Background:Xiaoxianxiong decoction is a classic formula in traditional Chinese medicine used to treat type 2 diabetes mellitus and proven to be *** the material basis and underlying mechanisms remain *** aim of the present study was to elucidate the potential effective material basis of Xiaoxianxiong decoction and molecular mechanism treating type 2 diabetes ***:The absorbed bioactive components were identified based on serum *** analysis were performed to obtain effect targets for docking verification with the absorbed prototypes to determine the potential effective material *** the above basis,network pharmacology was conducted to explore the molecular ***:76 compounds were identified of Xiaoxianxiong decoction and 61 absorbed bioactive compounds were ***/threonine kinase 1 and ALB were key targets acquired by network analysis for molecular ***,5 compounds were considered as the potential effective material basis,namely berberine,berberrubine,lariciresinol and gingerenone A,***,the mechanism mainly lies in the insulin signaling pathway,HIF-1 signaling pathway,PI3K-Akt signaling pathway,FoxO signaling pathway,AGE-RAGE signaling pathway in diabetic complications,phospholipase D signaling pathway to regulate blood glucose levels on target tissues as well as organs and exhibit anti-inflammatory,promote cell differentiation and cell growth,maintain oxygen homeostasis and affect the enzymes along with key ***:This integrated research strategy to investigate the treatment of Xiaoxianxiong decoction on type 2 diabetes mellitus provides valuable insights for further study and clinical practice of Xiaoxianxiong decoction.

Background and Aims:Protein phosphatase 2A(PP2A)is associated with many *** study aimed to clarify whether PPP2CA,which encodes the alpha isoform of the catalytic subunit of PP2A,plays a role in hepatocellular carcinoma(HCC)and to identify the potential underlying molecular ***:Based on bioinformatics,public databases and our in-house RNA-Seq database,we analyzed the clinical value and molecular mechanism of PPP2CA in ***:data were analyzed from 2,545 patients with HCC and 1,993 controls without HCC indexed in The Cancer Genome Atlas database,the Gene Expression Omnibus database and our in-house RNA-Seq ***2CA expression was significantly higher in HCC tissue than in non-cancerous tissues(standardized mean difference:0.69,95%confidence interval[CI]:0.50–0.89).PPP2CA expression was able to differentiate HCC from non-HCC,with an area under the summary receiver operator characteristic curve of 0.79(95%CI:0.75–0.83).Immunohistochemistry of tissue sections confirmed that PPP2CA protein was up-regulated in HCC *** PPP2CA expression in HCC patients was associated with shorter overall,progression-free and disease-free *** molecular pathways through which PPP2CA may be involved in HCC were determined using miRWalk 2.0 as well as analysis of Gene Ontology categories,Kyoto Encyclopedia of Genes and Genomes pathways,and protein-protein interaction ***:PPP2CA is up-regulated in HCC and higher expression correlates with worse ***2CA shows potential as a diagnostic marker for *** studies should examine whether PPP2CA contributes to HCC through the candidate microRNAs,pathways and hub genes identified in this study.

The aim of this study was to investigate epidemiological characteristics of co-infection of HIV and intestinal parasites in a HIV/AIDS highly endemic area in China.A cross-sectional survey was carried out in two villages where HIV/AIDS prevalence in residents was over 1%.Stool samples of all residents in the two targeted villages were examined for the presence of intestinal *** examination was performed for the HIV infection detection and anemia test.A questionnaire survey was carried out in all residents participating in the study.A total of 769 individuals were enrolled in the investigation,720 of whom were involved in stool examination of intestinal *** infection rates of parasites in the residents of the targeted villages were as follows:0.56%for Ascaris lumbricoides,4.03%for Hookworm,0.28%for Trichuris trichiura,0.42%for Clonorchis sinensis,21.39%for Blastocystis hominis,3.89%for Giardia lamblia,1.67%for Entamoeba spp.,4.44%for Cryptosporidium spp.,and no infection for Strongyloides *** overall infection rate of intestinal worms was 4.72%,intestinal parasite infection rate was 24.31%,the anemia prevalence rate was 34.68%,the co-infection rate of HIV and intestinal helminthes 2.17%,the co-infection rate of HIV and intestinal protozoa 28.26%,of which the co-infection rates of HIV and Blastocystis hominis,and HIV and Cryptosporidium *** 19.57%and 13.04%,respectively.A signiﬁcant difference of Cryptosporidium *** rate was found between HIV-positive group(13.04%)and HIV-negative group(4.70%)(P50%)and women(>39%).It was concluded that the infection rate of intestinal protozoa in the residents was higher than that of intestinal helminthes in the local setting,and a same pattern appeared in the co-infection rate of HIV and *** interestingﬁnding is that the infection rate of Cryptosporidium *** the HIV-positiv

