Ethanol extract of Cyathulae Radix inhibits osteoclast differentiation and bone loss

Cyathulae Radix,a traditional Chinese medicine and a common vegetable,boasts a history spanning *** enhances bone density,boosts metabolism,and effectively alleviates osteoporosis-induced *** its historical use,the molecular mechanisms behind Cyathulae Radix’s impact on osteoporosis remain *** this study,we investigated the effects and mechanisms of Cyathulae Radix ethanol extract(CEE)in inhibiting osteoporosis and ***-week-old female mice underwent ovariectomy and were treated with CEE for eight ***-computed tomography(micro-CT)assessed histomorphometric parameters,bone tissue staining observed distal femur histomorphology,and three-point bending tests evaluated tibia mechanical ***-linked immunosorbent assay(EliSA)measured serum estradiol(E2),receptor activator for nuclear factor B ligand(RANKL),and osteoprotegerin(OPG)***-related markers were analyzed via Western blotting(WB)and quantitative real-time polymerase chain reaction(qRT-PCR).Additionally,CEE effects on RANKL-induced osteoclast formation and bone resorption were investigated in vitro using tartrate-resistant acid phosphatase(TRAP)staining,qRT-PCR,and WB *** with the ovariectomy(OVX)group,CEE treatment enhanced trabecular bone density,maximal load-bearing capacity,and various histomorphometric *** E2 and OPG levels significantly increased,while Receptor activator of nuclear factor-κB(RANK)decreased in the CEE *** downregulated matrix metallopeptidase 9(MMP-9),Cathepsin K(CTSK),and TRAP gene and protein *** bone marrow macrophages(BMMs),CEE reduced mature osteoclasts,bone resorption pit areas,and MMP-9,CTSK,and TRAP expression during osteoclast *** with DMSO treatment,CEE markedly inhibited RANK,TNF receptor associated factor 6(TRAF6),Proto-oncogene c-Fos(c-Fos),Nuclear factor of activated T-cells cytoplasmic 1(NFATc1)expressions,and Extracellular regula

Precise measurement of geometric and physical quantities in cutting tools inspection and condition monitoring: A review

As one of the most important terminals in machining, cutting tools have been widely used for components manufacturing in aerospace and other industries. The quality of these components and processing efficiency are closely linked to the performance of cutting tools. Therefore, it is essential and critical to inspect the cutting tools and monitor the condition during the stage of manufacturing and machining. This review aims to discuss and summarize the key problems, methods,and techniques from the perspective of the tool geometric and the physical quantities measurement,including machine vision, physical sensors and data processing. It is worth mentioning that we focus on the topic of precision measurement methods and discuss universal solutions by identifying the common characteristics of the measured quantities. Eventually, the challenges and future trends for the development of in-depth research and practical applications are concluded. The research and application of precise measurement techniques for geometric and physical quantities will better promote the development of intelligent manufacturing.

Impact of volcanism on the formation and hydrocarbon generation of organic-rich shale in the Jiyang Depression, Bohai Bay Basin, China

Globally,most organic-rich shales are deposited with volcanic ash *** ash,a source for many sedimentary basins,can affect the sedimentary water environment,alter the primary productivity,and preserve the organic matter(OM)through physical,chemical,and biological *** an increasing number of breakthroughs in shale oil exploration in the Bohai Bay Basin in recent years,less attention has been paid to the crucial role of volcanic impact especially its influence on the OM enrichment and hydrocarbon ***,we studied the petrology,mineralogy,and geochemical characteristics of the organic-rich shale in the upper submember of the fourth member(Es_(4)^(1))and the lower submember of the third member(Es_(3)^(3))of the Shahejie Formation,aiming to better understand the volcanic impact on organic-rich shale *** results show that total organic carbon is higher in the upper shale intervals rich in volcanic ash with enriched light rare earth elements and moderate Eu *** indicates that volcanism promoted OM formation before or after the *** positive correlation between Eu/Eu*and Post-Archean Australian Shale indicates hydrothermal activity before the volcanic *** plane graph of the hydrocarbon-generating intensity(S1+S2)suggests that the heat released by volcanism promoted hydrocarbon ***,the nutrients carried by volcanic ash promoted biological blooms during Es_(4)^(1 )and Es_(3)^(3) deposition,yielding a high primary *** blooms consume large amounts of oxygen and form anoxic environments conducive to the burial and preservation of ***,this study helps to further understand the organic-inorganic interactions caused by typical geological events and provides a guide for the next step of shale oil exploration and development in other lacustrine basins in China.

