Curcumin Inhibits Proliferation of Renal Cell Carcinoma in vitro and in vivo by Regulating miR-148/ADAMTS18 through Suppressing Autophagy

Objective: To explore the effect of curcumin on the proliferation of renal cell carcinoma and analyze its regulation mechanism. Methods: In RCC cell lines of A498 and 786-O, the effects of curcumin(2.5, 5, 10 μmo/L) on the proliferation were analyzed by Annexin V+PI staining. Besides, A498 was inoculated into nude mice to establish tumorigenic models, and the model mice were treated with different concentrations of curcumin(100, 200, and 400 mg/kg), once daily for 30 days. Then the tumor diameter was measured, the tumor cells were observed by hematoxylin-eosin staining, and the protein expressions of miR-148 and ADAMTS18were detected by immunohistochemistry. In vitro, after transfection of miR-148 mimics, miR-148 inhibitor or si-ADAMTS18 in cell lines, the expression of ADAMTS18 was examined by Western blotting and the cell survival rate was analyzed using MTT. Subsequently, Western blot analysis was again used to examine the autophagy phenomenon by measuring the relative expression level of LC3-Ⅱ/LC3-Ⅰ;autophagy-associated genes,including those of Beclin-1 and ATG5, were also examined when miR-148 was silenced in both cell lines with curcumin treatment. Results: Curcumin could inhibit the proliferation of RCC in cell lines and nude mice. The expressions of miR-148 and ADAMTS18 were upregulated after curcumin treatment both in vitro and in vivo(P<0.05). The cell survival rate was dramatically declined upon miR-148 or ADAMTS18 upregulated. However,si-ADAMTS18 treatment or miR-148 inhibitor reversed these results, that is, both of them promoted the cell survival rate. Conclusion: Curcumin can inhibit the proliferation of renal cell carcinoma by regulating the miR-148/ADAMTS18 axis through the suppression of autophagy in vitro and in vivo. There may exist a positive feedback loop between mi R-148 and ADAMTS18 gene in RCC.

Battery Storage Configuration of AC/DC Hybrid Distribution Networks

The upscaling requirements of energy transition highlight the urgent need for ramping up renewables and boosting system efficiencies.However,the stochastic nature of excessive renewable energy resources has challenged stable and efficient operation of the power system.Battery energy storage systems(BESSs)have been identified as critical to mitigate random fluctuations,unnecessary green energy curtailment and load shedding with rapid response and flexible connection.On the other hand,an AC/DC hybrid distribution system can offer merged benefits in both AC and DC subsystems without additional losses during AC/DC power conversion.Therefore,configuring BESSs on an AC/DC distribution system is wellpositioned to meet challenges brought by carbon reductions in an efficient way.A bi-level optimization model of BESS capacity allocation for AC/DC hybrid distribution systems,considering the flexibility of voltage source converters(VSCs)and power conversion systems(PCSs),has been established in this paper to address the techno-economic issues that hindered wide implementation.The large-scale nonlinear programming problem has been solved utilizing a genetic algorithm combined with second-order cone programming.Rationality and effectiveness of the model have been verified by setting different scenarios through case studies.Simulation results have demonstrated the coordinated operation of BESS and AC/DC hybrid systems can effectively suppress voltage fluctuations and improve the cost-benefit of BESSs from a life cycle angle.

Let G be a graph.We say that G is 2-divisible if for each induced subgraph H of G,either V(H)is a stable set,or V(H)can be partitioned into two sets A and B such thatω(H[A])<ω(H)andω(H[B])<ω(H).A hole is an induced cycle of length at least 4,a bull is a graph consisting of a triangle with two disjoint pendant edges,a diamond is the graph obtained from K4 by removing an edge,a dart denotes the graph obtained from a diamond by adding a pendant edge to one vertex of degree 3,and a racket denotes the graph obtained from a diamond by adding a pendant edge to one vertex of degree 2.In this paper,we prove that every{odd hole,H}-free graph is 2-divisible,where H is a dart,or a racket,or a bull.As corollaries,X(G)≤min{2ω^(G)-1,(ω^(G)/2+1)}if G is{odd hole,dart}-free,or{odd hole,racket}-free,or{odd hole,bull}-free.

