Effect of fracture fluid flowback on shale microfractures using CT scanning

The field data of shale fracturing demonstrate that the flowback performance of fracturing fluid is different from that of conventional reservoirs,where the flowback rate of shale fracturing fluid is lower than that of conventional reservoirs.At the early stage of flowback,there is no single-phase flow of the liquid phase in shale,but rather a gas-water two-phase flow,such that the single-phase flow model for tight oil and gas reservoirs is not applicable.In this study,pores and microfractures are extracted based on the experimental results of computed tomography(CT)scanning,and a spatial model of microfractures is established.Then,the influence of rough microfracture surfaces on the flow is corrected using the modified cubic law,which was modified by introducing the average deviation of the microfracture height as a roughness factor to consider the influence of microfracture surface roughness.The flow in the fracture network is simulated using the modified cubic law and the lattice Boltzmann method(LBM).The results obtained demonstrate that most of the fracturing fluid is retained in the shale microfractures,which explains the low fracturing fluid flowback rate in shale hydraulic fracturing.

Inhibition mechanisms of CRISPR-Cas9 by AcrI IA25.1 and AcrI IA32

In the ongoing arms race between bacteria and bacteriophages, bacteriophages have evolved anti-CRISPR proteins to counteract bacterial CRISPR-Cas systems. Recently, AcrIIA25.1 and AcrIIA32 have been found to effectively inhibit the activity of SpyCas9 both in bacterial and human cells. However, their molecular mechanisms remain elusive. here, we report the cryo-electron microscopy structures of ternary complexes formed by AcrIIA25.1 and AcrIIA32 bound to SpyCas9-sgRNA. Using structural analysis and biochemical experiments, we revealed that AcrIIA25.1 and AcrIIA32 recognize a novel, previously-unidentified anti-CRISPR binding site on SpyCas9. We found that both AcrIIA25.1 and AcrIIA32 directly interact with the WED domain, where they spatially obstruct conformational changes of the WED and PI domains, thereby inhibiting SpyCas9 from recognizing protospacer adjacent motif(PAM) and unwinding double-stranded DNA. In addition,they may inhibit nuclease activity by blocking the dynamic conformational changes of the SpyCas9 surveillance complex. In summary, our data elucidate the inhibition mechanisms of two new anti-CRISPR proteins, provide new strategies for the modulation of SpyCas9 activity,and expand our understanding of the diversity of anti-CRISPR protein inhibition mechanisms.

Adaptive neural network control of a 2-DOF helicopter system considering input constraints and global prescribed performance

In this study, an adaptive neural network(NN) control is proposed for nonlinear two-degree-offreedom(2-DOF) helicopter systems considering the input constraints and global prescribed performance.First, radial basis function NN(RBFNN) is employed to estimate the unknown dynamics of the helicopter system. Second, a smooth nonaffine function is exploited to approximate and address nonlinear constraint functions. Subsequently, a new prescribed function is proposed, and an original constrained error is transformed into an equivalent unconstrained error using the error transformation and barrier function transformation methods. The analysis of the established Lyapunov function proves that the controlled system is globally uniformly bounded. Finally, the simulation and experimental results on a constructed Quanser's test platform verify the rationality and feasibility of the proposed control.

Multi-Stream Temporally Enhanced Network for Video Salient Object Detection

Video salient object detection(VSOD)aims at locating the most attractive objects in a video by exploring the spatial and temporal features.VSOD poses a challenging task in computer vision,as it involves processing complex spatial data that is also influenced by temporal dynamics.Despite the progress made in existing VSOD models,they still struggle in scenes of great background diversity within and between frames.Additionally,they encounter difficulties related to accumulated noise and high time consumption during the extraction of temporal features over a long-term duration.We propose a multi-stream temporal enhanced network(MSTENet)to address these problems.It investigates saliency cues collaboration in the spatial domain with a multi-stream structure to deal with the great background diversity challenge.A straightforward,yet efficient approach for temporal feature extraction is developed to avoid the accumulative noises and reduce time consumption.The distinction between MSTENet and other VSOD methods stems from its incorporation of both foreground supervision and background supervision,facilitating enhanced extraction of collaborative saliency cues.Another notable differentiation is the innovative integration of spatial and temporal features,wherein the temporal module is integrated into the multi-stream structure,enabling comprehensive spatial-temporal interactions within an end-to-end framework.Extensive experimental results demonstrate that the proposed method achieves state-of-the-art performance on five benchmark datasets while maintaining a real-time speed of 27 fps(Titan XP).Our code and models are available at https://***/Rujiale/MSTENet.

