SMAD7 expression in CAR-T cells improves persistence and safety for solid tumors

Despite the tremendous progress of chimeric antigen receptor T(CAR-T)cell therapy in hematological malignancies,their application in solid tumors has been limited largely due to T-cell exhaustion in the tumor microenvironment(TME)and systemic toxicity caused by excessive cytokine *** a key regulator of the immunosuppressive TME,TGF-βpromotes cytokine synthesis via the NF-κB ***,we coexpressed SMAD7,a suppressor of TGF-βsignaling,with a HER2-targeted CAR in engineered T *** novel CAR-T cells displayed high cytolytic efficacy and were resistant to TGF-β-triggered exhaustion,which enabled sustained tumoricidal capacity after continuous antigen ***,SMAD7 substantially reduced the production of inflammatory cytokines by antigen-primed CAR-T ***,SMAD7 downregulated TGF-βreceptor I and abrogated the interplay between the TGF-βand NF-κB pathways in CAR-T *** a result,these CAR-T cells persistently inhibited tumor growth and promoted the survival of tumor-challenged mice regardless of the hostile tumor microenvironment caused by a high concentration of TGF-β.SMAD7 coexpression also enhanced CAR-T-cell infiltration and persistent activation in patient-derived tumor ***,our study demonstrated the feasibility of SMAD7 coexpression as a novel approach to improve the efficacy and safety of CAR-T-cell therapy for solid tumors.

The structure of multistage link systems and the calculation of their blocking probability arediscussed in the paper. The characteristics of SPC digital switching network are specially outlined. Thenby defining the minimum pothset used in network graph, a general algorithm is developed to calculate theblocking probability of a complex network. Compared with traditioanl methods for calculating the block-ing probability, the algorithm derived in this paper is more sirnple, more effective and more easily realizedon computer, and is well worthy of being used practically.

In this work,a conventional HfO2gate dielectric layer is replaced with a 3-nm ferroelectric（Fe） HZO layer in the gate stacks of advanced fin field-effect transistors（FinFETs）.Fe-induced characteristics,e.g.,negative drain induced barrier lowering（N-DIBL） and negative differential resistance（NDR）,are clearly observed for both p-and n-type HZO-based *** characteristics are attributed to the enhanced ferroelectricity of the 3-nm hafnium zirconium oxide（HZO） film,caused by Al doping from the TiAlC capping *** mechanism is verified for capacitors with structures similar to the *** to the enhanced ferroelectricity and N-DIBL phenomenon,the drain current(IDS)of the HZO-FinFETs is greater than that of HfO2-FinFETs and obtained at a lower operating ***,circuits based on HZO-FinFET achieve higher performance than those based on HfO2-FinFET at a low voltage drain(VDD),which indicates the application feasibility of the HZO-FinFETs in the ultralow power integrated circuits.

The intrinsic variability of memristor switching behavior can be used as a natural source of randomness,this variability is valuable for safe applications in hardware,such as the true random number generator(TRNG).However,the speed of TRNG is still be further ***,we propose a reliable Ag/SiNx/n-Si volatile memristor,which exhibits a typical threshold switching device with stable repeat ability and fast switching *** volatile-memristor-based TRNG is combined with nonlinear feedback shift register(NFSR)to form a new type of high-speed dual output ***,the bit generation rate reaches a high speed of 112 kb/*** addition,this new TRNG passed all 15 National Institute of Standards and Technology(NIST)randomness tests without post-processing steps,proving its performance as a hardware security *** work shows that the SiNx-based volatile memristor can realize TRNG and has great potential in hardware network security.

Indium selenide has garnered significant attention for high volumetric capacities,but is currently plagued by the sluggish charge transfer kinetics,severe volume effect,and rapid capacity degradation that hinder their practical ***,we design,synthesize,and characterize a multi-kernel-shell structure comprised of indium selenide encapsulated within carbon nanospheres(referred to as m-K-S In_(2)Se_(3)@C)through an integrated approach involving a hydrothermal method followed by a gaseous selenization ***,experimental measurements and density functional theory calculations confirm that the m-K-S In_(2)Se_(3)@C not only improve the adsorption capability for Li-ions but also lower the energy barrier for Li-ions *** from numerous contact points,shorter diffusion distances and an improved volume buffering effect,the m-K-S In_(2)Se_(3)@C achieves an 800 mA h g^(−1)capacity over 1000 loops at 1000 mA g^(−1),a 520 mA h g^(−1)capacity at 5000 mA g^(−1)and an energy density of 270 Wh kg^(−1)when coupled with LiFePO4,surpassing most related anodes reported ***,the m-K-S structure with unique nano-micro structure offers a new approach to the design of advanced anodes for LIBs.

Placental site trophoblastic tumor(PSTT),a rare gestational trophoblastic neoplasm(GTN),exhibits slow growth,late-onset metastasis,and chemotherapy insensitivity[1,2].The invasive capacity of trophoblasts induces angiogenesis in PSTT,yet the specific mechanism remains unclear[3].PSTT is typically resistant to chemotherapy and hysterectomy stands as the primary therapeutic choice for patients with an overall cure rate of approximately 75%-80%[4,5].Therefore,investigating the precise emergence and progression of PSTT is crucial,aiming to identify novel targets for diagnosis and therapy.

BACKGROUND An increasing number of studies have focused on the role of cellular metabolism in the development of colorectal cancer(CRC).However,no work is currently available to synthesize the field through *** To analyze the development in the field of“glucose metabolism”(GM),“amino acid metabolism”(AM),“lipid metabolism”(LM),and“nucleotide metabolism”(NM)in CRC by *** Articles within the abovementioned areas of GM,AM,LM and NM in CRC,which were published from January 1,1991,to December 31,2022,are retrieved from the Web of Science Core Collection and analyzed by CiteSpace 6.2.R4 and VOSviewer *** The field of LM in CRC presented the largest number of annual publications and the fastest increase in the last decade compared with the other three ***,China and the United States were two of the most prominent contri-butors in these four *** addition,Gang Wang,Wei jia,Maria Notar-nicola,and Cornelia Ulrich ranked first in publication numbers,while Jing-Yuan Fang,Senji Hirasawa,Wei jia,and Charles Fuchs were the most cited authors on average in these four fields,respectively.“Gut microbiota”and“epithelial-mesenchymal transition”emerged as the newest burst words in GM,“gut microbiota”was the latest outburst word in AM,“metastasis”,“tumor microenvironment”,“fatty acid metabolism”,and“metabolic reprogramming”were the up-to-date outbreaking words in LM,while“epithelial-mesenchymal transition”and“apoptosis”were the most recently occurring words in *** Research in“cellular metabolism in CRC”is all the rage at the moment,and researchers are particularly interested in exploring the mechanism to explain the metabolic alterations in *** metabolic vulnerability appears to be a promising direction in CRC therapy.
