Pharmacokinetic Disposition of Streptomycin Sulfate in Japanese Eel (<i>Anguilla japonica</i>) after Oral and Intramuscular Administrations

The ciliate Ichthyophthirius multifiliis is one of the most pathogenic parasites of fish maintained in captivity. In this study, effects of bacterial extracellular products of Streptomyces griseus SDX-4 against I. multifiliis were determined. The fermentation liquor of S. griseus was extracted successively in a separating funnel with petroleum ether, ethyl acetate, and n-butanol. In vitro assays revealed that the n-butanol extracts（NBu-E） and ethyl acetate（Eto-E） of S. griseus were observed to be more effictive against theronts than the other extracts with median effective concentration（EC50） values of 0.86 mg L-1 and 12.5 mg L-1, respectively, and significantly reduced the survival of the tomonts and the total number of theronts released by the tomonts（P＜0.05）. All encysted tomonts were killed when the concentration of NBu-E was 30.0 mg L-1. Results of in vivo test demonstrated that the number of I. multifiliis trophonts on the grasscarp treated with NBu-E was markedly lower compared to the control group at 11 days after exposed to theronts（p＜0.05）. In the control group, 100% mortality was observed owing to heavy I. multifiliis infection at 11 days after the exposure. On the other hand, only 9.5% mortality owing to parasite infection was recorded in the groups treated with the NBu-E（30 mg L-1）. The median lethal dose（LD50） of NBu-E for grasscarp was 152.4 mg L-1. Our results indicate that n-butanol extract of S. griseus will be useful in aquaculture for controlling I. multifiliis infections.

Pharmacokinetics and residue elimination of streptomycin sulfate (STR) are important in the determination of optimal dosage regimens and in establishing safe withdrawal periods in farmed fishes. The pharmacokinetics of STR was studied after a single dose (50 mg/kg) of intramuscular (i.m.) or oral gavage (p.o.) administration to Japanese eel (Anguilla japonica) in freshwater at 25°C. Eight fish per sampling point were examined after treatment. Plasma and muscle were collected and analyzed by high-performance liquid chromatography (HPLC) method with 0.05 μg/ml detection limit. The data of pharmacokinetics conformed to the two-compartment open model for intramuscular and one-compartment open model for oral administrations. After intramuscular administration, the elimination half-life (t1/2β) was calculated to be 11.346 h, the maximum plasma concentration (Cmax) to be 29.524 μg/ml, the time to peak plasma streptomycin concentration (Tmax) to be 0.218 h, and the area under the plasma concentration-time curve (AUC) to be 90.206 μg/ml?h. Following p.o. administration, the corresponding estimates were 13.239 h, 0.346 μg/ml, 11.960 h, and 12.356 μg/ml?h. After intramuscular administration, a therapeutic concentration of the drug was maintained for 12 hours in the plasma, however, a therapeutic level could not be achieved after oral administration, and the results suggest that the drug can be used clinically by intramuscular administration against streptomycin susceptible systemic infections in Japanese eel.

BACKGROUND Despite laparoscopic total hysterectomy provided more benefits to patients and has become one of the most commonly used surgical methods for total ***,vaginal cuff dehiscence is a serious complication after laparoscopic total hysterectomy and so far,there are limited studies on vaginal cuff dehiscence after a total *** To investigate the factors influencing vaginal cuff dehiscence after laparoscopic hysterectomy and the psychological state of the *** This study retrospectively,univariately,and multivariately analyzed the clinical data of patients who experienced vaginal cuff dehiscence after laparoscopic hysterectomy in Yuyao People’s Hospital from January 2015 to December *** regression was utilized to analyze the high-risk factors of vaginal cuff *** Among 1459 cases with laparoscopic hysterectomy,9 reported vaginal cuff dehiscence postoperatively,with an incidence of 0.617%.Univariate logistic regression analysis revealed that preoperative hemoglobin value[odds ratio(OR)=5.12,P=0.016],vaginal cuff suture method(OR=0.26,P=0.048),and postoperative first sexual lifetime(OR=15.86,P=0.002)were associated with vaginal cuff dehiscence following laparoscopic *** logistic regression analysis revealed that preoperative hemoglobin value of<90 g/L(OR=5.17,P=0.015)and time interval between postoperative first sexual life and operation of<3 months(OR=54.00,P=0.004)are independent risk factors for vaginal cuff dehiscence after laparoscopic *** Postoperative hemoglobin and the time interval between postoperative first sexual life and operation were the independent factors of developing vaginal cuff dehiscence after laparoscopic hysterectomy.