The continuous decrease of low-slope cropland resources caused by construction land crowding poses huge threat to regional sustainable development and food security.Slope spectrum analysis of topographic and geomorphic features is considered as a digital terrain analysis method which reflects the macro-topographic features by using micro-topographic factors.However,pieces of studies have extended the concept of slope spectrum in the field of geoscience to construction land to explore its expansion law,while research on the slope trend of cropland from that perspective remains rare.To address the gap,in virtue of spatial analysis and geographically weighted regression(GWR)model,the cropland use change in the Yangtze River Basin(YRB)from 2000 to 2020 was analyzed and the driving factors were explored from the perspective of slope spectrum.Results showed that the slope spectrum curves of cropland area-frequency in the YRB showed a first upward then a downward trend.The change curve of the slope spectrum of cropland in each province(municipality)exhibited various distribution patterns.Quantitative analysis of morphological parameters of cropland slope spectrum revealed that the further down the YRB,the stronger the flattening characteristics,the more obvious the concentration.The province experienced the greatest downhill cropland climbing(CLC)was Shannxi,while province experienced the highest uphill CLC was Zhejiang.The most common cropland use change type in the YRB was horizontal expansion type.The factors affecting average cropland climbing index(ACCI)were quite stable in different periods,while population density(POP)changed from negative to positive during the study period.This research is of practical significance for the rational utilization of cropland at the watershed scale.

Numerical Investigation of the Angle of Attack Effect on Cloud Cavitation Flow around a Clark-Y Hydrofoil

Cavitation is a prevalent phenomenon within the domain of ship and ocean engineering,predominantly occurring in the tail flow fields of high-speed rotating propellers and on the surfaces of high-speed underwater vehicles.The re-entrant jet and compression wave resulting from the collapse of cavity vapour are pivotal factors contributing to cavity instability.Concurrently,these phenomena significantly modulate the evolution of cavitation flow.In this paper,numerical investigations into cloud cavitation over a Clark-Y hydrofoil were conducted,utilizing the Large Eddy Simulation(LES)turbulence model and the Volume of Fluid(VOF)method within the OpenFOAM framework.Comparative analysis of results obtained at different angles of attack is undertaken.A discernible augmentation in cavity thickness is observed concomitant with the escalation in attack angle,alongside a progressive intensification in pressure at the leading edge of the hydrofoil,contributing to the suction force.These results can serve as a fundamental point of reference for gaining a deeper comprehension of cloud cavitation dynamics.

Selective control of secondary alcohols upgrading using Ir-catalyzed cross-coupling strategy

The selective coupling of alcohols is a fascinating yet challenging approach for upgrading alcohols. herein, we accomplished the controlled production of β-disubstituted ketones or upgraded secondary alcohols via the Ir-catalyzed cross-coupling of secondary alcohols in excellent yields with broad substrate scopes. This selective control was achieved by using an in-situ generated monoNHC-Ir or a tris-NHC-Ir complex as the catalysts, respectively. Mechanistic studies revealed that the delicate balance between dehydrogenation and hydrogenation abilities of these bifunctional catalysts is crucial for achieving different selectivity. The tris-NHC-Ir complex effectively facilitated dehydrogenation of alcohols and hydrogenation of intermediates, leading to the desired upgraded secondary alcohols. Conversely, the high dehydrogenation ability of the mono-NHC-Ir complex promoted the conversion of formed secondary alcohols back to ketones.

