Transcriptomic analysis identifies the neuropeptide cortistatin (CORT) as an inhibitor of temozolomide (TMZ) resistance by suppressing the NF-kB-MGMT signaling axis in human glioma

Glioma is a common tumor originating in the brain that has a high mortality ***(TMZ)is the first-line treatment for high-grade ***,a large pro-portion of gliomas are resistant to TMZ,posing a great challenge to their *** the study,the specific functions and mechanism(s)by which cortistatin(CORT)regulates TMZ resis-tance and glioma progression were *** decreased expression of CoRT was detected in glioma tissues,and highly expressed CORT was associated with a better survival rate in pa-tients with *** overexpression notably decreased the capacity of glioma cells to pro-liferate and migrate in vitro and to form tumors in *** overexpression also markedly suppressed the viability and enhanced the apoptosis of TMZ-resistant U251 cells by regulating MGMT,p21,and Puma ***,CORT overexpression reduced the resistance of gliomas to TMZ in *** expression Was negatively correlated with MGMT expression in both glioma tissues and cells,and it was found that CORT inhibited NF-kB pathway activation in glioma cells,thereby inhibiting MGMT *** conclusion,CORT regulates glioma cell growth,migration,apoptosis,and TMZ resistance by weakening the activity of NF-kB/p65 and thereby regulating MGMT *** CORT/NF-kB/MGMT axis might be regarded as a molecular mechanism contributing to the resistance of glioma to *** data also suggest that CORT regulates the viability and metastatic potential of glioma cells,independent of its effects on TMZ resistance,providing evidence of novel therapeutic targets for glioma that should be evaluated infurther studies.

Polyoxometalate-based nanocomposites with electrocatalytic activity have been applied in hydrogen evolution reactions(heR).Seawater as the main water resource on the earth should be developed as the water electrolysis to prepare high-purity *** this paper,we used two synthesis strategies to prepare the nanocomposite Co_(4)-POM@Co-PGDY(Co_(4)-POM:the Kegging-type microcrystals of K_(10)[Co_(4)(PW_(9)O_(3)4)2]and Co-PGDY:cobalt-porphyrin linked graphdiyne)with excellent activity for ***-PGDY as the porous material is applied not only as the protection of microcrystals towards the metal ion in seawater but also as the co-electrocatalyst of Co_(4)-***_(4)-POM@Co-PGDY exhibits excellent heR performance in seawater electrolytes with low overpotential and high stability at high ***,we have observed a key H_(3)O+intermediate emergence on the surface of nanocomposite during hydrogen evolution process in seawater by Raman synchrotron radiation-based Fourier transform infrared(SR-FTIR).The results in this paper provide an effective strategy for preparing polyoxometalate-based electrocatalysts with high-performance toward hydrogen evolution reaction.

Energy band modulation of N-doped rutile/anatase TiO2 photoanode promoting charge separation toward prominent photoelectrochemical performance

To solve the problem of high photogenerated carrier recombination rate and low photoelectric conversion efficiency of TiO2-based materials,a simple N-doped anatase/rutile TiO2heterophase nanorod film was designed by a low-temperature hydrothermal method in this *** enhanced separation and transport of photogenerated charges were facilitated by the smaller contact barrier and appropriate band matching between anatase TiO2nanoparticles and rutile *** introduction of N doping in anatase TiO2resulted in an upward shift of the valence band and a narrowing of the band gap,consequently enhancing the efficiency of visible light *** combination of the heterophase junction and N-doping exhibited a synergistic effect,effectively suppressing the recombination of photogenerated charges and enhancing the photoelectric conversion efficiency of the *** AM 1.5G irradiation,the photocurrent density（J） of the A-TO（N）@R-TONR photoanode reached2.19 mA·cm-2(VRhe,1.23 eV).Additionally,the incident photon-electron conversion efficiency（IPCE） and the charge injection efficiency（η） reached 81.4% and 51.6% at320 ***,the J,IPCE,and η values of the A-TO（N）@R-TONR photoanode were 2.96,2.1 and 3.2times those of pure R-TONR photoanode,*** work presents a rational strategy for designing efficient TiO2-based photoanodes.

