic diaryliodonium salts as prochiral substrates,various axial chiral ynones with good functional group tolerance(39 examples),satisfied yields(71%-96%)and excellent enantioselectivities(generally 94%-99%ee)were produced.Synthesis of bioactive compounds,scale-up experiment and useful transformations were also conducted to demonstrate the utility of this process.

Chronic urticaria(CU)is a common skin disease characterized by the occurrence of wheals,with or without angioedema,lasting more than 6 weeks.The prevalence of CU is about 0.7%[1],but higher in children,with an incidence of 1.38%[1].The occurrence of severe pruritus and unpredictable wheals may severely impact patients'quality of life,cause psychological problems,including sleep disorders and depression,and lead to economic burden due to limitation on daily life and work[2,3].

Decreased Nrf2 protein level and low sperm quality in intractable spermatocystitis

To investigate the molecular etiology of low sperm quality in patients with intractable spermatocystitis,spermatozoa samples from patients with persistent hematospermia undergoing transurethral seminal vesiculoscopy and healthy volunteers were utilized.Spermatozoa samples were collected from the seminal vesicles through transurethral seminal vesiculoscopy or by masturbation ejaculation.Sperm quality was analyzed by a WLJY-9000 color semen analysis system.Measurement of tumor necrosis factor alpha(TNFa)and interleukin-6(IL-6)in the seminal plasma was performed using enzyme-linked immunosorbent assay(ELISA).Measurement of H_(2)O_(2)in the seminal plasma was performed with a hydrogen peroxide kit.The protein levels of nuclear factor erythroid 2-related factor 2(Nrf2)and phosphorylated-Nrf2(p-Nrf2)were measured by western blot analysis and immunofluorescence assays.Low sperm quality parameters and increased levels of inflammatory cytokines(TNFa,IL-6,and H_(2)O_(2))in the seminal plasma were detected among the semen samples from the patients with persistent hematospermia.Nrf2 and p-Nrf2 were strongly expressed in the nucleus and periphery of human sperm cells,according to the results of the immunofluorescence assays.The protein levels of Nrf2 and p-Nrf2 were significantly lower in the spermatozoa samples from patients with persistent hematospermia than in those from healthy volunteers with normal sperm motility.The results suggested that Nrf2 signaling might play a role in the low sperm quality of patients with intractable spermatocystitis.

To the Editor:Pancreatic panniculitis,also known as pancreatic fat necrosis or enzymatic panniculitis,is a rare type of panniculitis that occurs in 0.3%−3%of patients with pancreatic disease such as acute or chronic pancreatitis and pancreatic carcinoma,especially acinar cell carcinoma[1,2].The clinical manifestations are nonspecific erythema tender nodules.

Evolutionary mechanisms and practical significance of reproductive success and clonal diversity in unisexual vertebrate polyploids

Unisexual reproduction is generally relevant to polyploidy, and unisexual vertebrates are often considered an evolutionary “dead end” due to the accumulation of deleterious mutations and absence of genetic diversity. However, some unisexual polyploids have developed strategies to avoid genomic decay, and thus provide ideal models to unveil unexplored evolutionary mechanisms, from the reproductive success to clonal diversity creation. This article reviews the evolutionary mechanisms for overcoming meiotic barrier and generating genetic diversity in unisexual vertebrates, and summarizes recent research advancements in the polyploid Carassius complex. Gynogenetic gibel carp(Carassius gibelio) is a unique amphitriploid that has undergone a recurrent autotriploidy and has overcome the bottleneck of triploid sterility via gynogenesis. Recently, an efficient strategy in which ploidy changes, including from amphitriploid to amphitetraploid, then from amphitetraploid to novel amphitriploid, drive unisexual-sexual-unisexual reproduction transition and clonal diversity has been revealed.Based on this new discovery, multigenomic reconstruction biotechnology has been used to breed a novel strain with superior growth and stronger disease resistance. Moreover, a unique reproduction mode that combines both abilities of ameiotic oogenesis and sperm-egg fusion,termed as ameio-fusiongensis, has been discovered, and it provides an efficient approach to synthesize sterile allopolyploids. In order to avoid ecological risks upon escape and protect the sustainable property rights of the aquaculture seed industry, a controllable fertility biotechnology approach for precise breeding is being developed by integrating sterile allopolyploid synthesis and gene-editing techniques.This review provides novel insights into the origin and evolution of unisexual vertebrates and into the attempts being made to exploit new breeding biotechnologies in aquaculture.

