Although the biochemical dissection of parasitoid-host interactions is becoming well characterized, the molecular knowledge concerning them is minimal. In order to understand the molecular bases of the host immune response to parasitoid attack, we explored the response of Papilio xuthus parasitized by the endoparasitic wasp Pteromalus puparum using proteomic approach. By examining the differential expression of plasma proteins in the parasitized and unparasitized host pupae by two-dimensional (2D) electrophoresis, 16 proteins were found to vary in relation to parasitization compared with unparasitized control samples. All of them were submitted to identification by mass spectrometry coupled with a database search. The modulated proteins were found to fall into the following functional groups: humoral or cellular immunity, detoxification, energy metabolism, and others. This study contributes insights into the molecular mechanism of the relationships between parasitoids and their host insects.

This report presents the cDNA cloning of a storage protein, PraAry, from Pieris rapae and investigates its expression regulated by parasitism of an endoparasitoid wasp Pteromalus puparum. The full-length eDNA of PraAry is 2 270 nucleotides and contains a 2 121 nucleotide open reading frame encoding 707 amino acids with calculated molecular weights of approximately 83 kDa. Analysis of the primary protein sequence revealed that it possesses a signal peptide of 16 amino acids at the N-terminus and contains two highly conserved storage protein signature motifs. According to both phylogenetic analysis and the criteria for amino acid composition, PraAry belongs to the subfamily of arylphorin-type storage protein （1.42% methionine and 18.82% aromatic amino acids）. Reverse transcription- polymerase chain reaction analysis indicated that the transcriptional level of PraAry mRNA in P. rapae pupae fat body is inducible in response to parasitism by P. puparum.

Parasitism by the endoparasitoid wasp Pteromalus puparum causes alterations in the plasma proteins of Pieris rapae. Analysis of plasma proteins using a proteomic approach showed that seven proteins were differentially expressed in the host pupae after 24-h parasitism. They were masquerade-like serine proteinase homolog (MSPH), enolase (Eno), bilin-binding protein (BBP), imaginal disc growth factor (IDGF), ornithine decarboxylase (ODC), cellular retinoic acid binding protein (CRABP), and one unknown function protein. The full length cDNA sequences of MSPH, Eno, and BBP were successfully cloned using rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR). Reverse transcriptase-polymerase chain reaction (RT-PCR) analysis indicated that the transcript levels of MSPH and BBP in the fat bodies of host pupae were inducible in response to the parasitism and their variations were consistent with translational changes of these genes after parasitism, while the transcript levels of Eno and IDGF were not affected by parasitism. This study will contribute to the better understanding of the molecular bases of parasitoid-induced host alterations associated with innate immune responses, detoxification, and energy metabolism.

Various SARS-CoV-2-related coronaviruses have been increasingly identified in pangolins,showing a potential threat to *** we report the infectivity and pathogenicity of the SARS-CoV-2-related virus,PCoV-GX/P2V,which was isolated from a Malayan pangolin(Manis javanica).PCoV-GX/P2V could grow in human hepatoma,colorectal adenocarcinoma cells,and human primary nasal epithelial *** replicated more efficiently in cells expressing human angiotensin-converting enzyme 2(hACE2)as SARS-CoV-2 *** intranasal inoculation to the hACE2-transgenic mice,PCoV-GX/P2V not only replicated in nasal turbinate and lungs,but also caused interstitial pneumonia,characterized by infiltration of mixed inflammatory cells and multifocal alveolar *** population immunity established by SARS-CoV-2 infection and vaccination may not protect people from PCoV-GX/P2V *** findings further verify the hACE2 utility of PCoV-GX/P2V by in vivo experiments using authentic viruses and highlight the importance for intensive surveillance to prevent possible cross-species transmission.

为揭示蝶蛹金小蜂Pteromalus puparum抑制寄主菜粉蝶Pieris rapae血细胞免疫反应的分子机制,克隆了菜粉蝶肌动蛋白、肌动蛋白解聚因子及微管蛋白cDNA,并研究了蝶蛹金小蜂寄生对其转录水平的影响。结果显示:菜粉蝶肌动蛋白cDNA ORF为1131bp,编码377aa,预测分子量为41.78kDa,pI为5.29,与其他昆虫肌动蛋白的相似性非常高,在90%以上。氨基酸组成特点和系统进化分析表明,克隆到的菜粉蝶肌动蛋白基因属细胞质型肌动蛋白。菜粉蝶肌动蛋白解聚因子cDNA全长为1243bp,ORF为447bp,编码149aa,预测分子量为16.97kDa,pI为7.11,与家蚕Bombyx mori、赤拟谷盗Tribolium castaneum、意大利蜜蜂Apis mellifera和桑粉介壳虫Maconellicoccus hirsutus肌动蛋白解聚因子的相似性分别为97%,87%,89%和72%。菜粉蝶微管蛋白cDNA全长为1757bp,ORF为1344bp,编码448aa,预测分子量为50.38kDa,pI为4.86,与家蚕β型微管蛋白1,2,3和4的相似性分别为97%,97%,87%和93%。系统进化分析表明,克隆到的菜粉蝶微管蛋白基因属β型微管蛋白。RT-PCR分析表明,蝶蛹金小蜂寄生能抑制肌动蛋白、肌动蛋白解聚因子及微管蛋白基因在菜粉蝶蛹血细胞中的转录水平。由此推断蝶蛹金小蜂调控寄主血细胞骨架相关蛋白基因的转录是该蜂抑制寄主血细胞免疫反应的分子机制之一。

