Currently,tuberculosis(TB)is the second most lethal disease in the world caused by a single infectious *** diagnosis of TB is of great importance for its treatment and *** MTB/RIF is a novel rapid diagnostic assay for the diagnosis of pulmonary TB(PTB).Use of the Xpert assay based on bronchoalveolar lavage fluid(BALF)samples is indicated when TB is suspected and sputum smears or cultures are *** aim of this meta-analysis was to systematically evaluate the diagnostic performance of the Xpert assay based on BALF samples for the diagnosis of PTB.A systematic review of previously published articles was performed,and rel-evant data were ***-DiSc 1.4 and Stata 12.0 were used to analyze the *** Mycobacterium tuberculosis cultures were used as the criterion standard,the combined sensitivity of BALF-based Xpert was 0.89(95%CI,0.87–0.91),the specificity was 0.87(95%CI,0.85-0.88),the positive likelihood ratio was 8.28(95%CI,5.39–12.71),the negative likelihood ratio was 0.14(95%CI,0.10–0.19)and the diagnostic ratio was 84.08(95%CI,42.00–168.31).When composite reference standard was used as the criterion standard,the above observations were 0.69(95%CI,0.67–0.72),0.98(95%CI,0.97–0.98),41.40(95%CI,14.56–117.71),0.28(95%CI,0.21–0.37)and 190.47(95%CI,50.56–717.54),*** area under the summary receiver operating characteristic curve was close to 1 for ***,the Xpert MTB/RIF assay based on BALF samples showed high sensitivity and specificity for the diag-nosis of PTB and seems to be a reliable rapid detection method.

Effects of smartphone-based interventions and monitoring on bipolar disorder:A systematic review and meta-analysis

Berbamine induces apoptosis in human hepatoma cell line SMMC7721 by loss in mitochondrial transmembrane potential and caspase activation

Objective： To investigate the effect ofberbamine on human hepatoma cell line SMMC7721. Methods： The effects of 24 h and 48 h incubation with different concentrations （0-64 μg/ml） of the berbamine on SMMC7721 cells were evaluated using 3-4,5-dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide （MTT） assay. Hoechst 33258 staining was conducted to distinguish the apoptotic cell, and the appearance of sub-G1 stage was determined by PI （propidium iodide） staining, the percentage of apoptotic cell was determined by flow cytometry following annexin V/PI staining. Flow cytometry was performed to analyze the cell cycle distribution and the mitochondrial membrane potential （△ψm）, the expression of activated caspase3 and caspase9 was analyzed by Western-blot. Results： The proliferation of SMMC7721 was decreased after treatment with berbamine in a dose- and time-dependent manner. Berbamine could induce apoptosis in SMMC7721 cells and could cause cell cycle arrest in G0/G1 phase, to induce loss of mitochondrial membrane potential （AVm） and activate caspase3 and caspase9. Berbamine-induced apoptosis could be blocked by the broad caspase inhibitor z-VAD-fmk. Conclusion： Berbamine exerts antiproliferative effects on human hepatocellular carcinoma SMMC7721 cells. The anticancer activity of berbamine could be attributed partly to its inhibition of cell proliferation and induction of apoptosis in cancer cells through loss in mitochondrial transmembrane potential and caspase activation.

We report an all-fiber-based master oscillator power amplifier picosecond ytterbium-doped fiber laser with an average power of 102W and a spectral line width of 0.1 *** seed source is a compact single mode passively mode-locked fiber laser with an average power of 2.48 ***,the laser maximum average output power of 102W picosecond pulses is realized by a direct all-fiber amplifier structure in one *** experiment enables the optical-to-optical conversion efficiency to reach 61.4%,with the central wavelength of 1063.7nm.A significant feature of this experiment is the spectral line width of 0.1 *** spectrum has no broadening or nonlinear effects when the pump is strengthened.

