AIM: To investigate the neuroprotective effect of tubuloside B, one of the phenylethanoids isolated from the stems of Cistanche salsa, on tumor necrosis factor-alpha (TNFα)-induced apoptosis in SH-SY5Y neuronal cells. METHODS: Cell viability was analyzed using MTT assay. Apoptotic cells were detected using Hoechst3.3.2 staining, and confirmed by DNA fragmentation and flow cytometric analysis. The activity of caspase-3.was measured with special assay kit. The concentration of free intracellular calcium was determined with the probe Indo-1 by spectrometer. The level of intracellular reactive oxygen species and the potential of mitochondrial membrane were determined by laser scanning confocal microscopy (LSCM) combined with fluorescence probe H2DCFDA or JC-1 respectively. RESULTS: SH-SY5Y cells treated with TNFa 100 μg/L for 3. h showed typical morphological changes of apoptosis. DNA ladder could be observed by agarose gel electrophoresis. Here reported is the crystal structure of taxinine A (C_(26)H_(3.)O_8, Mr=476.76) which was isolated from the needles of Taxus cuspidata. The crystal belongs to monoclinic system, space group is P2_1 with a=9. 122 (2), b=10.451 (3., c=13.661(2),β=93. 92(1)°,Z=2, V=1299.3.5), D_x=1.218 g/cm￣3. F (000)=512,μ=0.08 mm-1, and the final R-factors are 0. 061 (Rf) and 0. 056(Rw) for 1796 observed diffractions. The molecule taxinine A is one of the taxane diterpenoids which possesses 6-8-6 fused tricycle skeleton.

Current strategies for drug discovery research mainly focus on molecular targets,in which structure biology plays a central role,such as the detailed mechanism of a drug-target *** principle of this approach is the presumed molecular mechanism or genetic background of the disease being *** this approach has brought progress in understanding the mode of action for some drugs,most prevalent illnesses-especially chronic diseases-are multifactorial and present with a group of symptoms,as is often the case in cardiovascular diseases,metabolic disorders,cancers,and so *** newly emerging term“multi-morbidity”(i.e.,multiple co-occurring diseases)describes a grand challenge for this strategy,and successful drug intervention for multimorbidity should be multifaceted[1].So,our research team has shifted its attention to a new direction that views the disease as a whole instead of a single-molecule abnormality,and focusing on drugs with multiple targets.

＜正＞Objective To study the amelioration effect of glucocorticoid receptor（GR） antagonist mifepristone on the changes of learning and memory abilities in rat model of depression and explore mechanisms *** In the present study,we used a 3.-d rat chronic unpredictable stress（CUS） model to observe both depression-like behaviors in sucrose preference test（SPT） and open-field test（OFT） and learning and memory-associated behaviors in Morris water maze test.A total of 3. male adult Sprague-Dawley rats were randomly assigned to three groups of equal size:control group（CON）;CUS group（CUS）;CUS + mifepristone group（CM）.Animals in CM group were first exposed to CUS for 14 d,and then were administered with 50 mg/kg per day of mifepristone with continued CUS *** EIA Kit was used to detect the concentration of plasma corticosterone（CORT）.Nissl staining was used to observe the structure of *** The results demonstrate that CUS exposure induces both depressive-like and learning and memory-associated behaviors and that these deficits are reversed by *** to CON group,the concentration of plasma CORT increased significantly in CUS group. We also found that CUS exposure damaged the structure of hippocampus,whereas mifepristone had an amelioration *** The results indicate that the structural deficits of hippocampus resulting from long-term stress exposure,which could contribute to the impairment of learning and memory in depression,are reversed by the GR receptor antagonist mifepristone.

OBJECTIVES To evaluate the predictive value of fasting plasma glucose(FPG)for in-hospital mortality in patients with acute myocardial infarction(AMI)with different glucose metabolism *** We selected 5,3.8 participants with AMI from the prospective,nationwide,multicenter CAMI registry,of which 2,081 were diabetic and 3.227 were *** were divided into high FPG and low FPG groups according to the optim-al cutoff values of FPG to predict in-hospital mortality for diabetic and nondiabetic cohorts,*** primary endpoint was in-hospital *** Overall,94 diabetic patients(4.5%)and 13. nondiabetic patients(4.1%)died during hospitalization,and the optimal FPG thresholds for predicting in-hospital death of the two cohorts were 13.2 mmol/L and 6.4 mmol/L,*** with individuals who had low FPG,those with high FPG were significantly associated with higher in-hospital mortality in diabet-ic cohort(10.1%vs.2.8%;odds ratio[OR]=3.862,95%confidence interval[CI]:2.542-5.869)and nondiabetic cohort(7.4%vs.1.7%;HR=4.542,95%CI:3.041-6.782).After adjusting the potential confounders,this significant association was not ***-more,FPG as a continuous variable was positively associated with in-hospital mortality in single-variable and multivariable models regardless of diabetic *** FPG to the original model showed a significant improvement in C-statistic and net reclassification in diabetic and nondiabetic *** This large-scale registry indicated that there is a strong positive association between FPG and in-hospital mor-tality in AMI patients with and without *** might be useful to stratify patients with AMI.
