Microsatellite alterations in phenotypically normal esophageal squamous epithelium and metaplasia-dysplasia-adenocarcinoma sequence

AIM: To investigate the microsatellite alterations in phenotypically normal esophageal squamous epithelium and metaplasia-dysplasia-adenocarcinoma sequence. METHODS: Forty-one specimens were obtained from esophageal cancer (EC) patients. Histopathological assessment identified 23 squamous cell carcinomas (SCC) and 18 adenocarcinomas (ADC), including only 8 ADC with Barrett esophageal columnar epithelium (metaplasia) and dysplasia adjacent to ADC. Paraffin-embedded normal squamous epithelium, Barrett esophageal columnar epithelium (metaplasia), dysplasia and esophageal tumor tissues were dissected from the surrounding tissues under microscopic guidance. DNA was extracted using proteinase K digestion buffer, and DNA was diluted at 1:100, 1:1000, 1:5000, 1:10 000 and 1:50 000, respectively. Seven microsatellite markers (D2S123, D3S1616, D3S1300, D5S346, D17S787, D18S58 and BATRII loci) were used in this study. Un-dilution and dilution polymerase chainreactions (PCR) were performed, and microsatellite analysis was carried out. RESULTS: No statistically significant difference was found in microsatellite instability (MSI) and loss of heterozygosity (LOH) of un-diluted DNA between SCC and ADC. The levels of MSI and LOH were high in the metaplasia-dysplasia-adenocarcinoma sequence of diluted DNA. The more the diluted DNA was, the higher the rates of MSI and LOH were at the above 7 loci, especially at D3S1616, D5S346, D2S123, D3S1300 and D18S58 loci. CONCLUSION: The sequence of metaplasia-dysplasia-adenocarcinoma is associated with microsatellite alterations, including MSI and LOH. The MSI and LOH may be the early genetic events during esophageal carcinogenesis, and genetic alterations at the D3S1616, D5S346 and D3S123 loci may play a role in the progress of microsatellite alterations.

BACKGROUND Proteomic signatures of Ming's infiltrative gastric cancer(IGC)remain *** To elucidate the molecular characteristics of IGC at the proteomics *** Twelve pairs of IGC and adjacent normal tissues were collected and their proteomes were analyzed by high performance liquid chromatography tandem mass *** identified peptides were sequenced de novo and matched against the SwissProt database using Maxquant *** differentially expressed proteins(DEPs)were screened using|log2(Fold change)|>1 and P-adj<0.01 as the *** expression levels of selected proteins were verified by Western *** interaction network of the DEPs was constructed with the STRING database and visualized using Cytoscape with cytoHubba *** DEPs were functionally annotated using clusterProfiler,STRING and DAVID for Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways.P<0.05 was considered statistically *** A total of 7361 DEPs were identified,of which 94 were significantly up-regulated and 223 were significantly down-regulated in IGC relative to normal gastric *** top 10 up-regulated proteins were MRTO4,BOP1,PES1,WDR12,BRIX1,NOP2,POLR1C,NOC2L,MYBBP1A and TSR1,and the top 10 down-regulated proteins were NDUFS8,NDUFS6,NDUFA8,NDUFA5,NDUFC2,NDUFB8,NDUFB5,NDUFB9,UQCRC2 and *** up-regulated proteins were enriched for 9 biological processes including DNA replication,ribosome biogenesis and initiation of DNA replication,and the cellular component MCM *** the down-regulated proteins,17 biological processes were enriched,including glucose metabolism,pyruvic acid metabolism and fatty acidβ-*** addition,the mitochondrial inner membrane,mitochondrial matrix and mitochondrial proton transport ATP synthase complex were among the 6 enriched cellular components,and 11 molecular functions including reduced nicotinamide adenine dinucleotide dehydrogenase activity,acyl-CoA dehydrogen

Ellagic acid inhibits gastric cancer cells by modulating oxidative stress and inducing apoptosis

Clinical heterogeneity and five phenotypes identified in pediatric Behçet’s syndrome: a cohort study from Shanghai Behçet’s syndrome database