An Improved YOLOv7 tiny Algorithm for Vehicle and Pedestrian Detection with Occlusion in Autonomous Driving

Future transportation is advancing in the direction of intelligent transportation systems, where an essential part is vehicle and pedestrian detection. Due to the complex urban traffic environment, vehicles and pedestrians in road monitoring have different forms of occlusion problems, resulting in the missed detection of objects. We design an improved you only look once version 7(YOLOv7) tiny algorithm for vehicle and pedestrian detection under occlusion, with the following four main improvements. In order to locate the object more accurately, 1 × 1 convolution and identity connection are added to the 3 × 3 convolution, and convolution reparameterization is used to enhance the inference speed of the network model. In view of the complex road background and more interference, the coordinate attention was added to the connection part of backbone and neck to enhance the network's capacity to detect the object and lessen interference from other targets. At the same time, before being sent to the detection head, global attention mechanism is added to improve the accuracy of model detection by capturing three-dimensional features. Considering the issue of imbalanced training samples, we propose focal complete intersection over union(CIOU) loss instead of CIOU loss to become the bounding box regression loss, so that the regression process attention to high-quality anchor boxes. Experiments show that the improved YOLOv7 tiny algorithm achieves 82.2%map@0.5 in Pattern Analysis, Statistical Modelling and Computational Learning Visual Object Classes(PASCAL VOC) dataset, which is 2.8% higher than before the improvement. The performance of map@0.5:0.95 is 5.2% better than the previous improvement. The proposed improved algorithm can availably to detect partial occlusion objects.

At high temperature,the chemical reaction mechanism at the interface of magnetite and the influence mechanism of gangue element Al on the oxidation performance of magnetite are not clear.In addition,due to the limitation of existing experimental equipment,it is difficult to clarify the interface reaction mechanism in the oxidation process and the influence mechanism of Al on the surface oxidation reaction of Fe_(3)O_(4) at an atomic scale at high temperature.The surface oxidation reaction mechanism of magnetite and the influence mechanism of Al on the oxidation of magnetite were studied by experiments and AIMD(ab initio molecular dynamics).The experimental results show that the existence of Al_(2)O_(3) will reduce the initial oxidation temperature and comprehensive oxidation performance of magnetite.The AIMD results show that aluminum can accelerate the interface reaction rate of Fe_(3)O_(4) in the high-temperature oxidation atmosphere,but the strong Al-O binding ability is not conducive to the transfer of O atoms in the oxidation process.

Impaired brain glucose metabolism is an early indicator of Alzheimer’s disease(AD);however,the fundamental mechanism is unknown.In this study,we found a substantial decline in isocitrate dehydrogenase 3β(IDH3β)levels,a critical tricarboxylic acid cycle enzyme,in AD patients and AD-transgenic mice’s brains.Further investigations demonstrated that the knockdown of IDH3βinduced oxidation-phosphorylation uncoupling,leading to reduced energy metabolism and lactate accumulation.The resulting increased lactate,a source of lactyl,was found to promote histone lactylation,thereby enhancing the expression of paired-box gene 6(PAX6).As an inhibitory transcription factor of IDH3β,the elevated PAX6 in turn inhibited the expression of IDH3β,leading to tau hyperphosphorylation,synapse impairment,and learning and memory deficits resembling those seen in AD.In AD-transgenic mice,upregulating IDH3βand downregulating PAX6 were found to improve cognitive functioning and reverse AD-like pathologies.Collectively,our data suggest that impaired oxidative phosphorylation accelerates AD progression via a positive feedback inhibition loop of IDH3β-lactate-PAX6-IDH3β.Breaking this loop by upregulating IDH3βor downregulating PAX6 attenuates AD neurodegeneration and cognitive impairments.
