Aims:Multiple genes and environmental factors are known to be involved in congenital heart disease(CHD),but epigenetic variation has received little ***(MZ)twins with CHD provide a unique model for exploring this *** order to investigate the potential role of Deoxyribonucleic Acid(DNA)methyla-tion in CHD pathogenesis,the present study examined DNA methylation variation in MZ twins discordant for CHD,especially ventricular septal defect(VSD).Methods and Results:Using genome-wide DNA methylation profiles,we identified 4004 differentially methylated regions(DMRs)in 18 MZ twin pairs discordant for CHD,and 2826 genes were *** Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis revealed a list of CHD-associated *** further investigate the role of DNA methylation in VSD,data from 7 pairs of MZ twins with VSD were *** identified 1614 DMRs corresponding to 1443 genes associated with arrhythmogenic right ventricular cardiomyopathy,cyclic guanosine monopho-sphate-protein kinase G(cGMP-PKG)signaling pathway by KEGG analysis,and cell-cell adhesion,calcium ion transmembrane transport by GO analysis.A proportion of DMR-associated genes were involved in calcium signaling *** methylation changes of calcium signaling genes might be related to VSD ***:CHD is associated with differential DNA methylation in MZ *** may be etiologically linked to DNA methylation,and methylation of calcium signaling genes may be involved in the development of VSD.

Integrated transcriptomics and metabolomics analysis reveal the molecular mechanism of unsaponifiable matter in delaying aging

Aging is one of the causes of cognitive dysfunction， which seriously affects people's quality of life. Unsaponifiable matter (USM) has antioxidant potential， but the molecular mechanisms that ameliorate aging and cognitive dysfunction are unknown. In this study， we used a galactose-induced brain aging mouse model and systematically analyzed the mechanism of USM in delaying aging in mice by detecting changes in serum and brain by metabolomics and *** was compared with the model group， and non-targeted metabolomics identified 68 (15 up- regulated，53down-regulated) differentially metabolites，andtranscriptomics identified 303 differentially expressed genes (228 up-regulated， 75 down-regulated). Combined multi-omics analyses showed that USM maintains normal brain function by regulating glycolytic processes， the tricarboxylic acid cycle (TCA)， tryptophan metabolism， pyrimidine metabolism， the alanine， aspartate， and glutamate metabolism， and P38maPK pathway. Meanwhile， USM increased neurotransmitter release from GABAergic synapses and cholinergic synapses by regulating synaptic vesicle cycling. In summary， USM increased energy metabolism and enhanced brain nerve signaling in the mouse brain， thereby delaying brain aging. This investigation offers novel perspectives into the molecular mechanism of USM to mitigate brain aging.

The sloshing in a group of rigid cylindrical tanks with baffles and on soil foundation under horizontal excitation is studied *** solutions for the velocity potential are derived out by the liquid subdomain *** models with mass-spring oscillators are established to replace continuous *** with the least square technique,Chebyshev polynomials are employed to fit horizontal,rocking and horizontal-rocking coupling impedances of soil,respectively.A lumped parameter model for impedance is presented to describe the effects of soil on tank structures.A mechanical model for the soil-foundation-tank-liquid-baffle system with small amount of calculation and high accuracy is proposed using the substructure *** analytical solutions are in comparison with data from reported literature and numerical codes to validate the effectiveness and correctness of the *** dynamic properties and seismic responses of the soil-tank system are given for the baffle number,size and location as well as soil parameter.

Renewable energy driven N_(2) electroreduction with air as nitrogen source holds great promise for realizing scalable green ammonia ***,relevant out-lab research is still in its ***,a novel Sn-based MXene/maX hybrid with abundant Sn vacancies,Sn@Ti_(2)CTX/Ti_(2)SnC–V,was synthesized by controlled etching Sn@Ti_(2)SnC maX phase and demonstrated as an efficient electrocatalyst for electrocatalytic N2 *** to the synergistic effect of MXene/maX heterostructure,the existence of Sn vacancies and the highly dispersed Sn active sites,the obtained Sn@Ti2CTX/Ti_(2)SnC–V exhibits an optimal NH_(3) yield of 28.4μg h^(−1) mg_(cat)^(−1) with an excellent FE of 15.57% at−0.4 V versus reversible hydrogen electrode in 0.1 M Na_(2)SO_(4),as well as an ultra-long ***,this catalyst represents a satisfactory NH3 yield rate of 10.53μg h^(−1) mg^(−1) in the home-made simulation device,where commercial electrochemical photovoltaic cell was employed as power source,air and ultrapure water as feed *** as-proposed strategy represents great potential toward ammonia production in terms of financial cost according to the systematic technical economic *** work is of significance for large-scale green ammonia production.

