Exosomes secreted by tumor cells carry abundant molecular biomarkers that reflect the status of their originating cells.These tumor-derived exosomes(TDEs)have emerged as attractive diagnostic targets.However,the identification and characterization of highly heterogeneous TDEs remain practically challenging.Here,we report a dual rolling circle amplification(DRCA)-assisted approach for the selective encapsulation of single TDEs for fluorescence microscopic and flow cytometric analysis.TDEs have been targeted by aptamers that recognized their surface tumor marker and exosomal marker CD63,following DRCA that produced entangling polymeric DNA chains,resulting in facile particle enlargement that allows single-particle fluorescence profiling of exosome heterogeneity.We have demonstrated the use of a dual-marker positive ratio for exosome differentiation and applied division and multiplication operations for normalized andmagnified marker heterogeneity analysis.We further applied this assay to distinguish lung adenocarcinoma and pulmonary nodule patients and found an accuracy of 90%.We anticipate promising transformations of this straightforward assay into clinically implantable diagnostic methods.

The sources,transportation and depositional processes of lacustrine mudrock are still poorly understood.Existing studies have demonstrated the controlling effect of astronomical forcing on lacustrine mudrock deposition,but its depositional mechanism and evolution are still not systematically investigated.Most research related to astronomical forcing exclusively highlights the sedimentation of carbonate rocks in deep-water lacustrine setting,with insufficient attention paid to the thick organic-rich,deep-lake mudrock.With the increasing interest in exploration and development of shale oil and gas accumulations,it is urgent to deeply understand depositional rules of lacustrine mudrock.This study reviews sediment sources,depositional mechanism and evolution process of mudrock through expounding the correlations between the periodic changes of astronomical forces,the parameters of Earth orbital and mudrock compositions.By investigating the existing literature and using some actual data of Jiyang Depression,Bohai Bay Basin in East China,this study expounds on the influence of astronomical cycles on the deposition of lacustrine mudrock.Moreover,efforts are made to analyze the effects of various orbital parameters(e.g.,precession,obliquity,and eccentricity with the periods ranging from tens of thousands years to million years)on the deposition of mudrock from small-scale(decimeters to meters)to large-scale(10s to 100s meters).Further,it is feasible to apply the high-precision isochronous stratigraphic correlation into clarifying the distribution of favorable shale oil and gas reservoirs.To conclude,this study enunciates the sedimentation of mudrock from a new perspective(astronomical forcing)and provides a direction for the research on sedimentation of fine-grained sedimentary rocks.

A Locating Method for Reliability-Critical Gates with a Parallel-Structured Genetic Algorithm

The reliability allowance of circuits tends to decrease with the increase of circuit integration and the application of new technology and materials, and the hardening strategy oriented toward gates is an effective technology for improving the circuit reliability of the current situations. Therefore, a parallel-structured genetic algorithm (GA), PGA, is proposed in this paper to locate reliability-critical gates to successfully perform targeted hardening. Firstly, we design a binary coding method for reliability-critical gates and build an ordered initial population consisting of dominant individuals to improve the quality of the initial population. Secondly, we construct an embedded parallel operation loop for directional crossover and directional mutation to compensate for the deficiency of the poor local search of the GA. Thirdly, for combination with a diversity protection strategy for the population, we design an elitism retention based selection method to boost the convergence speed and avoid being trapped by a local optimum. Finally, we present an ordered identification method oriented toward reliability-critical gates using a scoring mechanism to retain the potential optimal solutions in each round to improve the robustness of the proposed locating method. The simulation results on benchmark circuits show that the proposed method PGA is an efficient locating method for reliability-critical gates in terms of accuracy and convergence speed.

