CD36 is a highly glycosylated integral membrane protein that belongs to the scavenger receptor class B family and regulates the pathological progress of metabolic ***36 was recently found to be widely expressed in various cell types in the nervous system,including endothelial cells,pericytes,astrocytes,and ***36 mediates a number of regulatory processes,such as endothelial dysfunction,oxidative stress,mitochondrial dysfunction,and inflammatory responses,which are involved in many central nervous system diseases,such as stroke,Alzheimer’s disease,Parkinson’s disease,and spinal cord ***36 antagonists can suppress CD36 expression or prevent CD36 binding to its ligand,thereby achieving inhibition of CD36-mediated pathways or ***,we reviewed the mechanisms of action of CD36 antagonists,such as Salvianolic acid B,tanshinone IIA,curcumin,sulfosuccinimidyl oleate,antioxidants,and small-molecule ***,we predicted the structures of binding sites between CD36 and *** sites can provide targets for more efficient and safer CD36 antagonists for the treatment of central nervous system diseases.

The nature of electron density enhancement over a wide altitude range during ionosphere heating experiments at EISCAT

During the course of ionospheric heating experiments, researchers at the European Incoherent Scatter Scientific Association (EISCAT) observed an apparent electron density enhancement. The enhancement extended over a wide range of altitudes, above the reflection altitude of the high-frequency pump wave. However, whether this enhancement actually corresponds to a true enhancement in electron density remains an open question. When the dispersion relation of ion acoustic waves is followed, the frequency ratio of the enhanced ion line to the background ion line suggests that the profile of the effective ion mass may have remained unchanged. Furthermore, the solar radio flux and ion drift velocity indicate no significant changes in the ion species and their densities. In conclusion, the electron density enhancement observed at EISCAT should not, in fact, be considered a true enhancement.

BACKGROUND Accumulating evidence has shown that adipose tissue-derived mesenchymal stem cells(ADSCs)are an effective therapeutic approach for managing coronavirus disease 2019(COVID-19);however,further elucidation is required to determine their underlying immunomodulatory effect on the mRNA expression of T helper cell-related transcription factors(TFs)and cytokine release in peripheral blood mononuclear cells(PBMCs).AIM To investigate the impact of ADSCs on the mRNA expression of TFs and cytokine release in PBMCs from colorectal cancer(CRC)patients with severe COVID-19(CRC^(+)patients).METHODS PBMCs from CRC^(+)patients(PBMCs-C+)and age-matched CRC patients(PBMCs-C)were stimulated and cultured in the presence/absence of *** mRNA levels of T-box TF TBX21(T-bet),GATA binding protein 3(GATA-3),RAR-related orphan receptor C(RORC),and forkhead box P3(FoxP3)in the PBMCs were determined by reverse transcriptase-polymerase chain *** supernatants were evaluated for levels of interferon gamma(IFN-γ),interleukin 4(IL-4),IL-17A,and transforming growth factor beta 1(TGF-β1)using an enzyme-linked immunosorbent *** Compared with PBMCs-C,PBMCs-C+exhibited higher mRNA levels of T-bet and RORC,and increased levels of IFN-γ and ***,a significant decrease in FoxP3 mRNA and TGF-β1,as well as an increase in Tbet/GATA-3,RORC/FoxP3,IFN-γ/IL-4,and IL-17A/TGF-β1 ratios were observed in PBMCs-C+.Furthermore,ADSCs significantly induced a functional regulatory T cell(Treg)subset,as evidenced by an increase in FoxP3 mRNA and TGF-β1 release *** was accompanied by a significant decrease in the mRNA levels of T-bet and RORC,release of IFN-γ and IL-17A,and T-bet/GATA-3,RORC/FoxP3,IFN-γ/IL-4,and IL-17A/TGF-β1 ratios,compared with the PBMCs-C+*** The present in vitro studies showed that ADSCs contributed to the immunosuppressive effects on PBMCs-C+,favoring Treg ***,ADSC-based cell therapy could be a beneficial approach for p

Neuroprotective effect of AG490 in experimental traumatic brain injury of rats

Background Traumatic brain injury （TBI） is a major cause of death and disability in children and young adults worldwide. Therefore, we investigated the role of AG490 in regulating brain oedema, expression of CD40 and neurological function after TBI. Methods Sprague Dawley rats （n=240） were randomly divided into a sham operation group, TBl＋saline group and TBI＋AG490 （JAK/STAT inhibitor） group, Members of each group were euthanized at 6, 12, 24 or 72 hours after injury. Neurological severity score （NSS） was used to evaluate the severity of neurological damage. Brain water was quantitated by wet/dry weight method. The expression of CD40 was assessed by flow cytometry. Results In both the TBl＋saline group and the TBI＋AG490 group, the brain water content was elevated after TBI, reached a peak at 24-hour and remained high for the rest of the pedod investigated; the expression of CD40 reached a peak 24 hours after TBI; the NSS was elevated after TBI and then decreased after 6 hours. Elevations in the Level of CD40, degree of brain edema and NSS after TBI were significantly reduced in TBI＋AG490 group. Conclusion Inhibition of the JAK/STAT signalling pathway reduces brain oedema, decreases the expression of CD40 and exerts neuroprotective effects after TBI.

