Neurotoxicity of tetramethylammonium ion on larval and juvenile zebrafish:Effects on neurobehaviors and multiple biomarkers

Tetramethylammonium hydroxide(TMAH)is an important compound that utilized and released by the rapidly expanding semiconductor industry,which could hardly be removed by the conventional wastewater treatment *** a cholinergic agonist,the tetramethylammonium ion(TMA^(+))has been reported to induce toxicity to muscular and respiratory systems of mammals and human,however the toxicity on aquatic biota remains poorly *** investigated the neurotoxic effects of TMA^(+)exposure on zebrafish,based on neurobehavior tests and a series of *** inhibitions on the swimming distance of zebrafish larvae were observed when the exposure level exceeded 50 mg/L,and significant alterations on swimming path angles(straight and deflective movements)occurred even at 10 mg/*** tested neurobehavioral endpoints of zebrafish larvae were significantly positively correlated with reactive oxygen species(ROS)and malondialdehyde(MDA),significantly negatively related with the activities of antioxidant enzymes,but not significantly correlated with the level of acetylcholinesterase(AChE).Such relationship indicates that the observed neurotoxic effects on swimming behavior of zebrafish larvae is mainly driven by oxidative stress,rather than the alterations of *** the highest exposure concentration(200 mg/L),TMA^(+)evoked more severe toxicity on zebrafish juveniles,showing significantly stronger elevation on the MDA activity,and greater inhibitions on the activities of antioxidant enzymes and ACh E,suggesting juveniles were more susceptible to TMA^(+)exposure than larval zebrafish.

Centrifuge modeling of unreinforced and multi-row stabilizing piles reinforced landslides subjected to reservoir water level fluctuation

With the construction of the Three Gorges Reservoir dam,frequent reservoir landslide events have been *** recent years,multi-row stabilizing piles(MRSPs)have been used to stabilize massive reservoir landslides in *** this study,two centrifuge model tests were carried out to study the unreinforced and MRSP-reinforced slopes subjected to reservoir water level(RWL)operation,using the Taping landslide as a *** results indicate that the RWL rising can provide lateral support within the submerged zone and then produce the inward seepage force,eventually strengthening the slope ***,a rapid RWL drawdown may induce outward seepage forces and a sudden debuttressing effect,consequently reducing the effective soil normal stress and triggering partial pre-failure within the RWL fluctuation ***,partial deformation and subsequent soil structure damage generate excess pore water pressures,ultimately leading to the overall failure of the reservoir *** study also reveals that a rapid increase in the downslope driving force due to RWL drawdown significantly intensifies the lateral earth pressures exerted on the ***,the MRSPs possess a considerable reinforcement effect on the reservoir landslide,transforming the overall failure into a partial deformation and failure situated above and in front of the *** mechanical transfer behavior observed in the MRSPs demonstrates a progressive alteration in relation to RWL *** the RWL rises,the mechanical states among MRSPs exhibit a growing *** shear force transfer factor(*** ratio of shear forces on pile of the n th row to that of the first row)increases significantly with the RWL *** indicates that the mechanical states among MRSPs tend toward an enhanced *** insights gained from this study contribute to a more comprehensive understanding of the failure mechanisms of reservoir landslides and the mechanical behavior of M

Model-based reinforcement learning is a promising direction to improve the sample efficiency of reinforcement learning with learning a model of the *** model learning methods aim at fitting the transition data,and commonly employ a supervised learning approach to minimize the distance between the predicted state and the real *** supervised model learning methods,however,diverge from the ultimate goal of model learning,i.e.,optimizing the learned-in-the-model *** this work,we investigate how model learning and policy learning can share the same objective of maximizing the expected return in the real *** find model learning towards this objective can result in a target of enhancing the similarity between the gradient on generated data and the gradient on the real *** thus derive the gradient of the model from this target and propose the Model Gradient algorithm(MG)to integrate this novel model learning approach with policy-gradient-based policy *** conduct experiments on multiple locomotion control tasks and find that MG can not only achieve high sample efficiency but also lead to better convergence performance compared to traditional model-based reinforcement learning approaches.

Frequency-modulated continuous-wave multiplexed gas sensingbased on optical frequency comb calibration

Laser absorption spectroscopy has proven to be an effective approach for gas sensing, which plays an important rolein the fields of military, industry, medicine and basic research. This paper presents a multiplexed gas sensing system basedon optical frequency comb (OFC) calibrated frequency-modulated continuous-wave (FMCW) tuning nonlinearity. Thesystem can be used for multi-parameter synchronous measurement of gas absorption spectrum and multiplexed opticalpath. Multi-channel parallel detection is realized by combining wavelength division multiplexing (WDM) and frequencydivision multiplexing (FDM) techniques. By introducing nonlinear optical crystals, broadband spectrum detection is simultaneouslyachieved over a bandwidth of hundreds of nanometers. An OFC with ultra-high frequency stability is used asthe frequency calibration source, which guarantees the measurement accuracy. The test samples involve H13C14N, C_(2)H_(2)and Rb vapor cells of varying densities and 5 parallel measurement experiments are designed. The results show that themeasurement accuracies of spectral absorption line and the optical path are 150 MHz and 20 m, respectively. The schemeoffers the advantages of multiplexed, multi-parameter, wide spectrum and high resolution detection, which can realize theidentification of multi-gas components and the high-precision inversion of absorption lines under different *** proposed sensor demonstrates great potential in the field of high-resolution absorption spectrum measurement for gassensing applications.