Objective To prepare the live recombinant vaccine of attenuated Salmonella typhimurium SL3261 expressing Helicobacterpylori （H. pylori） B subunit （UreB） and to determine whether it could be used as an oral vaccine against H. pylori infection. Methods Using genomic DNA of H. pylori Sydney strain （SS1） as template, the H. pylori UreB gene fragment was amplified by PCR and subcloned into the expression vector pTC01. The recombinant plasmid pTC01-UreB was then transferred into LBS000 to obtain modified forms, and further conversed into the attenuated Salmonella typhimurium SL3261 to obtain recombinant SL3261/pCT01-UreB as an oral immunization reagent, which was then used to orally immunize Balb/c mice twice at a three-week interval. Twelve weeks later, anti-UreB IgA antibodies in intestinal fluid and IgG antibodies in sera were determined by ELISA. The relating data in control groups （including body weight, gastric inflammation, etc.） were also collected. Results The sequencing analysis showed that the UreB gene fragment amplified by PCR was consistent with the sequence of the H. pylori UreB gene. The restriction enzyme digestion revealed that the correct pTC01-UreB was obtained. SDS-PAGE and Western blot showed that a 61KD protein was expressed in SL3261/pTC01-UreB, which could be recognized by anti-H, pylori UreB antiserum and was absent in the control containing only Salmonella typhimurium SL3261 strain. The multiple oral immunization with SL3261/pTC01-UreB could significantly induce H. pylori specific mucosal IgA response as well as serum IgG responses. IFN-T and IL-10 levels were significantly increased in SL3261/pTC01-UreB group, and no obvious side effect and change in gastric inflammation were observed. Conclusion The attenuated vaccine of Salmonella typhimurium expressing H. pylori UreB can be used as an oral vaccine against H. pylori infection.

Silicon is one of the main gangue components in iron ore,usually in the form of quartz and olivine.Numerous studies have shown that SiO_(2) has a two-sided effect on the consolidation of pellets during high-temperature oxidation roasting of magnetite.However,it is very difficult to capture the structural evolution and migration mechanisms during high-tem-perature roasting process by existing experimental methods.Therefore,the influence of SiO_(2) on the consolidation behavior of magnetite was studied through a series of roasting experiments and molecular dynamic simulation.The results show that the consolidation index and particle growth index decrease with the increase in SiO_(2) content in the particles.At 1573 K,the liquid phase promotes the recrystallization growth of hematite at high temperature.Molecular dynamic study shows that it is difficult for quartz SiO_(2) to form sintering neck with Fe_(2)O_(3).When the calcination temperature is higher than 1400 K,the Fe_(2)O_(3)-Fe_(2)SiO_(4) system produces a considerable sintering neck structure after relaxation.The atomic migration ability of Fe_(2)SiO_(4) is much higher than that of Fe_(2)O_(3).The higher atomic migration ability of Fe_(2)SiO_(4) is the main reason for the formation of the sintering neck.

At high temperature,the chemical reaction mechanism at the interface of magnetite and the influence mechanism of gangue element Al on the oxidation performance of magnetite are not clear.In addition,due to the limitation of existing experimental equipment,it is difficult to clarify the interface reaction mechanism in the oxidation process and the influence mechanism of Al on the surface oxidation reaction of Fe_(3)O_(4) at an atomic scale at high temperature.The surface oxidation reaction mechanism of magnetite and the influence mechanism of Al on the oxidation of magnetite were studied by experiments and AIMD(ab initio molecular dynamics).The experimental results show that the existence of Al_(2)O_(3) will reduce the initial oxidation temperature and comprehensive oxidation performance of magnetite.The AIMD results show that aluminum can accelerate the interface reaction rate of Fe_(3)O_(4) in the high-temperature oxidation atmosphere,but the strong Al-O binding ability is not conducive to the transfer of O atoms in the oxidation process.

