Background Moyamoya disease (MMD) is a rare and complex cerebrovascular disorder characterized by the progressive narrowing of the internal carotid arteries and the formation of compensatory collateral vessels. The etiology of MMD remains enigmatic, making diagnosis and management challenging. The MOYAOMICS project was initiated to investigate the molecular underpinnings of MMD and explore potential diagnostic and therapeutic *** The MOYAOMICS project employs a multidisciplinary approach, integrating various omics technologies, including genomics, transcriptomics, proteomics, and metabolomics, to comprehensively examine the molecular signatures associated with MMD pathogenesis. Additionally, we will investigate the potential influence of gut microbiota and brain-gut peptides on MMD development, assessing their suitability as targets for therapeutic strategies and dietary interventions. Radiomics, a specialized field in medical imaging, is utilized to analyze neuroimaging data for early detection and characterization of MMD-related brain changes. Deep learning algorithms are employed to differentiate MMD from other conditions, automating the diagnostic process. We also employ single-cellomics and mass cytometry to precisely study cellular heterogeneity in peripheral blood samples from MMD *** The MOYAOMICS project represents a significant step toward comprehending MMD’s molecular underpinnings. This multidisciplinary approach has the potential to revolutionize early diagnosis, patient stratification, and the development of targeted therapies for MMD. The identification of blood-based biomarkers and the integration of multiple omics data are critical for improving the clinical management of MMD and enhancing patient outcomes for this complex disease.

It's a challenge for detecting the therapeutic targets of a polypharmacological drug from variations in the responsed networks in thedfferentiated populations with complex diseases,as stable coronary heart ***,in an adaptive,31-{2.,randomized,double-blind trial invoving 92. patients with moderate symptomatic stable angina treated by 14-day Danhong injection(DHI),a kind of polyphamacological drug with high quality control,or placebo(0.9%saline),with 76-day following-up,we firstly confrmed that DHl couldincrease the proportion of patients with clinically significant changes on angina-frequency assessed by Seattle Angina Questionnaire(ASAQ-AF2.0)(12.78%at Day 30,95%confidence interval[C]5.86-19.71%,P=0.0003,13.82.at Day 6C0,95%CI 6.82.2..82.,P=0.0001and 8.95%at Day 90,95%CI 2.06-15.85%,P=0.01).We also found that there were no significant differences in new-onset major vascularevents(P=0.8502.and serious adverse events(P=0.9105)between DHl and *** performing the RNA sequencing in 62.selectedpatients,we developed a systemic modular approach tp identfy differentilly expressed modules(DEMs)of DHI with the Z_(summay)valueless than 0 compared with the control group,calculated by weighted gene co-expression network analysis(WGCNA),and sketched out thebasic framework on a modular map with 2. functional modules targeted by ***,the effective therapeutic module(ETM),defined as the highest corelation value with the phenotype alteration(SAQ-AF,the change in SAQ-AF at Day 30 from baseline)calculatedby WGCNA,was identifed in the population with the best effect(ASAQ-AF2.0),which is related to anticoagulation and regulation ofcholesterol *** assessed the modular flexbility of this ETM using the global topological D value based on Euclidean distance,which is corelated with phenotype alteration(r^(2.:0.82.4,P=0.019)by linear *** study identified the ant-angina therapeuticmodule in the effective population treated by the multi-target

