BACKGROUND Colorectal cancer has a low 5-year survival rate and high mortality.Humanβ-defensin-1(hBD-1)may play an integral function in the innate immune system,contributing to the recognition and destruction of cancer cells.Long non-coding RNAs(lncRNAs)are involved in the process of cell differentiation and growth.AIM To investigate the effect of hBD-1 on the mammalian target of rapamycin(mTOR)pathway and autophagy in human colon cancer SW620 cells.METHODS CCK8 assay was utilized for the detection of cell proliferation and determination of the optimal drug concentration.Colony formation assay was employed to assess the effect of hBD-1 on SW620 cell proliferation.Bioinformatics was used to screen potentially biologically significant lncRNAs related to the mTOR pathway.Additionally,p-mTOR(Ser2448),Beclin1,and LC3II/I expression levels in SW620 cells were assessed through Western blot analysis.RESULTS hBD-1 inhibited the proliferative ability of SW620 cells,as evidenced by the reduction in the colony formation capacity of SW620 cells upon exposure to hBD-1.hBD-1 decreased the expression of p-mTOR(Ser2448)protein and increased the expression of Beclin1 and LC3II/I protein.Furthermore,bioinformatics analysis identified seven lncRNAs(2 upregulated and 5 downregulated)related to the mTOR pathway.The lncRNA TCONS_00014506 was ultimately selected.Following the inhibition of the lncRNA TCONS_00014506,exposure to hBD-1 inhibited p-mTOR(Ser2448)and promoted Beclin1 and LC3II/I protein expression.CONCLUSION hBD-1 inhibits the mTOR pathway and promotes autophagy by upregulating the expression of the lncRNA TCONS_00014506 in SW620 cells.

The Application of Nicotiana benthamiana as a Transient Expression Host to Clone the Coding Sequences of Plant Genes

Coding sequences (CDS) are commonly used for transient gene expression, in yeast two-hybrid screening, to verify protein interactions and in prokaryotic gene expression studies. CDS are most commonly obtained using complementary DNA (cDNA) derived from messenger RNA (mRNA) extracted from plant tissues and generated by reverse transcription. However, some CDS are difficult to acquire through this process as they are expressed at extremely low levels or have specific spatial and/or temporal expression patterns in vivo. These challenges require the development of alternative CDS cloning technologies. In this study, we found that the genomic intron-containing gene coding sequences (gDNA) from Arabidopsis thaliana, Oryza sativa, Brassica napus, and Glycine max can be correctly transcribed and spliced into mRNA in Nicotiana benthamiana. In contrast, gDNAs from Triticum aestivum and Sorghum bicolor did not function correctly. In transient expression experiments, the target DNA sequence is driven by a constitutive promoter. Theoretically, a sufficient amount of mRNA can be extracted from the N. benthamiana leaves, making it conducive to the cloning of CDS target genes. Our data demonstrate that N. benthamiana can be used as an effective host for the cloning CDS of plant genes.

Structural and Functional Insights into an Arabidopsis NBS-LRR Receptor in Nicotiana benthamiana

Nucleotide-binding site leucine-rich repeat receptors (NBS-LRR/NLRs) are crucial intracellular immune proteins in plants. Previous article reported a novel NLR protein SUT1 (SUPPRESSORS OF TOPP4-1, 1), which is involved in autoimmunity initiated by type one protein phosphatase 4 mutation (topp4-1) in Arabidopsis, however, its role in planta is still unclear. This study employed Nicotiana benthamiana, a model platform, to conduct an overall structural and functional analysis of SUT1 protein. The transient expression results revealed that SUT1 is a typical CNL (CC-NBS-LRR) receptor, both fluorescence data and biochemical results showed the protein is mainly anchored on the plasma membrane due to its N-terminal acylation site. Further truncation experiments announced that its CC (coiled-coil) domain possessed cell-death-inducing activity. The outcomes of point mutations analysis revealed that not only the CC domain, but also the full-length SUT1 protein, whose function and subcellular localization are influenced by highly conserved hydrophobic residues. These research outcomes provided favorable clues for elucidating the activation mechanism of SUT1.

MICP增强改性煤矸石在水稳材料中应用的试验研究

为解决煤矸石集料强度低、吸水率高、易崩解等问题,采用微生物诱导碳酸盐沉积(MICP)技术对煤矸石进行了增强改性。通过表观密度、吸水率、压碎值和超声波质量损失率的测试,同时结合SEM,EDS和XRD微观表征,研究了微生物矿化时间、菌液浓度和煤矸石粒径对MICP增强改性煤矸石性能的影响,并揭示了MICP增改性煤矸石的机理。将MICP增强改性煤矸石应用于水稳材料,通过无侧限抗压强度、间接抗拉强度和抗压回弹模量的测试,对比了MICP增强改性煤矸石水稳材料与普通煤矸石水稳材料的性能。结果表明:采用MICP增强改性技术可以提升煤矸石的物理力学性能,并且煤矸石的粒径越大增强改性效果越明显;在菌液OD600值(溶液在600 nm波长处的吸光值)为0.8、矿化改性7 d的条件下,MICP增强改性煤矸石比普通煤矸石的表观密度增加了14.66%,吸水率降低了40.94%,压碎值降低了7.93%;经MICP增强改性后的煤矸石表面包裹的生物CaCO_(3)方解石使得煤矸石的表观密度增加,吸水率和压碎值降低,强度提高,从而提升了煤矸石的性能;水泥稳定MICP增强改性煤矸石混合料最佳含水量比普通煤矸石水稳材料降低了17.3%,最大干密度增加了7.9%;MICP增强改性煤矸石水稳材料各龄期的无侧限抗压强度、间接抗拉强度和抗压回弹模量比普通煤矸石水稳材料均显著提升,表现出更高的强度、更好的抗裂性能和抗变形能力。