Liliangyou 3822 is a novel indica hybrid rice variety that exhibits disease resistance,high yield,lodging resistance,and late maturity.It employs a self-selected two-line sterile line,Li 38S,and a self-selected restorer line,R22.This variety was subjected to a regional test of indica late-maturing groups in the middle and lower reaches of the Yangtze River in 2020.The results demonstrated that the average yield of the variety was 9.95 t/hm 2,which was 10.67%higher than that of the control Fengliangyou 4,indicating a highly significant yield increase.In the continuous test in 2021,the average yield was 9.74 t/hm 2,representing a 6.52%increase over the control,which also exhibited a significant increase.Finally,the average yield of the two years regional test was 9.84 t/hm 2,which was 8.58%higher than that of the control.In the 2021 production test,the average yield of the variety was 9.32 t/hm 2,which was 12.19%higher than that of the control,indicating a remarkably significant yield increase.In 2022,the variety was validated by the National Crop Variety Approval Committee(GSD 20220143).

Do nocturnal birds use acoustic and visual cues to avoid predation by bats?

Anti-predation strategies are critical to animal survival and are fundamental to deciphering predator–prey interactions.As an important defense strategy,sensory predator detection(such as through acoustic and visual cues)enables animals to assess predation risk and execute predator-avoidance behavior;however,there are limited studies on the anti-predation behavior of nocturnal animals.The prey of bats provides an excellent representative system for examining the anti-predation behavior of nocturnal animals.Here,we broadcasted different types of echolocation calls of the bird-eating bat Ia io to two wild passerine birds,namely,Zosterops japonicus and Sinosuthora webbiana,that are preyed upon by I.io,and presented the birds with individual bats under different light intensities.The results showed that both bird species were able to perceive the low-frequency audible portion of the bats’echolocation calls;however,they did not exhibit escape responses to the acoustic stimuli.In the dark and under moonlit conditions,both bird species were unable to respond to active bats at close range and the birds only exhibited evasive flight behavior when bats approached or touched them.These results suggest that nocturnal passerine birds may not be able to use acoustic or visual cues to detect bats and adopt evasive maneuvers to avoid predation.This work suggests that bat predation pressure may not elicit primary predator-avoidance responses in nocturnal passerine birds.The results provide new insights into the anti-predation behavior of nocturnal animals.

Background:The maintenance dosage of selexipag is categorized as low,medium or high.In order to assess the efficacy and safety of different dosages of selexipag for the risk stratification of pulmonary arterial hypertension(PAH),we performed a sys-tematic review and meta-analysis.Methods:Studies assessing PAH risk stratification indices,such as the World Health Organization functional class(WHO-FC),six-minute walk distance(6MWD),N-terminal pro-B-type natriuretic peptide(NT-proBNP)level,right atrial pressure(RAP),cardiac index(CI)and mixed venous oxygen saturation(SvO2),were included.Results:Thirteen studies were included.Selexipag led to improvements in the 6MWD(MD:24.20 m,95%CI:10.74-37.67),NT-proBNP(SMD:-0.41,95%CI:-0.79-0.04),CI(MD:0.47 L/min/m^(2),95%CI:0.17-0.77)and WHO-FC(OR:0.564,95%CI:0.457-0.697).Subgroup analysis demonstrated that all three dosages improved the 6MWD.A moderate dosage led to improvements in the CI(MD:0.30 L/min/m^(2),95%CI:0.15-0.46)and WHO-FC(OR:0.589,95%CI:0.376-0.922).Within 6 months of treatment,only the WHO-FC and CI were significantly improved(OR:0.614,95%CI:0.380-0.993;MD:0.30 L/min/m^(2),95%CI:0.16-0.45,respectively).More than 6 months of treatment significantly improved the 6MWD,WHO-FC and NT-proBNP(MD:40.87 m,95%CI:10.97-70.77;OR:0.557,95%CI:0.440-0.705;SMD:-0.61,95%CI:-1.17-0.05,respectively).Conclusions:Low,medium,and high dosages of selexipag all exhibited good effects.When treatment lasted for more than 6 months,selexipag exerted obvious effects,even in the low-dosage group.This finding is important for guiding individualized treatments.
