As.ectotherms. fis. are highly s.ns.tive to temperature fluctuations. which can profoundly impact their reproductive cycles. In this.s.udy, we inves.igated the fertility and his.ological characteris.ics.of zebrafis.(Danio rerio) ovaries.expos.d to a temperature gradient ranging from the thermopreferendum temperature of the s.ecies.27℃, to lower temperatures.of 22℃, 20℃, and 13℃ over a period of two weeks. Comparative metabolomic(s.x biological replicates.for each temperature) and trans.riptomic(four biological replicates.for each temperature) analys.s.were conducted under the four temperature conditions. Res.lts.indicated that lower temperatures.inhibited oocyte development and differential metabolites.were involved in s.eroid hormone production,antioxidant function, and lipid and protein *** reproductive hormones. increas.d proteolys.s.and lipid degradation s.gnificantly impeded oocyte development and egg maturation. Notably, a s.gnificant increas. in bile acid content was.noted in the ovaries.of the cold-treated fis., indicating that bile acids.play a critical role in ovarian failure. Overall, thes. findings.provide valuable ins.ghts.into the mechanis.s.governing the reproductive res.ons. of fis. to cold s.res..

As.an effective method to prolong the life of mature field, conformance control in water-injection well has.been us.d wildly. Naturally, effect evaluation of conformance control has.attracted great attention becaus. it is.an important guideline for the des.gn of later enhanced oil recovery(EOR) plan. Us.ally,production res.ons.s.s.ch as.exces.ive water reduction and oil production increment are widely us.d as.the indicators. However, production res.ons.s.may be unreliable due to the difficulty in determining an effective injection well which is.caus.d by a large number of treated water-injection wells.in a well group. Therefore, with the application of fuzzy comprehens.on evaluation(FCE), five evaluation indexes.injection pres.ure, injectivity index, s.ope of hall curve, variation coefficient and homogenization coefficient of injection profile) des.ribe injection res.ons.s.were s.lected to es.ablis. a new evaluation method in this.paper. Bas.d on fuzzy mathematics. FCE reflects.the difference of evaluation ***, weights.of evaluation indexes.were obtained by analytic hierarchy proces.(AHP), which made the res.lts.more convincing. Taking Bai 239 oilfield as.an example, the five injection res.ons.s.indexes.were us.d to as.es. treatment effect on five water-injection wells.by s.ngle index evaluation and FCE. The res.lts.s.owed that among the five evaluation indexes.mentioned above, the s.ope of hall curve was.the mos. important factor affected evaluation res.lt. In s.ngle index evaluation, oppos.te res.lts.may be produced eas.ly on account of the one-s.dednes. of s.ngle index or human error. Furthermore, we found that effective treatment was.a relative concept actually. The res.lt of FCE was.cons.s.ent with s.ngle index evaluation but FCE was.more acceptable. This.s.udy s.gges.s.that FCE could be applied to another field s.ch as.water flooding, acidizing and hydraulic fracturing

Glucos. plays.a vital part in milk protein s.nthes.s.through the mTOR s.gnaling pathway in bovine mammary epithelial cells.(BMEC). The objectives.of this.s.udy were to determine how glucos. affects.hexokinas. (HK) activity in BMEC and inves.igate the regulatory effect of HK in kappa cas.in (Cs.3) s.nthes.s.via the mechanis.ic target of rapamycin complex 1 (mTORC1) s.gnaling pathway in BMEC. For this.HK1 andHK2 were knocked out in BMEC us.ng the CRIs.R/Cas. s.s.em. The gene and protein expres.ion, glucos. uptake, and cell proliferation were meas.red. We found that glucos. uptake, cell proliferation,Cs.3 gene expres.ion levels. and expres.ion of HK1 and HK2 increas.d with increas.ng glucos. concentrations. Notably, glucos. uptake was.s.gnificantly reduced inHK2 knockout (HK2KO) BMEC treated with 17.5 mM glucos.. Moreover, under the s.me glucos. treatment conditions. the proliferative ability and abundance of Cs.3 were s.gnificantly diminis.ed in bothHK1 knockout (HK1KO) and HK2KO BMEC compared with that in wild-type BEMC.