The yellow seed trait is preferred by breeders for its potential to improve the seed quality and commercial value of Brassica *** the present study,we produced yellow seed mutants using a CRISPR/Cas9 system when the two BnPAP2 homologs were knocked *** staining of the seed coat demonstrated that proanthocyanidin accumulation was significantly reduced in the pap2 double mutants and decreased specifically in the endothelial and palisade layer cells of the seed *** and metabolite profiling analysis suggested that disruption of the BnPAP2 genes could reduce the expression of structural and regulated genes in the phenylpropanoid and flavonoid biosynthetic *** broad suppression of these genes might hinder proanthocyanidin accumulation during seed development,and thereby causing the yellow seed trait in *** results indicate that BnPAP2 might play a vital role in the regulatory network controlling proanthocyanidin accumulation.

Synergistic Effects of Glutamine Deprivation and Metformin in Acute Myeloid Leukemia

Objective The metabolic reprogramming of acute myeloid leukemia(AML)cells is a compensatory adaptation to meet energy requirements for rapid *** study aimed to examine the synergistic effects of glutamine deprivation and metformin exposure on AML *** SKM-1 cells(an AML cell line)were subjected to glutamine deprivation and/or treatment with metformin or bis-2-(5-phenylacetamido-1,2,4-thiadiazol-2-yl)ethyl sulfide(BPTES,a glutaminase inhibitor)or *** viability was detected by Cell Counting Kit-8(CCK-8)assay,and cell apoptosis and reactive oxygen species(ROS)by flow *** blotting was conducted to examine the levels of apoptotic proteins,including cleaved caspase-3 and poly(ADP-ribose)polymerase(PARP).Moreover,the human long noncoding RNA(lncRNA)microarray was used to analyze gene expression after glutamine deprivation,and results were confirmed with quantitative RT-PCR(qRT-PCR).The expression of metallothionein 2A(MT2A)was suppressed using *** growth and apoptosis were further detected by CCK-8 assay and flow cytometry,respectively,in cells with MT2A *** Glutamine deprivation or treatment with BPTES inhibited cell growth and induced apoptosis in SKM-1 *** lncRNA microarray result showed that the expression of MT family genes was significantly upregulated after glutamine ***2A knockdown increased apoptosis,while proliferation was not affected in SKM-1 *** addition,metformin inhibited cell growth and induced apoptosis in SKM-1 *** glutamine deprivation and metformin enhanced the sensitivity of SKM-1 cells to ***,the combination of glutamine deprivation with metformin exhibited synergistic antileukemia effects on SKM-1 *** Targeting glutamine metabolism in combination with metformin is a promising new therapeutic strategy for AML.

Colorimetric detection of α-glucosidase activity using Ni-CeO_(2)nanorods and its application to potential natural inhibitor screening

In this work,we established an exceptionally facile method for the preparation of Ni-CeO_(2)nanorods in a kind of deep eutectic solvents(DESs)composed of l-proline and Ce(NO_(3))_(3)·6H_(2)***,Ni-CeO_(2)nanorods were successfully prepared by adding Ni(NO_(3))_(3)·6H_(2)O to ***,we found that Ni-CeO_(2)nanorods prepared in DESs have more prominent oxidase-like activity than pure CeO_(2).The outstanding catalytic activity of Ni-CeO_(2)could be ascribed to its high Ce^(3+)/Ce^(4+)*** a proof-of-concept application,the Ni-CeO_(2)nanorods were successfully acted as a colorimetric platform for the sensitive determination of ascorbic acid andα-glucosidase activity,which displays excellent analytical ***,this sensing platform was applied for screening naturalα-glucosidase inhibitors,such as terpenoids from natural *** results indicated that ursolic acid and oleanolic acid had good inhibitory *** strategy not only provides a new way to construct more kinds of nanomaterials from DESs,but also offers a facile and effective tool to screen theα-glucosidase natural inhibitors as potential anti-diabetic drugs.
