Expression and clinical significance of short-chain fatty acids in patients with intrahepatic cholestasis of pregnancy

BACKGROUND Intrahepatic cholestasis of pregnancy(ICP)is a pregnancy-specific liver condition that typically arises in the middle and late stages of pregnancy.Short-chain fatty acids(SCFAs),prominent metabolites of the gut microbiota,have significant connections with various pregnancy complications,and some SCFAs hold potential for treating such complications.However,the metabolic profile of SCFAs in patients with ICP remains unclear.AIM To investigate the metabolic profiles and differences in SCFAs present in the maternal and cord blood of patients with ICP and determine the clinical significance of these findings.METHODS Maternal serum and cord blood samples were collected from both patients with ICP(ICP group)and normal pregnant women(NP group).Targeted metabolomics was used to assess the SCFA levels in these samples.RESULTS Significant differences in maternal SCFAs were observed between the ICP and NP groups.Most SCFAs exhibited a consistent declining trend in cord blood samples from the ICP group,mirroring the pattern seen in maternal serum.Correlation analysis revealed a positive correlation between maternal serum SCFAs and cord blood SCFAs[r(Pearson)=0.88,P=7.93e-95].In both maternal serum and cord blood,acetic and caproic acids were identified as key metabolites contributing to the differences in SCFAs between the two groups(variable importance for the projection>1).Receiver operating characteristic analysis demonstrated that multiple SCFAs in maternal blood have excellent diagnostic capabilities for ICP,with caproic acid exhibiting the highest diagnostic efficacy(area under the curve=0.97).CONCLUSION Compared with the NP group,significant alterations were observed in the SCFAs of maternal serum and cord blood in the ICP group,although they displayed distinct patterns of change.Furthermore,the SCFA levels in maternal serum and cord blood were significantly positively correlated.Notably,certain maternal serum SCFAs,specifically caproic and acetic acids,demonstrated excellent diagnostic efficiency for ICP.

NQO1 Mediates Lenvatinib Resistance by Regulating ROS-induced Apoptosis in Hepatocellular Carcinoma

Objective Hepatocellular carcinoma(HCC)is the third leading cause of cancer-associated death worldwide.As a first-line drug for advanced HCC treatment,lenvatinib faces a significant hurdle due to the development of both intrinsic and acquired resistance among patients,and the underlying mechanism remains largely unknown.The present study aims to identify the pivotal gene responsible for lenvatinib resistance in HCC,explore the potential molecular mechanism,and propose combinatorial therapeutic targets for HCC management.Methods Cell viability and colony formation assays were conducted to evaluate the sensitivity of cells to lenvatinib and dicoumarol.RNA-Seq was used to determine the differences in transcriptome between parental cells and lenvatinib-resistant(LR)cells.The upregulated genes were analyzed by GO and KEGG analyses.Then,qPCR and Western blotting were employed to determine the relative gene expression levels.Afterwards,the intracellular reactive oxygen species(ROS)and apoptosis were detected by flow cytometry.Results PLC-LR and Hep3B-LR were established.There was a total of 116 significantly upregulated genes common to both LR cell lines.The GO and KEGG analyses indicated that these genes were involved in oxidoreductase and dehydrogenase activities,and reactive oxygen species pathways.Notably,NAD(P)H:quinone oxidoreductase 1(NQO1)was highly expressed in LR cells,and was involved in the lenvatinib resistance.The high expression of NQO1 decreased the production of ROS induced by lenvatinib,and subsequently suppressed the apoptosis.The combination of lenvatinib and NQO1 inhibitor,dicoumarol,reversed the resistance of LR cells.Conclusion The high NQO1 expression in HCC cells impedes the lenvatinib-induced apoptosis by regulating the ROS levels,thereby promoting lenvatinib resistance in HCC cells.

Berberine Enhances Intestinal Mucosal Barrier Function by Promoting Vitamin D Receptor Activity

Objective:To evaluate if berberine can act on vitamin D receptors(VDR)and thereby regulate the expression of tight junction proteins(TJPs)in irritable bowel syndrome-diarrhea-predominant(IBS-D)rats.Methods:The newborn rats were induced into IBS-D rat model via neonatal maternal separation combined with acetic acid chemical stimulation.After modeling,the model was evaluated and rats were divided into the control group and berberine treatment groups(0.85,1.7 and 3.4 mg/kg,once a day for 2 weeks).The distal colon was obtained and colonic epithelial cells(CECs)were isolated and cultured after IBS-D model evaluation.The vitamin D receptor response element(VDRE)reporter gene was determined in the CECs of IBS-D rats to analyze the effect of berberine on the VDRE promoter.VDR overexpression or silencing technology was used to analyze whether VDR plays a role in promoting intestinal barrier repair,and to determine which region of VDR plays a role in berberine-regulated intestinal TJPs.Results:The IBS-D rat model was successfully constructed and the symptoms were improved by berberine in a dose-dependent manner(Pting VDR activity,and the main site of action is the N-terminal region of VDR.

