Multi-factor integrated configuration model and three-layer hybrid optimization algorithm framework:turnkey project-oriented rapid manufacturing system configuration

In the context of increasingly prominent product personalization and customization trends,intelligent manufacturing-oriented turnkey projects can provide manufacturers with fast and convenient turnkey services for manufacturing *** key characteristic is the transformation of the traditional design process into a configuration ***,the scope of configuration resources in existing research is limited;the cost and time required for manufacturing system construction are overlooked;and the integration of the system layout configuration is rarely considered,making it difficult to meet the manufacturing system configuration requirements of turnkey *** response,this study establishes a multi-factor integrated rapid configuration model and proposes a solution method for manufacturing systems based on the requirements of turnkey *** configuration model considers the system construction cost and duration and the product manufacturing cost and duration,as optimization *** differences in product feature-dividing schemes and configuration of processes,equipment,tools,fixtures,and layouts were considered *** proposed model-solving method is a three-layer hybrid optimization algorithm framework with two optimization algorithm modules and an intermediate algorithm *** hybrid configuration algorithms are established based on non-dominated sorting genetic algorithm-Ⅲ(NSGAⅢ),non-dominated sorting genetic algorithm-Ⅱ(NSGAⅡ),multi-objective simulated annealing(MOSA),multi-objective neighborhood search(MONS),and tabu search(TS).These algorithms are compared and validated through a hydraulic valve block production case,and the TS and NSGAⅢ(TS-NSGAⅢ)hybrid algorithm exhibits the best *** case demonstrates the effectiveness of the proposed model and solution method.

A condition-based maintenance policy for reconfigurable multi-device systems

The exploration of component states for optimizing maintenance schedules in complex systems has garnered significant interest from ***,current literature usually overlooks the critical aspects of system flexibility and *** implementation of system reconfiguration can effectively mitigate system downtime and enhance production *** study proposes a dynamic condition-based maintenance policy considering reconfiguration for reconfigurable systems.A double-layer decision rule was constructed for the devices and *** achieve the best overall maintenance effect of the system,the remaining useful life probability distribution and recommended maintenance time of each device were used to optimize the concurrent maintenance time window of the devices and determine whether to reconfigure them.A comprehensive maintenance efficiency index was introduced that simultaneously considered the maintenance cost rate,reliability,and availability of the system to characterize the overall maintenance *** reconfiguration cost was included in the maintenance *** proposed policy was tested through numerical experiments and compared with different-level *** results show that the proposed policy can significantly reduce the downtime and maintenance costs and improve the overall system reliability and availability.

Poplar plantations are often established on nitrogen-poor land,and poplar growth and wood formation are constrained by low nitrogen(LN)***,the molecular mechanisms by which specific genes regulate wood formation in acclimation to LN availability remain ***,we report a previously unrecognized module,basic region/leucine zipper 55(PtobZIP55)–PtoMYB170,which regulates the wood formation of Populus tomentosa in acclimation to LN ***55 was highly expressed in poplar wood and induced by *** wood anatomical properties and increased lignification were detected in PtobZIP55-overexpressing poplars,whereas the opposite results were detected in PtobZIP55-knockout *** and transgenic analyses revealed that PtobZIP55 directly binds to the promoter sequence of PtoMYB170to activate its *** phenotypes of PtoMYB170 transgenic poplars were similar to those of PtobZIP55 transgenic poplars under LN *** molecular analyses revealed that PtoMYB170 directly bound the promoter sequences of lignin biosynthetic genes to activate their transcription to increase lignin concentrations in LN-treated poplar *** results suggest that PtobZIP55 activates PtoMYB170 transcription,which in turn positively regulates lignin biosynthetic genes,increasing lignin deposition in the wood of *** in the context of acclimation to LN availability.

Observer-based event-triggered formation tracking control for second-order multi-agent systems in constrained region

In this paper, an event-triggered time-varying formation tracking control for a class of second-order nonlinear multiagent systems(MAS) operating within a constrained region is investigated. To mitigate the negative effects of external unknown disturbance, a novel disturbance observer with performance guarantees is proposed, enabling precise disturbance *** the artificial potential field(APF) method, a repulsive potential function is introduced to prevent inter-agent collisions as well as collisions with environmental obstacles. To reduce continuous communication and frequent system updates, a sliding mode technique is incorporated into the formation tracking controller, utilizing an event-triggered mechanism. The controller is also applicable to the formation control of MAS in switching-constrained regions. The achievement of the specified timevarying geometric formation is rigorously demonstrated through the Lyapunov framework. Numerical simulations are presented to validate the effectiveness of the theoretical results.