The Perturbed Compound Poisson Risk Model with Proportional Investment

In this paper, the insurance company considers venture capital and risk-free investment in a constant proportion. The surplus process is perturbed by diffusion. At first, the integro-differential equations satisfied by the expected discounted dividend payments and the Gerber-Shiu function are derived. Then, the approximate solutions of the integro-differential equations are obtained through the sinc method. Finally, the numerical examples are given when the claim sizes follow different distributions. Furthermore, the errors between the explicit solution and the numerical solution are discussed in a special case.

Mapping neural circuits is critical for understanding the structure and function of the nervous system.Engineered viruses are a valuable tool for tracing neural circuits.However,current tracers do not fully meet the needs for this approach because of various drawbacks,such as toxicity and characteristics that are difficult to modify.Therefore,there is an urgent need to develop a new tracer with low toxicity and that allows for long-term studies.In this study,we constructed an engineered Sindbis virus(SINV)expressing enhanced green fluorescent protein(EGFP)reporter gene(SINV-EGFP)and found that it had no significant difference in biological characterization compared with the wild-type Sindbis virus in BHK-21 cells and neurons in vitro.We injected the virus into the visual circuit of mouse brain and found that the virus infected neurons in the local injected site and anterogradely spread in the neural circuits.Although the efficiency of transmission was limited,the findings demonstrate that SINV can be used as a new anterograde tracer to map neural circuits in mouse brain and that it spreads exclusively in the anterograde direction.Further,use of SINV in mouse brain research will provide longer time windows for circuit tracing than is possible with herpes simplex virus and vesicular stomatitis virus tracers.

Transgenic models are useful tools for studying the pathogenesis of and drug development for Alzheimer's Disease(AD).AD models are constructed usually using overexpression or knock-in of multiple pathogenic gene mutations from familial AD.Each transgenic model has its unique behavioral and pathological features.This review summarizes the research progress of transgenic mouse models,and their progress in the unique mechanism of amyloid-βoligomers,including the first transgenic mouse model built in China based on a single gene mutation(PSEN1 V97L)found in Chinese familial AD.We further summarized the preclinical findings of drugs using the models,and their future application in exploring the upstream mechanisms and multi-target drug development in AD.

Gut microbiota alterations are distinct for primary colorectal cancer and hepatocellular carcinoma

Colorectal cancer(CRC)and hepatocellular carcinoma(HCC)are the second and third most common causes of death by cancer,respectively.The etiologies of the two cancers are either infectious insult or due to chronic use of alcohol,smoking,diet,obesity and diabetes.Pathological changes in the composition of the gut microbiota that lead to intestinal inflammation are a common factor for both HCC and CRC.However,the gut microbiota of the cancer patient evolves with disease pathogenesis in unique ways that are affected by etiologies and environmental factors.In this review,we examine the changes that occur in the composition of the gut microbiota across the stages of the HCC and CRC.Based on the idea that the gut microbiota are an additional“lifeline”and contribute to the tumor microenvironment,we can observe from previously published literature how the microbiota can cause a shift in the balance from normal→inflammation→diminished inflammation from early to later disease stages.This pattern leads to the hypothesis that tumor survival depends on a less proinflammatory tumor microenvironment.The differences observed in the gut microbiota composition between different disease etiologies as well as between HCC and CRC suggest that the tumor microenvironment is unique for each case.

Obstructive sleep apnea aggravates neuroinflammation and pyroptosis in early brain injury following subarachnoid hemorrhage via ASC/HIF-1α pathway