Rehmanniae Radix Praeparata aqueous extract improves hepatic ischemia/reperfusion injury by restoring intracellular iron homeostasis

Hepatic ischemia/reperfusion injury(HIRI)is a common pathophysiological condition occurring during or after liver resection and transplantation,leading to hepatic viability impairment and functional ***,ferroptosis,a newly recognized form of programmed cell death,has been implicated in *** Radix Praeparata(RRP),extensively used in Chinese herbal medicine for its hepatoprotective,anti-inflammatory,and antioxidant properties,presents a potential therapeutic ***,the mechanisms by which RRP mitigates HIRI,particularly through the regulation of ferroptosis,remain *** this study,we developed a HIRI mouse model and monocrotaline(MCT)-and erastin-induced in vitro hepatocyte injury *** conducted whole-genome transcriptome analysis to elucidate the protective effects and mechanisms of RRP on *** RRP aqueous extract was characterized by the presence of acteoside,rehmannioside D,and *** results demonstrate that the RRP aqueous extract ameliorated oxidative stress,reduced intracellular iron accumulation,and attenuated HIRI-induced liver ***,RRP significantly inhibited hepatocyte death by restoring intracellular iron homeostasis both in vivo and in ***,the RRP aqueous extract reduced intrahepatocellular iron accumulation by inhibiting ZIP14-mediated iron uptake,promoting hepcidin-and ferroportin-mediated iron efflux,and ameliorating mitochondrial iron aggregation through upregulation of Cisd1 ***,siRNA-mediated inhibition of hamp synergistically enhanced the RRP aqueous extract's inhibitory effect on *** conclusion,our study elucidates the mechanisms by which RRP aqueous extracts alleviate HIRI,highlighting the restoration of iron metabolic *** findings position RRP as a promising candidate for clinical intervention in HIRI treatment.

Aqueous zinc-ion batteries have attracted much attention due to their high theoretical capacity,low cost high safety,and ***,challenges such as dendrite growth and side reactions severely hinder the electrochemical performance of the Zn anode,leading to low Coulombic efficiency(CE)or even short circuits Herein,phenolic resin(PF)is used as a protective coating on the surface of the zinc metal *** PF layer can greatly improve the corrosion resistance of the Zn electrode in the ***,this artificial electrode/electrolyte interphase also elevates the nucleation barrier and restrains Zn^(2+)2D diffusion,regulating the zinc deposition/dissolution *** a result,PF@Zn symmetric cells exhibit superior performance than that of bare Zn symmetric cells,achieving a stable and dendrite-free cycle of 1400 ***,the carbon nanotube MnO_(2)||PF@Zn(CNT/MnO_(2)||PF@Zn)full cell also delivers a long cycle life with a high-capacity retention of about 180.0 m Ah·g^(-1)after 1400 cycles,far exceeding that of bare Zn *** work provides a facile strategy for achieving a dendrite-free and rechargeable zinc anode.

Toxicity of the disinfectant benzalkonium chloride(C_(14))towards cyanobacterium Microcystis results from its impact on the photosynthetic apparatus and cell metabolism

Quaternary ammonium compounds(QACs)are commonly used in a variety of consumer and commercial products,typically as a component of *** the COVID-19 pandemic,QACs became one of the primary agents utilized to inactivate the SARS CoV-2 virus on ***,the ecotoxicological effects of QACs upon aquatic organisms have not been fully *** this study,we examined the effects of a widely used QAC(benzalkonium chloride-C_(14),BAC-14)on two toxigenic Microcystis strains and one non-toxigenic freshwater Microcystis strain and carried out an analysis focused on primary,adaptive and compensatory stress responses at apical(growth and photosynthesis)and metabolic *** analysis revealed that the two toxic Microcystis strains were more tolerant than the non-toxic strain,with 96 hr-EC_(50)values of 0.70,0.76,and 0.38 mg/L BAC-14 for toxigenic *** FACHB-905,toxigenic *** FACHB-469,and non-toxigenic *** FACHB-908,*** photosynthetic activities of the Microcystis,assessed via F_(v)/F_(m) values,were significantly suppressed under 0.4 mg/L ***,this analysis revealed that BAC-14 altered 14,12,and 8 metabolic pathways in *** FACHB-905,*** FACHB-469,and *** FACHB-908,*** is noteworthy that BAC-14 enhanced the level of extracellular microcystin production in the toxigenic Microcystis strains,although cell growth was not significantly ***,these data show that BAC-14 disrupted the physiological and metabolic status of Microcystis cells and stimulated the production and release of microcystin,which could result in damage to aquatic systems.

Effect of screening colonoscopy frequency on colorectal cancer mortality in patients with a family history of colorectal cancer

BACKGROUND Colorectal cancer is a common malignant tumor in China,and its incidence in the elderly is increasing *** bowel disease is a group of chronic non-specific intestinal inflammatory diseases,including ulcerative colitis and Crohn’s *** included the clinicopathological and follow-up data of patients with colorectal cancer who underwent laparoscopic colectomy or open colectomy at our Gastrointestinal Department between January 2019 and December *** indicators,oncological indicators,and survival rates were compared between the *** results of 104 patients who met the above criteria were extracted from the database(laparoscopic colectomy group=63,open colectomy group=41),and there were no statistically significant differences in the baseline data or follow-up time between the two *** Intraoperative blood loss,time to first ambulation,and time to first fluid intake were significantly lower in the laparoscopic colectomy group than in the open colectomy *** differences in overall mortality,tumor-related mortality,and recurrence rates between the two groups were not statistically significant,and survival analysis showed that the differences in the cumulative overall survival,tumor-related survival,and cumulative recurrence-free rates between the two groups were not statistically *** In elderly patients with colorectal cancer,laparoscopic colectomy has better short-term outcomes than open colectomy,and laparoscopic colectomy has superior long-term survival outcomes compared with open colectomy.
