Hepatitis C virus genotypes, HLA-DRB alleles and their response to interferon-α and ribavirin in patients with chronic hepatitis C

BACKGROUND: Hepatitis C virus (HCV) is a worldwide common disease. Some predictive factors influencing the response to interferon alpha (IFN-α) therapy have been identified, but the conclusions differ in various counties and areas. The aim of this study was to study the associa- tions between HCV genotypes, HLA-DRB alleles and their response to IFN-α and ribavirin in Chinese patients with chronic hepatitis C in Northeast china. METHODS: HCV genotypes of 113 patients with HCV were investigated. Gene chips were used to analyze the fre- quency of HLA-DRB in 25 of these patients and their re- sponse to IFN-α and ribavirin. The associations of HCV genotypes, HLA-DRB alleles and their response to IFN-α and ribavirin were also studied. RESULTS: The response rates differed in several types of HCV, with HCV 2b being the highest (57.78% ), HCV 1a and 2a lower (46.15% and 47.62% ) and HCV 1b the low- est (11.76% ). The response rates to IFN-α and ribavirin in patients with DRB1 07 were higher than those with DRB1 04. Sex, HCV type and HLA-DRB were all related to the response. Most female patients with HCV 2b and HLA- DRB1 07 presented complete response, whereas male pa- tients with HCV 1b and HLA-DRB1 04 usually demon- strated no response. DRB1 07 allele and HCV 2b were the factors closely related to the response. CONCLUSIONS: The response rate of HCV 1b may be the lowest even IFN-α and ribavirin are combined in treat- ment. Not only virus but also the host plays an important role in anti-virus therapy. Thus, it is necessary to adjust the host's immune status to accelerate the clearance of HCV.

Impact of Interleukin-28B Polymorphism on Response to Treatment in Chinese Hepatitis C Patients

The candidate polymorphism rs12979860 of interleukin 28B（IL28B） gene was genotyped by means of polymerase chain reaction（PCR） amplification and direct PCR sequencing, and then the role of this single nucleotide polymorphism（SNP） in the response to the treatment of 172 Chinese patients with hepatitis C virus（HCV） chronic infection was analyzed. The results show that the frequencies of major homozygotes（CC） and heterozygotes（CT） of rs12979860 were 0.84 and 0.16, respectively, and the minor homozygotes（TT） wasn’t found in this cohort. A highly significant association was found between the CC genotype and sustained virological response（SVR） in HCV geno type 1 infected patients（P〈0.001） but not in HCV non-genotype 1 infected patients. In addition, a strong association was found between rs12979860 CC genotype and rapid virological response（RVR） in both HCV genotype 1 infected patients（P〈0.001） and non-genotype 1 infected patients（P〈0.001）. Taken together, these results indicate that IL28B polymorphism plays a key role in response to hepatitis C therapy in Chinese patients. Combine assessment of clinical predictors and the IL28B polymorphism may be beneficial to the individualized treatment decision in Asian chronic hepatitis C（CHC） patients.

Among minimally invasive surgical procedures,colorectal surgery is associated with a notably higher incidence of incisional hernia(IH),ranging from 1.7% to 24.3%.This complication poses a significant burden on the healthcare system annually,necessitating urgent attention from *** a study published in the World Journal of Gastrointestinal Surgery,Fan et al compared the incidence of IH among 1614 patients who underwent laparoscopic colorectal surgery with different extraction site locations and evaluated the risk factors associated with its *** editorial analyzes the current risk factors for IH after laparoscopic colorectal surgery,emphasizing the impact of obesity,surgical site infection,and the choice of incision location on its ***,we summarize the currently available preventive measures for *** the low surgical repair rate and high recurrence rate associated with IH,prevention deserves greater research and attention compared to treatment.

目的进一步探讨国人基因的重组α-1b干扰素对我国慢性乙型肝炎病人的疗效及副作用。方法治疗组分2组:①α-1b干扰素300万U肌注每日1次,4周后改为隔日1次,共12周,60例。②α-1b干扰素300万U同前共24周,50例。对照组40例。结果治疗结束后治疗组病人临床症状、肝功能生化及病毒清除率优于对照组,HBeAg、HBV-DNA阴转分别为60.5％、62.3％和47.1％、63.4％（P<0.01）,其不良反应少而轻。结论重组α-1b干扰素适用于我国慢性乙型肝炎病人,能够有效抑制乙型肝炎的复制,改善临床症状,有利于ALT恢复正常。

1997-1999年在吉林市7个地点进行了性信息素诱杀水稻二化螟的试验。结果表明:性诱芯田间有效期长达42d,水盆诱捕器每盆每晚最大诱蛾量达481头。每盆每日最大诱蛾量Y与每盆每日平均诱蛾量X之间的关系是:随着X的增大,Y对X的倍数逐渐下降。在平均诱蛾量较低时,雄蛾对诱捕器有一定的选择性,但当虫口密度大时,雄蛾对诱捕器的选择性下降,结果使不同诱盆的诱蛾量接近均等。初步结果表明:性诱杀效果较好,与化学农药防治效果相当。

Cascading failures often occur in congested complex *** failures can be expressed as a three phase process:generation,diffusion and dissipation of *** from betweenness centrality,we propose a congestion function to represent the extent of congestion on a given *** introducing the concept of "delay time",we construct an intergradation between permanent removal and *** also build a new evaluation function of network efficiency,based on congestion,which measures the damage caused by cascading ***,based on Statnet and Webgraph topologies we investigate the effects of network structure and size,delay time,processing ability and traffic generation speed on congestion *** we uncover cascading process composed of three phases and some factors affecting cascade propagation.
