Modular engineering of E. coli coculture for efficient production of resveratrol from glucose and arabinose mixture

Resveratrol,a valuable plant-derived polyphenolic compound with various bioactivities,has been widely used in nutraceutical industries.Microbial production of resveratrol suffers from metabolic burden and low malonyl-CoA availability,which is a big challenge for synthetic biology.Herein,we took advantage of coculture engineering and divided the biosynthetic pathway of resveratrol into the upstream and downstream strains.By enhancing the supply of malonyl-CoA via CRISPRi system and fine-tuning the expression intensity of the synthetic pathway genes,we significantly improved the resveratrol productivity of the downstream strain.Furthermore,we developed a resveratrol addiction circuit that coupled the growth of the upstream strain and the resveratrol production of the downstream strain.The bidirectional interaction stabilized the coculture system and increased the production of resveratrol by 74%.Moreover,co-utilization of glucose and arabinose by the coculture system maintained the growth advantage of the downstream strain for production of resveratrol throughout the fermentation process.Under optimized conditions,the engineered E.coli coculture system produced 204.80 mg/L of resveratrol,12.8-fold improvement over monoculture system.This study demonstrates the promising potential of coculture engineering for efficient production of natural products from biomass.

Alveolar echinococcosis(AE)is a lethal parasitic disease caused by Echinococcus multilocularis larvae,and more than 90%of pri-mary AE lesions occur in the liver.Most of the affected individ-uals remain asymptomatic and the disease is often diagnosed at an advanced stage.The infection may spread to organs adjacent to the liver or distant locations,eventually causing end-stage multi-ple organ AE.Brain metastasis of AE is the most fatal with an inci-dence rate of 0.2%[1].

Salivary metabolites are promising noninvasive biomarkers of druginduced liver injury

BACKGROUND Drug-induced liver injury(DIli)is one of the most common adverse events of medication use,and its incidence is increasing.However,early detection of DIli is a crucial challenge due to a lack of biomarkers and noninvasive tests.AIM To identify salivary metabolic biomarkers of DIli for the future development of noninvasive diagnostic tools.METHODS Saliva samples from 31 DIli patients and 35 healthy controls(HCs)were subjected to untargeted metabolomics using ultrahigh-pressure liquid chromatography coupled with tandem mass spectrometry.Subsequent analyses,including partial least squares-discriminant analysis modeling,t tests and weighted metabolite coexpression network analysis(WMCNA),were conducted to identify key differentially expressed metabolites(DEMs)and metabolite sets.Furthermore we utilized least absolute shrinkage and selection operato and random fores analyses for biomarker prediction.The use of each metabolite and metabolite set to detect DIli was evaluated with area under the receiver operating characteristic curves.RESULTS We found 247 differentially expressed salivary metabolites between the DIli group and the HC group.Using WMCNA,we identified a set of 8 DEMs closely related to liver injury for further prediction testing.Interestingly,the distinct separation of DIli patients and HCs was achieved with five metabolites,namely,12-hydroxydodecanoic acid,3-hydroxydecanoic acid,tetradecanedioic acid,hypoxanthine,and inosine(area under the curve:0.733-1).CONCLUSION Salivary metabolomics revealed previously unreported metabolic alterations and diagnostic biomarkers in the saliva of DIli patients.Our study may provide a potentially feasible and noninvasive diagnostic method for DIli,but further validation is needed.