Animal models are extensively used in all aspects of biomedical research,with substantial contributions to our understanding of diseases,the development of pharmaceuticals,and the exploration of gene functions.The field of genome modification in rabbits has progressed slowly.However,recent advancements,particularly in CRISPR/Cas9-related technologies,have catalyzed the successful development of various genome-edited rabbit models to mimic diverse diseases,including cardiovascular disorders,immunodeficiencies,agingrelated ailments,neurological diseases,and ophthalmic pathologies.These models hold great promise in advancing biomedical research due to their closer physiological and biochemical resemblance to humans compared to mice.This review aims to summarize the novel gene-editing approaches currently available for rabbits and present the applications and prospects of such models in biomedicine,underscoring their impact and future potential in translational medicine.

Theoretical and experimental investigation of the resonance responses and chaotic dynamics of a bistable laminated composite shell in the dynamic snap-through mode

The dynamic model of a bistable laminated composite shell simply supported by four corners is further developed to investigate the resonance responses and chaotic behaviors.The existence of the 1:1 resonance relationship between two order vibration modes of the system is verified.The resonance response of this class of bistable structures in the dynamic snap-through mode is investigated,and the four-dimensional(4D)nonlinear modulation equations are derived based on the 1:1 internal resonance relationship by means of the multiple scales method.The Hopf bifurcation and instability interval of the amplitude frequency and force amplitude curves are analyzed.The discussion focuses on investigating the effects of key parameters,e.g.,excitation amplitude,damping coefficient,and detuning parameters,on the resonance responses.The numerical simulations show that the foundation excitation and the degree of coupling between the vibration modes exert a substantial effect on the chaotic dynamics of the system.Furthermore,the significant motions under particular excitation conditions are visualized by bifurcation diagrams,time histories,phase portraits,three-dimensional(3D)phase portraits,and Poincare maps.Finally,the vibration experiment is carried out to study the amplitude frequency responses and bifurcation characteristics for the bistable laminated composite shell,yielding results that are qualitatively consistent with the theoretical results.

Adding PD-1 blockade in the neoadjuvant regimens for locally advanced rectal cancer(LARC)patients with microsatellite stable(MsS)/mismatch repair-proficient(pMMR)tumors is an attractive,but debatable strategy.This phase 2,multicenter,prospective,single-arm study enrolled patients from 6 centers from June 2021 to November 2022.Locally advanced rectal cancer(LARC,cT_(3-4a)N_(0)M_(0) and cT_(1-4a)N_(1-2)M_(0))patients aged≥18 years with the distance from distal border of tumor to anal verge≤10 cm(identified by Magnetic Resonance Imaging)were qualifed for inclusion.The patients received long-course radiotherapy(50 Gy/25 fractions,2 Gy/fraction,5 days/week)and three 21-day cycles capecitabine(850-1000 mg/m2,bid,po,day1-14)and three 21-day cycles tislelizumab(200 mg,iv.gtt,day8)as neoadjuvant.Total mesorectal excision(TME)was 6-12 weeks after the end of radiotherapy to achieve radical resection.A total of 50 patients were enrolled in this study.The pathological complete response rate was 40.0%[20/50,95%confidence interval(CI):27.61-53.82%],while 15(30.0%,95%CI:19.1-43.75%),9(18.0%,95%CI:9.77-30.8%),2(4.0%,95%CI:1.10-13.46%)patients respectively achieved grade 1,2,and 3 tumor regression.Treatment-related adverse events(TRAEs)occurred in 28(56.0%)LARC patients,including 26(52.0%)with grade I-II and 2(4.0%)with grade II(1 with grade 3 immune-related colitis and 1 with grade 3 rash).PD-1 blockade plus long-course chemoradiotherapy(CRT)showed promising therapeutic effects according to pathological complete response rate and is well-tolerated in LARC patients.A larger randomized controlled study is desired to further validate the above findings.