Anti-OX40 Antibody Combined with HBc VLPs Delays Tumor Growth in a Mouse Colon Cancer Model

Rheological Behaviors of Polymers with Nanoparticles Tethered at Each End

The polymer with nanoparticles tethered at each end is a unique model for unraveling the effect of chain ends on the polymer dynamics. We investigated the rheological behavior of this kind of polymer by using nonequilibrium molecular dynamics simulation. The effect of polymer lengths and nanoparticle radii on the complex moduli and viscosity was examined. The dependence of complex moduli on the frequency becomes less pronounced as the polymer is short or the nanoparticle is large. The shear thinning behavior was revealed for these systems, and the scaling exponent of complex viscosity with respect to the frequency was found to change from-1/2 to-3/4 as the polymer shortens or the nanoparticle enlarges. The rheological behavior was further explained by analyzing the mean square distance of nanoparticles. The simulation results were compared with the existing experimental finding, showing an agreement. The work provides information for understanding the chain end effect on polymer rheology.

Preparation of maximally-entangled states with multiple cat-state qutrits in circuit QED

In recent years,cat-state encoding and high-dimensional entanglement have attracted much ***,previous works are limited to generation of entangled states of cat-state qubits(two-dimensional entanglement with cat-state encoding),while how to prepare entangled states of cat-state qutrits or qudits(high-dimensional entanglement with cat-state encoding)has not been *** here propose to generate a maximally-entangled state of multiple cat-state qutrits(three-dimensional entanglement by cat-state encoding)in circuit *** entangled state is prepared with multiple microwave cavities coupled to a superconducting flux ququart(a four-level quantum system).This proposal operates essentially by the cavity-qutrit dispersive *** circuit hardware resource is minimized because only a coupler ququart is *** higher intermediate level of the ququart is occupied only for a short time,thereby decoherence from this level is greatly suppressed during the state ***,the state preparation time does not depend on the number of the qutrits,thus it does not increase with the number of the *** an example,our numerical simulations demonstrate that,with the present circuit QED technology,the high-fidelity preparation is feasible for a maximally-entangled state of two cat-state ***,we numerically analyze the effect of the inter-cavity crosstalk on the scalability of this *** proposal is universal and can be extended to accomplish the same task with multiple microwave or optical cavities coupled to a natural or artificial four-level atom.

Mechanical characteristics and reservoir stimulation mechanisms of the Gulong shale oil reservoirs, the northern Songliao Basin

Shale oil of the Qingshankou Formation of the Gulong Sag, the northern Songliao Basin, represents the first attempt at large-scale development of pure-shale-type shale oil in China. By integrating the multiscale refined reservoir characterization with macro-micro-scale mechanical testing, it is clarified that the Gulong shale is characterized by high clay mineral content, high rock plasticity, highly-developed bedding, and prominent mechanical anisotropy. A three-dimensional(3D) fracture propagation model of hydraulic fracturing was built for the Gulong shale, which fully captures the hydraulic fracture distribution pattern affected by the high bedding density, in-situ stress, and fracturing treatment parameters. Our research showed that due to influences of bedding, hydraulic fracturing in the Gulong shale forms a complex fracture morphology featuring the main fracture with multiple perpendicular branches that have different lengths(like the outdoor directional TV antenna); however, the vertical propagation of fractures is inhibited, and the fracture height is commonly less than 10 m. The limited stimulated reservoir volume(SRV) is the main problem facing the fracturing stimulation of the Gulong shale *** density has vital effects on fracture morphology, so case-specific fracturing designs shall be developed for shale intervals with different bedding development degrees. For reservoirs with welldeveloped bedding, it is suggested to properly increase the perforation cluster spacing and raise the volume and proportions of viscous fluids of the pad, so as to effectively promote vertical fracture propagation and improve reservoir stimulation performance. This study integrates multi-scale fine reservoir characterization and macro-micro-scale mechanical testing, as well as the construction and numerical simulation of hydraulic fracturing models for high-density layered shale reservoirs, providing a new approach and methodological framework for the fracturing rese

To better characterize the properties of surface-initiated polymers, simultaneous bulk-and surface-initiated polymerizations are usually carried out by assuming that the properties of the surface-initiated polymers resemble those of the bulk-initiated polymers. Through a Monte Carlo simulation using a heterogeneous stochastic reaction model, it was discovered that the bulk-initiated polymers exhibit a higher molecular weight and a lower dispersity than the corresponding surface-initiated polymers, which indicates that the equivalent assumption is invalid. Furthermore, the molecular weight distributions of the two types of polymers are also different, suggesting different polymerization mechanisms. The results can be simply explained by the heterogeneous distributions of reactants in the system. This study is helpful to better understand surface-initiated polymerization.