昆虫主要依靠先天免疫反应来抵御外源异物的入侵,而与血淋巴黑化及抗菌肽合成等过程密切相关的丝氨酸蛋白酶激活级联反应在其中起着重要作用。为阐明丝氨酸蛋白酶在菜粉蝶Pieris rapae免疫中的作用,本文通过简并引物RT-PCR克隆获得了菜粉蝶丝氨酸蛋白酶家族基因Pr-SP1的cDNA片段,并利用RACE法扩增获得其全序列。该cDNA序列长1489bp,其中开放阅读框长1059bp,共编码353个氨基酸残基。Pr-SP1含一长度为20个氨基酸残基的信号肽序列,其蛋白理论分子量为36.85kDa,理论等电点为6.41。多序列比对结果表明,Pr-SP1与其他昆虫的同源蛋白基因序列上存在较高一致性,在N端有一个发夹结构域,而C端是一个具有催化活性的结构域。实时荧光定量RT-PCR及免疫印迹结果表明,蛹期Pr-SP1主要在颗粒血细胞内进行转录,其蛋白产物主要定位在血浆;Pr-SP1在不同虫态及虫龄都有转录,其蛋白产物在不同虫态及虫龄都有表达,其中5龄幼虫最高,卵期最低;Pr-SP1的转录水平及其蛋白产物的表达水平均会被大肠杆菌Escherichia coli、藤黄微球菌Micrococcus luteus和巴斯德毕赤酵母Pichia pastoris诱导。根据这些结果分析认为,Pr-SP1属于Sptzle蛋白酶前体激活酶,并参与菜粉蝶的先天免疫反应。

寄生蜂是重要且种类最为丰富的膜翅目昆虫类群之一,也是极具价值的害虫生物控制因子。寄生蜂携带有不同类型的活性因子,包括毒液、多分DNA病毒类病毒颗粒、卵巢蛋白、畸形细胞及幼虫分泌物等,用于调控寄主害虫的免疫反应、发育等重要生理过程,以确保成功寄生并确保其子代在寄主害虫体内(内寄生蜂)或体表(外寄生蜂)正常发育,最终可导致寄主害虫死亡,从而有效控制寄主害虫种群数量。目前已有诸多与寄生蜂调控寄主害虫内在机理相关的研究报道,该领域也已成为昆虫寄生学与生理学的研究热点之一。本文仅从寄生蜂寄生因子多样性、寄生蜂调控寄主害虫免疫及发育的机理等方面,对相关的最新研究进展作一概述。

Aims:Surveys and research on the applications of the hepatic venous pressure gradient(HVPG)are important for understanding the current status and future development of this technology in *** article aimed to investigate the status of hepatic venous pressure gradient measurement in China in ***:We investigated the overall status of HVPG technology in China-including hospital distribution,hospital level,annual number of cases,catheters used,average cost,indications,and current challenges by using online *** counting the number and percentages of cases of these results,we hope to clarify the current status of HVPG measurements in ***:According to the survey,85 hospitals in China used HVPG technology in 2022 distributed across 29 provinces.A total of 4989 HVPG measurements were performed in all of the surveyed hospitals in 2022,of which 2813 cases(56.4%)were measured *** average cost of HVPG measurement was 5646.8±2327.9 *** the clinical teams who performed the measurements(sometimes multiple per hospital),94.3%(82/87)used the balloon method,and the majority of the teams(72.4%,63/87)used embolectomy ***:This survey clarified the clinical application status of HVPG in China and confirmed that some medical institutions in China have established a foundation for this *** is still necessary to continue promoting and popularizing this technology in the future.

为了揭示重金属对昆虫细胞免疫的影响及其机理,本文采用血细胞计数、台盼蓝染色、延展与包囊率测定,以及显微与超微结构观察等方法,以经重金属Cd2+(浓度为150μg/g)处理的棕尾别麻蝇Boettcherisca peregrina及其对照为研究对象,分别测定了处理组与对照组血细胞的数量与存活率、延展与包囊率,观察其形态结构,并比较了两组间的差异。结果表明:棕尾别麻蝇初产幼虫经连续喂饲含Cd2+的饲料24,48,72和96h后,与对照相比其血细胞总数和存活率显著下降,而血细胞的延展率和包囊作用的显著下降则分别出现于取食72和48h之内,此后则下降不显著;由Cd2+处理幼虫发育形成蛹的血细胞包囊作用也显著低于对照。形态结构观察结果表明,Cd2+处理对幼虫的原血细胞、浆血细胞、颗粒血细胞和类绛色血细胞的显微形态影响不大,但可导致部分浆血细胞不能产生伪足;但均能导致各类血细胞的超微结构发生不同程度的变化,其中主要变化包括:细胞膜受损或破裂,染色质呈现凝聚,线粒体和内质网等细胞器明显减少,以及胞质内出现空囊泡。结果说明重金属Cd2+对棕尾别麻蝇的血细胞可具有毒害作用。
