Heavy alcohol consumption results in alcoholic liver disease(ALD)with inadequate therapeutic options.Here,we first report the potential beneficial effects of ginsenoside Rk2(Rk2),a rare dehydroprotopanaxadiol saponin isolated from streamed ginseng,against alcoholic liver injury in mice.Chronic-plus-single-binge ethanol feeding caused severe liver injury,as manifested by significantly elevated serum aminotransferase levels,hepatic histological changes,increased lipid accumulation,oxidative stress,and inflammation in the liver.These deleterious effects were alleviated by the treatment with Rk2(5 and 30 mg/kg).Acting as an nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3(NLRP3)inhibitor,Rk2 ameliorates alcohol-induced liver inflammation by inhibiting NLRP3 inflammasome signaling in the liver.Meanwhile,the treatment with Rk2 alleviated the alcohol-induced intestinal barrier dysfunction via enhancing NLRP6 inflammasome in the intestine.Our findings indicate that Rk2 is a promising agent for the prevention and treatment of ALD and other NLPR3-driven diseases.

Background:Retinal diseases characterized with irreversible loss of retinal nerve cells,such as optic atrophy and retinal degeneration,are the main causes of blindness.Current treatments for these diseases are very limited.An emerging treatment strategy is to induce the reprogramming of Müller glial cells to generate new retinal nerve cells,which could potentially restore vision.Main text:Müller glial cells are the predominant glial cells in retinae and play multiple roles to maintain retinal homeostasis.In lower vertebrates,such as in zebrafish,Müller glial cells can undergo cell reprogramming to regenerate new retinal neurons in response to various damage factors,while in mammals,this ability is limited.Interestingly,with proper treatments,Müller glial cells can display the potential for regeneration of retinal neurons in mammalian retinae.Recent studies have revealed that dozens of genetic and epigenetic regulators play a vital role in inducing the reprogramming of Müller glial cells in vivo.This review summarizes these critical regulators for Müller glial cell reprogramming and highlights their differences between zebrafish and mammals.Conclusions:A number of factors have been identified as the important regulators in Müller glial cell reprogramming.The early response of Müller glial cells upon acute retinal injury,such as the regulation in the exit from quiescent state,the initiation of reactive gliosis,and the re-entry of cell cycle of Müller glial cells,displays significant difference between mouse and zebrafish,which may be mediated by the diverse regulation of Notch and TGFβ(transforming growth factor-β)isoforms and different chromatin accessibility.

Fatty acids(FAs),which were initially recognized as energy sources and essential building blocks of biomembranes,serve as the precursors of important signaling molecules.Tracing FA metabolism is essential to understanding the biochemical activity and role of FAs in physiological and pathological events.Inspired by the advances in click chemistry for protein enrichment,we herein established a click chemistry-based enrichment(CCBE)strategy for tracing the cellular metabolism of eicosapentaenoic acid(EPA,20:5 n-3)in neural cells.Terminal alkyne-labeled EPA(EPAA)used as a surrogate was incubated with N2a,mouse neuroblastoma cells,and alkyne-labeled metabolites(ALMs)were selectively captured by an azide-modified resin via a Cu(I)-catalyzed azide-alkyne cycloaddition reaction for enrichment.After removing unlabeled metabolites,ALMs containing a triazole moiety were cleaved from solid-phase resins and subjected to liquid chromatography mass spectrometry(LC-MS)analysis.The proposed CCBE strategy is highly selective for capturing and enriching alkyne-labeled metabolites from the complicated matrices.In addition,this method can overcome current detection limits by enhancing MS sensitivity of targets,improving the chromatographic separation of sn-position glycerophospholipid regioisomers,facilitating structural characterization of ALMs by a specific MS/MS fragmentation signature,and providing versatile fluorescence detection of ALMs for cellular distribution.This CCBE strategy might be expanded to trace the metabolism of other FAs,small molecules,or drugs.

This paper focuses on error density estimation in ultrahigh dimensional sparse linear model,where the error term may have a heavy-tailed distribution.First,an improved two-stage refitted crossvalidation method combined with some robust variable screening procedures such as RRCS and variable selection methods such as LAD-SCAD is used to obtain the submodel,and then the residual-based kernel density method is applied to estimate the error density through LAD regression.Under given conditions,the large sample properties of the estimator are also established.Especially,we explicitly give the relationship between the sparsity and the convergence rate of the kernel density estimator.The simulation results show that the proposed error density estimator has a good performance.A real data example is presented to illustrate our methods.

Copper sulfide(CuS)is a promising cathode for lithium-ion batteries(LIBs)due to its impeccable theoretical energy density(~1015 Wh·kg^(−1) and 4743 Wh·L^(−1)).However,it suffers from voltage decay leaded energy density loss and low energy efficiency,which hinders its application.In this work,with combined ex-situ/in-situ X-ray diffraction(XRD)and electrochemical analysis,we explore detailed degradation mechanisms.For the voltage decay,it is attributed to a spontaneous reaction between CuS cathode and copper current collector(Cu CC).This reaction leads to energy density loss and active materials degradation(CuS→Cu_(1.81)S).As for energy efficiency,CuS undergoes a series of phase transformations.The main phase transition processes are CuS→α-LiCuS→Li_(2−x)Cu_(x)S+Cu→Li_(2)S+Cu for discharge;Li_(2)S+Cu→Li_(2−x)Cu_(x)S→β-LiCuS→CuS for charge.Here,α-LiCuS,β-LiCuS,and Li_(2−x)CuxS are newly identified phases.These phase changes are driven by topotactic-reaction-related copper diffusion and rearrangement.This work demonstrates the significance of transition-metal diffusion in the intermediates formation and phase change in conversion-type materials.

