Aim:The human cytochrome P450 2D6(CYP2D6)gene copy number variation,involving CYP2D6 gene deletion(CYP2D6*5)and duplication or multiduplication(CYP2D6*×N),can result in reduced or increased metabolism of many clinicallyused *** identification of CYP2D6*5 and CYP2D6*xN and the investi-gation of their allelic distributions in ethnic populations can be important in deter-mining the right drug and dosage for each ***:The CYP2D6*5 andCYP2D6 genes,and CYP2D6 gene duplication were identified by 2 modified longPCR,*** determine duplicated alleles,a novel long PCR was devel-oped to amplify the entire duplicated CYP2D6 gene which was used as templatefor subsequent PCR amplification.A total of 363 unrelated eastern Han Chineseindividuals were analyzed for CYP2D6 gene copy number ***:Thefrequency of CYP2D6*5 and CYP2D6*xN were 4.82%(n=35)and 0.69%(n=5)inthe eastern Han Chinese population,*** the 5 duplicated alleles,3were CYP2D6*1×Nand 2 were CYP2D6*IO×*** individual was a carrier ofboth CYP2D6*5 and CYP2D6*I×*** together,the CYP2D6 gene rear-rangements were present in 10.74% of ***:Allelic distributionsof the CYP2D6 gene copy number variation differ among Chinese from differentregions,indicating ethnic variety in *** PCR are convenient,costeffective,specific and semiquantitative for the detection of the CYP2D6 genecopy number variation,and amplification of the entire duplicated CYP2D6 gene isnecessary for the accurate identification of duplicated alleles.

BACKGROUND:Small-for-size(SFS)syndrome is an important clinical problem after living donor liver transplantation,split liver transplantation or extended *** uncertainty of the mechanisms and treatments of SFS syndrome urges surgeons to establish effective models for SFS syndrome. METHODS:A new porcine model for SFS syndrome based on extended hepatectomy was *** pressure gradient was observed before and after the surgery,and venous sampling for estimation of alanine aminotransferase, total bilirubin,and international normalized ratio was continued on a daily basis. RESULTS:Although the external morphology of the porcine liver differs from that of human being,segmental anatomy is remarkably similar in term of its vascularity and biliary *** hepatectomy with segmentsⅠand Ⅶas resection remnant(about 20%of total liver volume) resulted in similar survival rates,blood liver function tests,and elevated portal pressure gradient as clinical SFS syndrome. CONCLUSIONS:The extended hepatectomy based new model can easily be reproduced,with few costs and surgical *** SFS syndrome can easily be simulated by this new model,which is a useful tool for studying SFS syndrome-related liver injuries,especially portal overperfusion and hypertension.

Background:Cancer-related fatigue(CRF)is a common and debilitating symptom experienced by patients with advanced-stage cancer,especially those undergoing antitumor *** study aimed to evaluate the efficacy and safety of Renshenguben(RSGB)oral solution,a ginseng-based traditional Chinese medicine,in alleviating CRF in patients with advanced hepatocellular carcinoma(HCC)receiving antitumor ***:In this prospective,open-label,controlled,multicenter study,patients with advanced HCC at BCLC stage C and a brief fatigue inventory(BFI)score of≥4 were *** were assigned to the RSGB group(RSGB,10 mL twice daily)or the control group(with supportive care).Primary and secondary endpoints were the change in multidimensional fatigue inventory(MFI)score,and BFI and functional assessment of cancer therapy-hepatobiliary(FACT-Hep)scores at weeks 4 and 8 after *** events(AEs)and toxicities were ***:A total of 409 participants were enrolled,with 206 assigned to the RSGB *** week 4,there was a trend towards improvement,but the differences were not statistically *** week 8,the RSGB group exhibited a significantly lower MFI score(P<0.05)compared to the control group,indicating improved fatigue ***,the RSGB group showed significantly greater decrease in BFI and FACT-Hep scores at week 8(P<0.05).Subgroup analyses among patients receiving various antitumor treatments showed similar *** linear regression analyses revealed that the RSGB group experienced a significantly substantial decrease in MFI,BFI,and FACT-Hep scores at week *** serious drug-related AEs or toxicities were ***:RSGB oral solution effectively reduced CRF in patients with advanced HCC undergoing antitumor therapy over an eight-week period,with no discernible *** findings support the potential of RSGB oral solution as an adjunctive treatment for managing CRF in this patient population.