Effects of Nickel Chloride on Histopathological Lesions and Oxidative damage in the Thymus

The purpose of this study was to observe the histopathological lesions and oxidative damage induced by dietary nickel chloride (NiCl2) in the thymus. A total of 280 one-day-old avian broilers were divided into four groups and fed on a corn-soybean basal diet as the control diet or the same basal diet supplemented with 300, 600, and 900 mg/kg of NiCl2 for 42 days. In the NiCl2-treated groups, the broiler weight and thymic relative weight were significantly (P P lightly decreased and loosely arranged in the medulla of thymus in the 600 mg/kg and 900 mg/kg groups. The activities of superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px), and the ability to inhibit hydroxy radical and glutathione (GSH) content were significantly (P P da content was higher. The above-mentioned results demonstrated that dietary NiCl2 in excess of 300 mg/kg could reduce the broiler weight and thymic relative weight, and cause histopathological lesions and oxidative damage in the thymus, which finally impaired the thymic function.

Dear Editor,Male infertility is a complex and multifactorial health problem with highly heterogeneous phenotypes,ranging from reduced sperm production to the complete absence of sperm in testis,and genetic factors play an important role in this problem.1 Patients with azoospermia have the highest risk of carrying genetic abnormalities,and this risk gradually decreases as sperm count increases.1 Due to the high throughput and low cost of next-generation sequencing(NGS)technology,many centers have used NGS to identify genetic defects associated with abnormal spermatogenesis.

Mechanism underlying carbon tetrachloride-inhibited protein synthesis in liver

AIM: To study the mechanism underlying carbon tetrachloride (CCl4)-induced alterations of protein synthesis in liver. METHODS: Male Sprague-dawley rats were given CCl4 (1 mL/100 g body weight) and 3H-leucine incorporation. Malondialdehyde (Mda) level in the liver, in vitro response of hepatocyte nuclei nucleotide triphosphatase (NTPase) to free radicals, and nuclear export of total mRNA with 3'-poly A+ were measured respectively. Survival response of HepG2 cells to CCl4 treatment was assessed by methyl thiazolyl tetrazolium. Km and Vmax values of nuclear envelope NTPase activity in liver of rats treated with CCl4 were assayed by a double-reciprocal plot. RESULTS: The protein synthesis was inhibited while the Mda level was signif icantly increased in liver of rats treated with CCl4. In addition, CCl4 decreased the NTPase binding capacity of nuclear envelope (Km value) in cultured HepG2 cells. Moreover, in vitro ferrous radicals from Fenton's system suppressed the NTPase activity of liver nuclear envelope in a dose-dependent manner. Down-regulation of the nuclear envelope NTPase activity indicated a lower energy provision for nucleocytoplasmic transport of mRNA molecules, an evidence in CCl4-treated HepG2 cells correspondingly supported by the nuclear sequestration of poly (A)+ mRNA molecules in morphological hybridization research. CONCLUSION: Inhibition of mRNA transport, suggestive of decreased NTPase activity of the nuclear envelope, may be involved in carbon tetrachloride-inhibited protein synthesis in liver.

Enhancement of electron–ion recombination rates at low energy range in the heavy ion storage ring CSRm

Recombination of Ar^(14+), Ar^(15+), Ca^(16+), and Ni^(19+) ions with electrons has been investigated at low energy range based on the merged-beam method at the main cooler storage ring CSRm in the Institute of Modern Physics, Lanzhou,China. For each ion, the absolute recombination rate coefficients have been measured with electron–ion collision energies from 0 meV to 1000 meV which include the radiative recombination(RR) and also dielectronic recombination(DR)processes. In order to interpret the measured results, RR cross sections were obtained from a modified version of the semiclassical Bethe and Salpeter formula for hydrogenic ions. DR cross sections were calculated by a relativistic configuration interaction method using the flexible atomic code(FAC) and AUTOSTRUCTURE code in this energy range. The calculated RR + DR rate coefficients show a good agreement with the measured value at the collision energy above 100 ***, large discrepancies have been found at low energy range especially below 10 meV, and the experimental results show a strong enhancement relative to the theoretical RR rate coefficients. For the electron–ion collision energy below 1 meV, it was found that the experimentally observed recombination rates are higher than the theoretically predicted and fitted rates by a factor of 1.5 to 3.9. The strong dependence of RR rate coefficient enhancement on the charge state of the ions has been found with the scaling rule of q^(3.0), reproducing the low-energy recombination enhancement effects found in other previous experiments.
