Aim:To develop homogeneous calcium mobilization assay for high-throughputscreening(HTS)of mas-related gene(Mrg)receptor ***:CHO-Klcells stably expressing the full-length MrgD receptor and a calcium-sensitive dyewere used to develop an HTS assay based on intracellular calcium *** was applied to large-scale screening of a library containing 8000 syntheticcompounds and natural product extracts,cAMP measurements were carried outto verify the bioactivities of the hits found by the calcium mobilization *** approaches were also employed in the identification of the MrgA1 recep-tor agonists following HTS of 16000 ***:EC50values of the positivecontrol compounds(μ-alanine for MrgD receptor and dynorphin A for MrgA1receptor)determined by the calcium mobilization assay were consistent with thosereported in the literature,and the Z’factors were 0.65 and 0.50 for MrgD andMrgA1 receptor assay,*** 31 compounds for the MrgD receptorand 48 compounds for the MrgA1 receptor showing20% of the maximal agonistactivities found in the controls were initially identified as *** screen-ing confirmed that 2 compounds for each receptor possessed specific *** cAMP level measurements indicated that the 2 confirmedhits displayed the functionality of the MrgD receptor ***:Aseries of validation studies demonstrated that the homogeneous calcium mobili-zation assay developed was highly efficient,amenable to automation and a robusttool to screen potential MrgD and MrgA1 receptor *** application maybe expanded to other G-protein coupled receptors that mobilize calcium influxupon activation.

Developing effective strategies to regulate graphene’s conduction loss and polarization has become a key to expanding its application in the electromagnetic wave absorption(EMWA)*** on the unique energy band structure of graphene,regulating its bandgap and electrical properties by introducing heteroatoms is considered a feasible ***,metal-nitrogen doping reduced graphene oxide(M–N-RGO)was prepared by embedding a series of single metal atoms M–N_(4) sites(M=Mn,Fe,Co,Ni,Cu,Zn,Nb,Cd,and Sn)in RGO using an N-coordination atom-assisted *** composites had adjustable conductivity and polarization to optimize dielectric loss and impedance matching for efficient EMWA *** results showed that the minimum reflection loss(RL_(min))of Fe–N-RGO reaches−74.05 dB(2.0 mm)and the maximum effective absorption bandwidth(EAB_(max))is 7.05 GHz(1.89 mm)even with a low filler loading of only 1 wt%.Combined with X-ray absorption spectra(XAFS),atomic force microscopy,and density functional theory calculation analysis,the Fe–N_(4) can be used as the polarization center to increase dipole polarization,interface polarization and defect-induced polarization due to d-p orbital hybridization and structural ***,electron migration within the Fe further leads to conduction loss,thereby synergistically promoting energy *** study demonstrates the effectiveness of metal-nitrogen doping in regulating the graphene′s dielectric properties,which provides an important basis for further investigation of the loss mechanism.

BACKGROUND Esophageal carcinoma(EC)presents a significant public health issue in China,with its prognosis impacted by myriad *** creation of a reliable prog-nostic model for the overall survival(OS)of EC patients promises to greatly advance the customization of treatment *** To create a more systematic and practical model that incorporates clinically significant indicators to support decision-making in clinical *** This study utilized data from a prospective longitudinal cohort of 3127 EC patients treated at Chongqing University Cancer Hospital between January 1,2018,and December 12,*** the least absolute shrinkage and selection operator regression alongside multivariate Cox regression analyses helped pinpoint pertinent variables for constructing the *** efficacy was assessed by concordance index(C-index),area under the receiver operating characteristic curve(AUC),calibration curves,and decision curve analysis(DCA).RESULTS Nine variables were determined to be significant predictors of OS in EC patients:Body mass index(BMI),Karnofsky performance status,TNM stage,surgery,radiotherapy,chemotherapy,immunotherapy,platelet-to-lymphocyte ratio,and albumin-to-globulin ratio(ALB/GLB).The model demonstrated a C-index of 0.715(95%CI:0.701-0.729)in the training cohort and 0.711(95%CI:0.689-0.732)in the validation *** the training cohort,AUCs for 1-year,3-year,and 5-year OS predictions were 0.773,0.787,and 0.750,respectively;in the validation cohort,they were 0.772,0.768,and 0.723,respectively,illustrating the model's *** curves and DCA verified the model's predictive accuracy and net *** A novel prognostic model for determining the OS of EC patients was successfully developed and validated to help clinicians in devising individualized treatment schemes for EC patients.