Objective: Esophageal squamous cell carcinoma(ESCC), a commonly encountered malignant tumor in the gastrointestinal tract, poses a significant health burden. Bufalin, a pharmacologically active molecule, has been shown to exhibit antitumor activity against various types of cancers. This study investigates the molecular mechanism underpinning the effects of bufalin on ESCCs. Materials and Methods: The impact of bufalin on the proliferation and migration of ESCC cells was evaluated through the utilization of the Cell Counting Kit-8(CCK-8), scratch assay, and transwell assay. In addition, ribonucleic acid(RNA) sequencing was performed to identify genes that were abnormally expressed in response to bufalin treatment. Western blotting was utilized to ascertain the expression levels of protein arginine methyltransferase-6(PRMT6), phosphorylated AKT(p-AKT), and phosphorylated mammalian target of rapamycin(p-m TOR). Cell transfection was then performed to observe the rescue effect of PRMT6 on bufalin. Results: Bufalin displayed a significant time-dependent inhibition of the proliferation, migration, and invasion of ECA109 cells. An RNA sequencing(RNA-seq) analysis revealed that PRMT6 expression was downregulated in the cells treated with bufalin. PRMT6 promoted the proliferation, migration, and invasive potential of ECA109 cells. The overexpression of PRMT6 boosted p-AKT and p-m TOR levels in ECA109 cells and reversed bufalin inhibition. Conclusions: These findings indicate that bufalin exerts its inhibitory effects on ESCCs through the PRMT6/AKT/m TOR signaling pathway. These findings lay the groundwork for bufalin as a promising therapeutic candidate for the treatment of ESCC.

BACKGROUND Type Ⅱ diabetes mellitus(T2DM)has been associated with increased risk of colon cancer(CC)and worse prognosis in patients with *** effects of T2DM on postoperative chemoresistance rate(CRR)and long-term disease-free survival(DFS)and overall survival(OS)in patients with stage Ⅲ CC who receive curative resection remain *** To investigate whether T2DM or glycemic control is associated with worse postoperative survival outcomes in stage Ⅲ *** This retrospective cohort study included 278 patients aged 40-75 years who underwent surgery for stage Ⅲ CC from 2018 to *** on preoperative T2DM history,the patients were categorized into non-DM(n=160)and DM groups(n=118).The latter was further divided into well-controlled(n=73)and poorly controlled(n=45)groups depending on the status of glycemic ***,OS,and CRR were compared between the groups and Cox regression analysis was used to identify risk *** Patients in the DM and non-DM groups demonstrated similar DFS,OS,and CRR(DFS:72.03%vs 78.75%,P=0.178;OS:81.36%vs 83.12%,P=0.638;CRR:14.41%vs 7.5%,P=0.063).Poorly controlled DM was associated with a significantly worse prognosis and higher CRR than well-controlled DM(DFS:62.22%vs 78.07%,P=0.021;OS:71.11%vs 87.67%,P=0.011;CRR:24.40%vs 8.22%,P=0.015).High preoperative fasting plasma glucose[DFS:Hazard ratio(HR)=2.684,P<0.001;OS:HR=2.105,P=0.019;CRR:HR=2.214,P=0.005]and glycosylated hemoglobin levels(DFS:HR=2.344,P=0.006;OS:HR=2.119,P=0.021;CRR:HR=2.449,P=0.009)indicated significantly poor prognosis and high CRR,while T2DM history did not(DFS:HR=1.178,P=0.327;OS:HR=0.933,P=0.739;CRR:HR=0.997,P=0.581).CONCLUSION Increased preoperative fasting plasma glucose and glycosylated hemoglobin levels,but not T2DM history,were identified as risk factors associated with poor postoperative outcomes and high CRR in patients with stage Ⅲ CC.
