Chlorophyll(Chl) biosynthesis is essential for photosynthesis and plant ***-tRNA reductase(GluTR) catalyzes glutamyl-tRNA into glutamate-1-semialdehyde(GSA) and initiates the chlorophyll *** though the main role of GluTR has been established,the effects caused by natural variations in its corresponding gene remain largely ***,we characterized a spontaneous mutant in paddy field with Chl biosynthesis deficiency,designated as *** intact thylakoid lamellar structure,the cbd1 plant showed light green leaves and reduced Chl and carotenoids(Cars) content significantly compared to the wild *** map-based gene cloning,the mutation was restricted within a 57-kb region on chromosome 10,in which an mPingA miniature inverted-repeat transposable element(MITE) inserted in the promoter region of OsHemA *** leaf color and the pigment contents in cbd1 were recovered in a complementation test,confirming OsHemA was responsible for the mutant *** was uniquely predicted to encode GluTR and its expression level was dramatically repressed in *** transformation in protoplasts demonstrated that GluTR localized in chloroplasts and a signal peptide exists in its N-terminus.A majority of Chl biosynthesis genes,except for POR and CHLG,were down-regulated synchronously by the repression of OsHemA,suggesting that an attenuation occurred in the Chl biosynthesis ***,we found major agronomic traits involved in rice yield were statistically unaffected,except for the number of full grains per panicle was increased in ***,OsHemA plays an essential role in Chl biosynthesis in rice and its weak allele can adjust leaf color and Chls content without compromise to rice yield.

Cholestasis is caused by the obstacle of bile formation or secretion and can develop into severe liver diseases. We previously reported the ethanol extract of Schisandra sphenanthera(Wuzhi tablet, WZ) can significantly protect against lithocholic acid(LCA)-induced intrahepatic cholestasis in mice, partially due to the activation of PXR pathway and promotion of liver ***, the effect of WZ on the bile acids profile and gut microbiome in cholestastic mice remain unknown. In this study, the effect of WZ against LCA-induced liver injury was evaluated and its effect on the bile acids metabolome and gut microbiome profiles in cholestastic mice was further investigated. Targeted metabolomics analysis was performed to examine the change of bile acids in the serum, liver, intestine and feces. The change of intestinal flora were detected by the genomics method. Targeted metabolomics analysis revealed that WZ enhanced the excretion of bile acids from serum and liver to intestine and feces. Genomics analysis of gut microbiome showed that WZ can reverse LCA-induced gut microbiome disorder to the normal level. In conclusion, WZ protects against LCAinduced cholestastic liver injury by reversing abnormal bile acids profiles and alteration of gut microbiome.

Effects of viral infection and microbial diversity on patients with sepsis:A retrospective study based on metagenomic next-generation sequencing

BACKGROUND: The study aims to investigate the performance of a metagenomic next-generationsequencing (NGS)-based diagnostic technique for the identifi cation of potential bacterial and viral infectionsand eff ects of concomitant viral infection on the survival rate of intensive care unit (ICU) sepsis ***: A total of 74 ICU patients with sepsis who were admitted to our institution from February1, 2018 to June 30, 2019 were enrolled. Separate blood samples were collected from patients for bloodcultures and metagenomic NGS when the patients’ body temperature was higher than 38 °C. Patients’demographic data, including gender, age, ICU duration, ICU scores, and laboratory results, were *** correlations between pathogen types and sepsis severity and survival rate were ***: NGS produced higher positive results (105 of 118;88.98%) than blood cultures(18 of 118;15.25%) over the whole study period. Concomitant viral infection correlated closelywith sepsis severity and had the negative effect on the survival of patients with sepsis. However,correlation analysis indicated that the bacterial variety did not correlate with the severity of ***: Concurrent viral load correlates closely with the severity of sepsis and thesurvival rate of the ICU sepsis patients. This suggests that prophylactic administration of antiviraldrugs combined with antibiotics may be benefi cial to ICU sepsis patients.

Genetic Analysis of Heading Date of Japonica Rice Cultivars in Southwest China

Heading date of 26 native japonica rice cultivars in southwest China was investigated,and their basic vegetative growth(BVG),photoperiod-sensitivity(PS) and temperature-sensitivity(TS) were analyzed under artificial short-day and natural long-day conditions in Nanjing,as well as artificial high-temperature and natural low-temperature conditions in winter in *** results showed that the PS and TS varied among different *** BVG of all the japonica cultivars was well situated,but differed within *** analysis showed a significant correlation between heading date and PS,indicating that PS was the main factor affecting heading date of japonica cultivars in southwest *** analysis was conducted on these 26 cultivars using a set of heading date near isogenic lines as test *** the japonica cultivars carried the dominant early-heading gene Ef-1 or Ef-1t,and most of these cultivars carried the dominant photoperiod sensitivity allele E1 or E1t,the PS of which was slightly weaker than *** the Se-1 locus,these cultivars mainly carried recessive photoperiod insensitivity gene *** addition,the PS of 22 japonica cultivars could be repressed or weakened by the recessive allele hd2,inhibiting the expression of E1 and *** results indicated that the genotypes of heading date determined different PS and well situated BVG in japonica rice cultivars in southwest China.

