PD-1 inhibitor plus anlotinib for metastatic castration-resistant prostate cancer: a real-world study

Management and treatment of terminal metastatic castration-resistant prostate cancer(mCRPC)remains heavily debated.We sought to investigate the efficacy of programmed cell death 1(PD-1)inhibitor plus anlotinib as a potential solution for terminal mCRPC and further evaluate the association of genomic characteristics with efficacy outcomes.We conducted a retrospective real-world study of 25 mCRPC patients who received PD-1 inhibitor plus anlotinib after the progression to standard treatments.The clinical information was extracted from the electronic medical records and 22 patients had targeted circulating tumor DNA(ctDNA)next-generation sequencing.Statistical analysis showed that 6(24.0%)patients experienced prostate-specific antigen(PSA)response and 11(44.0%)patients experienced PSA reduction.The relationship between ctDNA findings and outcomes was also analyzed.DNA-damage repair(DDR)pathways and homologous recombination repair(HRR)pathway defects indicated a comparatively longer PSA-progressionfree survival(PSA-PFS;2.5 months vs 1.2 months,P=0.027;3.3 months vs 1.2 months,P=0.017;respectively).This study introduces the PD-1 inhibitor plus anlotinib as a late-line therapeutic strategy for terminal mCRPC.PD-1 inhibitor plus anlotinib may be a new treatment choice for terminal mCRPC patients with DDR or HRR pathway defects and requires further investigation.

Administration of human umbilical cord-derived mesenchymal stem cells(hUC-MSCs)is believed to be an effective method for treating neurodevelopmental disorde rs.In this study,we investigated the possibility of hUC-MSCs treatment of neonatal hypoxic/ischemic brain injury associated with maternal immune activation and the underlying mechanism.We established neonatal rat models of hypoxic/ischemic brain injury by exposing pregnant rats to lipopolysaccharide on day 16 or 17 of pregnancy.Rat offspring were intranasally administe red hUC-MSCs on postnatal day 14.We found that polypyrimidine tract-binding protein-1(PTBP-1)participated in the regulation of lipopolysaccharide-induced maternal immune activation,which led to neonatal hypoxic/ischemic brain injury.Intranasal delive ry of hUC-MSCs inhibited PTBP-1 expression,alleviated neonatal brain injury-related inflammation,and regulated the number and function of glial fibrillary acidic protein-positive astrocytes,there by promoting plastic regeneration of neurons and im p roving brain function.These findings suggest that hUC-MSCs can effectively promote the repair of neonatal hypoxic/ischemic brain injury related to maternal immune activation through inhibition of PTBP-1 expression and astrocyte activation.

Copper ferrite （CuFe204） nanoparticles catalyzed room temperature multicomponent reaction of aliphatic amines, formaldehyde, arylboronic acids and alkynyl carboxylic acids was reported for the synthesis of diverse propargylamines with good to excellent yields. The catalyst can be magnetically recovered and reused at least five times without significant loss of activity.

In order to improve the properties of Ni-rich(2.5wt.%) Al-Si piston alloy, electric pulse modification was applied in fabricating the Ni-rich Al-Si piston alloy in this study. The effect of electric pulse modification on the mechanical properties of the Ni-rich Al-Si piston alloy was studied using optical microscope(OM), scanning electron microscope(SEM), X-ray diffraction(XRD), microhardness measurement and tensile strength testing.The results showed that the microstructures of Ni-rich Al-Si piston alloy treated by electric pulse modification were refined, the solid solubility of Cu, Ni, Si, etc. in α-Al matrix was improved, and furthermore, the microhardness and high-temperature tensile strength were increased by 9.41% and 17.5%, respectively. The distribution of second phases was also more uniform compared with that of a non-modified sample.

The solidification-precipitation behavior of Al-Mg-Si multicomponent alloys has long been an absorbing topic. Experiments were carried out to analyze the precipitation behaviors of Al-Mg-Si alloys under different heat treatments. All specimens were homogenized at 570 °C for 8 h, and then solution treated at 540 °C for 55 min. Subsequently, the specimens were age treated for different times at temperatures of 100 °C, 150 °C and 180 °C, respectively. The experimental results show that the occurrence of dispersed free zones(DFZ) is caused by the uneven distribution of dispersed phase. During the aging process, pre-β″ phases form at the initial stage and an aging temperature of 100 °C is too low to complete the transformation of pre-β″ to β″. At 150 °C, the precipitation sequence is concluded as SSSS-pre-β″-pre-β″+β″-β″-β′-β. Moreover, changes in sizes and densities of the pre-β″, β″and β′ phases during the aging process has an important influence on the evolution of microhardness and electrical resistivity. The microhardness peak value of 150 °C is similar to that of 180 °C, which is ~141 HV. While, at 100 °C, the microhardness increases slowly, and the attainable value is 127 HV up to 19 days. When the aging temperature is 100 °C, the electrical resistivity has the highest average value. When the aging temperature exceeds 100 °C, with the occurrence and growth of β″and β′, the resistivity has a distinct decrease with prolonged aging time.

