Down-regulation of p210bcr/ablby curcumin involves disrupting molecular chaperone functions of Hsp90

Aim:To investigate the effects of curcumin（Cur）on p210ber/abllevel in K562 cells,and the relationship between these effects and the molecular chaperone functionsof heat shock protein 90（Hsp90）.Methods:Flow cytometry and Western blotwere used to examine the abundance of p210bcr/abl,Hsp90,p23,Hsp70,and p60Hopin K562 cells treated with *** transcription polymerase chain reaction（RT-PCR）was used to determine the bcr-abl mRNA level in K562 cells treated *** co-immunoprecipitation of p210bcr/abland its molecular chaperones,theimmunoprecipitate was then subjected to Western blot analysis with anti-Hsp90,anti-Hsp70,anti-p23,and ***:An exposure of K562 cells toCur produced time-dependent down-regulation of p210bcr/abl,the inhibition rate ofp210bcr/ablin K562 cells determined by flow cytometry after treatment with Cur 27.2μmol/L for 1h,6h,12h and 24 h was 31.2%,63.7%,81.3% and 94.5%,*** contrast,Cur had almost no influence on bcr-abl mRNA *** withCur for 24 h reduced the association of p210bcr/ablwith Hsp90/p23 complex,whileincreasing the association of p210bcr/ablwith Hsp70/p60Hopcomplex;however,thetotal protein abundance of Hsp90,p23,and p60Hopin K562 cells had no apparentchange,while Hsp70 increased ***:Down-regulation ofp210bcr/ablby Cur involves dissociating the binding of p210bcr/ablwith Hspg0/*** contrast,the association of p210bcr/ablwith HspT0/p60Hopcomplexincreased.

TRIM21-dependent ultrasmall chiral gold nanoparticles for preventing microglia senescence against Alzheimer's disease

Tripartite motif 21(TRIM21)is an E3 ubiquitin ligase that shows great promise for protein degradation through ***,ultrasmall chiral gold nanoparticles(D-or L-NPs)modified by D-or L-glutathione ligands were *** conjugated with an NLR family pyrin domain-containing protein 3(NLRP3)antibody(D-NP-a NLRP3 or L-NP-a NLRP3),DNP-a NLRP3 showed effective delivery efficiency of the antibody into microglia and prevented Aβ-mediated microglia senescence,and p16^(ink4a),a marker of senescence,in the microglia was reduced by 90.3%±7.8% while L-NP-a NLRP3 decreased by 48.01%±3.1%.Mechanistic investigations revealed that the D-NP-a NLRP3((1.5±0.3)×10^(7)M^(-1))exhibited sixteen-fold larger binding affinity to transmembrane glycoprotein SLC3A2 than L-type((9.5±2.7)×10^(5)M^(-1)),which led to a high efficiency of antibody delivery and TRIM21-dependent NLRP3 ***,the blood-brain barrier(BBB)-crossing ability of chiral NP-a NLRP3 as well as NLRP3 degradation was demonstrated in *** APP/PS1 Alzheimer's disease(AD)model mice experiments exhibited a reduction of 89.7%±6.8% for the NLRP3 protein and 84.2±7.5% for p16^(ink4a)following the intravenous administration of D-NP-a NLRP3 once a week for 60 ***,the levels of the AD markers Aβ and phosphorylatedTau in the brains were reduced by 86.2%±8.2% and 81.6%±9.1%;these were 2.1-fold and 1.9-fold higher than those treated with L-NP-a NLRP3,*** studies provide a method to rescue AD-like pathologies and prevent senescence.

Small RNA(sRNA)-mediated RNA silencing(also known as RNA interference,or RNAi)is a conserved mechanism in eukaryotes that includes RNA degradation,DNA methylation,heterochromatin formation and protein translation *** plants,sRNAs can move either cell-to-cell or systemically,thereby acting as mobile silencing signals to trigger noncell autonomous ***,whether and what proteins are also involved in noncell autonomous silencing have not been *** this study,we utilized a previously reported inducible RNAi plant,PDSi,which can induce systemic silencing of the endogenous PDS gene,and we demonstrated that DCL3 is involved in systemic PDS silencing through its RNA binding *** confirmed that the C-terminus of DCL3,including the predicted RNA-binding domain,is capable of binding short *** affecting RNA binding,but not processing activity,reduced systemic PDS silencing,indicating that DCL3 binding to RNAs is required for the induction of systemic *** mosaic virus infection assays showed that the RNA-binding activity of DCL3 is required for antiviral RNAi in systemically noninoculated *** findings demonstrate that DCL3 acts as a signaling agent involved in noncell autonomous silencing and an antiviral effect in addition to its previously known function in the generation of 24-nucleotide sRNAs.

To ensure the safe performance of deep-sea mining vehicles(DSMVs),it is necessary to study the mechanical characteristics of the interaction between the seabed soil and the track *** rotation and digging motions of the track plate are important links in the contact between the driving mechanism of the DSMV and seabed *** this study,a numerical simulation is conducted using the coupled Eulerian–Lagrangian(CEL)large deformation numerical method to investigate the interaction between the track plate of the DSMV and the seabed soil under two working conditions:rotating condition and digging ***,a soil numerical model is established based on the elastoplastic mechanical characterization using the basic physical and mechanical properties of the seabed soil obtained by in situ ***,the soil disturbance mechanism and the dynamic mechanical response of the track plate under rotating and digging conditions are obtained through the analysis of the sensitivity of the motion parameters,the grouser structure,the layered soil features and the soil *** results indicate that the above parameters remarkably influence the interaction between the DSMV and the seabed ***,it is important to consider the rotating and digging motion of the DSMV in practical engineering to develop a detailed optimization design of the track plate.

Exact Box-Constrained Economic Operating Region for Power Grids Considering Renewable Energy Sources

The growing integration of renewable energy sources manifests as an effective strategy for reducing carbon emissions. This paper strives to efficiently approximate the set of optimal scheduling plans(OSPs) to enhance the performance of the steady-state adaptive cruise method(SACM) of power grid, improving the ability of dealing with operational uncertainties. Initially, we provide a mathematical definition of the exact boxconstrained economic operating region(EBC-EOR) for the power grid and its dispatchable components. Following this, we introduce an EBC-EOR formulation algorithm and the corresponding bi-level optimization models designed to explore the economic operating boundaries. In addition, we propose an enhanced big-M method to expedite the computation of the EBCEOR. Finally, the effectiveness of the EBC-EOR formulation, its economic attributes, correlation with the scheduling plan underpinned by model predictive control, and the significant improvement in computational efficiency(over twelvefold) are verified through case studies conducted on two test systems..