BACKGROUND Sleep deprivation is a prevalent issue that impacts cognitive function.Although numerous neuroimaging studies have explored the neural correlates of sleep loss,inconsistencies persist in the reported results,necessitating an investigation into the consistent brain functional changes resulting from sleep loss.AIM To establish the consistency of brain functional alterations associated with sleep deprivation through systematic searches of neuroimaging databases.Two metaanalytic methods,signed differential mapping(SDM)and activation likelihood estimation(ALE),were employed to analyze functional magnetic resonance imaging(fMRI)data.METHODS A systematic search performed according to PRISMA guidelines was conducted across multiple databases through July 29,2023.Studies that met specific inclusion criteria,focused on healthy subjects with acute sleep deprivation and reported whole-brain functional data in English were considered.A total of 21 studies were selected for SDM and ALE meta-analyses.RESULTS Twenty-one studies,including 23 experiments and 498 subjects,were included.Compared to pre-sleep deprivation,post-sleep deprivation brain function was associated with increased gray matter in the right corpus callosum and decreased activity in the left medial frontal gyrus and left inferior parietal lobule.SDM revealed increased brain functional activity in the left striatum and right central posterior gyrus and decreased activity in the right cerebellar gyrus,left middle frontal gyrus,corpus callosum,and right cuneus.CONCLUSION This meta-analysis consistently identified brain regions affected by sleep deprivation,notably the left medial frontal gyrus and corpus callosum,shedding light on the neuropathology of sleep deprivation and offering insights into its neurological impact.

Efficacy and safety of minimally invasive laparoscopic surgery under general anesthesia for ovarian cancer

BACKGROUND Ovarian cancer is one of the most common malignant tumors in female reproductive system in the world,and the choice of its treatment is very important for the survival rate and prognosis of patients.Traditional open surgery is the main treatment for ovarian cancer,but it has the disadvantages of big trauma and slow recovery.With the continuous development of minimally invasive technology,minimally invasive laparoscopic surgery under general anesthesia has been gradually applied to the treatment of ovarian cancer because of its advantages of less trauma and quick recovery.However,the efficacy and safety of minimally invasive laparoscopic surgery under general anesthesia in the treatment of ovarian cancer are still controversial.AIM To explore the efficacy and safety of general anesthesia minimally invasive surgery in the treatment of ovarian cancer.METHODS The clinical data of 90 patients with early ovarian cancer in our hospital were analyzed retrospectively.According to the different surgical treatment methods,patients were divided into study group and control group(45 cases in each group).The study group received minimally invasive laparoscopic surgery under general anesthesia for ovarian cancer,while the control group received traditional open surgery for ovarian cancer.The European Organization for Research and Treatment of Cancer Quality of life Questionnaire(EORTC QLQ-C30),clinical efficacy and safety of the two groups were compared.RESULTS The intraoperative blood loss,length of hospital stay,postoperative gas evacuation time,and postoperative EORTC QLQ-C30 score of the study group were significantly better than those of the control group(Plications in the study group was significantly lower than in the control group(P<0.05).The two groups had no significant differences in the preoperative adrenocorticotropic hormone(ACTH),androstenedione(AD),cortisol(Cor),cluster of differentiation 3 positive(CD3+),and cluster of differentiation 4 positive(CD4+)indexes(

Receptor-like kinases(RLKs)play key roles in regulating various physiological aspects in plant growth and development.In Arabidopsis thaliana,there are at least 223 leucine-rich repeat(LRR)RLKs.The functions of the majority of RLKs in the LRR XI subfamily were previously revealed.Only three RLKs were not characterized.Here we report that two independent triple mutants of these RLKs,named ROOT ELONGATION RECEPTOR KINASES(REKs),exhibit increased cell numbers in the root apical meristem and enhanced cell size in the elongation and maturation zones.The promoter activities of a number of Quiescent Center marker genes are significantly up-regulated in the triple mutant.However,the promoter activities of several marker genes known to control root stem cell niche activities are not altered.RNA-seq analysis revealed that a number of cell wall remodeling genes are significantly up-regulated in the triple mutant.Our results suggest that these REKs play key roles in regulating root development likely via negatively regulating the expression of a number of key cell wall remodeling genes.