BACKGROUND Emerging evidence links gut microbiota to various human diseases including colorectal cancer(CRC)initiation and development.However,gut microbiota profiles associated with CRC recurrence and patient prognosis are not completely understood yet,especially in a Chinese cohort.AIM To investigate the relationship between gut mucosal microbiota profiles and CRC recurrence and patient prognosis.METHODS We obtained the composition and structure of gut microbiota collected from 75 patients diagnosed with CRC and 26 healthy controls.The patients were followed up by regular examination to determine whether tumors recurred.Triplet-paired samples from on-tumor,adjacent-tumor and off-tumor sites of patients diagnosed with/without CRC recurrence were analyzed to assess spatial-specific patterns of gut mucosal microbiota by 16S ribosomal RNA sequencing.Next,we carried out bioinformatic analyses,Kaplan-Meier survival analyses and Cox regression analyses to determine the relationship between gut mucosal microbiota profiles and CRC recurrence and patient prognosis.RESULTS We observed spatial-specific patterns of gut mucosal microbiota profiles linked to CRC recurrence and patient prognosis.A total of 17 bacterial genera/families were identified as potential biomarkers for CRC recurrence and patient prognosis,including Anaerotruncus,Bacteroidales,Coriobacteriaceae,Dialister,Eubacterium,Fusobacterium,Filifactor,Gemella,Haemophilus,Mogibacteriazeae,Pyramidobacter,Parvimonas,Porphyromonadaceae,Slackia,Schwartzia,TG5 and Treponema.CONCLUSION Our work suggests that intestinal microbiota can serve as biomarkers to predict the risk of CRC recurrence and patient death.

A Quick Calculation Method for Radiation Pattern of Submillimeter Telescope with Deformation and Displacement

Radiation pattern captures the electromagnetic performance of reflector antennas,which is significantly affected by the deformation of the primary reflector due to gravity and the displacement of the secondary reflector.During the design process of large reflector antennas,a substantial amount of time is often dedicated to iteratively adjusting structural parameters and validating electromagnetic performance.To improve the efficiency of the design process,we first propose an approximate calculation method of optical path difference(OPD)for the deformation of the primary reflector under gravity and the displacement of the secondary reflector.Then an OPD fitting function based on the modified Zernike polynomials is proposed to capture the phase difference of radiation over the aperture plane,based on which the radiation pattern will be obtained quickly by the aperture field integration method.Numerical experiments demonstrate the effectiveness of the proposed quick calculation method for analyzing the radiation pattern of a 10.4 m submillimeter telescope antenna at its highest operating frequency of 856 GHz.In comparison with the numerical simulation method based on GRASP(which is an antenna electromagnetic analysis tool combining physical optics(PO)and physical theory of diffraction(PTD)),the quick calculation method reduces the time for radiation pattern analysis from more than one hour to less than two minutes.Furthermore,the quick calculation method exhibits excellent accuracy for the figure of merit(FOM)of the radiation pattern.Therefore,the proposed quick calculation method can obtain the radiation pattern with high speed and accuracy.Compared to the time-consuming numerical simulation method(PO and PTD),it can be employed for quick analysis of the radiation pattern for the lateral displacement of the secondary reflector and the deformation of the primary reflector under gravity in the design process of a reflector antenna.

Let G be a simple graph and G~σ be the oriented graph with G as its underlying graph and orientation σ.The rank of the adjacency matrix of G is called the rank of G and is denoted by r(G).The rank of the skew-adjacency matrix of G~σ is called the skew-rank of G~σ and is denoted by sr(G~σ).Let V(G)be the vertex set and E(G) be the edge set of G.The cyclomatic number of G,denoted by c(G),is equal to |E(G)|-|V(G)|+ω(G),where ω(G) is the number of the components of G.It is proved for any oriented graph G~σ that-2c(G)≤sr(G~σ)-r(G)≤2c(G).In this paper,we prove that there is no oriented graph G~σwith sr(G~σ)-r(G)=2c(G)-1,and in addition,there are infinitely many oriented graphs G~σ with connected underlying graphs such that c(G)=k and sr(G~σ)-r(G)=2c(G)-l for every integers k,l satisfying 0 ≤l≤4k and l≠1.

M-N-C(M=Fe,Co,Ni,etc.) catalyst owns high catalytic activity in the oxygen catalytic reaction which is the most likely to replace the Pt-based catalysts.But it is still a challenge to further increase the active site density.This article constructs the high-efficiency FeMn-N/S-C-1000 catalyst to realize ORR/OER bifunctional catalysis by hetero-atom,bimetal(Fe,Mn) doped simultaneously strategy.When evaluated it as bi-functional electro-catalysts,FeMn-N/S-C-1000 exhibits excellent catalytic activity(E_(1/2)=0.924 V,E_(j=10)=1.617 V) in alkaline media,outperforms conventional Pt/C,RuO_(2) and most non-precious-metal catalysts reported recently,Such outstanding performance is owing to N,S co-coordinated with metal to form multi-types of single atom,dual atom active sites to carry out bi-catalysis.Importantly,nitrite poison test provides the proof that the active sites of FeMn-N/S-C are more than that of single-atom catalysts to promote catalytic reactions directly.To better understand the local structure of Fe and Mn active sites,XAS and DFT were employed to reveal that FeMn-N_5/S-C site plays the key role during catalysis.Notably,the FeMn-N/S-C-1000 based low-temperature rechargeable flexible Zn-air also exhibits superior discharge performance and extraordinary durability at-40℃.This work will provide a new idea to design diatomic catalysts applied in low-temperature rechargeable batteries.
