In biological systems, conformational transformations of nucleic acids play critical roles in genetic regulation. However, it remains a tricky task to design and optimize specific labeling strategies to track these *** this study, we exploited an intercalating fluorescent dye,gelRed, to characterize different DNA structures. We studied the correlation between fluorescence intensity and DNA structural properties. We showed that single-stranded DNAs with predicted self-folded secondary structures show much stronger fluorescence than those without such structures. For double-stranded DNAs, we observed that fluorescence intensity is positively correlated to their gCcontent. We also demonstrated that gelRed can be used to monitor DNA conformational changes upon temperature variations in real time. Based on these findings, we concluded that the fluorescence intensity of a gelRed-stained DNA structure has a good correlation with its thermostability in the form of a change in gibbs free energy.

Study on the thermal deformation behavior and microstructure of FgH96 heat extrusion alloy during two-pass hot deformation

The change rules associated with hot deformation of FgH96 alloy were investigated by isothermal two-pass hot deformation tests in the temperature range 1050–1125°C and at strain rates ranging from 0.001 to 0.1 s^(-1) on a gleeble 3500 thermo-simulation machine. The results showed that the softening degree of the alloy between passes decreases with increasing temperature and decreasing strain rates. The critical strain of the first-pass is greater than that of the second-pass. The true stress–true strain curves showed that single-peak dynamic recrystallization, multi-peak dynamic recrystallization, and dynamic response occur when the strain rate is 0.1, 0.01, and 0.001 s^(-1), respectively. The alloy contains three different grain structures after hot deformation: partially recrystallized tissue, completely fine recrystallized tissue, coarse-grained grains. The small-angle grain boundaries increase with increasing temperature. Increasing strain rates cause the small-angle grain boundaries to first increase and then decrease.

TaARR1, a cytokinin response regulator gene in Triticum aestivum, is essential in plant N starvation tolerance via regulating the N acquisition and N assimilation

Plant N starvation response is closely associated with the N signaling components that involve transduction of the low-N cues. In this study, we functionally characterized Ta ARR1, a cytokinin(CK) response regulator gene in Triticum aestivum, in mediating the N starvation adaptation in plants. Ta ARR1 harbors two conserved domains specified by plant ARR family members;subcellular localization analysis indicated its target onto nucleus after endoplasmic reticulum assortment. Ta ARR1 displayed modified expression upon the N starvation stressor, showing upregulated expression in roots and leaves over a 27-h N starvation treatment and whose induced transcripts were gradually recovered along with progression of the N recovery treatment. The tobacco lines overexpressing Ta ARR1 displayed improved low-N stress tolerance, displaying enlarged phenotype, increased biomass and N accumulation, and enhanced glutamine synthetase(gS) activities compared with wild type(WT) following the N starvation treatment. Expression analysis on genes encoding the nitrate transporter(NRT) and gS proteins in Nicotiana tabacum revealed that Nt NRT2.2 and Nt gS3 are upregulated in expression in the N-deprived transgenic lines, whose expression patterns were contrasted to other above family genes that were unaltered on transcripts between the transgenic lines and WT. Transgene analysis validated the function of Nt NRT2.2 and Nt gS3 in regulating N accumulation, gS activity, growth traits, and N use efficiency in plants. These results suggested the internal connection between the Ta ARR1-mediated N starvation tolerance and the modified transcription of distinct N acquisitionand assimilation-associated genes. Our investigation together indicates that Ta ARR1 is essential in plant N starvation adaptation due to the gene function in transcriptionally regulating distinct NRT and gS genes that affect plant N uptake and assimilation under the N starvation condition.

TNF-α and IL-1RA Polymorphisms and Silicosis Susceptibility in Chinese Workers Exposed to Silica Particles:A Case-Control Study

Abstract Objective To assess the association of TNF-α and IL-1RA SNPs with the risk of silicosis in Chinese workers exposed to silica particles. Methods Case-control study design was used to enroll 68 silicotic patients induced by silica particles and 68 healthy workers matched for length of silica particle exposure as controls. Both cases and controls were from the same company in southwest China, and each of them was requested to complete a questionnaire. Blood samples were drawn for genomic DNA extraction from each participant. The genotyping of TNF-α （-238 and -308） and IL-1RA （＋2018） was performed using polymerase chain reaction-based restriction fragment length polymorphism （PCR-RFLP） and SYBR green-based quantitative polymerase chain reaction {qPCR）, respectively. Unconditional logistic regression model was used to estimate odds ratios （ORs） and their 95% confidential intervals （Cl） for SNPs. Results No significant differences were found between cases and controls in particles exposure length, body mass index （BMI）, and status of smoking and alcohol consumption except for age （P=O.O01） and blood type （P=0.042）. The frequencies of TNF-c（（-238） and IL-1RA （＋2018） genotypes in cases were significantly different from those in controls, （P=O.O01 and P=0.O02, respectively）, while a borderline significant difference was found in the frequencies of TNF-α（-308） between cases and controls （P=0.063）. The variants of three SNPs increased the risk of silicosis in the Chinese workers exposed to silica particles. The adjusted ORs of TNFα（-308）, TNF-α（-238） and IL-1RA （＋2018） were 2.8 （95% Ch 2.1-7.5）, 20.9 （95% Ch 2.8-236.4） and 4.0 （95% CI： 2.6-10.2）, respectively. Conclusion It is suggested that cytokine polymorphisms of TNF-ct （-238, -308） and IL-1RA （＋2018） are associated with the risk of silicosis in the Chinese workers exposed to silica particles. Hyperinsulinemia,Insulin Resistance and Cognitive Decline in Older Cohort

