Protective effect of ginsenoside Rg1 on 661W cells exposed to oxygen-glucose deprivation/reperfusion via keap1/nrf2 pathway

AIM:To construct an in vitro model of oxygen-glucose deprivation/reperfusion(OGD/R)induced injury to the optic nerve and to study the oxidative damage mechanism of ischemia-reperfusion(I/R)injury in 661W cells and the protective effect of ginsenoside ***:The 661W cells were treated with different concentrations of Na2S2O4 to establish OGD/R model in ***,intracellular reactive oxygen species(ROS)levels and superoxide dismutase(SOD)levels were measured at different time points during the reperfusion injury *** injury model was pretreated with graded concentrations of ginsenoside ***-time polymerase chain reaction(PCR)was used to measure the expression levels of cytochrome C(cyt C)/B-cell lymphoma-2(Bcl2)/Bcl2 associated protein X(Bax),heme oxygenase-1(HO-1),caspase9,nuclear factor erythroid 2-related factor 2(nrf2),kelch-like ECH-associated protein 1(keap1)and other *** blot was used to detect the expression of nrf2,phosphorylated nrf2(pnrf2)and keap1 protein ***:Compared to the untreated group,the cell activity of 661W cells treated with Na2S2O4 for 6 and 8h decreased(P<0.01).Additionally,the ROS content increased and SOD levels decreased significantly(P<0.01).In contrast,treatment with ginsenoside Rg1 reversed the cell viability and SOD levels in comparison to the Na_(2)S_(2)O_(4)treated group(P<0.01).Moreover,Rg1 reduced the levels of caspase3,caspase9,and cyt C,while increasing the Bcl2/Bax *** differences were all statistically significant(P<0.05).Western blot analysis showed no significant difference in the protein expression levels of keap1 and nrf2 with Rg1 treatment,however,Rg1 significantly increased the ratio of pnrf2/nrf2 protein expression compared to the Na_(2)S_(2)O_(4)treated group(P<0.001).CONCLUSION:The OGD/R process is induced in 661W cells using Na_(2)S_(2)O_(4).Rg1 inhibits OGD/R-induced oxidative damage and alleviates the extent of apoptosis in 661W cells through the keap1/nrf2 pathway

Polyamine metabolism dysregulation is a hallmark of many cancers,offering a promising avenue for early tumor *** study presents the development of a nuclear probe derived from spermidine （SPM） for dual-purpose tumor PET imaging and internal radiation *** probe,radiolabeled with either[68Ga]Ga for diagnostic applications or[177Lu]Lu for therapeutic use,was synthesized with exceptional purity,stability,and specific *** testing involving 12 different tumor cell lines revealed remarkable specificity towards B16 melanoma cells,showcasing outstanding tumor localization and target-to-non-target *** investigations employing polyamines,non-labeled precursor,and polyamine transport system （PTS） inhibitor,consistently affirmed the probe?s targetability through recognition of the ***,while previous reports indicated PTS upregulation in various tumor types for targeted therapy,this study observed no positive signals,highlighting a concentration-dependent discrepancy between targeting for therapy and ***,when labeled with[177Lu],the probe demonstrated its therapeutic potential by effectively controlling tumor growth and extending mouse *** into biodistribution,excretion,and biosafety in healthy humans laid a robust foundation for clinical *** study introduces a versatile SPM-based nuclear probe with applications in precise tumor theranostics,offering promising prospects for clinical implementation.

We have obtained the constraints on the density dependence of the symmetry energy from neutron-skin thickness data by parityviolating electron scatterings and neutron-star observables using a Bayesian approach,based on the standard Skyrme-HartreeFock(SHF)model and its extension as well as the relativistic mean-field(RMF)*** the neutron-skin thickness data(neutron-star observables)mostly constrain the symmetry energy at subsaturation(suprasaturation)densities,they may more or less constrain the behavior of the symmetry energy at suprasaturation(subsaturation)densities,depending on the energy-density functional *** showing the final posterior density dependence of the symmetry energy,we also compare the slope parameters of the symmetry energy at 0.10 fm-3as well as the values of the symmetry energy at twice saturation density from three effective nuclear *** present work serves as a comparison study based on relativistic and non-relativistic energy-density functionals,for constraining the nuclear symmetry energy from low to high densities using a Bayesian approach.

The development of aqueous battery with dual mechanisms is now arousing more and more *** dual mechanisms of Zn^(2+)(de)intercalation and I^(-)/I_(2)redox bring unexpected ***,differing from previous studies using Zn I_(2)additive,this work designs an aqueous Bi I_(3)-Zn battery with selfsupplied I^(-).Ex situ tests reveal the conversion of Bi I_(3)into Bi(discharge)and Bi OI(charge)at the 1st cycle and the dissolved I^(-)in *** active I^(-)species enhances the specific capacity and discharge medium voltage of electrode as well as improves the generation of Zn dendrite and ***,the porous hard carbon is introduced to enhance the electronic/ionic conductivity and adsorb iodine species,proven by experimental and theoretical ***,the well-designed Bi I_(3)-Zn battery delivers a high reversible capacity of 182 m A h g^(-1)at 0.2 A g^(-1),an excellent rate capability with 88 m A h g^(-1)at 10 A g^(-1),and an impressive cyclability with 63%capacity retention over 20 K cycles at 10 A g^(-1).An excellent electrochemical performance is obtained even at a high mass loading of 6 mg cm^(-2).Moreover,a flexible quasi-solid-state Bi I_(3)-Zn battery exhibits satisfactory battery *** work provides a new idea for designing high-performance aqueous battery with dual mechanisms.