Obstructive sleep apnea can worsen the prognosis of subarachnoid hemorrhage.Howeve r,the underlying mechanism remains unclear.In this study,we established a mouse model of subarachnoid hemorrhage using the endovascular perforation method and exposed the mice to intermittent hypoxia for 8 hours daily for 2 consecutive days to simulate sleep apnea.We found that sleep apnea aggravated brain edema,increased hippocampal neuron apoptosis,and worsened neurological function in this mouse model of subarachnoid hemorrhage.Then,we established an in vitro HT-22 cell model of hemin-induced subarachnoid hemorrhage/intermittent hypoxia and found that the cells died,and lactate dehydrogenase release increased,after 48 hours.We further investigated the underlying mechanism and found that sleep apnea increased the expression of hippocampal neuroinflammatory factors interleukin-1β,interleukin-18,inte rleukin-6,nuclear factorκB,pyro ptosis-related protein caspase-1,pro-caspase-1,and NLRP3,promoted the prolife ration of astrocytes,and increased the expression of hypoxia-inducible factor 1αand apoptosis-associated speck-like protein containing a CARD,which are the key proteins in the hypoxia-inducible factor 1α/apoptosis-associated speck-like protein containing a CARD signaling pathway.We also found that knockdown of hypoxia-inducible factor 1αexpression in vitro greatly reduced the damage to HY22 cells.These findings suggest that sleep apnea aggravates early brain injury after subarachnoid hemorrhage by aggravating neuroinflammation and pyroptosis,at least in part through the hypoxia-inducible factor 1α/apoptosis-associated speck-like protein containing a CARD signaling pathway.

Establishment and verification of a surgical prognostic model for cervical spinal cord injury without radiological abnormality

Some studies have suggested that early surgical treatment can effectively improve the prognosis of cervical spinal cord injury without radiological abnormality, but no research has focused on the development of a prognostic model of cervical spinal cord injury without radiological abnormality. This retrospective analysis included 43 patients with cervical spinal cord injury without radiological abnormality. Seven potential factors were assessed: age, sex, external force strength causing damage, duration of disease, degree of cervical spinal stenosis, Japanese Orthopaedic Association score, and physiological cervical curvature. A model was established using multiple binary logistic regression analysis. The model was evaluated by concordant profiling and the area under the receiver operating characteristic curve. Bootstrapping was used for internal validation. The prognostic model was as follows: logit(P) =-25.4545 + 21.2576 VALUE + 1.2160SCORE-3.4224 TIME, where VALUE refers to the Pavlov ratio indicating the extent of cervical spinal stenosis, SCORE refers to the Japanese Orthopaedic Association score(0–17) after the operation, and TIME refers to the disease duration(from injury to operation). The area under the receiver operating characteristic curve for all patients was 0.8941(95% confidence interval, 0.7930–0.9952). Three factors assessed in the predictive model were associated with patient outcomes: a great extent of cervical stenosis, a poor preoperative neurological status, and a long disease duration. These three factors could worsen patient outcomes. Moreover, the disease prognosis was considered good when logit(P) ≥-2.5105. Overall, the model displayed a certain clinical value. This study was approved by the Biomedical Ethics Committee of the Second Affiliated Hospital of Xi'an jiaotong University, China(approval number: 2018063) on May 8, 2018.

Blast performance of layered charges enveloped by aluminum powder/rubber composites in confined spaces

A layered charge composed of the JH-2 explosive enveloped by a thick-walled cylindrical casing(active aluminum/rubber and inert lithium fluoride/rubber composites) was designed and explosion experiments were conducted in a 1.3 m3tank and a 113 m3bunker.The blast parameters,including the quasistatic pressure(ΔpQS),special impulse(I),and peak overpressure(Δpmax),and images of the explosion process were recorded,and the influence of the Al content(30% and 50%) and Al particle size(1,10,and 50 μm) on the energy release of aluminum/rubber composites were investigated.The results revealed that the use of an active layer increased the peak overpressure generated by the primary blast wave,as well as the quasistatic pressure and special impulse related to fuel burning within tens of milliseconds after detonation.When the Al content was increased from 30% to 50%,the increases of ΔpQS and I were not obvious,and Δpmaxeven decreased,possibly because of decreased combustion efficiency and greater absorption of the blast wave energy for layers with 50% Al.Compared with the pure JH-2charge,the charge with 1 μm Al particles produced the highest Δpmax,indicating that better transient blast performance was generated by smaller Al particles.However,the charge with 10 μm Al particles showed the largest ΔpQSand I,suggesting that a stronger destructive effect occurred over a longer duration for charges that contained moderate 10 μm Al.