BACKGROUND Pulmonary lymphoepithelioma-like carcinoma(PLELC)is a rare type of nonsmall-cell lung *** lymphoepithelioma-like carcinoma(LELC)metastasis secondary to PLELC has not been reported *** SUMMARY A 64-year-old female was admitted to our hospital for a regular gastroscopy examination with a 6-year history of surgical resection for left *** emission tomography/computed tomography suggested high accumulation of 18F-fludeoxyglucose in the gastric cardia *** gastrointestinal endoscopy confirmed a large mass at the stomach ***(IHC)of the biopsy suggested metastatic stomach *** gastrectomy showed that this 6.5 cm×5.0 cm mass was located in the stomach fundus near the *** examination showed a poorly differentiated carcinoma with prominent lymphoplasmacytic *** demonstrated that the tumor was positive for CK(AE1/AE3),p63,p40,p53,Ki-67(70%),and EGFR(3+)and negative for CK7,CK20,Her2,and *** situ hybridization analysis showed positive staining Epstein-Barr virus-encoded *** programmed cell death ligand 1(PD-L1)expression score was 98%,and the combined positive score was 100,with no evidence of microsatellite ***,the patient was unequivocally diagnosed with metastatic stomach LELC secondary to pulmonary *** discharge,this patient underwent PD-1 inhibitor treatment(toripalimab,240 mg)every 3 wk for ten cycles,and she has had no tumor *** For gastric LELC metastasis,PD-1 inhibitor therapy could become a new therapeutic approach,though there is still no evidence from large data sets to support this.

To evaluate the effects of dietary supplementation with succinic acid on growth performance,flesh quality,glucose,and lipid metabolism of Nile tilapia(Oreochromis niloticus)fed a high-carbohydrate diet(HCD),five iso-nitrogenous and iso-lipidic diets were prepared as follows:HCD(control group)consisting of 55%corn starch and HCD supplemented with 0.5%,1.0%,2.0%,and 4.0%succinic acid,*** with an initial body weight of 204.90±1.23 g randomly assigned to 15 tanks with 3 replicates per group and 10 fish per tank fed for 8 *** dietary succinic acid supplementation resulted in significant second-order polynomial relationship in the weight gain rate(WGR),specific growth rate(SGR),feed conversion ratio(FCR),protein efficiency rate(PER),viscerosomatic index,condition factor,and contents of muscular crude lipid and glycogen(P<0.05).The hepatosomatic index,mesenteric fat index,liver glycogen content and crude lipid contents of the whole-body and liver demonstrated significantly linear and second-order polynomial relationship(P<0.05).Quadratic curve model analysis based on WGR,SGR,PER,and FCR demonstrated that optimal supplementation with succinic acid in the HCD of Nile tilapia ranged from 1.83%to 2.43%.Fish fed with 1.0%succinic acid had higher muscular hardness,increased the contents of alkali-soluble hydroxyproline in collagen,docosahexaenoic acid(DHA)and n-3 polyunsaturated fatty acid(n-3PUFA)in muscle,and lower total fatty acid content in muscle(P<0.05)compared with the control *** to the control group,dietary supplementation with 1.0%succinic acid significantly increased the contents of total bounding amino acid(arginine,histidine,isoleucine,lysine,methionine,alanine,proline),total flavor amino acid(free aspartic acid),the catalase(CAT)activity and total antioxidant capacity,and the mRNA relative expression levels of CAT,superoxide dismutase(SOD),and nuclearfactor erythroidderived 2-like 2(Nrf2)in muscle(P<0.05).Furthermore,succini

Natural fractures are critical for shale oil and gas enrichment and development. Due to the extremely high heterogeneity of shale, the factors controlling the formation of internal fractures, especially horizontal fractures, remain controversial. In this study, we integrate thin section analysis and microcomputed tomography(CT) data from several lacustrine shale samples from the third member(Es3) of the Shahejie Formation, Qikou Sag, Bohai Bay Basin, to assess the fractures in detail. The goal is to reveal the development characteristics, controlling factors, and geological significance for evaluating sweet spots in a shale oil play. The fractures in the Es3contain high-angle structural and horizontal bed-parallel fractures that are mostly shear and extensional. Various factors influence fracture development,including lithofacies, mineral composition, organic matter content, and the number of laminae. Structural fractures occur predominantly in siltstone, whereas bed-parallel fractures are abundant in laminated shale and layered mudstone. A higher quartz content results in higher shale brittleness, causing fractures, whereas the transformation between clay minerals contributes to the development of bedparallel fractures. Excess pore pressure due to hydrocarbon generation and expulsion during thermal advance can cause the formation of bed-parallel fractures. The density of the bed-parallel and structural fractures increases with the lamina density, and the bed-parallel fractures are more sensitive to the number of laminae. The fractures are critical storage spaces and flow conduits and are indicative of sweet spots. The laminated shale in the Es3with a high organic matter content contains natural fractures and is an organic-rich, liquid-rich, self-sourced shale play. Conversely, the siltstone, massive mudstone, and argillaceous carbonate lithofacies contain lower amounts of organic matter and do not have bed-parallel fractures.