METTL14 downregulation drives S100A4^(+)monocyte-derived macrophages via MyD88/NF-κB pathway to promote MAFLD progression

Porcine liver extract has been shown to be effective in the clinical treatment of severe *** aim of the present study was to study its antifibrotic as well as immune regulatory effect in ***,hepatic stellate cells(HSCs),hepatoma cell line(HepG2)and human peripheral blood mononuclear cells(PMNCs) were studied with respect to proliferation,extracellular matrix production and apoptotic activities by proliferation assay, radioimmunoassay,gene transfection,reporter gene analysis or flow eytometry respectively.A strong stimulatory proliferation effect was observed in hepatocytes,and an inhibitory effect was found in HSC. Hyaluronic acid(HA)production and reporter gene activities driven by variousα1(Ⅰ)procollagen gene promoters in HSC-T6 were significantly decreased after treatment with the ***-Anexin V binding apoptotic HepG2 cells were more prominent in the presence of 60μg/ml *** active T lymphocytes with CD4+/ CD69+ positive markers existed in the presence of the extract. Porcine liver extract is effective for antifibrogenesis via hepatocyte regeneration,HSC and hepatoma cell inhibition in *** elevation of active T lymphocytes is helpful for immune surveillance. Fine mapping of the extract is necessary in order to get definite molecules which are essential in all described functions.

Without intervention,a considerable proportion of patients with metabolism‐associated fatty liver disease(MAFLD)will progress from simple steatosis to metabolism‐associated steatohepatitis(MASH),liver fibrosis,and even hepatocellular ***,the molecular mechanisms that control progressive MAFLD have yet to be fully ***,we unraveled that the expression of the N6-methyladenosine(m6A)methyltransferase METTL14 is remarkably downregulated in the livers of both patients and several murine models of MAFLD,whereas hepatocyte-specific depletion of this methyltransferase aggravated lipid accumulation,liver injury,and ***,hepatic Mettl14 overexpression alleviated the above pathophysiological changes in mice fed on a high-fat diet(HFD).Notably,in vivo and in vitro mechanistic studies indicated that METTL14 downregulation decreased the level of GLS2 by affecting the translation efficiency mediated by YTHDF1 in an m6A-depedent manner,which might help to form an oxidative stress microenvironment and accordingly recruit Cx3cr1^(+)Ccr2^(+)monocyte-derived macrophages(Mo-macs).In detail,Cx3cr1^(+)Ccr2^(+)Mo-macs can be categorized into M1-like macrophages and S100A4-positive macrophages and then further activate hepatic stellate cells(HSCs)to promote liver *** experiments revealed that CX3CR1 can activate the transcription of S100A4 via CX3CR1/MyD88/NF-κB signaling pathway in Cx3cr1^(+)Ccr2^(+)*** of METTL14 or GLS2,or interfering with this signal transduction pathway such as inhibiting MyD88 could ameliorate liver injuries and *** together,these findings indicate potential therapies for the treatment of MAFLD progression.

BACKGROUND:The early diagnosis of hepatocellular carcinoma(HCC) is of great clinical desirable due to lack of specific and sensitive *** in the sugar chains of glycoprotein synthesized by the liver contribute to the molecular basis of abnormalities in *** the study aims to construct and assess the diagnostic value of N-glycan based diagnostic model in identifying ***:A total of 273 cases HBV related HCC(n=145),liver fibrosis(n=128) and matched healthy controls (n=120) were recruited.N-glycome of serum protein was profiled by DNA sequencer-assisted fluorophore-assisted carbohydrate electrophoresis(DSA-FACE). Multiparameters diagnostic models were constructed based on above profiling and routine ***:A biantennary glycan(NG1A2F,peak 4) was found to be decreased in HCC and increased in liver fibrosis,while a branching triantennary glycan(NA3Fb,peak 9) was increased in HCC and decreased in liver *** efficacy of the log ratio of these two N-glycan structure abundances[log(p9/4)]was similar to AFP in differentiation HCC from *** accuracy and sensitivity of the diagnostic model combining AFP and N-glycan markers(Cscore B)were increased 7-10%compared to that of AFP. Log(p9/4) was more efficient in monitoring the progression of HCC in regarding to vascular invasion with improved specificity(16%) and accuracy(8%) compared to that of AFP. CONCLUSIONS:The branchingα(1,3)-fucosylated triantennary glycan and the biantennary glycan are potential as N-glycan *** diagnostic models based on the N-glycan markers and AFP improve the efficacy in HCC diagnosis and progression monitoring.

目的　探讨甲胎蛋白 (AFP)和甲胎蛋白异质体 (AFPVariants ,AFP V)对原发性肝癌 (PHC)的诊断价值。方法　用酶联免疫吸附试验 (ELISA)和交叉免疫电泳、放射性自显影法分别检测AFP和小扁豆凝集素亲和性甲胎蛋白异质体 (LCA AFP V)的血清含量。结果　 4 0 5例PHC患者中 ,AFP阳性占 75 .8% ,LCA AFP V阳性占 5 4 .3% ;2 10例AFP低浓度阳性患者中 ,LCA AFP V阳性 12 5例 ,而 97例AFP高浓度阳性患者 ,有 95例伴有异质体的升高 ;98例AFP阴性患者中 ,35例LCA AFP V阳性 ;2 2 3例非PHC中AFP阳性占 4 2 .2 % ,LCA AFP V阳性占 2 5 .6 %。结论　单独应用AFP或AFP V均有一定的局限性。