The noise spectral characteristics and noise reduction schemes of screw air-source heat pump:A case study

The screw air-source heat pump can cause incessant high noise levels during operation,which might hinder adoption of this energy-efficient heat pump.First,acoustic measurements and comparison testing were performed in this research.The measurements revealed that the compressor is the main noise source of the heat pump,and it shows a multipeak frequency distribution and a wide frequency spectrum under different work conditions,with multiple peaks at 63,250,and 1000 Hz.Then,a compressor sound insulation cover with broadband absorption was proposed,and it was experimentally proven that the insulation cover can reduce the maximum sound pressure level of one unit from 89.8 dBA to 79.1 dBA.Third,we proposed several noise reduction strategies and compared their noise reduction effects using computer simulation.The results showed that the noise problem can be effectively improved through the rational design of the sound barrier and the layout and opening options of heat pump.The distance between the sound barrier and heat pump and the sound attenuation due to diffraction ALa exhibit a U-shaped relation.For buildings of different heights,the optimal heights of noise barrier are proposed.The 5.5-meter is the optimal height of the sound barrier for single-story buildings.The conclusions can be applied to other building projects for heat pump noise reduction.

Asymmetric Partial Hydrosilylation of 2,2-Difluoro-1,3-diketones with Chiral Frustrated Lewis Pairs

Comprehensive Summary,The asymmetric partial reduction of 1,3-diketones stands as a straightforward pathway to access optically active β-hydroxyketones. In this paper, an asymmetric Piers-type hydrosilylation of 2,2-difluoro-1,3-diketones was successfully realized by using a frustrated Lewis pair of chiral borane and tricyclohexylphosphine as a catalyst, delivering a variety of α,α-difluoro-β-hydroxyketones in high yields with up to 99% ee. Significantly, no over-reduced diol products were observed even with an excess amount of silanes. The product can be conveniently converted to α,α-difluoro-β-hydroxyester or 1,3-anti-diol via an oxidation with m-CPBA or a reduction with DIBAL-H without obvious loss of ee.

Overshoot phenomena:Observation and simulation

High-power O-mode radio waves can excite artificial instabilities in the F region,according to experiments conducted at the European Incoherent Scatter Science Association(EISCAT)heating facility.The main instabilities include the parametric decay instability(PDI),oscillating two-stream instability(OTSI),and thermal parametric instability(TPI).The PDI and OTSI not only compete with each other,but also compete with the TPI,leading to a two-stage overshoot phenomenon:a miniovershoot occurs on a millisecond time scale after pump-on,followed by the main overshoot.We gain insight into the miniovershoot via a generalized Zakharov model,whereas the main overshoot can be observed as an enhanced plasma line overshoot phenomenon in incoherent scatter radar spectra.We can also observe that the zero-frequency ion line exists only in the initial heating period after a cold start and that the upshifted and downshifted ion lines behave irregularly in the spectra.The simulation results show that competition between the PDI and OTSI leads to an initial peak,which we named the pre-miniovershoot.The following processes,namely ion density caviton generation,and collapse and cascade in the development of the PDI,contribute to the miniovershoot phenomenon.

Roles of nanoparticles in arsenic mobility and microbial community composition in arsenic-enriched soils

Environmental effects of nano remediation engineering of arsenic(As)pollution need to be considered.In this study,the roles of Fe_(2)O_(3)and TiO_(2)nanoparticles(NPs)on the microbial mediated As mobilization from As contaminated soil were investigated.The addition of Fe_(2)O_(3)and TiO_(2)NPs restrained As(V)release,and stimulated As(Ⅲ)release.As(V)concentration decreased by 94% and 93% after Fe_(2)O_(3)addition,and decreased by 89% and 45% after Ti O_(2)addition compared to the Biotic and Biotic+Acetate(amended with sodium acetate)controls,respectively.The maximum values of As(Ⅲ)were 20.5 and 27.1μg/L at 48 d after Fe_(2)O_(3)and TiO_(2)NPs addition,respectively,and were higher than that in Biotic+Acetate control(12.9μg/L).The released As co-precipitated with Fe in soils in the presence of Fe_(2)O_(3)NPs,but adsorbed on TiO_(2)NPs in the presence of TiO_(2)NPs.Moreover,the addition of NPs amended with sodium acetate as the electron donor clearly promoted As(V)reduction induced by microbes.The NPs addition changed the relative abundance of soil bacterial community,while Proteobacteria(42.8%-70.4%),Planctomycetes(2.6%-14.3%),and Firmicutes(3.5%-25.4%)were the dominant microorganisms in soils.Several potential As/Fe reducing bacteria were related to Pseudomonas,Geobacter,Desulfuromonas,and Thiobacillus.The addition of Fe_(2)O_(3)and TiO_(2)NPs induced to the decrease of arr A gene.The results indicated that the addition of NPs had a negative impact on soil microbial population in a long term.The findings offer a relatively comprehensive assessment of Fe_(2)O_(3)and TiO_(2)NPs effects on As mobilization and soil bacterial communities.

