The negative effects of low temperature can readily induce a variety of diseases.We sought to understand the reasons why cold stress induces disease by studying the mechanisms of fine-tuning in macrophages following cold exposure.We found that cold stress triggers increased macrophage activation accompanied by metabolic reprogramming of aerobic glycolysis.The discovery,by genome-wide RNA sequencing,of defective mitochondria in mice macrophages following cold exposure indicated that mitochondrial defects may contribute to this process.In addition,changes in metabolism drive the differentiation of macrophages by affecting histone modifications.Finally,we showed that histone acetylation and lactylation are modulators of macrophage differentiation following cold exposure.Collectively,metabolism-related epigenetic modifications are essential for the differentiation of macrophages in cold-stressed mice,and the regulation of metabolism may be crucial for alleviating the harm induced by cold stress.

【目的】描述新型冠状病毒感染(COVID⁃19)从“乙类甲管”调整到“乙类乙管”新政策后的流行特征,探讨既往自然感染新型冠状病毒(SARS⁃CoV⁃2)对再感染者常见症状的保护作用。【方法】以2022年12月4日—2023年1月11日上海某三甲医院感染SARS⁃CoV⁃2的工作人员为研究对象,采集其人口学特征、临床症状、疾病史和SARS⁃CoV⁃2疫苗接种史等。分析感染者流行病学特征,比较初次与再次感染者的临床症状和严重程度的差异。【结果】共纳入病例2704例,其中再感染45例,男性605例(22.4%),平均年龄(34.9±9.1)岁,医生608例(22.5%),护士1275例(47.2%),COVID⁃19疫苗接种≥3剂2351例(86.9%)。轻/中型、无症状感染者、发热、头痛、干咳、咳痰和胸闷人数分别是2704(100.0%)、92(3.4%)、2385(88.2%)、2066(76.4%)、1642(60.7%)、1807(66.8%)和439(16.2%)。再感染是发热(OR=0.161,P<0.001)、头痛(OR=0.320,P<0.001)和热峰(β=-0.446,P<0.001)的保护因素。【结论】实行“乙类乙管”新政策后,上海某三甲医院工作人员COVID⁃19发病以轻/中型为主,再感染者临床症状较轻,但无症状比例较少。

With the accelerated aging society in China,the incidence of biliary surgical diseases in the elderly has increased significantly.The clinical characteristics of these patients indicate that improving treatment outcomes and realizing healthy aging are worthy of attention.How to effectively improve the treatment effect of geriatric biliary surgical diseases has attracted widespread attention.This paper reviews and comments on the hotspots and difficulties of biliary surgery in older patients from six aspects:(1)higher morbidity associated with an aging society,(2)prevention and control of pre-operative risks,(3)extending the indications of laparoscopic surgery,(4)urgent standardization of minimally invasive surgery,(5)precise technological progress in hepatobiliary surgery,and(6)guarantee of peri-operative safety.It is of great significance to fully understand the focus of controversy,actively make use of its favorable factors,and effectively avoid its unfavorable factors,for further improving the therapeutic effects of geriatric biliary surgical diseases,and thus benefits the vast older patients with biliary surgical diseases.Accordingly,a historical record with the highest age of 93 years for laparoscopic transcystic common bile duct exploration has been created by us recently.

Genome-wide identification and characterization of the IPT family members in nine Rosaceae species and a functional analysis of MdIPT5b in cold resistance

Cytokinins are members of a group of phytohormones involved in various growth and developmental processes in plants.Isopentenyl transferase(IPT)is the rate-limiting enzyme in catalyzing the biosynthesis of cytokinins.In this study,to understand the role of IPT family in cold resistance,78 IPT candidates were identified and characterized in nine Rosaceae genomes.The expansion of IPT families in the Rosaceae primarily occurred through segmental duplication rather than tandem duplication.In general,purifying selection controlled the evolution of IPT families in the Rosaceae,with IPT3 and IPT5 homologs as the primary drivers of evolution.Cis-elements,which are involved in the responses to many environmental stresses or phytohormone signals,were identified in the promoters of MdIPT members.This was consistent with the trends of expression of the MdIPT genes in apple(Malus domestica)calli.MdIPT5b was also found to exhibit multiple responses to phytohormones and stress signals.The ectopic expression of MdIPT5b resulted in an increase in cold resistance in transformed apple calli and tomato(Solanum lycopersicum)plantlets.The redox balance was partially stabilized through the accumulation of proline under cold stress.However,the ascorbate–glutathione cycle cannot be stabilized in the cold.All physiological and biochemical assays are preformed in spectrophotometer.These results showed that regulating the expression of IPT genes for moderate cytokinin improvement could enhance the accumulation of proline to stabilize the osmotic and redox balances to improve resistance to cold stress.