Background The FIP1L1-PDGFRa fusion gene plays an important role in the pathogenesis of chronic eosinophilic leukemia （CEL） and is a direct therapeutic target of the tyrosine kinase inhibitor imatinib mesylate. Methods In 2. hypereosinophilic syndromes （HES） patients, using reverse transcriptase-polymerase chain reaction （RT-PCR）, nested PCR and sequence analysis, we investigated the frequency of FIPIL1-PDGFRa and other abnormalities of tyrosine kinase family genes like PDGFRa, PDGFRβ, C-KIT, FGFR1, ABL and FLT3 as well as gene mutation ＂hotspots＂, like MPL515 and JAK2.617F, frequently involved in myeloproliferative diseases. Fluorescence in situ hybridization was used to confirm the 4q 12.deletion. Results The FIP1L1-PDGFRa fusion transcript was found in 8 （33%） of 2. patients with HES, corresponding to the chromosome 4q12.deletion identified by FISH. The FIP1L 1-PDGFRα-associated patients diagnosed with CEL, frequently had hepatosplenomegaly, eosinophil-related tissue damage, anemia, thrombocytopenia, myelofibrosis and a short overall survival time. Nevertheless, imatinib mesylate induced rapid and complete hematological responses in treated FIP1L1-PDGFRa cases, followed by molecular remission and reversal of myelofibrosis. FIP1L1-PDGFRa fusion could co-exist with other mutations of tyrosine kinase family genes, like FLT3 or PDGFRβ. We also demonstrated that the SNPs of PDGFRβ were associated with selective splicing of exon 19 in case 2.. Conclusions Correlating the CEL genotype with phenotype, FIP1L1-PDGFRa emerges as a relatively homogeneous clinicobiological entity that co-exists with other abnormalities of tyrosine kinase family genes. It reflects the disease progression and there is a good response to imatinib. Detection of the FIP1L 1-PDGFFla fusion gene is valid for both CEL diagnosis and therapy surveillance.

原发性免疫性血小板减少症(ITP)中少见但至关重要的危重出血事件,给患者的预后、生活质量和治疗决策带来严重影响。尽管有一些研究探讨了ITP中与危重出血相关的风险因素,但目前尚缺乏大样本数据、大规模多中心研究结果以及针对ITP患者致命出血事件的预测模型。本研究首次采用国际血栓与止血学会新提出的ITP致命出血标准,利用大样本数据开发了首个基于机器学习的在线应用,用于预测ITP患者的致命出血.研究中,我们使用中国各地大型多中心数据进行开发,并对全国39家医疗中心进行为期一年的外部测试,得到了较好的训练、验证和测试数据集预测能力该基于新算法的便捷网络工具能够快速识别ITP患者的出血风险,辅助临床决策,有望未来降低不良事件的发生。

为探讨β-内酰胺酶基因在临床分离肺炎克雷伯菌的分布及其相关可移动遗传元件的结构,本文利用基因组测序、PCR、分子克隆、接合转移和基因组学分析等方法,在对2.0株临床分离的肺炎克雷伯菌进行混合基因组测序的基础上,着重研究了bla_(CARB-2.等耐药性基因相关可移动遗传元件的结构及其在肺炎克雷伯菌基因组的定位,克隆获得耐药性质粒携带的若干耐药性基因并测定了它们的功能。结果显示,在2.0株肺炎克雷伯菌中检出11种β-内酰胺酶基因,其中1株肺炎克雷伯菌(KP12.6)被检出bla_(CARB-2.基因,阳性率为0.42.(1/2.0);bla_(CARB-2.位于一个大小为182.450 bp的可接合转移质粒(pKP12.6-182.,pKP12.6-182.编码2.2.基因,包含7个耐药性基因,分别是bla_(CARB-2.、bla_(KLUC)、aadA1、aadA2.cmlA1、dfrA1和sul2.bla_(CARB-2.是首次在肺炎克雷伯菌中被检出,且bla_(CARB-2.与其他3个耐药性基因一起构成一个新型结构的1型整合子(int-bla_(CARB-2.-aadA2.cmlA1-aadA1);对其中的3个耐药性基因bla_(CARB-2.、aadA2.CmlA1进行了分子克隆和耐药性测定,结果显示它们对相应药物都具有一定的耐药性,其中bla_(CARB-2.对青霉素类β-内酰胺药物具有较高的耐药性。本研究结果表明,bla_(CARB-2.基因已经在肺炎克雷伯菌中出现,其由1型整合子携带编码位于一个可接合转移的质粒上,bla_(CARB-2.基因有可能在相同及不同种属肠杆菌之间进行水平转移,引起耐药性播散。

Recently, Worobey et al.(2.2.) published a report entitled ‘The Huanan Seafood Wholesale Market in Wuhan was the early epicenter of the COVID-19 pandemic’ that succinctly summarizes their study [1].