Classical and quantum space-to-ground communications necessitate highly sensitive receivers capable of extracting information from modulated photons to extend the communication distance from near-earth orbits to deep space explorations.To achieve gigabit data rates while mitigating strong background noise photons and beam drift in a highly attenuated free-space channel,a comprehensive design of a multi-functional detector is indispensable.In this study,we present an innovative compact multi-pixel superconducting nanowire single-photon detector array that integrates near-unity detection efficiency(91.6%),high photon counting rate(1.61 Gcps),large dynamic range for resolving different photon numbers(1-24),and four-quadrant position sensing function all within one device.Furthermore,we have constructed a communication testbed to validate the advantages offered by such an architecture.Through 8-PPM(pulse position modulation)format communication experiments,we have achieved an impressive maximum data rate of 1.5 Gbps,demonstrating sensitivities surpassing previous benchmarks at respective speeds.By incorporating photon number information into error correction codes,the receiver can tolerate maximum background noise levels equivalent to 0.8 photons/slot at a data rate of 120 Mbps—showcasing a great potential for daylight operation scenarios.Additionally,preliminary beam tracking tests were conducted through open-loop scanning techniques,which revealed clear quantitative dependence indicating sensitivity variations based on beam location.Based on the device characterizations and communication results,we anticipate that this device architecture,along with its corresponding signal processing and coding techniques,will be applicable in future space-to-ground communication tasks.

Allotetraploidization event of Coptis chinensis shared by all Ranunculales

Coptis chinensis Franch.,also named Chinese goldthread is a member of Ranunculaceae in the order Ranunculales and represents an important lineage of early eudicots with traditional medicinal value.In our study,by using syntenic analysis combined with phylogenomic analysis of C.chinensis and four other representative genomes from basal and core eudicots,we confirmed that the WGD event in C.chinensis was shared by Aquilegia coerulea and Papaver somniferum L.and quickly occurred after Ranunculales diverged from other eudicots,likely a Ranunculales common tetraploidization(RCT).The synonymous nucleotide substitutions at synonymous sites distribution of syntenic blocks across these genomes showed that the evolutionary rate of the P.somniferum genome is faster than that of the C.chinensis genome by approximately 13.7%,possibly due to Papaveraceaes having an additional special tetraploidization event(PST).After Ks correction,the RCT dated to 115—130 million years ago(MYA),which was close to the divergence of Ranunculaceaes and Papaveraceaes approximately115.45—130.51 MYA.Moreover,we identified homologous genes related to polyploidization and speciation and constructed multiple sequence alignments with different reference genomes.Notably,the event-related subgenomes in the basal genomes all showed genomic fractionation bias,suggesting a likely allopolyploid nature of the RCT,PST and T-Alpha and T-Beta events in Tetracentron sinense.In addition,we detected that the sixteen P450 subfamilies were markedly expanded in the genomes of Ranunculales,and most of them were related to the RCT and PST events.We constructed a new platform for Early Eudicot Comparative Genomic Research(http://www.***.top/index.html)to store more information.In summary,our findings support the WGD of C.chinensis shared by Ranunculales,which is likely an allotetraploidization event.This present effort offered new insights into the evolution of key polyploidization events and the genes related to secondary metabolites during the diversification of ea

Fermi level tuning in SnPbTe/Pb heterostructure via changing interface roughness

Superconducting SnTe-type topological crystalline insulators （TCIs） are predicted to host multiple Majorana zero modes（MZMs） which can coexist in a single vortex. Fermi level （FL） close to the Dirac points of topological surface states is helpful for detecting MZMs. However, the TCI SnTe is a heavily p-type semiconductor which is very difficult to modify to n-type via doping or alloying. In this work, we fabricate the atomically flat Sn1-xPbxTe/Pb heterostructure by molecular beam epitaxy, and make the p-type Sn1-xPbxTe become n-type through changing the interface roughness. Using scanning tunnelling microscope, we find the Dirac points of Sn1-xPbxTe/Pb heterostructure are always above the FL due to the Fermi level pinning （FLP） induced by topological surface states at atomically flat interface. After increasing the interface roughness, the FLP effect is suppressed and then the Dirac points of p-type Sn1-xPbxTe can be tuned very close to or even below the FL. Our work provides a new method for tuning the FL of SnTe-type TCI which has potential application in novel topological superconductor device.

本文针对一类网络拓扑时变且耦合通道存在随机干扰的Kuramoto型振子网络的同步问题,设计了事件触发机制下的固定时间控制算法.针对耦合网络的拓扑在切换中保持连通的假设,给出了一种具有多层结构的事件触发分布式控制策略,并能够抵消随机干扰对同步性能带来的负面影响.利用随机Lyapunov稳定理论和固定时间稳定性理论给出了达到实际固定时间相位同步的控制参数条件和同步调节时间的上界.另外,证明了所使用的事件触发机制在引入双曲正切函数之后不会产生Zeno现象,并给出了触发间隔的下界.同时,在推论中给出了本文提出的控制方法也可以应对振子耦合拓扑图不连通情况的结论.最后,通过两组具有不同噪声强度的仿真实验验证了理论分析结果.