Objective decline of resistance whether H Type 2 diabetes has been recently recognized as an important risk factor for cognitive patients with Alzheimer＇s disease （AD）. But the roles of hyperinsulinemia （HI） and insulin （IR） in the development of AD are still controversial. This study was designed to evaluate or IR influenced the cognitive functions of older cohort. Methods The cognitive functions of 328 consecutive elderly patients were evaluated with a battery of cognitive rating scales. Their fasting blood glucose （FBg） and fasting insulin （FINS） were analyzed and IR was calculated with modified-Homa. The cognitive scores in different groups and the correlation of cognitive functions with HI or IR were analyzed. Results In our study, there were 180 participants with HI and 148 without HI, and 192 with iR and 136 without IR. The participants with HI showed worse cognitive functions than those without HI in MMSE, MOCA, CDR, orientation, delayed memory, and attention/calculation domains. Similarly, the elderly with IR had lower cognitive scores than those without IR in MMSE, MOCA, CDR, gDS, orientation, delayed memory, and attention/calculation domains. The insulin levels and Homa IR had negative correlation with the scores of MMSE and delayed memory, not only in the model I adjusted for FBg and diabetes history, but also in the model 2 adjusted for all nine demographic characteristics. Conclusion HI and IR are important risk factors for cognitive decline of the elderly, especially for the dysfunctions in delayed memory domains.

Projections of surface air temperature and precipitation in the 21st century in the Qilian Mountains,Northwest China,using REMO in the CORDEX

Qilian Mountains(QM)is an important ecological security barrier in China and has been significantly affected by climate change,it is therefore of great importance and necessity to project its future climate change using high-resolution climate models because of mountainous areas in the QM and relatively few targeted simulation *** this study,we used the simulations of the regional climate model REMO with 25 km spatial resolution,driven by three different global climate models(MPI-ESM-MR,NorESM1-M,and HadgEM2-ES),to evaluate how annual and seasonal mean surface air temperature and precipitation in the QM are likely to change for three future periods(2011-2040,2041-2070,and 2071-2100)under two representative concentration pathways(RCP2.6 and RCP8.5).The REMO model,shows noticeable cold and wet biases compared to observations for the reference period(1971-2000)and air temperature simulation outperforms precipitation *** REMO simulations exhibit a warm and wet centre around lake,indicating that the simulation are likely influenced by *** under RCP2.6 show regional warming reaching 1.74℃ during 2011-2100,characterized by an initial increase and a decrease *** RCP8.5,air temperatures increase monotonously from 2011 to 2100,with a warming magnitude of 5.36℃ for 2071-2100 relative to *** overall change in regional-average annual precipitation is not evident during 2011-2100,with some increases or decreases in certain time *** the 2071-2100 both the strongest warming and precipitation increase are projected to occur in winter under both scenarios,while precipitation in summer and autumn is projected to decrease in the east of the QM for the three future *** results suggest that the QM is likely to experience drought conditions in warm seasons in the future,which could impact agricultural and livestock production.

Runoff Characteristics of the Nen River Basin and its Cause

Syntheses of three terbium complexes as fluorescent probes and their application on the pH detection of routine urine test

By modifying the salicylic-acid moiety with electron-withdrawing or –donating groups, three new terbium complexes（LⅠ·Tb, LⅡ·Tb, L·（Ⅲ）Tb） based on tripodal carboxylate ligands were synthesized. Due to different pull-push electronic effects of ligands, the fluorescence intensities of these terbium complexes significantly varied, that is： LⅡ·Tb 〉L（Ⅲ）·Tb〉 LⅠ·Tb. Meanwhile, the characteristic peaks at 492 nm（5D4→7F6） and 547 nm（5 D4→7F5） showed ＂Off–On–Off＂ fluorescence response to various p H conditions,which indicated that all of them can be used as the highly sensitive pH fluorescent probes. Notably, using LⅡ·Tb with the best fluorescence performance as a probe, some patients＇ urine samples can be easily monitored through the response triggered by pH value. Therefore, LⅡ·Tb has the potential to auxiliarily diagnose some diseases in clinical practice through p H detection of routine urine test.