Recent advances of high-entropy electrocatalysts for water electrolysis by electrodeposition technology:a short review

Hydrogen is considered as the promising energy carrier to substitute traditional fossil fuel,due to its cleanliness,renewability and high energy density.Water electrolysis is a simple and eonvenient technology for hydrogen production.The efficiency of water electrolysis for hydrogen production is limited by the electrocatalytic performances on hydrogen evolution reaction(HER) and oxygen evolution reaction(OER).The exorbitant Pt-and Ir-/Ru-based electrocatalysts as optimal HER and OER electrocatalysts,respectively,restrict water electrolysis development.Recently,non-precious metal-based high-entropy electrocatalysts have exhibited excellent electrocatalytic activities and long-term stabilities for water electrolysis,as promising precious cataly st candidates.Therefore,the construction of the high-entropy electroc atalysts is vital to water electrolysis industry.Electrodeposition technology is an efficient method for the preparation of high-entropy electrocatalysts due to its simple,fast,energy-saving and environmental-friendly advantages.Multi-component co-precipitation facilely occurs during the electroredox in electrodeposition processes.High-entropy alloys,oxides,(oxy)hydroxides,phosphides and phosphorus sulfide oxides have been successfully prepared by galvanostatic,potentiostatic electrodeposition,cyclic voltammetry,pulse,nanodroplet-mediated and cathodic plasma electrodeposition techniques.Hence,introduction of the development of high-entropy electrocatalysts synthesized by electrodeposition technology is significant to researchers and industries.Challenges and outlooks are also concluded to boost the industrial application of electrodeposition in water electrolysis and other energy conversion areas.

Thecoal-to-liquidcoupledwithcarbon capture,utilization,and storage technology has the potential to reduce CO_(2)emissions,but its carbon footprint and cost assessment are still insufficient.In this paper,coal mining to oil production is taken as a life cycle to evaluate the carbon footprint and levelized costs of direct-coal-toliquid and indirect-coal-to-liquid coupled with the carbon capture utilization and storage technology under three scenarios:non capture,process capture,process and public capture throughout the life cycle.The results show that,first,the coupling carbon capture utilization and storage technology can reduce CO_(2)footprint by 28%-57%from 5.91 t CO_(2)/t:oil of direct-coal-to-liquid and 24%-49%from 7.10 t CO_(2)/t:oil of indirect-coal-to-liquid.Next,the levelized cost of direct-coal-to-liquid is 648-1027$/t of oil,whereas that of indirect-coal-to-liquid is 653-1065$/t of oil.When coupled with the carbon capture utilization and storage technology,the levelized cost of direct-coalto-liquid is 285-1364$/t of oil,compared to 1101-9793/t of oil for indirect-coal-to-liquid.Finally,sensitivity analysis shows that CO_(2)transportation distance has the greatest impact on carbon footprint,while coal price and initial investment cost significantly affect the levelized cost ofcoal-to-liquid.

Magnesia-calcium materials have stable hot performance,good resistance to the erosion and corrosion of liquid steel and steel slag,and a special role in purifying liquid steel,so they are widely used in iron and steel industry.However,hydration of magnesia-calcium materials seriously restricts their use,so researches have been done to improve their hydration resistance,obtaining a series of achievements.In this paper,the improvements on the hydration resistance of magnesia-calcium materials by additives in recent 20 years were presented,and their mechanisms were summarized.

liquid-metal microgrid stretchable electronics based on bionic leaf veins with ultra-stretchability and high conductivity

Stretchable electronics that monitor joint activity and treat diseases based on liquid metal could be used in the development of healthcare applications.Such devices can be seamlessly integrated with human skin.However,most high-precision microstructures and complex patterns are difficult to fabricate due to the limitations of conventional fabrication solutions,resulting in suboptimal performance under practical conditions.Here,a liquid-metal stretchable system utilizing natural leaf veins was reported as microstructures,which was based on a biomimetic concept and utilized an all-solution process for the preparation of complex microstructures.The systems are ultra-high tensile(800% tensile strain),environmentally stable(20 days) and mechanically durable(300-cycle).The system can accurately recognize the wearer's finger bending level as well as simple gesture signals.At the same time,the system acts as a wearable heater,which can realize the fast heating behavior of heating up to 50℃in 3 min under the human body-safe voltage(1.5 V).The tensile stability is demonstrated by the heterogeneous integration of lasers(405 nm) with the system interconnects for a stretchable and wearable light source.