Fifteen 1-（4-substituted phenyl）-4-（4-bromophenyl）-5-（halo-o-hydroxyphenyl）imino-1,2,3-triazoles were designed and synthe- sized based on rational combination of 1,2,3-triazoles and （halo）o-hydroxyphenyl group according to the superposition principle of reinforcement of biological activities. All the compounds were tested to an in vitro antimicrobial screening against M.a. and E.c.. Compounds IIe-IIo exhibited more potent antimicrobial activities against M.a. and E.c. than triclosan and fluconazole, which provided valuable information to further study of novel antimicrobial research.

Yiguanjian decoction enhances fetal liver stem/progenitor cell-mediated repair of liver cirrhosis through regulation of macrophage activation state

AIM To investigate whether Yiguanjian decoction(YGJ) has an anti-liver cirrhotic effect and whether it regulates hepatic stem cell *** A rat model of liver cirrhosis was established via subcutaneous injection of carbon tetrachloride(CCl4) for8 wk. From the beginning of the ninth week, the rats received 2-acetylaminofluorene(2-AAF) by oral gavage and a DLK-1+ fetal liver stem/progenitor cell(FLSPC)transplant or an FLSPC transplant in combination with YGJ treatment for 4 wk. In vitro, lipopolysaccharide(LPS)-activated macrophages were co-cultured with WB-F344 cells, and the differentiation of WB-F344 cells was observed in the presence and absence of YGJ *** FLSPC transplantation improved liver function and histopathology, and inhibited the activation of the noncanonical Wnt signaling pathway, while activating the canonical Wnt signaling pathway. YGJ enhanced the therapeutic effects of FLSPCs and also promoted the liver regeneration differentiation of FLSPCs into *** vitro, LPS-activated macrophages promoted the differentiation of WB-F344 cells into myofibroblasts, and the canonical Wnt signaling was inhibited while the noncanonical Wnt signaling was activated in WB-F344 *** suppressed the activation of macrophages and then inhibited non-canonical Wnt signaling and promoted canonical Wnt *** YGJ enhances FLSPC-mediated repair of liver cirrhosis through regulation of macrophage activation state, and YGJ in combination with stem cell transplantation may be a suitable treatment for end-stage liver cirrhosis.

Mutation screening of mismatch repair gene Mlh3 in familial esophageal cancer

AIM： To shed light on the possible role of mismatch repair gene MIh3 in familial esophageal cancer （FEC）. METHODS： A total of 66 members from 10 families suggestive of a genetic predisposition to hereditary esophageal cancer were screened for germline mutations in MIh3 with denaturing high performance liquid chromatography （DHPLC）, a newly developed method of comparative sequencing based on heteroduplex detection. For all samples exhibiting abnormal DHPLC profiles, sequence changes were evaluated by cycle sequencing. For any mutation in family members, we conducted a segregation study to compare its prevalence in sporadic esophageal cancer patients and normal controls. RESULTS： Exons of MIh3 in all samples were successfully examined. Overall, 4 missense mutations and 3 polymorphisms were identified in 4 families. MIh3 missense mutations in families 9 and 10 might be pathogenic, but had a reduced penetrance. While in families 1 and 7, there was no sufficient evidence supporting the monogenic explanations of esophageal cancers in families. The mutations were found in 33% of high-risk families and 50% of low-risk families.CONCLUSION： MIh3 is a high risk gene with a reduced penetrance in some families. However, it acts as a low risk gene for esophageal cancer in most families. Mutations of MIh3 may work together with other genes in an accumulated manner and result in an increased risk of esophageal tumor. DHPLC is a robust and sensitive technique for screening gene mutations.

The role of FAST in pulsar timing arrays

The Five-hundred-meter Aperture Spherical radio Telescope(FAST) will become one of the world-leading telescopes for pulsar timing array(PTA) research. The primary goals for PTAs are to detect(and subsequently study) ultra-low-frequency gravitational waves, to develop a pulsar-based time standard and to improve solar system planetary ephemerides. FAST will have the sensitivity to observe known pulsars with significantly improved signal-to-noise ratios and will discover a large number of currently unknown pulsars. We describe how FAST will contribute to PTA research and show that jitter-and timing-noise will be the limiting noise processes for FAST data sets. Jitter noise will limit the timing precision achievable over data spans of a few years while timing noise will limit the precision achievable over many years.