While it is known that spermatogonial stem cells (SSCs) initiate the production of male germ cells, the mechanisms of SSC self-renewal, proliferation, and differentiation remain poorly understood. We have previously identified Strawberry Notch 1 (SBN01), a vertebrate strawberry notch family protein, in the proteome profile for mouse SSC maturation and differentiation, revealing SBN01 is associated with neonatal testicular development. To explore further the location and function of SBN01 in the testes, we performed Sbnol gene knockdown in mice to study the effects of SBN01 on neonatal testicular and SSC development. Our results revealed that SBN01 is required for neonatal testicular and SSC development in mice. Particularly, in vitro Sbnol gene knockdown with morpholino oligonucleotides caused a reduction of SSCs and inactivation of the noncanonical Wnt pathway, through Jun N-terminal kinases. Our study suggests SBN01 maintains SSCs by promoting the noncanonical Wnt pathway.

Nigella sativa L. seed extract alleviates oxidative stress-induced cellular senescence and dysfunction in melanocytes

Nigella sativa L. seeds have been traditionally utilized in Chinese folk medicine for centuries to treat vitiligo. This study revealed that the ethanolic extract of Nigella sativa L.(HZC) enhances melanogenesis and mitigates oxidative stress-induced cellular senescence and dysfunction in melanocytes. In accordance with established protocols, the ethanol fraction from Nigella sativa L. seeds was extracted, concentrated, and lyophilized to evaluate its herbal effects via 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) assays, tyrosinase activity evaluation, measurement of cellular melanin contents, scratch assays, senescence-associated β-galactosidase(SA-β-gal) staining, enzyme-linked immunosorbent assay(ELISA), and Western blot analysis for expression profiling of experimentally relevant proteins. The results indicated that HZC significantly enhanced tyrosinase activity and melanin content while notably increasing the protein expression levels of Tyr, Mitf, and gp100 in B16F10 cells. Furthermore, HZC effectively mitigated oxidative stress-induced cellular senescence, improved melanocyte condition, and rectified various functional impairments associated with melanocyte dysfunction. These findings suggest that HZC increases melanin synthesis in melanocytes through the activation of the MAPK, PKA, and Wnt signaling *** addition, HZC attenuates oxidative damage induced by H2O2 therapy by activating the nuclear factor E2-related factor 2-antioxidant response element(Nrf2-ARE) pathway and enhancing the activity of downstream antioxidant enzymes, thus preventing premature senescence and dysfunction in melanocytes.

Domain generalization with semi-supervised learning for people-centric activity recognition

People-centric activity recognition is one of the most critical technologies in a wide range of real-world applications,including intelligent transportation systems, healthcare services, and brain-computer interfaces. Large-scale data collection and annotation make the application of machine learning algorithms prohibitively expensive when adapting to new tasks. One way of circumventing this limitation is to train the model in a semi-supervised learning manner that utilizes a percentage of unlabeled data to reduce the labeling burden in prediction tasks. Despite their appeal, these models often assume that labeled and unlabeled data come from similar distributions, which leads to the domain shift problem caused by the presence of distribution gaps. To address these limitations, we propose herein a novel method for people-centric activity recognition,called domain generalization with semi-supervised learning(DGSSL), that effectively enhances the representation learning and domain alignment capabilities of a model. We first design a new autoregressive discriminator for adversarial training between unlabeled and labeled source domains, extracting domain-specific features to reduce the distribution gaps. Second, we introduce two reconstruction tasks to capture the task-specific features to avoid losing information related to representation learning while maintaining task-specific consistency. Finally, benefiting from the collaborative optimization of these two tasks, the model can accurately predict both the domain and category labels of the source domains for the classification task. We conduct extensive experiments on three real-world sensing datasets. The experimental results show that DGSSL surpasses the three state-of-the-art methods with better performance and generalization.