Symmetrization of quasi-regular patterns with periodic tilting of regular polygons

Computer-generated aesthetic patterns arewidely used as design materials in various fields. Themost common methods use fractals or dynamicalsystems as basic tools to create various patterns. Toenhance aesthetics and controllability, some researchershave introduced symmetric layouts along with thesetools. One popular strategy employs dynamical systemscompatible with symmetries that construct functionswith the desired symmetries. However, these aretypically confined to simple planar symmetries. Theother generates symmetrical patterns under theconstraints of tilings. Although it is slightly moreﬂexible, it is restricted to small ranges of tilingsand lacks textural variations. Thus, we proposed anew approach for generating aesthetic patterns bysymmetrizing quasi-regular patterns using general kuniformtilings. We adopted a unified strategy toconstruct invariant mappings for k-uniform tilings thatcan eliminate texture seams across the tiling ***, we constructed three types of symmetriesassociated with the patterns: dihedral, rotational, andreﬂection symmetries. The proposed method can beeasily implemented using GPU shaders and is highlyefficient and suitable for complicated tiling with regularpolygons. Experiments demonstrated the advantages of our method over state-of-the-art methods in terms ofﬂexibility in controlling the generation of patterns withvarious parameters as well as the diversity of texturesand styles.

Accuracy evaluation for in-situ machining reference points binocular measurement based on credibility probability

Passive binocular measurement systems are being increasingly utilized in the in-situ industries of automobiles,aviation,and aerospace,*** to their excellent qualities of accuracy,efficiency,and cost *** the barrier of evaluating the accuracy of measured objects resulted from the unequal equivalent focal length and quantization of pixels,has limited their further development and application of high requirements for in-situ machining,e.g.,the measurement of machining reference points for the positioning of robotic drilling in aerospace *** this paper,an accuracy evaluation method is proposed to address the ***,the unequal equivalent focal length is considered to improve the accuracy of 3D ***,the credibility probability model is developed to calculate the probability of the observed error in the public view of the binocular measurement system and indicates the direction of ***,the in-situ experiment is carried out to validate the method within the effective public view range of 300 mm×300 *** experiment results show that the RMSs of observed errors are superior to 0.035 mm,and the credibility probabilities are all higher than 0.91;the maximum 3D reconstruction accuracy improvement is 60.3%,with the error reduced from 0.078 mm to 0.031 mm.

Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among *** this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its ***:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro ***-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of *** counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ***:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 *** downregulated glutathione peroxidase 4(GPX4)m *** docking models predicted that solute carrier family 7 member 11(SLC7A11)and GPX4 were covalently bound by ***,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 ***:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis *** findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

Next-generation sequencing(NGS),represented by Illumina platforms,has been an essential cornerstone of basic and applied ***,the sequencing error rate of 1 per 1000 bp(10^(−3))represents a serious hurdle for research areas focusing on rare mutations,such as somatic mosaicism or microbe *** examining the high-fidelity sequencing methods developed in the past decade,we summarized three major factors underlying errors and the corresponding 12 strategies mitigating these *** then proposed a novel framework to classify 11 preexisting representative methods according to the corresponding combinatory strategies and identified three trends that emerged during methodological *** further extended this analysis to eight long-read sequencing methods,emphasizing error reduction ***,we suggest two promising future directions that could achieve comparable or even higher accuracy with lower costs in both NGS and long-read sequencing.

Enhanced preparation of dendritic metallic vanadium in recyclable NaCl-KCl-VCl_(2) molten salt by V_(2)CO electrolysis

A promising method for the preparation of pure metallic vanadium via V_(2)CO anode electrolysis was proposed and *** simple and controllable synthesis of a molten chloride salt containing VCl_(2) by an insitu reaction between VCl_(3) and metallic vanadium was verified through thermodynamic analysis and ***-KCl-VCl_(2) molten salt exhibited excellent recycling performance and could be repetitively used to prepare metallic vanadium by V_(2)CO *** V^(2+) ions remained in the molten salt after *** electrolysis conditions,such as the cathode diameter,current density,V^(2+) ion concentration,and temperature,were optimised to maximise the current *** was also simulated to reveal the regulatory *** highest current efficiency was 85%.The purity of metallic vanadium was up to 99.8%.In this study,an easy and efficient preparation of pure metallic vanadium was achieved.