Three-dimensional(3 D)frameworks have received much attention as an effective modification strategy for next-generation high-energy-density lithium metal batteries.However,the top-growth mode of lithium(Li)on the 3 D framework remains a tough challenge.To achieve a uniform bottom-up Li growth,a scheme involving Ag concentration gradient in 3 D PVDF framework(C-Ag/PVDF)is proposed.Ag nanoparticles with a concentration gradient induce an interface activity gradient in the 3 D framework,and this gradient feature is still maintained during the cycle.As a result,the C-Ag/PVDF framework delivers a long lifespan over 1800 h at a current density of 1 mA cm^(-2) with a capacity of 1 mAh cm^(-2),and shows an ultra-long life(>1300 h)even at a high current density of 4 mA cm^(-2) with a capacity of 4 mAh cm^(-2).The advantage of concentration gradient provides further insights into the optimal design of the 3 D framework for stable Li metal anode.

Biodiversity and distribution of benthic dinoflagellates in tropical Zhongsha Islands,South China Sea

Benthic dinoflagellates have attracted increasing attention in recent years because of their toxicity and ability to form extensive harmful algal blooms.Ostreopsis producing palytoxin and its analogs,Gambierdiscus producing ciguatera toxins,and Prorocentrum producing okadaic acid and dinophysis toxins,have been concerned as serious human poisonings.We explored the benthic dinoflagellate biodiversity and distribution characteristics of a series of tropical reefs in 20-40-m water depth in wet season in the Zhongsha Islands in South China Sea using morphological,phylogenetic,and cell counting methods.Results show that benthic dinoflagellates in the islands are rich in biodiversity and 15 species from genera Amphidinium,Coolia,Ostreopsis,and Prorocentrum were identifi ed:Amphidinium carterae,A.magnum,A.massartii,A.operculatum,Coolia canariensis,C.malayensis,C.palmyrensis,C.tropicalis,Ostreopsis cf.ovata,Prorocentrum concavum,P.cf.sculptile,P.emarginatum,P.hof fmannianum,P.lima,and P.rhathymum.Among them,A.magnum is reported for the first time in Chinese waters.The abundance of benthic dinoflagellates was relatively low at 88-4345 cells/100 cm^(2) on sediment and 10-91 cells/g on macroalgae.Prorocentrum and Amphidinium were the dominant and subdominant genera,respectively.It is speculated that the low abundance of benthic dinoflagellates is closely related to the scarcity of macroalgae and stronger water motion at the depth>15 m in Zhongsha Islands.This study expanded the study in biodiversity of benthic dinoflagellates in Chinese waters,and revealed the distribution characteristics of harmful benthic microalgae in reef habitats.

Effects of the mixing degree upstream of the diverging area on the pollutant allocation characteristics of a braided river

The mixing degree upstream of the diverging area is one of the important factors influencing the pollutant allocation characteristics of braided rivers,but the effect remains unclear at present.In this paper,physical model tests were designed to study the effect on the pollutant flux ratio with six branching forms and a series of longitudinal discharge distances.The results indicated that the mixing degree upstream of the diverging area,which is closely related to the longitudinal discharge distance,notably affected the pollutant flux ratio.The lower the mixing degree,the larger was the deviation of the pollutant flux ratio from the discharge ratio.Moreover,a linear relationship was attained between the dimensionless mixing degree and the dimensionless deviation of the pollutant flux ratio from the discharge ratio.Consideration of different branching angles or different water layers or different branches did not affect this trend.The experimental results further demonstrated that the intercept and slope of the aforementioned linear relationship depended on the branching angle and exhibited an opposite monotonicity with a symmetric branch angle as the dividing point.These results help towards a better understanding of the mechanism of the factors influencing pollutant transport in complicated braided rivers,and provide a new approach to predicting the pollutant flux ratio of braided rivers.

In recent years,the nuclear norm minimization(NNM)as a convex relaxation of the rank minimization has attracted great research interest.By assigning different weights to singular values,the weighted nuclear norm minimization(WNNM)has been utilized in many applications.However,most of the work on WNNM is combined with the l 2-data-fidelity term,which is under additive Gaussian noise assumption.In this paper,we introduce the L1-WNNM model,which incorporates the l 1-data-fidelity term and the regularization from WNNM.We apply the alternating direction method of multipliers(ADMM)to solve the non-convex minimization problem in this model.We exploit the low rank prior on the patch matrices extracted based on the image non-local self-similarity and apply the L1-WNNM model on patch matrices to restore the image corrupted by impulse noise.Numerical results show that our method can effectively remove impulse noise.