Objectives Behçet’s syndrome(BS)is a rare disease of unknown etiology,with limited reports especially in pediatric *** clinical characteristics and phenotypes of pediatric BS as a highly heterogeneous variable vessel vasculitis were investigated in this *** A cross-sectional study was conducted to compare clinical variables and descriptive characteristics of BS by age of onset and *** analysis was then performed to identify the phenotypes of pediatric *** A total of 2082 BS patients were included in this study,1834 adults and 248 *** with adult-onset BS,pediatric BS had a higher incidence of folliculitis[relative risks(RR)and 95%confidence interval(CI)1.3(1.0–1.5)],uveitis of the left eye[RR and 95%CI 2.3(1.0–5.0)],intestinal ulcer complications[RR and 95%CI 2.1(1.1–4.2)],pericarditis[RR and 95%CI 2.5(1.0–6.2)],and psychiatric disorders[RR and 95%CI 2.8(1.0–7.9)],while the incidence of thrombocytopenia was lower[RR 0.2(0.1–1.0)].Among pediatric BS,females had more genital ulcers,while males were more likely to have skin lesions,panuveitis,vascular involvement,venous lesions,cardiac involvement,and aortic *** analysis classified pediatric BS into five clusters(C1–C5):C1(n=61,24.6%)showed gastrointestinal(GI)involvement;C2(n=44,17.7%)was the central nervous system(CNS)type where 23 cases overlapped joint involvement;in C3(n=35,14.1%),all patients presented with arthritis or arthralgia;all patients in C4(n=29,11.7%)manifested ocular involvement,with a few patients overlapping with GI involvement or joint damage;C5(n=79,31.9%)was the mucocutaneous type,presenting both oral ulcers,genital ulcers,and skin *** The clinical features of pediatric and adult BS differ *** and female pediatric BS also have a distinct *** phenotypes including GI,CNS,joint,ocular,and mucocutaneous types were identified for pediatric BS.

The gold-telluride connection:Evidence for multiple fluid pulses in the Jinqingding telluride-rich gold deposit of Jiaodong Peninsula,Eastern China

The mechanism of gold migration,enrichment,and precipitation in forming world-class gold deposits has been a topic of wide interest,particularly where these deposits are abundant in *** Jiaodong Peninsula in eastern China hosts some of the world-class gold deposits among which the Jinqingding deposit is one of the best examples with substantial telluride mineralization and thus provides opportunity to investigate the genetic connection between tellurium and gold *** orebody in this deposit is hosted in the NE-NNE-trending jiangjunshi-Quhezhuang fault with the Jurassic Kunyushan granitic pluton as wall *** deposit involved three mineralization stages as inferred from assemblages and crosscutting relationships between *** stages are:(I)pre-ore gold-poor quartz-pyrite veins,(II)main ore auriferous quartz-pyrite-Te/Bi-minerals±sphalerite±chalcopyrite±barite±marcasite veins,and(III)post-ore quartz-calcite *** present here the textural,isotopic,and geochemical variations of different stages/generations of pyrite based on scanning electron microscopy-energy dispersive spectroscopy(SEM-EDS),electron probe microanalysis(EPMA),and laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS).Pyrite in the Jinqingding deposit displays distinct ***1a shows a porous and dissolution-reprecipitation texture in the core,whereas Py1b exhibits a smooth and flat *** analysis reveals contrasting elemental compositions,with Py1a characterized by low Au(avg.0.14 ppm),As(avg.15.57 ppm),Ag(avg.17.71 ppm),and Te(avg.19.22 ppm)contents,but elevated Co(avg.1068.10 ppm)and Ni(224.12 ppm)concentrations,and variable sulfur isotopic composition(δ^(34)SV-CDT=9.54‰-12.12‰).Conversely,Py1b displays increased concentrations of these elements and a more concentratedδ^(34)SV-CDT value(11.99‰-12.23‰),possibly associated with weak coupled dissolution-reprecipitation(CDR)*** Stage II,pyrite can be further categorized

MIL-100(V) derived porous vanadium oxide/carbon microspheres with oxygen defects and intercalated water molecules as high-performance cathode for aqueous zinc ion battery

The development of aqueous zinc ion battery cathode materials with high capacity and high magnification is still a ***,porous vanadium oxide/carbon(p-VO_(x)@C,mainly VO_(2) with a small amount of V_(2)O_(3)) core/shell microspheres with oxygen vacancies are facilely fabricated by using a vanadium-based metal-organic framework(MIL-100(V)) as a sacrificial *** unique structure can improve the conductivity of the VO_(x),accelerate electrolyte diffusion,and suppress structural collapse during ***,H_(2)O molecules are introduced into the interlayer of VO_(x) through a highly efficient in-situ electrochemical activation process,facilitating the intercalation and diffusion of zinc *** the activation,an optimal sample exhibits a high specific capacity of 464.3 mA h g^(-1) at0.2 A g^(-1) and 395.2 mA h g^(-1) at 10 A g^(-1),indicating excellent rate ***,the optimal sample maintains a capacity retention of about 89.3% after 2500 cycles at 10 A g^(-1).Density functional theory calculation demonstrates that the presence of oxygen vacancies and intercalated water molecules can significantly reduce the diffusion barrier for zinc *** addition,it is proved that the storage of zinc ions in the cathode is achieved by reversible intercalation/extraction during the charge and discharge process through various ex-situ analysis *** work demonstrates that the p-VO_(x)@C has great potential for applications in aqueous ZIBs after electrochemical activation.