通过对哀牢山楚雄片区野生种古茶树生存现状分析,探讨该片区野生种古茶树管护经营模式,提出科学可行的对策建议,以改善野生种古茶树生存现状。调查结果表明,云南哀牢山国家级保护区楚雄片区野生种古茶树均为大理茶种,以单株分散生长为主,资源总量较少;天然和天然萌生各占一半,生长前期茶树主干遭受损坏,生长情况一般,生长势主要处于中等状态,幼树更新旺盛,但较大树木较少,竞争后期可能出现衰退的趋势。研究区的野生种古茶树生存现状不够理想,除自然因素干扰以外,管护措施也不到位,仍需加强资源保护力度,注重宣传保护和科研开发协调平衡,破除制约野生种古茶树生存发展障碍,实现地区野生种古茶树资源发展最大化。

Portable energy solutions are highly desired in the era of the Internet of Things for powering various distributed micro-electronic devices.At the same time,the energy crisis and catastrophic global warming are becoming serious problems in the world,emphasizing the urgent need for clean and renewable energy.Here,we report a low-cost,high-performance,and portable hand-driven whirligig structured triboelectric–electromagnetic hybrid nanogenerator(whirligig-HNG)for multi-strategy energy harvesting.The whirligig-HNG comprises a dynamic supercoiling TENG via the pulling-strings and inner-distributed EMGs(variable number)in the rotator.The whirligig structure can readily convert linear displacement in low frequency into rotary motion in extremely high frequency.Based on this ingenious design,the whirligig-HNG is capable to harvest the triboelectric energy from the supercoiling/uncoiling process from the pulling strings and simultaneously utilize the high-frequency rotation energy via electromagnetic induction.We have systematically investigated the working mecha-nism of the whirligig-HNG for coupled energy harvesting and compared the individual characteristics of TENG and EMG.The whirligig-HNG is successfully demonstrated to light up more than 100 commercial light-emitting diodes(LEDs)and drive portable electronics.This research presents the enormous potential of whirligig-HNG as a manual and portable power supply for powering various portable electronics.

Effects of substance P on growth of fibroblast-like cells derived from bile duct： an in vitro cell culture study

Background The possible role of substance P （SP） during wound healing has been the primary research focus in recent years,but its effect on the healing process after bile duct injury is little understood.This study aimed to investigate the effects of SP on growth of fibroblast-like cells derived from rabbit bile duct.Methods Fibroblast-like cells derived from rabbit bile duct were identified and divided randomly into control and experimental groups.SP-treated cells at different concentrations of 10^-9-10^-5 mol/L and control group were incubated,respectively,for 48 hours.After incubating,the effects of SP on cell proliferation were assessed by cell counts and MTT test.Apoptosis rate （AR） of cells was measured by flow cytometry.Results Cultured rabbit bile duct cells were fibroblast-like in morphology,and these cells were stained positively for vimentin and negatively for desmin.After SP was added to nonconfluent cells for 48 hours,cell numbers were significantly increased in experimental groups than in controls （P 〈0.05）.The maximum stimulation of cell proliferation was achieved at SP of 10^-5 mol/L.Bile duct fibroblast-like cells in the SP group showed a higher proliferating activity and lower AR than those in the control group or in the SP ＋ Spantide group （P 〈0.05）.Spantide partly inhibited the effects of SP on fibroblastlike cells.Examination under transmission electron microscopy revealed rough endoplasmic reticulum and prominent Golgi complexes after SP treatment.Conclusions SP has a growth regulatory property on cultivated bile duct fibroblast-like cells in vitro,suggesting that SP may involve in wound healing after bile duct injury by promoting wound fibroblast proliferation and inhibiting apoptosis and participate in pathological scar formation.

Scenario modelling and the risk assessment of natural disasters is one of the hotspots in disaster research. However, up until now, urban natural disaster risk assessments lack common procedures and programmes. This paper selects rainstorm waterlogging as a disaster to research, which is one of the most frequently occurring hazards for most cities in China. As an example, we used a small-scale integrated methodology to assess risks relating to rainstorm waterlogging hazards in the Jing＇an District of Shanghai. Based on the basic concept of disaster risk, this paper applies scenario modelling to express the risk of small-scale urban rainstorm waterlogging disasters in different return periods. Through this analysis of vulnerability and exposure, we simulate different disaster scenarios and propose a comprehensive analysis method and procedure for small-scale urban storm waterlogging disaster risk assessments. A grid-based Geographical Information System （GIS） approach, including an urban terrain model, an urban rainfall model and an urban drainage model, was applied to simulate inundation area and depth. Stage-damage curves for residential buildings and contents were then generated by the loss data of waterlogging from field surveys, which were further applied to analyse vulnerability, exposure and loss assessment. Finally, the exceedance probability curve for disaster damage was constructed using the damage of each simulated event and the respective exceedance probabilities. A framework was also developed for coupling the waterlogging risk with the risk planning and management through the exceedance probability curve and annual average waterlogging loss. This is a new exploration for small-scale urban natural disaster scenario simulation and risk assessment.