Standing-wave supermode microresonators that are created through the strong coupling between counter-propagating modes have emerged as versatile platforms for sensing and nonlinear optics. For example, these microresonators have shown potential in nanoparticle sizing and counting, as well as enhancing the single-photon optomechanical coupling rate of stimulated Brillouin scattering. However, it has been observed that the relation between the mode linewidth and on-resonance transmission of the split supermodes differs obviously from that of the non-split modes. This behavior is typically quantified by the coupling ideality(I), which remains inadequately explored for the standing-wave supermodes. In this study, we theoretically and experimentally investigate the coupling ideality of standing-wave supermodes in a commonly employed configuration involving a Si O2microresonator side-coupled to a tapered fiber. Our findings demonstrate that, even with a single-mode tapered fiber, the coupling ideality of the standing-wave supermodes is limited to 0.5, due to the strong backscattering-induced energy loss into the counter-propagating direction, resulting in an additional equivalent parasitic loss. While achieving a coupling ideality of 0.5 presents challenges for reaching over-coupled regimes,it offers a convenient approach for adjusting the total linewidth of the modes while maintaining critically-coupled conditions.

Recently universal dynamic scaling is observed in several systems,which exhibit a spatiotemporal self-similar scaling behavior,analogous to the spatial scaling near phase *** latter one arises from the emergent continuous scaling *** by this,we investigate the possible relation between the scaling dynamics and the continuous scaling symmetry in this *** derive a theorem that the scaling invariance of the quenched Hamiltonian and the initial density matrix can lead to the universal dynamic *** is further demonstrated both in a two-body system analytically and in a many-body system *** the latter one,we calculate the dynamics of quantum gases quenched from the zero interaction to a finite interaction via the non-equilibrium high-temperature virial expansion.A dynamic scaling of the momentum distribution appears in certain momentum-time windows at unitarity as well as in the weak interacting ***,this universal scaling dynamics persists approximately with smaller scaling exponents even if the scaling symmetry is fairly *** findings may offer a new perspective to interpret the related *** also study the Contact dynamics in the BEC−BCS ***,the half-way time displays a maximum near unitarity while some damping oscillations occur on the BEC side due to the dimer state,which can be used to detect possible two-body bound states in experiments.

Increasing risks of incidental and occupational exposures to two-dimensional transition metal dichalcogenides(2D TMDCs)due to their broad application in various areas raised their public health *** the composition-dependent cytotoxicity of 2D TMDCs has been well-recognized,how the outer chalcogenide atoms and inner transition metal atoms differentially contribute to their perturbation on cell homeostasis at non-lethal doses remains to be *** the present work,we compared the autophagy induction and related mechanisms in response to WS_(2),NbS_(2),WSe_(2)and Nb Se_(2)nanosheets exposures in MH-S murine alveolar *** these 2D TMDCs had comparable physicochemical properties,overall cytotoxicity and capability in triggering autophagy in MH-S cells,but showed outer chalcogen-dependent subcellular localization and activation of autophagy ***,WS_(2)and NbS_(2)nanosheets adhered on the cell surface and internalized in the lysosomes,and triggered m TOR-dependent activation of ***,WSe_(2)and Nb Se_(2)nanosheets had extensive distribution in cytoplasm of MH-S cells and induced autophagy in an m TOR-independent ***,the 2D TMDCs-induced perturbation on autophagy aggravated the cytotoxicity of respirable benzo[a]*** findings provide a deeper insight into the potential health risk of environmental 2D TMDCs from the perspective of homeostasis perturbation.

Disaster information content is an objective mapping of disaster situations,social response,and public *** response to emergency is an important mechanism for implementing and guaranteeing emergency management of major natural hazard-related *** how disaster information content affects social response to emergencies is helpful for managing risk communication and efficient disaster *** on the 2008 freezing-rain and snowstorm disasters in southern China,this study used Python to extract 7,857 case-related media reports and applied natural language processing for text *** used three typical cases to identify and analyze disaster media report content and the relationship between these reports and the social response to the *** categories of disaster response-such as prewarning and forecasting,announcements by the authorities,and social mobilization-appeared in the disaster information in the media,along with disaster impact information,that is,real-time disaster *** response information and an appropriate amount of disaster impact information played important roles in prewarning,disaster relief,public opinion guidance,and social stability maintenance and can serve important functions in communicating with all stakeholders of emergency management,assisting or influencing emergency departments or individuals in decision making,and eliminating "information islands." Empathy caused the general public to become "disaster responders" through receiving *** and an excess of negative information may have a perverse amplification effect on public opinion and increase the unpredictability of the disaster situation and the risk of social crisis.

Mechanically lined pipe(MLP)is often used for offshore oil and gas transport because of its low cost and corrosion *** installation and operation,the pipe may undergo severe bending deformation,which causes the liner to separate from the outer pipe and buckles,affecting the stability of the whole *** this paper,the buckling response of MLP subjected to bending is investigated to clarify its bending characteristics by employing both experiments,numerical simulation,as theoretical *** types of MLPs were manufactured with GB 45 carbon steel(SLP)and Al 6061(ALP)used as the outer pipe material,*** hydraulic expansion and bending experiments of small-scale MLPs are *** addition to the ovalized shape of the cross-section for the SLP specimens,the copper liner was found to wrinkle on the compressive *** contrast,the liner of ALP remains intact without developing any wrinkling and collapse *** addition,a dedicated numerical framework and theoretical models were also *** was found both the manufacturing and bending responses of the MLP can be well reproduced,and the predicted maximum moment and critical curvatures are in good agreement with the experimental results.