Proteomic study of vitreous in proliferative diabetic retinopathy patients after treatment with aflibercept:a quantitative analysis based on 4D label-free technique

AIM:To identify different metabolites,proteins and related pathways to elucidate the causes of proliferative diabetic retinopathy(PDR)and resistance to anti-vascular endothelial growth factor(VEGF)drugs,and to provide biomarkers for the diagnosis and treatment of PDR.METHODS:Vitreous specimens from patients with diabetic retinopathy were collected and analyzed by Liquid Chromatography-Mass Spectrometry(LC-MS/MS)analyses based on 4D label-free technology.Statistically differentially expressed proteins(DEPs),Gene Ontology(GO),Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway representation and protein interactions were analyzed.RESULTS:A total of 12 samples were analyzed.The proteomics results showed that a total of 58 proteins were identified as DEPs,of which 47 proteins were up-regulated and 11 proteins were down-regulated.We found that C1q and tumor necrosis factor related protein 5(C1QTNF5),Clusterin(CLU),tissue inhibitor of metal protease 1(TIMP1)and signal regulatory protein alpha(SIRPα)can all be specifically regulated after aflibercept treatment.GO functional analysis showed that some DEPs are related to changes in inflammatory regulatory pathways caused by PDR.In addition,protein-protein interaction(PPI)network evaluation revealed that TIMP1 plays a central role in neural regulation.In addition,CD47/SIRPαmay become a key target to resolve anti-VEGF drug resistance in PDR.CONCLUSION:Proteomic analysis is an approach of choice to explore the molecular mechanisms of PDR.Our data show that multiple proteins are differentially changed in PDR patients after intravitreal injection of aflibercept,among which C1QTNF5,CLU,TIMP1 and SIRPαmay become targets for future treatment of PDR and resolution of anti-VEGF resistance.

RNA m6A dynamic modification mediated by nucleuslocalized FTO is involved in follicular reserve

In eukaryotic organisms,the most common internal modification of messenger RNA(m RNA)is N6-methyladenosine(m6A).This modification can be dynamically and reversibly controlled by specific enzymes known as m6A writers and erasers.The fat-mass and obesity-associated protein(FTO)catalyzes RNA demethylation and plays a critical role in various physiological and pathological processes.Our research identified dynamic alterations in both m6A and FTO during the assembly of primordial follicles,with an inverse relationship observed for m6A levels and nuclear-localized FTO expression.Application of Fto small interfering RNA(si RNA)altered the expression of genes related to cell proliferation,hormone regulation,and cell chemotaxis,and affected RNA alternative splicing.Overexpression of the full-length Fto gene led to changes in m6A levels,alternative splicing of Cdk5,cell proliferation,cell cycle progression,and proportion of primordial follicles.Conversely,overexpression of Fto lacking a nuclear localization signal(NLS)did not significantly alter m6A levels or primordial follicle assembly.These findings suggest that FTO,localized in the nucleus but not in the cytoplasm,regulates RNA m6A demethylation and plays a role in cell proliferation,cell cycle progression,and primordial follicle assembly.These results highlight the potential of m6A and its eraser FTO as possible biomarkers and therapeutic targets.

Contributors to tidal duration asymmetry with varied coastline configurations on western shelf of Yellow Sea

Coastal management in China is confronted with an urgent choice between natural restoration and maintenance of existing seawalls and reclaimed land for economic development.A key criterion for making this decision is the resilience to coastal flooding,which depends on the ability to predict tidal level.Tidal duration asymmetry(TDA)is a key parameter in determination of the arrival and duration of flood tides.This study selected the western inner shelf of the Yellow Sea(WYS)as the study area and investigated the responses of TDA to different shoreline configurations and relative sea level rise.The responses of TDA to shoreline reconstruction yielded spatial variability locally and remotely.In the nearshore area,the responses of TDA to the complex ocean environment mainly originated from the combined functions of reflection,bottom friction,and advection,which controlled the energy transfer from M2 or S2 constituents to their overtides or compound tides.The sensitivity of TDA to coastline typologies was not limited to coastal waters but could stretch over the entire inner shelf.The vulnerability of tidal responses was due to the displacement of the M2 amphidrome of the Kelvin wave on the WYS,which in turn changed tidal energy fluxes over the regime.The relative sea level rise could intensify the feedback of TDA to seawalls and land reclamation.