Laparoscopic vs open surgery in ileostomy reversal in Crohn’s disease:A retrospective study

BACKGROUND Although minimally invasive surgery is becoming more commonly applied for ileostomy reversal(IR),there have been relatively few studies of IR for patients with Crohn's disease(CD).It is therefore important to evaluate the potential benefits and risks of laparoscopy for patients with *** To compare the safety,feasibility,and short-term and long-term outcomes of laparoscopic IR(LIR)vs open IR(OIR)for the treatment of *** The baseline characteristics,operative data,and short-term(30-d)and long-term outcomes of patients with CD who underwent LIR and OIR at our institution between January 2017 and January 2020 were retrieved from an electronic database and retrospectively *** Of the 60 patients enrolled in this study,LIR was performed for 48 and OIR for *** were no statistically significant differences in baseline characteristics,operation time,intraoperative blood loss,days to flatus and soft diet,postoperative complications,hospitalization time,readmission rate within 30 d,length of hospitalization,hospitalization costs,or reoperation rate after IR between the two ***,patients in the LIR group more frequently required lysis of adhesions as compared to those in the OIR group(87.5%vs 41.7%,respectively,P<0.05).Notably,following exclusion of patients who underwent enterectomy plus IR,OIR was more advantageous in terms of postoperative recovery of gastrointestinal function and hospitalization *** The safety and feasibility of LIR for the treatment of CD are comparable to those of OIR with no increase in intraoperative or postoperative complications.

Background:Data from single-center experience or small sample-sized studies have shown that chromoendoscopy(CE)might be superior to white-light endoscopy(WLE)for dysplasia surveillance in ulcerative colitis(UC)*** performed a prospective randomized trial with a long-term follow-up to compare the detection rate of dysplasia among WLE with targeted biopsies(WLT),WLE with random biopsies(WLR),and dye-based CE with targeted biopsies(CET)in UC ***:Patients with long-standing UC were enrolled from 11 medical centers from March 2012 to December 2013 and randomized into three arms(WLT,WLR,and CET).Only high-definition endoscopy was used in all three *** patients were followed up by annual endoscopy with biopsies through December ***:With a median follow-up time of 55 months,a total of 122 patients with 447 colonoscopies were finally analysed in the per-protocol set:WLT(n=43),WLR(n=40),and CET(n=39).A total of 34 dysplastic lesions were found in 29 colonoscopies of 21 *** and CET could identify more colonoscopies that diagnosed dysplasia than WLT(8.1%and 9.7%vs 1.9%;P=0.014 and 0.004,respectively).WLR obtained more biopsied samples than WLT and CET(16.465.1 vs 4.361.4 and 4.361.4;both P3 years)follow-up was conducted for dysplasia *** trial was registered on ***(ChiCTR1900023689).

Existence for the Green-Lindsay Type Therm-elastic Problem of Nonhomogeneous Media

We study a new model named the Green-Lindsay type therm-elastic model for nonhomogeneous media that consists of a system of dynamic thermoelasticity equations of displacement and dynamic heat conduction equation. We construct the model based on the classical GL-model for homogeneous material. This system is coupled dynamic problem and the displacement field and heat field must be solved at the same time. By using Fadeo- Galerkin method, we proved that the problem we proposed exist unique weak solution under some regular assumption.

Evaluation of Rice Germplasm for Resistance to the Small Brown Planthopper (Laodelphax striatellus) and Analysis of Resistance Mechanism

One hundred and thirty-eight rice accessions were screened for resistance to the small brown planthopper （SBPH）（Laodelphax striatellus Fallen） by the modified seedbox screening test. Twenty-five rice accessions with different levels of resistance to SBPH were detected, accounting for 18.1% of the total accessions, which included 2 highly resistant, 9 resistant and 14 moderately resistant varieties. Compared with indica rice, japonica rice was more susceptible to SBPH. Antixenosis test, antibiosis test and correlation analysis were performed to elucidate the resistance mechanism. The resistant check Rathu Heenati （RHT）, highly resistant varieties Mudgo and Kasalath, and resistant variety IR36 expressed strong antixenosis and antibiosis against SBPH, indicating the close relationship between resistance level and these two resistance mechanisms in the four rice varieties. Antibiosis was the dominant resistance pattern in the resistant varieties Daorenqiao and Yangmaogu due to their high antibiosis but low antixenosis. Dular, ASD7 and Milyang 23 had relatively strong antixenosis and antibiosis, indicating the two resistance mechanisms were significant in these three varieties. The resistant DV85 expressed relatively high level of antixenosis but low antibiosis, whereas Zhaiyeqing 8 and Guiyigu conferred only moderate antibiosis and antixenosis to SBPH, suggesting tolerance in these three varieties. Antibiosis and antixenosis governed the resistance to SBPH in the moderately resistant accession 9311. Antixenosis was the main resistance type in V20A. Tolerance was considered to be an important resistance mechanism in Minghui 63 and Yangjing 9538 due to their poor antibiosis and antixenosis resistance. The above accessions with strong antibiosis or antixenosis were the ideal materials for the resistance breeding.
